1.Effect of hypoxia on expression of angiopoietin-2 in human retinal pigment epithelium

Background Choroidal neovascularization(CNV)has been descrihed as a main reason of visual loss in a lot of ocular diseases.Researches showed that local hypoxia and retinal pigment epithelial(RPE) cells play an important role in the formation of CNV.A closely relationship of angiopoietin-2 (Ang-2) and angiogenesis has been proved.However,whether the expression of Aog-2 in hypoxic cultured human RPE cells is associated with the pathogenesis of CNV is still below understood.Objective This study was to investigate the effects of hypoxia on expression of Ang-2 in cultured human RPE cells in vitro,and discuss the possible effects of Ang-2 in the formation of CNV.Methods Human RPE cells were cultured and passaged,and 4-7 generation of cells were used in the experiment.The cells were incubated in cultural plate at the density of 5×107 cells/L.The culture medium containing 200 μmol/L CoCl2 was used to establish the hypoxia model of human RPE cells culturecd in vitro for 0.5,1,2,4,6,12and 24 hours,and the RPE cells cultured under normoxia were as controls.Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of Ang-2 mRNA in cultured human RPE cells,and enzymelinked immunosorbnent assay(ELISA) was used to assay the content of Ang-2 protein in supernatant of cultured human RPE cells.Results The survival rate of human RPE cells was 90% after resoscitation.The fourth generation of cells showed the fusiform with the less pigment in them.The Ang-2 mRNA/β-actin mRNA value in human RPE cells was significantly different among various groups(F=1086.30,P=0.00),The Ang-2 mRN A/β-actin mRNA value in hypoxia cultured for 0.5 hours group began to increase and peaked in hypoxia culture for 4-6 hours group,with the significant differences in comparison with normoxia control group(P<0.05).The Ang-2 mRNA/β-actin mRNA value decreased to the baseline level at hypoxia for 24 hours.The ELISA analysis showed that the concentration of Ang-2 protein in supernatant of RPE cells showed significant difference among groups(F=1034.00,P=0.00).The concentration of Ang-2 protein increased at hypoxia culture for 0.5 hours and peaked at 6 hours,showing significant differences in comparison with the control group (P<0.05).Conclusions Hypoxia could significantly up-regulate the expression of Ang-2 in human RPE cells cultured in vitro.Ang-2 expresses highly in the early stage of hypoxia,implying that Ang-2 participates in the formation of CNV.
2.Kidney transplantation for treating lower urinary tract abnormality A follow-up in 4 cases

BACKGROUND: Lower urinary tract abnormality (LUTA) can lead to end-stage renal disease (ESRD). However, insufficient attention has been paid to these patients in China, and they are usually thought unsuitable for kidney transplantation because of high incidences of graft dysfunction and septic complication.OBJECTIVE: To explore the characteristics and postoperative follow-up after kidney transplantation in four patients with LUTA.DESIGN, TIME AND SETTING: A retrospective analysis of LUTA cases was performed from July 2002 to June 2006 at the Organ Transplantation Center of Chinese PLA, Second Affiliated Hospital of General Hospital of Chinese PLA.PARTICIPANTS: Four ESRD cases of LUTA, who received allograft kidney transplantation.METHODS: Four cases of LUTA experienced detail evaluation before kidney transplantation. Ureter was reimplanted into the original bladder during transplantation and suprapubic cystostomy was performed as urinary drainage.MAIN OUTCOME MEASURES: Patient and graft survival, graft function, urinary tract infection (UTI) and graft rejection were supervised after surgery and quality of life was evaluated.RESULTS: Two patients recovered well with normal renal function and no rejection. Despite the moderate but easily controlled UTI, they handled the urinary diversion well and were satisfied with the quality of life. One patient experienced an acute rejection 17 days after transplantation and survived with functional graft until now after the rejection was reversed. One patient lost the graft for repeated hemorrhage at the site of vascular anastomosis, and then returned to hematodialysis.CONCLUSION: Kidney transplantation is suitable but special for patients with LUTA. Careful evaluation before transplantation, suitable urinary drainage and rigorous follow-up after surgery are keys to the success of kidney transplantation for this subgroup of patients.

6.Analysis on the Laws of Acupoint Selection in Acupuncture Treatment of Headache

Objective:To explore the application of data mining in the literature study on acupuncture treatment Of headache,and to summarize the laws between TCM pattern identification of headache and acupoint selection,acupoint combination.Methods:In the study of the clinical literature about acupuncture treatment for headache,published domestically from 1950 to 2008,the number and frequency of the acupoints in the literature,and the selecting laws and frequency of the main acupoints in pattern identification,and the meridian-tropism relationship between the main acupoints and combined acupoints,and the distributing rules of commonly used acupoints on the body surface were investigated by frequency-analyzing method.Resuits:The main acupoints in acupuncture treatment of headache ale primarily the acupoints on the head and specific acupoints from yang meridians,with the acupoints combined in accordance with pattern identification by meridian theory for different paaerns.Conclusion:By analyzing the laws of acupoint selection in the acupuncture treatment of headache with data mining,its new laws can be obtained.
7.Mitigation of safflor yellow injection on acute lung injury of rats induced by oleic acid

Objective To investigate the effects of safflor yellow (SY) injection on acute lung injury (ALI) rats induced by oleic acid (OA) and its potential mechanism. Methods Wistar rats were randomly divided into six groups,including control group,OA group,OA+10 mg/kg anisodamine group,OA+8 mg/kg SY group,OA+16 mg/kg SY group,and OA+32 mg/kg SY group. Normal saline or anisodamine or SY were pretreated before 0.18 g/kg OA iv injection. The arterial partial pressure of oxygen,the pulmonary water content index,and MPO activity in lung tissue were determined; The mRNA level of TNF-?,IL-1?,IL-6,ICAM-1,and VCAM-1 were measured by RT-PCR; And NF-?B p65 protein in nucleus observed by immunohistochemical technology,while the level of p38 MAPK protein phosphorylation in lung tissue was analyzed by Western blotting. Results The arterial partial pressure of oxygen in all SY groups was higher than that in OA group,while the pulmonary water content index and MPO activity were lower than those in OA group,as well as the mRNA level of the above inflammatory cytokines and the NF-?B p65 in nucleus and level of p38 MAPK phosphorylation. Conclusion SY could alleviate the lung tissue edema,increase the arterial partial pressure of oxygen,and depress MPO activity in ALI rat induced by OA. The SY mechanism of attenuating the acute lung injury may be associated with inhibiting the p38 phosphorylation and NF-?B activation and reducing inflammatory factors expression.

9.Effects of piracetam on chronic epilepsy in childhood rats and quantitative effects on content of acetylcholine and activity of cholinacetyltranslase in hippocampus in learning-memory complex animal model

BACKGROUND: Cholinergic system projected in brain and hippocampal structure is relevant with learning and memory. Piracetam acts on protecting and repairing cerebral neural cell, resisting cerebral functional injury due to physical and chemical factors and improving learning-memory capacity.OBJECTIVE: Chronic epilepsy in childhood animal and learning-memory complex animal model were self-prepared to observe the changes in content of acetylcholine and activity of cholinacetyltranslase in cerebral hippocampus and the intervention of piracetam.DESIGN: Randomized control experiment and non-blind evaluation were designed.SETTING: Department of Pediatrics of Second Hospital of Hebei Medical University and Institute of Traditional Chinese Medicine of Hebei Medical UniversityMATERIALS: The experiment was performed in Hebei Medical University and College of Life Sciences of Hebei Normal University from July to December 2004, in which, 50 Wistar childhood rats of clean grade and either sex were employed.METHODS: Coriamyrthin injection was administrated muscularly to duplicated chronic epileptic grand mal model in rats. Muscular injection was repeated once every three days. During modeling, those with general paroxysmal convulsion with posterior extremities standing or falling with standing or general stiffness-paroxysmal attack continuously for 3 times, the injection was changed to be once every 14 days. Ten rats were selected to be in normal control without modeling. The rest 40 rats after 3 months of modeling were randomized into 4 groups, named piracetam of 2.4 g/'L group (Group A), piracetam of 4.8 g/L group (Group B), dilantin 6 g/L +piracetam 4.8 g/L group (Group C) and model group (Group D), 10 rats in each. In each group, gastric infusion was performed continuously in 3 months after modeling, once per day, 10 mL/kg. In Group A and Group B,piracetam mixed solution of 2.4 g/L and 4.8 g/L was administrated for infusion respectively. In Group C, dilantin 6 g/L and piracetam 4.8 g/L were infused. In group D and the control group, normal saline 10 mL/kg was administrated. Relevant index determination was done 1 month after medication. Morris water maze test was performed to discover platform time and searching distance of epileptic rats, continuously for 3 days, twice per day. After test, the rats were sacrificed to collect brains to determine the content of acetylcholine in bilateral hippocampus. In Group C and Group B, the searching time of 6 tests was decreased of various degrees compared with Group D [(44±9) s, (45±9) s;(43±9) s, (42±8) s; (42±7) s,(42±7) s; (40±9) s, (39±9) s; (38±7) s, (35±9) s; (35±6) s, (34±8) s,t=2.352-4.029, P < 0.05-0.01]. In every medication group, the average searching time was decreased gradually by the increased frequency of erage searching distance in Group D was remarkably increased compared with the control [(793±74) cm, (420±81) cm;(763±89) cm, (418±57) cm;(690±67) cm, (382±69) cm; (623±81) cm, (356±71) cm;(592±98) cm,(330±69) cm;(550±54) cm,(301±97) cm,P< 0.01]. In Group C and Group B, the average searching distance of 6 tests was decreased of various degrees compared with Group D [(586±91) cm, (510±89) cm;(566±70) cm,(497 ±76) cm; (521 ±84) cm, (455 ±56) cm; (480 ±74) cm, (421 ±63) cm;(437±51) cm, (396±79) cm;(392±79) cm, (385±48) cm, t=2.364-4.230, P < 0.05-0.01]. In every medication group, the average searching distance tent of acetylcholine in brain hippocampus and the activities of cholinacetyltranslase and acetylcholinesterase of rats in each group: those in Group D were all remarkably reduced compared with the control [(2.2±0.7) nmol/g,(3.8±0.9) nmol/g;(503.3±103.3) pkat/g, (778.3±125.0) pkat/g;(190.0±51.7) μkat/g, (368.3±86.7) μkat/g, P < 0.01]. In mixed group and Group B, the content of acetylcholine and activity of acetylcholinesterase were remarkably higher than the Group D [(2.7±0.6) nmol/g, (2.9±0.6) nmol/g;(256.7±58.3) μ kat/g, (306.7±88.3) μkat/g, t=3.445-4.148, P < 0.01]. In Group B, the activity of cholinacetyltranslase [(668.3±118.3) kat/g] was remarkably higher than those in the Group D(P < 0.01). Every index in group A was basically same as model group.CONCLUSION: Grand mal of chronic epileptic rat model is characterized as declined capacity of spatial learning and memory and associated with decreased content of acetylcholine and the activities of cholinacetyltranslase and acetylcholinesterase in brain hippocampus, explaining the successful complex model of learning and memory disturbance. Piracetam 4.8 g/L may increase content of acetylcholine and the activities of cholinacetyltranslase and acetylcholinesterase in brain hippocampus and improve learning-memory capacity, but its effect at 2.4 g/L was not remarkable.
10.Exploratory factor analysis of "The Compliance Scale Among Kidney Transplantation Recipients"

Objective To explore the structure of self-developed "The Compliance Scale Among Kidney Transplantation Recipients" in order to provide effective tool to evaluate kidney transplantation recipients' compliance. Methods 886 follow-up kidney transplantation recipients of six organ transplantation centers in Shanghai were surveyed by "The Compliance Scale Among Kidney Transplantation Recipients". Results The exploratory factor analysis yielded a 25-item four-factor model termed medication compliance, life habits compliance, self-monitoring compliance, follow-up compliance. Conclusions The self-developed "The Compliance Scale Among Kidney Transplantation Recipients" could be a useful tool for evaluating compliance among kidney transplantation recipients.
