1.Associations of fibrinogen genetic variation rs1049636 withγ′fibrinogen level and ischemic stroke

Objective To explore the association between fibrinogen gene polymorphism(rs1049636) and serum γ′ fibrinogen level and ischemic stroke (IS) .Methods 421 IS patients and 421 age‐and gender‐ matched healthy controls ,including 283 males and 138 females ,were recruited in this assay .The plasma γ′fibrinogen concentration was measured by enzyme‐linked immunosor‐bent assay (ELISA) .Fibrinogen gene polymorphism(rs1049636) were genotyped by using PCR‐LDR assay .Results γ′fibrinogen concentrations in IS patients[(159 .4 ± 97 .4)U/dL] were significantly higher than that in control group[(114 .2 ± 73 .0)U/dL] with statistically significant difference(P<0 .001) .Single nucleotide polymorphism(SNP) analysis showed that rs1049636 C allele was significantly associated withγ′fibrinogen level ,but not associated with increased risk of IS(P=0 .077) .Conclusion An associ‐ation between increasedγ′fibrinogen level and IS existed in Chinese Han population .However ,no association between rs1049636 C allele and IS risk was observed in our study .
2.Significance of combined detection of peripheral blood free Septin9 SDC2 and BCAT1 gene methylation in the diagnosis of colorectal cancer

Objective:To explore the clinical significance of combined detection of the promoter methylation of plasma free Septin9, SDC2 and BCAT1 genes in peripheral blood for the diagnosis of colorectal cancer. Methods The data of patients admitted to the Department of Gastroenterology, Shanghai East Hospital Affiliated to Tongji University from January to September 2019 were retrospectively analyzed. They were divided into colorectal cancer group (62 cases of colon cancer, 59 cases of rectal cancer), precancerous lesions group (77 cases of colorectal adenoma, 5 cases of high-grade intraepithelial neoplasia), interference group (61 cases of colorectal cancer and advanced adenoma negative but suffered other intestinal lesions, 17 cases of non-colorectal cancer) and healthy group (94 cases). The methylation status of three genes (Septin9, SDC2 and BCAT1) in peripheral blood plasma was detected simultaneously by fluorescence PCR. The relationship between the positive rate of three genes detected jointly and the clinic pathological characteristics of colorectal cancer was analyzed and compared with serum carcinoembryonic antigen (CEA) positive rate. The colorectal cancer group was divided into stage Ⅰ, Ⅱ, Ⅲ and Ⅳ according to TNM stage, and the colorectal cancer group was analyzed and counted by grade. The diagnostic efficiency of detection methods was analyzed by receiver operating characteristic (ROC) curve, and the area under ROC curve (AUC) was compared.Results:The positive rate of combined detection of SDC2 and BCAT1 gene methylation was higher than other three groups (χ 2 =237.246, P<0.001). The positive rate of combined detection of plasma Septin9, SDC2 and BCAT1 gene methylation was higher than CEA in colorectal cancer group ( P<0.001). The positive rates of the combined detection of plasma Septin9, SDC2 and BCAT1 gene methylation in stage Ⅰ-Ⅳ of colorectal cancer group were 73%(16/22), 87%(34/39), 86%(30/35) and 96%(24/25), respectively. Compared with CEA group, the positive rate of combined detection of plasma Septin9, SDC2 and BCAT1 gene methylation in stage Ⅰ-Ⅲ of colorectal cancer group was higher than serum CEA ( P<0.001), but the positive rate of stage Ⅳ was not statistically significant compared with CEA group ( P>0.05). ROC curve analysis showed that the AUC of Septin9, SDC2 and BCAT1 was 0.857(95% CI 0.810-0.903),0.819(95% CI 0.768-0.871)and 0.862(95% CI 0.816-0.909), respectively. The AUC of combined detection of three gene methylations was 0.889 (95% CI 0.846-0.933), and the AUC of combined detection with serum CEA was 0.913 (95% CI 0.874-0.951). There was no significant difference in the positive rate of combined detection of Septin9, SDC2 and BCAT1 gene methylation among different gender, age and cancerous site of colon cancer patients (all P>0.05). Conclusion:The combined detection of the promoter methylation of plasma free Septin9, SDC2 and BCAT1 genes in peripheral blood plasma is helpful for the early diagnosis of colorectal cancer. The positive rate in stage Ⅰ-Ⅲ of colorectal cancer group is higher than serum CEA. The combined diagnosis of the three genes can improve the diagnostic efficiency.

In this study, we explored the mechanism of Huganning tablet (HGNP) in the treatment of nonalcoholic fatty liver disease (NAFLD) based on network pharmacology and computer-aided drug design. Firstly, the potential ingredients and targets of HGNP were identified from TCMSP database, Swiss Target Prediction database, Chinese pharmacopoeia (2015) and literatures, and then the targets of HGNP intersected with NAFLD disease targets that obtained in GeneCards database to acquired potential targets. The bioconductor bioinformatics package of R software was used for gene ontology (GO) enrichment and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis. The network of “potential ingredient-key target-pathway” was formed in Cytoscape software to study the interactions between potential ingredients of HGNP, key targets, pathways and NAFLD. Based on the results of network pharmacology, the molecular docking analysis of the key targets and potential active ingredients in HGNP tablets with top degree in the network was conducted using Discovery Studio 2020 software, followed by molecular dynamics simulations, binding free energy calculation, drug-likeness properties analysis and ADMET (absorption, distribution, metabolism, excretion and toxicity) properties prediction.
4.The influence of COVID-19 prevention and control measures on the transmission and epidemic of influenza in Chongqing

Abstract: Objective To evaluate the influence of coronavirus disease 2019 (COVID-19) prevention and control measures on the transmission and epidemic of influenza in Chongqing, so as to provide references for formulating targeted influenza prevention and control strategies. Methods The influenza surveillance data, during the year 2018 to 2020, were collected through the "China Influenza Surveillance Information System", and the seasonal characteristics of influenza epidemic were analyzed. The percentage of influenza like cases (ILI%) and influenza virus positive rate between 2020 and 2018-2019 were compared, so as to evaluate the impact of COVID-19 prevention and control measures on influenza epidemic characteristics. Results The annual proportions of ILI cases in Chongqing were respectively 3.53%, 2.23% and 1.2% from 2018 to 2020, while the positive rates of influenza virus were respectively 13.97%, 23.81% and 2.65%. The distribution trend of ILI% from 2018 to 2019 fluctuated were similar, but it continued to drop and remain at a low level since February 2020. The positive rate of influenza virus showed an epidemic peak from December to March in 2018-2019, also peaked from November 2019 to January 2020, but decreased to 0 in March. ILI% was positively correlated with the positive rate of influenza virus (r=0.404 8, P<0.05). In 2020, compared with the same period of 2018-2019, the growth rate of ILI% was -66.09% and -46.32%, respectively. The positive rate of influenza virus in 2020 decreased by 81.03% and 88.87% compared with the same period of 2018-2019, respectively. The growth rates of influenza virus positive rate in January 2020 were decreased with a small rate of about 39.87%, and with a significantly decline of more than 93.65% from February. No influenza epidemic was found after March. Conclusions Since COVID-19 prevention and control measures were implemented in January 2020 in Chongqing, the ILI% and the positive rate of influenza virus in sentinel hospitals decreased significantly. In the season of high incidence of respiratory infectious diseases, personal protection and other measures can effectively reduce influenza virus infection.
5.The clinical analysis on the effects of different difficulty in root canal preparation.

OBJECTIVETo investigate the effects of root canal preparation treated by endodontists of different levels in different difficulty associated with the root canal anatomy factors.
METHODSTotally 422 teeth with 901 root canals were randomly selected from the Conservative Dentistry Department, West China Stomatology Hospital, and these teeth were respectively treated by junior specialist postgraduates, senior specialist postgraduates and endodontic specialists. Grading criteria of root canal risk factor and root canal therapy difficulty assessment were found based on 6 items: Tooth type, working length, root curvature, calcification, number of canals, and the previous endodontic treatment. The effects of root canal preparation with different difficulty in different groups were analyzed with RxC Chi-square test.
RESULTSSuccess rates of root canal preparation in junior group and senior group during three difficulty groups were statistically different (P < 0.05), and the root canal therapy difficulty group I > group II > group III. The operator factors in root canal therapy difficulty group I and group II were not significantly different P > 0.05). But in root canal therapy difficulty group III, the endodontic specialist group have highest success rate in root canal preparation.
CONCLUSIONThe cases of root canal treatment should be treated by endodontist of corresponding level according to the difficulty. In difficult cases, endodontic specialist can provide better root canal preparation result.

6.Value of lymphocyte count in assessing cellular immune function in patients with community-acquired pneumonia.

OBJECTIVETo investigate the value of lymphocyte count in assessing cellular immune function in patients with community-acquired pneumonia.
METHODSNinety-three patients with community-acquired pneumonia (including 53 non-severe and 40 severe cases) and 52 healthy adults were examined for routine blood test and T lymphocyte count. Blood lymphocyte counts and absolute T lymphocyte counts were compared among the 3 groups and their correlation was analyzed.
RESULTSCompared with the healthy control subjects, patients with community-acquired pneumonia showed significantly lower blood lymphocyte counts and CD3(+), CD4(+), and CD8(+) levels (P<0.05). CD3(+), CD4(+), and CD8(+) levels were positively correlated with blood lymphocyte counts. With blood lymphocyte count as the independent variable (L), and the regression equations for CD3(+), CD4(+), and CD8(+) levels were CD3(+)=485.45L+313.48 (F=59.68, P<0.01), CD4(+)=192.57L+290.11 (F=24.62, P<0.01), and CD8(+)=275.14L+18.04 (F=23.46, P<0.01) in the control group; CD3(+)=564.15L+25.04 (F=96.56, P<0.01), CD4(+)=381.91L-37.45 (F=68.60, P<0.01), and CD8(+)=165.61L+61.83 (F=55.47, P<0.01) in non-severe pneumonia group; and CD3(+)=565.44L+49.09 (F=31.87, P<0.01), CD4(+)=332.34L-17.37 (F=43.64, P<0.01), and CD8(+)=223.46L+54.39 (F=13.90, P<0.01) in severe pneumonia group.
CONCLUSIONPatients with community-acquired pneumonia have decreased cellular immune function. Absolute T lymphocyte count can be estimated by blood lymphocyte count to save the cost of laboratory tests.

7.Subcutaneous implantation of embryonic skin cells to construct a model of hair development in nude mice.

OBJECTIVETo observe the hair development after subcutaneous implantation of embryonic skin cells in nude mice, and construct a model of hair development.
METHODSDermal and epidermal cells isolated from embryonic mouse skin were mixed at a given ratio and injected subcutaneously in nude mice, and hair formation after the implantation was observed under a microscope.
RESULTSFormation of the hair follicles and fibers under the skin of the recipient nude mice and emergence of the hair shaft were observed microscopically.
CONCLUSIONEmbryonic skin cells be used to construct a complete model of hair development after implantation in vivo.

8.Effects of different parts of the follicle-unit grafts subjected to controlled injury.

OBJECTIVETo study the shaft elongation and morphological changes of follicle-unit (FUs) grafts subjected to controlled injury in different parts.
METHODSHuman FUs were isolated by microdissection under a dissecting microscope. The single hair of anagen FUs were randomly divided into A, B and C groups, and A and B groups were subjected to controlled injury with microsurgery imposed to the dermal papilla and the bulge of FUs, respectively, with C as the control group without any treatment. HE staining was used to detect the histological changes of the cells, and organ culture for 10 days was conducted to observe the morphological changes and elongation of FUs.
RESULTSThere were no histological or morphological changes in A, B and C groups. The average elongation of hair shaft was 1.293-/+0.245, 2.116-/+0.423 and 2.235-/+0.379 mm, respectively. There were significant differences between groups A and B (P<0.05) and between groups A and C (P<0.05). No significant difference was found between groups B and C (P>0.05).
CONCLUSIONDamage of the dermal papilla should be avoided in hair transplantation surgery.

9.Comparison between 64-slice computed tomography and coronary angiography in the diagnosis of coronary atherosclerosis.

OBJECTIVETo compare the accuracy of 64-slice computed tomography and coronary angiography in the diagnosis of coronary atherosclerosis.
METHODSThe imaging data of both 64-slice computed tomography and coronary angiography in 65 patients were analyzed retrospectively.
RESULTSAmong the 455 coronary arteries evaluated, the sensitivity, specificity and accuracy of 64-slice computed tomography were 54.6%, 95.1%, and 85.5%, respectively.
CONCLUSION64-slice computed tomography has a good potential in the diagnosis of coronary atherosclerosis with excellent sensitivity and specificity.
