Objective To explore the possible association of oxidative stress reaction with the mRNA expression of podocin in rats with adriarnycin(ADR)-induced nephrotic syndrome.Methods mRNA expression of podocin in renal cortex were investigated by in situ hybridization staining and semi-quantitative RT-PCR in ADR-induced nephrotic rats,the level of malondialdehyde(MDA), superoxide dismutase(SOD)and total antioxidative capacity(T-AOC) in renal cortex were measured, then the relationship between them was evaluated.Results In ADR rats, MDA increased at d7 and reached a significant higher level at d14 and d21;SOD decreased at d14 and persisted to d21; T-AOC decreased at d21.The podocin mRNA mostly expressed in cytoplasm of glomerular cells. Both the number and the intensity of positive cells increased notably as time progressed in ADR rats.The level of podocin mRNA expression showed no obvious changes at d7, while prominently increased at d14 and pesisted to d21 in ADR rats.There was significant correlativity between the mRNA expression of podocin and the level of MDA,SOD,T-AOC.Conclusion The mRNA expression of podocin is correlated with oxidative stress reaction in rats with ADR-induced nephrotic syndrome.

Background Dynamic observation in morphology and histology of conjunctival filtering bleb in rats can contribute to form ideal filtering blebs by interferencing the process of scarring.Objective This study was designed to observe the survival time,morphological and histopathological changes of conjunctival filtering bleb after filtering surgery.Methods Filtering surgeries were performed on 60 male adult Sprague-Dawley rats by implanting drainage tubes into the anterior chamber.The shape of filtering blebs was examined by slit lamp microscope 1 day,3,5,7,14,28 days after surgery.The filtering was typed according to the criteria of Kronfeld.Areas of the filtering bleb were measured and compared at various time points after surgery.Conjunctival and subconjunctival tissues at filtering zone were obtained 1 day,3,5,7,14,28 days after surgery,respectively,and hematoxylin and eosin staining was used for the histopathological evaluation.Results Well-elevated filtering blebs formed on the first day after surgery in 60 rat eyes.The survival time of filtering bleb was 7-17 days with the mean time of (11.93±2.23) days.Survival rate of the filtering blebs was 100％ in 1-5 days,77％ in 7 days,20％ in 14 days and 0 in 28 days after operation.The filtering blebs were functional (Ⅰ type) in 1-5 days after operation,23 eyes were functional in 7 days after operation (13 eyes were Ⅰ type,10 eyes were Ⅱ type),4 eyes were functional in 14 days after operation (2 eyes were Ⅰ type,2 eyes wereⅡtype).The filtering area shrunken from (7.50±1.08) mm2 to (1.83±0.35) mm2during the observational duration.The filtering blebs area of different days after surgery were statistically significant (F =180.056,P=0.000),and significant differences were found between any two timepionts after surgery (all at P =0.000).Histological examination showed that inflammatory finding was dominant in postoperative day 1-3,such as infiltration of inflammatory cells,vasodilatation and edema of tissue.However,proliferating manifestation became the main signs 5 days later,showing the increase of fibroblasts,neovascularization and collagen formation.Conclusions The model of conjunctival filtering blebs in rats can be successfully established by implanting silicone tube into anterior chamber.The inflammatory response,proliferation of fibroblasts and the accumulation of collagen fibers are involved in the scarring process of filtering blebs.

Human CD3AK cells were prepared from peripheral blood mononuclear cells by culturing with recombinant IL-2 and antiCD3AK McAb. The mechanism and regulation of CD3AK cytotoxic activity with cytokines (rhIFN-?, rhIFN-?, TNF) and chernotherapeutic agents (CDDP or ADM) were observed by LDH-release assay, ABC-CELISA and the flow cytometric assay. The results showed: (1) Adhesion molecules ICAM-l/LFA-1 participated in CD3AK-mediated killing of tumor cells, hrlFN-? and TNF enhanced cytotoxicity of CD3AK through this pathway. (2) CD3AK could indirectly kill tumor cells by releasing soluable cytotoxic factors. (3) The membrane-associated TNF may be involved in CD3AK-mediated cytotoxicity. (4) CD3AK cells could induce the apoptosis of tumor cells. (5) Pretreatment of tumor cells with CDDP or ADM resulted in the increased vulnerability of tumor cells to CD3AK-mediated killing, the enhancement of CD3AK-mediated cytotoxicity by CDDP was relative to the increased expression of ICAM-1, HLA-ABC on tumor cell membrane.

Objective To investigate the in vitro susceptibility of Minocycline and polymycin B against clinical islates of pan-drug resistant Acinetobacter baumanii ,to provide laboratory support for clinical treatment for drug selection.Methods The susceptible test of Minocycline and polymycin against 39 isolates of pan-drug resistant Acinetobacter baumanii were determined by K-B meth-od.Results 38 strains of pan-resistant Acinetobacter baumanii were sensitive to polymyxin B,sensitive rate was 97.4%,20 strains sensitive to minocycline,the sensitivity rate was 51.3%.polymyxin B sensitivity was more sensitive than Minocycline (P < 0.05). Conclusion Polymycin B had strong activit ies against pan-drug resistant Acinetobacter baumanii .

Objective To investigate the effect of the genotypic analysis of donor from cardiac death donation on the initial dose of Tac for liver transplant recipients and provide individualized administration for the early use of Tac in liver transplantation patients.Method Thirty recipients with a different genotype of CYP3A5 from cardiac death donors were collected from March 2010 to February 2013.The matched recipients were randomly divided into experiment group and control group.There was an adjustment of initial doses of Tac according to the donors' different CYP3A5 genotypes in experiment group but not in control group.Result In experiment and control groups,the average Tac blood concentrations at the 7th day after operation were (7.47 ± 1.83) and (8.68 ± 5.14) ng/mL,and the percent of recipeints reaching the optimal Tac concentrations was 72.2％ and 38.9％,respectively (P＜0.05).In experiment and control groups,22.2％ and 55.6％ recipients needed adjustments of Tac concentrations respectively (P＜0.05).Conclusion Individualized adjustment of Tac initial doses of recipients according to cardiac death donors' different CYP3A5 genotypes was benefit for reaching optimal concentrations as soon as possible and could decrease the rate of rejection,and reduce the side effects of Tac.

In this paper , the causes of patients in emergency ICU were analyzed .The authors used classical theory of ethics to illuminate and put forward the ethical principles that both doctors and patients should follow and the conception of patients′moderate right , as well as the relevant suggestions and strategies , with the aim to make the application of emergency ICU medical resources more reasonable and make the treatment and cure of the emer -gency and critical patients more conform to the requirement of the ethics .
