3.Antitumor activity of novel tetrahydro-beta-carboline derivatives as KSP inhibitors.

Inhibitors of kinesin spindle protein (KSP) are a promising class of anticancer agents that cause mitotic arrest and induce apoptosis of tumor cells. A series of novel tetrahydro-beta-carboline derivatives were synthesized as kinesin spindle protein inhibitor and evaluated as potential antitumor agents. All compounds showed promising KSP inhibitiory activity. Compounds 8 and 9 exhibited better antitumor activity (Lung/A549, Stomach/AGS) than CK0106023 with GI50/IC50 values (1.07/1.62 and 1.46/3.27 micromol x L(-1), 1.09/>10 and 1.22/6.33 micromol x L(-1), respectively).
4.Hepatoprotective Effect of Alcohol-Extract Propolis Against Acetaminophen-induced Acute Hepatic Injury in Mice

Objective To investigate the hepatoprotective effect of Propolis alcohol-extract (PAE) against acetaminophen (APAP)-induced acute hepatic damage in mice and to explore the possible mechanism.Methods Sixty-three C57BL/6 MT(-/-)mice were equally randomized into 9 groups.The normal control group and the model group received gastric gavage of normal saline (20 mL?kg-1),four PAE groups were given PAE in the dose of 12.5,25,50 and 100 mg?kg-1 respectively,alcohol +APAP group and alcohol control group received 20 %alcohol 20 mL?kg-1 and PAE control group was given PAE in the dose of 100 mg?kg-1 qd,for 4 consective days.Thirty miniutes after last administration,the mice in the normal control group,PAE control group and alcohol control group received saline 10 mL?kg-1,and the mice in other groups received APAP 380 mg?kg-1 to induce acute hepatic injury.The activities of serum alanine aminotransferase (ALT),aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) were determined,and the liver tissues were collected for histopathological assessment by HE staining under light microscope.The ratio of glutathione (GSH) and oxidized glutathione (GSSG),and the content of GSH,GSSG and malondialdehyde (MDA) in liver homogenate were also measured.Results Compared with the model group,PAE could markedly decrease serum ALT,AST and LDH activity,reduce the MDA level in liver homogenate,and increase hepatic GSH content and the ratio of GSH/GSSG in the liver homogenate.The hepatic histopathological changes in liver were also significantly ameliorated.In PAE control group,GSH content and the ratio of GSH/GSSG were also increased.However,the above indexes remained unchanged in alcohol control group.Conclusion The propolis alcohol-extract can prevent the liver from PAP-induced acute hepatic injury.
5.The occasion and clinical efficacy of radiofrequency ablation in the treatment of elderly patients with paroxysmal supraventricular tachycardia

Objective To explore the clinical pathological data of the elderly patients with paroxysmal supraventricular tachycardia(PSVT) and the timing of the radiofrequency ablation treatment for PSVT and its clinical efficacy.Methods The clinical pathological data of 45 elderly patients with PSVT for radiofrequency ablation were retrospectivly analyzed,and the information of cardiovascular and cerebrovascular diseases,the duration of PSVT and frequency of attacks,the reason of patients had to accept radiofrequency catheter ablation,the treatment effective of electrophysiological study and radiofrequency ablation were recorded.Results The duration and frequency of attacks with PSVT were positively related with pathogenesis in the elderly patients.Elderly PSVT patients with severe cardiovascular disease accounted for 28.89%.The first radiofrequency ablation treatment success rate was 100% in this research and the recurrence rate was 4.4%.Conclusion It is recommended that elderly PSVT patients early access to the radiofrequency ablation.
6.Effect of ginsenoside Rg1 on long-term potentiation impaired by lead in CA1 region of rat hippocampus

AIM To investigate whether ginsenoside Rg1 can reverse chronic lead-induced impairment of long-term potentiation (LTP) in the CA1 region of rat hippocampus. METHODS Neonatal Wistar rats were exposed to lead from parturition to weaning via milk of dams whose drinking water (20 mL per day) contained 0.2% lead acetate. Field excitatory postsynaptic potentials (fEPSP) were recorded and LTP was induced in the CA1 region in rat hippocampal slices on postnatal 20-25 d. RESULTS In hippocampal slices from both control and lead-exposed rats, perfusion with ginsenoside Rg1 50 μmol·L-1 for 20 min induced enhancement of fEPSP (LTP), while the amplitude of LTP in lead-exposed rats was lower than that of controls. In hippocampal slices from chronic lead-exposed rats, LTP induced by high-frequency stimulation (HFS, 1s, 100 Hz) was significantly reduced, while perfusing with ginsenoside Rg1 (50 μmol·L-1) for 20 min increased the amplitudes of LTP induced by HFS by 47.1%. CONCLUSION Rg1 can increase basic synaptic transmission and partially reverse chronic lead-induced impairment of HFS-LTP.
8.Beta-HIVS combined cisplatin inhibited activities of human ovarian cancer cell line SKOV3 in vitro.

OBJECTIVETo study the effect of beta-hydroxyisovaleryl shikonin (beta-HIVS) combined cisplatin on activities of ovarian cancer cell line SKOV3 in vivo and its possible mechanisms.
METHODSCells were divided into the blank control group and six beta-HIVS groups (2 - 30 micromol/L). Effect of beta-HIVS at different concentrations on the activities of ovarian cancer cell line SKOV3 was detected using MTT assay. SKOV3 cells were treated with cisplatin (10, 20, and 40 micromol/L) and beta-HIVS (0.25, 1, and 2.5 micromol/L) combined cisplatin. Effect of beta-HIVS combined cisplatin on the activities of ovarian cancer cell line SKOV3 was determined by MTT assay. The expression of Bcl-2 and Bax after treated by different concentrations of beta-HIVS was detected by Western blot.
RESULTSThe activities of SKOV3 were inhibited by different concentrations of beta-HIVS dose-dependently. The 50% inhibition rate (IC50) was 7.37 micromol/L. There was statistical difference in IC50 between each concentration beta-HIVS group and the blank control group (P < 0.05). There was statistical difference in IC50 between the beta-HIVS (1 and 2.5 micromol/L) combined cisplatin groups and the cisplatin group (P < 0.05, P < 0.01). The synergistic effect on beta-HIVS showed dose-dependent manner. Results of Western blot showed beta-HIVS at different concentrations (5, 7.5, and 10 micromol/L) could obviously up-regulate the expression level of Bax protein and inhibit the expression level of Bcl-2 protein, showing statistical difference when compared with the control group (P < 0.01). CONCLUSIONS; HIVS could obviously inhibit in vitro growth of SKOV3 in a dose-dependent manner. With the range of concentration, beta-HIVS showed synergetic effect with cisplatin. Besides, along with increasing beta-HIVS concentrations, the synergetic effect was more significant. The synergetic effect might accelerate the apoptosis of SKOV3 through up-regulating Bax expression and inhibiting Bcl-2 expression.

9. Chemical constituents from Guanxinning Injection

Objective: To investigate the chemical constituents from Guanxinning Injection. Methods: The chemical constituents were repeatedly isolated by column chromatographic techniques including silica gel, ODS, Sephadex LH-20, and macroporous resin, and were purified by preparation HPLC chromatography and recrystallization, and their structures were identified by physicochemical properties and spectroscopic data. Results: Nineteen compounds were isolated and identified as protocatechuic aldehyde (1), vanillin (2), ferulic acid (3), 2-furoic acid (4), senkyunolide I (5), caffeic acid (6), salvianolic acid A (7), isosalvianolic acid A (8), salvianolic acid B (9), isosalvianolic acid C (10), rosmarinic acid (11), 1, 3-dicaffeoylquinic acid (12), senkyunolide H (13), 4-hydroxybenzoic acid (14), m-hydroxybenzoic acid (15), o-hydroxybenzoic acid (16), 4-hydroxycinnamic acid (17), methyl rosmarinate (18), and salvianolic acid N methyl ester (19). Conclusion: Nineteen compounds are isolated from the Guanxinning Injection for the first time, and all HPLC analyses and literature data show that compounds 2, 3, 5, 13-16 are originated from Ligusticum chuanxiong, compounds 7-11, and 18-19 from Salvia miltiorrhiza, and compounds 1, 4, 6, 12, and 17 from L. chuanxiong and S. miltiorrhiza.
10. Investigation on organism pollution of various water resources after devastating earthquake in Yingxin Town of Wenchuan County

Objective: To investigate the organism pollution of various water resources in Yingxiu Town after earthquake, so as to provide evidence for safe and permanent drinking water resources. Methods: From the 32nd to the 37th day after Wenchuan Earthquake, the water samples were collected from different water resources around the camping areas of the disaster refugees and military rescuers in Yingxiu Town; the resources included the mountain spring (6 sites), valley water (4 sites), surface water (Min river, Pitiao river and the junction of the former 2 rivers) rain water (18 sites) water purified by military vehicles (7 sites) and chlorine-disinfected water (7 sites). The 85-type water examination kits were used to examine ammonia nitrogen and nitrite nitrogen contents, pH value, turbidity (observation with naked eye), smelling, and residual chlorine of the water samples. Bottled drinking water served as control. Average values and chi-square test were used for analysis of difference. Results: Two of the 4 valley water samples and all the surface water samples had a ammonia nitrogen content of 0. 10-0. 18 mg/L; the contents of ammonia nitrogen and nitrite nitrogen were similar between other water resources except for the above mentioned 2 valley water samples and the surface water samples. However, the average ammonia nitrogen contents in the directly harvested rain samples and rain samples dropped from the tents were 0. 625 mg/L and 0. 913 mg/L, respectively, both significantly higher than that in the control (0 mg/mL) and exceeding the safe standard (
