1.Hereditary spastic paraplegia with simultaneous mutations of Atlastin and Nipa1 genes in one family

Objective To analyze the simultaneous mutations in Atlastin and Nipa1 gene carried by a family with dominantly inherited HSP.Methods The Atlastin exons and Nipa1 exons were analyzed for the family members by ABI 3100 Genetic Analyzer.Results Genetics analysis revealed that both probands and her daughter were heterozygous for disease-specific missense mutations in both SPG3A/Atlastin(SPG3A P344L)and SPG6/Nipa1 genes(SPG6 T100A).Neither of the proband's parents had the SPG3A/Atlastin mutation.But the proband's father was heterozygous for the SPG6/Nipa1 gene mutation.Conclusion The simuhaneous mutations in both SPG3A/Atlastin and SPG6/Nipa1 are coexistent in the patients of this family.SPG3A/Atlastin is the de novo non-inherited mutation.
2.Status quo of medical tourism and its implication in China

Medical tourism, which is a new type of tourism activity with the combination of health and sightseeing, has become the new economic growth point of the tourism industry.Through a systematic literature review, the present paper analyzed the influencing factors and effects of medical tourism, as well as the behavior of medical tourists and relevant stakeholders of medical tourism.Further researches could combine qualitative and quantitative methods, and emphasize on the behavioral characteristics of medical tourists.In order to promote the development of medical tourism, we should increase the medical service qualities, promote the internationalization of medical tourism, introduction of special medical tourism products;improve medical tourism policies, regulations and laws;promote medical tourism through a strategic marketing and the role of relevant stakeholders of medical tourism should be paid attention on, and other measures to promote the healthy development of China''s medical tourism.
3.Analysis on status and determinants of self-treatment of rural floating population in Beijing

Objective:To understand the status and identify the determinants of self-treatment of rural floating population in Beijing. Methods: The data were derived from the Fourth National Health Service Investigation of China, part of which was about rural floating population in Beijing. According to Andersen health services utilization model, Binary Logistic Regression model was used to analyze the influencing fac-tors. Results:In the study, 51. 7% of the research objects were inclined to self-treatment when they ever felt uncomfortable or sick in Beijing. The Binary Logistic Regression model suggested that, some factors from predisposing, enabling and need, were statistically significant determinants of utilization of self-treat-ment. Conclusion:There exists a certain percentage of self-treatment behavior for rural floating population in Beijing, and we should take some measures to promote the efficacy and safety of self-treatment.
4.The influence of silencing miRNA-155 on proliferation and apoptosis of human cutaneous squamous cell carcinoma cell line A431

Objective To investigate the effects of antisense oligonucleotide against miRNA-155 (AS-miRNA-155) on proliferation,apoptosis and invasion and migration abilities of human cutaneous squamous cell carcinoma cell line A431. Methods AS-miRNA-155 was transfected into human cutaneous squamous cell carcinoma A431 cells by using LipofectamineTM 2000. Blank control without transfection and transfected with non-sense sequence were used as non-sense sequence control. Quantitative real-time polymerase chain reaction (qRT-PCR) was performed to detect the expression of miRNA-155 in A431 cells. Cell proliferation was analyzed using dimethyl thiazolyldiphenyl tetrazolium (MTT) assay. Cell cycle arrest and apoptosis were studied by flow cytometry (FCM). Invasion and migration were measured by Transwell chamber assays. Results The relative expression of miRNA-155 mRNA was lower in the transfection group than that in the blank control group and the negative control group (F=634.57, P<0.01), but there was no significant difference between the blank control group and the negative control group. After 72 h transfection, the survival rate was significantly lower in the transfection group than that of the blank control group and the negative control group, and the transfection rate decreased significantly by 120 h (P<0.05). Cells of G0/G1 phase increased, Cells of S phase reduced, the overall PI value decreased in transfection group, and the apoptosis rate of A431 cells, migration and invasion of cells increased (P<0.05). There was no significant difference in G2/M cycle between transfection group, blank control group and negative control group. There were no significant differences in A431 cell apoptosis rate, cell migration and invasive ability between blank control group and negative control group. Conclusion Antisense oligonucleotide against miRNA-155 can inhibit the expression of miRNA-155, the proliferation and promote the apoptosis of human cutaneous squamous cell carcinoma A431 cells, which indicates that miRNA-155 may become a new target for the regulation of gene expression in cutaneous squamous cell carcinoma.
5.Study on the status quo of medical personnel in the rural health clinics of underdeveloped areas -Taking Fengjie County as an example

Objective To investigate the status quo of the medical staff in 6 village clinics in Fengjie County, and to put forward countermeasures and suggestions for improving the quality and overall quality of medical personnel in village clinics. Methods The research literature and questionnaires were used to investigate the basic situation of the village doctors, the role of the village doctors in the residents' health care, and the needs and satisfaction of the residents in the village clinics. The data were analyzed by SPSS 19.0, and the samples were analyzed by descriptive statistics. Results The average age of the medi-cal staff in village clinics was (46.74±10.17) years old, and there was no medical staff under 30 years of age. The medical staff of the village health clinics had an irrational age structure, and the educational back-ground was generally low; the medical staff generally thought that their income was not high. They hope to participate in more training in medical technology; residents generally believe that the doctors' ability to diagnose and treat the disease needs to be improved. Conclusion It is necessary to strengthen the leading role of the government and bring into play the positive work of the medical staff in the village health clinic. It is also necessary to strengthen the training of talents, lay a solid foundation for development, take the demand as the guide to change the service concept and model, to improve the relevant mechanisms to create good surroundings for the development of medical staff in village clinics.
6.Research on nosocomial infection and drug resistance of Pseudomonas aeruginosa

Objective To investigate the distribution and drug resistance state of Pseudomonas aeruginosa, so as to offer reasonable experimental data for clinical therapy. Methods A statistic analy- sis for the specimen souree, distribution of infected departments and drug resistance state to 441 iso- lates was conducted in a retrospective way. Results Among 441 strains, 357 (80. 95%) were isolated from sputum species. The department with the highest isolating rate was ICU (26.5%), followed by department of respiratory disease (20. 63 %), department of neurosurgery (19.05 %) and department of geriatrics (17. 01%). The most sensitive antibiotic was Meropenem, which total drug resistance rate was 13. 61%; the most insensitive antibiotic was ampicillin-sulbactam, which total drug resistance rate was 91.38%. The drug resistance rate of P. Aeruginosa was increasing year by year. Conclusion Clinical isolates of P. Aeruginosa mostly come from sputum samples and ICU, and they are highly and multidrug resistant to various antibiotics.

9. mRNA expression of BCSG1, BTF, Bcl-2, Erα and ERβ in osteosarcoma and its significance

Objective:To observe the mRNA expression of BCSG1 (breast carcinoma specific gene, BCSG1), BTF(Bcl-2-associated transcription factor, BTF), Bcl-2 (B cell lymphoma/leukemia-2, Bcl-2) and two subtypes of estrogen receptors in osteosarcoma and their implication. Methods: The expression of BCSG1, ERα, BTF, ERβ, and Bcl-2 mRNA were examined in 16 poorly differentiated osteosarcoma samples and a cell line hFOB 1.19 by quantitative RT-PCR. The relationship between their expression was analyzed. Results: The mRNA expression of BCSG1, Bcl-2 and two subtypes of estrogen receptors in the osteosarcoma tissues was significantly higher than that in the osteoblast cell line of hFOB 1.19; the expression of BTF was not significantly different. ERα/ERβ in the osteosarcoma tissues was significantly higher than that in the hFOB 1.19 cells (P< 0.01). BCSG1, ERα, ERβ, Bcl-2 and BTF expression was not associated with the tumor size or patients gender. Correlation analysis showed there was a linear relationship between expression of BCSG1 and ERα, ERβ and Bcl-2, and ERα/ERβ ratio and Bcl-2. Conclusion: BCSG1 may enhance the activation of estrogen receptor-α gene transcription, and ERβ may influence the expression of Bcl-2 in osteosarcoma.
10.The correlation between serum levels of IL-33,TNF-α and disease severity,prognosis in patients with acute pancreatitis

Objective To explore the reference value of interleukin(IL)-33 and tumor necrosis factor-alpha(TNF)-α in early diagnosis,illness evaluation and prognosis of acute pancreatitis(AP) by dynamically monitoring the serum levels of IL-33 and TNF-α.Methods A total of 86 patients with AP were selected as objectives which were divided into two groups according the severity of illness which include 59 cases of mild AP group(MAP) and 27 cases of severe AP group(SAP),52 cases of systemic inflammatory response syndrome(SIRS) group and 34 cases of non SIRS group distinguished by SIRS,75 cases of survival group and 11 cases of death group identified by prognosis of AP.All of patients were assessed by Bedside Index of Severity in Acute Pancreatitis(BISAP) score within 48 h after admission and detected the serum levels of IL-33 and TNF-α at the first day,third day,seventh day and fourteenth day during hospital.A total of 63 healthy persons were recruited into control group.Results The serum levels of IL-33,TNF-α and BISAP score of SAP group and MAP group were higher than those of the control group in different monitoring time,but the SAP group increased more obviously than MAP group.The serum levels of IL-33 and TNF-α of SAP group were higher than those of MAP group at the first day,third day,seventh day and fourteenth day during hospital,and the differences between them had statistical significance(P<0.05).The serum levels of IL-33 and TNF-α of SAP group increased to peak at the third day and reduced gradually after conventional treatment(the seventh day in the hospital).The serum levels of IL-33 and TNF-α of SIRS group were significant higher than those of non SIRS group(P<0.05).The serum levels of IL-33,TNF-α and BISAP score of death group were significant higher than those of survival group(P<0.05).Spearman correlation analysis suggested that there were positive correlations between BISAP and the serum levels of IL-33 and TNF-α of AP patients.Conclusion There is an important clinical value to the early diagnosis,severity classification,guiding clinical treatment and the assessment of prognosis of AP by dynamically monitoring the serum levels of IL-33 and TNF-αof AP patients.
