1.Comparison on the influence of different incision cataract phacoemulsification on the postoperative ocular surface

AlM:To observe and compare the influence of different incision cataract phacoemulsification on comprehensive state in patients. METHODS: Sixty patients with cataract who were treated with phacoemulsification in our hospital from November 2012 to May 2014 were selected as research object, and they were divided into group A ( corneal incision group) 30 cases and group B ( corneoscleral rim incision group) 30 cases according to the disease state. Epithelial and incision repair time, tear break-up time, tear secretion, corneal staining score and tear related indexes before and after the treatment of two groups were respectively analyzed and compared. RESULTS: The epithelial and incision repair time constitution of group B were better than those of group A, the incision repair time, tear break - up time, tear secretion, corneal staining score and tear related indexes at 1,7, 14 and 28d after the treatment were all significantly better than those of group A too, there were all significant differences (all P<0. 05).CONCLUSlON: The application value of corneoscleral rim incision in the patients with cataract phacoemulsification is relatively higher,and bad influence of this method for the patients is smaller, the postoperative recovery of comprehensive state are faster.

3.The Role of E3 Ubiquitin Ligase Smurfs in BMP and TGF-bata Signaling in Bone Cells

The ubiquitin-proteasome system is composed by multiple enzymes which are ubiquitously expressed in mammalian cells and plays an essential role in a variety of biological processes. As key members of the protein degradation enzymatic system, the function of E3 ubiquitin ligases has been extensively investigated. BMP and TGF-? are critical molecules that regulate proliferation, differentiation and apoptosis of osteoblasts and chondrocytes through different signaling pathways. Resent findings indicate that the ubiquitin-proteasome system functions as a key regulator in bone cells and the E3 ligase mediates the proteolytic degradation of critical molecules in BMP and TGF-? signaling pathways. Recent progress on studies of HECT domain E3 ligase, Smurf, in osteoblasts and chondrocytes were summarized. The regulatory role of Smurf1 and Smurf2 in BMP and TGF-? signaling and osteoblast and chondrocyte function has been reviewed.

AIM: To identify an unknown peak in Guanxinning Injection(Radix et Rhizome Salviae Miltiorrhizae,Rhizoma Chuanxiong) chromatographic fingerprint. METHODS: The elution(44-46 min) separated and collected by HPLC was analyzed by GC-MS and HPLC-PDA-MS-MS. RESULTS: The unknown peak was corresponding to senkyunolidel I. CONCLUSION: It is quick method to identify the reported chemical in herbal medicine based on a small quantity of sample by GC-MS and HPLC-PDA-MS-MS.
7.Isolation and Identification of Endophytic Bacteria H-6 with High Antifungal Activity from Huperzia serrata

In this study,nearly 200 endophytic bacteria were isolated from different part of Huperzia serrata, over 60 bacterium with clear antifungal activity were selected from those cultures.Among them,strain H-6 exhibited the highest antifungal activity which was strongly inhibits the growth of many plant pathogenic fungi such as Sclerotinia scleroliorum,Fusarium graminearumt,Sclerotinia libertiana,Phytophthora capsici Leonia and Sesame fusarium wilt.According to the characteristics of morphology,physiology and biochem- istry tests and the comparison of 16S rDNA sequence,the strain H-6 was similar to the Burkholderia.So strain H-6 was identified as Burkholderia sp.H-6.The results also showed that Burkholderia sp.H-6 was markedly different from Burkholderia cepacia that was applied widely in agriculture as antagonistic bacteria. The medium and culture conditions of the strain all were optimized by single factor and orthogonal experiments.In the investigation of the culture condition,growth was carried out in a basal medium(potato juice)and gradually supplemented with the various ingredients to be investigated.The major ingredients be- ing investigated included carbon sources and nitrogen sources.The optimal antifungal activity production condition is growth in a medium(potato juice with 2.5%mannitol and 0.1%NaNO3),initial pH 4.0 at 28℃.
8.Analysis of immunophenotype characteristics in 109 cases of B lineage chronic B lymphoid leukemia.

This study was aimed to investigate the immunophenotypic characteristics of 109 cases of B-cell chronic lymphoid leukemia (B-CLL) so as to provide evidences for the diagnosis and therapy of B-CLL, and for the detection of the minimal residual disease and its prognosis. Immunophenotyping was performed in 109 patients of B-CLL by two/three color multiparameter flow cytometry analysis using a panel of monoclonal antibodies. The results showed that in 109 cases of B-CLL, all cases expressed CD19, the positive ratios of other B lineage antigen such as CD20, CD22 and CD23 were 95.40%, 94.50%, 86.20% respectively. None of the B-CLL cases expressed CD10. The expression ratio of FMC-7 and CD38 in 105 cases of B-CLL were 28.60% and 36.20%. Among the B-CLL cases the CD5(+) cells amounted to 86.23%, CD5(-) cells amounted to 13.76%, ZAP-70 protein was expressed in 12 out of 50 patients. It is concluded that immunophenotypic data are very useful for the diagnosis and detection of minimal residual disease of B-CLL, and the relationship between the immunophenotypic characteristics and the prognosis of B-CLL needs further study.

9.Beneficial effect of berberine on atherosclerosis based on attenuating vascular inflammation and calcification

OBJECTIVE To investigate the beneficial effect of berberine (BBR) on atherosclerosis in Apo-/-E mice and explore the underlying mechanisms based on attenuating vascular inflammation and modulating calcification in human umbilical vein endothelial cells (HUVECs) and smooth muscle cells(SMCs). METHODS 48 Apo-/-E mice,at 6-8 weeks old,were randomly allocated into 4 groups:normal, model, bbr and atorvastatin (positive control) groups with 12 mice in each group. They were fed with high-fat diet for 4 weeks except those in Normal group and then treated with indicated drugs or solvent for another 4 weeks. The morphology and inflammation infiltration of aortic were examined with HE staining. The expression of BMP- 2 in aortic was examined by immumohistochemical staining. Blood lipid levels were examined by automatic biochemical analyzer. The expression of IL-6, TNF-αand BMP-2 in serum and tissues was detected by ELISA method. The expression of ALP and the content of calcium were detected by commercially-available kits. HUVEC cells were stimulated with TNF-α and incubated with various concentrations of BBR for 24 h. The contents of intercellular cell adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule (VCAM-1), matrix metalloprotein-9 (MMP-9) in the culture supernatant were detected by ELISA method. Calcification was induced with β-glycerophos?phate in SMC cells and the effect of BBR on the content of calcium was examined. RESULTS 4-week berberine treatment markedly lowered serum TC and LDL-c levels and improved the plaquestabilityin Apo-/- E mice fed with a high- fat diet (P<0.05 or P<0.01) which was comparable with the effect of atorvastatin. Berberine also significantly decreased the levels of IL-6 and TNF-α in mice serum and aortic tissues (P<0.05 or P<0.001). Berberine tended to decrease ALP, BMP-2 levels and the content of calcium in mice serum and aortic tissues (P<0.05, P<0.01 or P<0.001) which were not observed in atorvastatin group. Berberine significantly lowered the levels of ICAM-1, VCAM-1, and MMP-9 in TNF-α-stimulated HUVECs. It can also lowered the content of calcium in SMCs. CONCLUSION BBR can profitably regulate the levels of blood lipid in mice fed with a high-fat diet, decrease the injury caused by inflammation, and attenuate vascular calcification. It may improve atherosclerosis and play a role in cardiovascular protection.
