Sepsis, the most common panthogeny, is characterized as multi-system injuries like immune system, inflammatory system and coagulation system, caused by systemic inflammatory response syndrome ( SIRS) and excessive oxidative stress after the invasion of pathogen.Severe cases can lead to multiple organ dysfunction syndrome and even death.Acute kidney injury is an independent risk factor which has an influence on patients'prognosis.Currently, the research of biomarkers of AKI is a significant field. Some new biomarkers are found to be charaterised by early appearance in blood or urine, non-traumatic and sensitivity in recent years, which contribute to their applications in early diagnosis of AKI.At present, the biomarkers of early diagnosis of AKI, such as calprotectin, interleukin-18, neutrophil gelatinase associated lipocalin (NGAL), cysteine protease inhibitor C, kidney injury molecule-1 (KIM-1) and cysteine rich 61, are under study. The current basic research and a few clinical studies show that these indicators may have a better sensitivity .

Objective To study the regulatory T cells (Treg-cell) frequencies in patients with childhood acute leukemia and evaluate its clinical application value by investigating the relationship between the increasing numbers of Treg cells and minimal residual disease of leukemia (MRD), Methods Foxp3-FITC/CD25-PE/CD4-PerCP/CD3-APC four-color staining flow cytometry was established to identify Treg cells. Treg cells frequencies both in 10 healthy controls and in 33 patients with newly diagnosed childhood acute leukemia ( B-ALL 17 cases, T-ALL 9 cases, AML 7 cases) were detected. The possibility of the accumulation of Treg cells being the prognostic marker for acute leukemia was evaluated by comparing the results of Treg cells frequency with that of MRD. Results The percentage of Treg cells in CD+4 CD+3 T cells was M = 8. 09% in normal bone marrows, which was significantly different from the results in the bone marrows of newly diagnosed childhood acute leukemia ( M = 12.77% , U = 3.41, P < 0.01 ), but it showed no significantly differences among B-ALL, T-ALL and AML groups. No association was observed between the expression of Treg cells and clinical-biologic characteristics studied. In addition, Treg cells frequency in MRD positive group was significantly different from that in MRD continuously negative group ( M = 14. 74% vs M=11.3%, t =252.5,P<0.05). Conclusions Consistent with results from solid tumor, the study identifies a significantly increased numbers of Treg cells in patients with childhood acute leukemia. The situation of accumulation of Treg cells is closely associated with MRD results during chnical remission. High level of Treg cells may cause poor prognosis and increase the possibility of relapse.

In order to investigate the effect of malic enzyme on anaerobic mixed acid fermentation, the gene encoding NAD+-dependent malic enzyme was amplified from E.coli DH5? genome by using PCR and cloned into vector pTrc99a to give an expression vector pTrc99a-sfcA.Malic enzyme was overproduced by E.coli FMJ39(ldh,pfl)harboring pTrc99a-sfcA under the condition of IPTG induction. The results of anaerobic fermentation indicated overexpression of malic enzyme would have influence on the pathway of formate, acetate and succinate of FMJ39.The concentration of formate and acetate was 17.58% and 15.27% higher than FMJ39,the succinate concentration was reduced by 26.87%.There was no obvious citrate concentration change. The experimental results also indicated that high concentration of L-Thr and L-Ser would induce the Tdc operon to convert pyruvate into formate and acetate even when pfl gene encoding the pyruvate formate lyase was inactivated.

Objective To investigate the effects and mechanisms of selective angiotensinⅡtype 1 receptor antagonist ZD7155 on the inhibition of pancreatic cancer in vitro. Methods MTT assays were used to determine the inhibition of pancreatic cancer cell line PaTu8988s by ZD7155 in different concen- trations and at different time.PaTu8988s cell cycle and cell apoptosis were detected by flow cytometry. Transmission electron microscope was used to investigate the apoptosis of PaTu8988s before and after the incubation with ZD7155 under different concentrations.PaTu8988s cell morphology was observed be- fore and after the incubation with ZD7155.Results MTT showed that the increase of inhibition of pan- creatic cancer cell by ZD7155 was in agreement with the increase of the concentrations of ZD7155 and the time of the incubation with ZD7155.The inhibition rates of PaTu8988s cells were 9%,18%,30%, 51%,60% and 78% by ZD7155 with the concentrations of 5?10~(-11),5?10~(-10),5?10~(-9),5?10~(-8),5?10~(-7) and 5?10~(-6) mol/L,respectively.The inhibition rates of PaTu8988s cells were 15%,25%, 36%,51%,67% and 85% by ZD7155 with the same concentration(5.0?10~(-8) mol/L)at 12,24,36, 48,60 and 72 hours,respectively.ZD7155 could also inhibit PaTu8988s cell cycle significantly and was dose-dependent.Cell electron microscopy showed that there were chromatin margination and apoptotic body in the cell nucleus when the cells were incubated with ZD7155,and these changes were increase with the concentrations of ZD7155.The morphology of PaTu8988s cell didn't have any change after in- cubation with ZD7155.Conclusions ZD7155 can inhibit the growth of pancreatic cancer cells in vitro by suppressing the S-phase of cell cycle and induce cell apoptosis without visible cell toxic effects.

·AIM: To investigate whether Erigeron Breviscapus (vant) Hand-Mazz (EBHM) EBHM has neuroprotective effect against N-methyl-D-aspartate (NMDA)-induced neuron death in retinal ganglion cell layer (RGCL).· METHODS: 60 healthy SD rats were randomly divided into four groups. 6 animals were normal control group (group A). The others were divided as group B (EBHM group), group C (normal saline+NMDA group) and group D (EBHM + NMDA group). Each group had 18 rats.10nmol NMDA was intravitreally injected to induce partial damage of the neurons in RGCL in the right eyes of Groups C and D. Same volume PBS was intravitreally injected into the left eyes as self-control. Groups B and D were pre-treated intraperitoneally with 6g/L EBHM solution at a dose of 150mg/kg body weight/day seven days before and after NMDA treatment. Group C were administrated intraperitoneally with 9g/L normal saline at the same time of EBHM injection. Rats were sacrificed at 4,7,14d after NMDA treatment. Flat whole retinas were stained with 5g/L cresyl violet and neuron counting in RGCL from both eyes were observed. Each subgroup had 6 rats.· RESULTS: There was no significant difference of neuron counting in RGCL between the right eye and the left eye in group A (P=0.200). There was no significant difference between normal control group and EBHM group either in the right eyes or in the left eyes at 4, 7 and 14 d respectively after intravitreal injection of 10nmol NMDA in group C and group D. (P=0.636, P=0.193). Neuron counting of RGCL in group C and D was significantly decreased in the NMDA-treated eyes at 4, 7 and 14d after intravitreal injection (P<0.001). There was no significant difference between self-control eyes group and normai control group(P>0.05). However, neuron counting was significantly higher in the EBHM+NMDA group than normal saline +NMDA group at 14days after intravitreal injection (P=0.044), but was lowered than normal control group (P<0.05).· CONCLUSION: EBHM has no effect on neuron counting of RGCL when administered alone in normal rats. The results indicates that EBHM plays a partial protective role in NMDA-induced neuron loss in RGCL in the rats.
