Objective To investigate the availability and mechanism of recombinant adenovirus vector expressing wild type p53 in gene therapy of prostate cancer. Methods Prostate cancer cell line PC3M was cultivated and recombinant adenovirus vector expressing wild type p53 was added to the media. Then the levels of proliferation and apoptosis were detected and changes of bax were exa mined . Results AdCMVp53 is effectively transfected to the cells. The transfecting ratio is 95%. In the cells transfected with AdCMVp53, the level of proliferation decreased. Tested by BrdU, the absorbance rate decreased from 1.454 in the control group to 0.496 in the experimental group( P

Radiotherapy is an important treatment on gynecological tumors,especially for brachyther- apy.~(137)Cs,~(192)Ir were usually used in the past,but the local control and survival rates were not increased obvi- ously.The machines,that was made in China,of ~(252)Cf neutrons brachytherapy were used already by many hos- pitals in our country and played a preponderant role more and more.It can increase the local control and sur- vival rates effectively on gynecological tumors.
4. Study on preparation of butyryl galactose ester-modified coix component microemulsions and their anticancer activity in vitro

Objective: To synthesize butyryl galactose ester (But-Gal) and prepare butyryl galactose ester-modified coix component microemulsions (But-Gal-CMEs) and to evaluate its physicochemical properties and anticancer activity in vitro. Methods: But-Gal was synthesized by enzyme-catalyzed reaction and the structure of the product was confirmed by 1H-NMR and FT-IR. The CMEs and But-Gal-CMEs were prepared by aqueous titration method using coix seed oil, Cremophor RH40, PEG400, But-Gal, and coixan solution as oil phase, surfactant, cosurfactant, target ligand, and aqueous phase, respectively. The average particle size, polydispersity index (PDI), and Zeta potential were detected by dynamic light scattering (DLS). The cytotoxicity of CMEs and But-Gal-CMEs aganist HepG2 cells was determined by MTT assay. The cellular uptake of CMEs and But-Gal-CMEs was detected by fluorescence microscopy. Results: The structure of But-Gal was confirmed by 1H-NMR and FT-IR. The But-Gal-CMEs displayed the spherical surface with mean droplet size of (57.68 ± 6.65) nm, PDI of 0.070 ± 0.006, and Zeta potential of (-2.95 ± 0.23) mV, respectively. MTT experiments showed that the half of HepG2 cell proliferation inhibition concentration (IC50) of But-Gal-CMEs and CMEs was 62.55 and 71.23 μg/mL. The HepG2 cell uptake results suggested that the fluorescence intensity of But-Gal-CMEs group was higher than that of CMEs group. Conclusion: The But-Gal-CMEs presents small particle size, good roundness, and good stability. In addition, But-Gal could increase the uptake rate of CMEs in HepG2 cells and enhance the inhibition of HepG2 cell proliferation.
5. Correlation analysis of seminal plasma free testosterone and L-carnitine with semen parameters

Objective: To analyze the correlation of seminal plasma free testosterone and free L-carnitine with semen density, vitality, and motility, so as to investigate their effects on male fertility and their roles in the examination and treatment of male infertility. Methods: Using chemiluminescence immunoassay method, LC-MS-MS and computer-assisted semen analysis system, we examined the seminal levels of free testosterone, free L-carnitine and sperm density, vitality, and motility in 192 infertile males (normozoospermia, n = 36; Oligozoospermie, n = 48; asthenozoospermia, n= 60; and oligoasthenozoospermia, n = 48). The difference of free testosterone and free L-carnitine levels between different groups was analyzed, and the correlation of their levels with semen density, vitality, and motility was also analyzed using the SPSS 13.0 statistical software. Results: The levels of free testosterone and free L-carnitine in the seminal plasma of the normozoospermia men were significantly higher than those of others (P<0.05). Correlation analysis showed that there was positive correlation between seminal free testosterone with the sperm density (r=0. 427, P<0.05), seminal free L-carnitine with the sperm density (r=0.318, P<0.01), and seminal free L-carnitine with sperm vitality, motility (r=0.641, P<0.01; r=0.568, P<0.01). Conclusion: Seminal free testosterone level is correlated with the sperm density, and seminal free L-carnitine level is correlated with sperm density, motility and vitality; both levels can be taken as a biochemical index for the clinical diagnosis and treatment of male infertility.
7.Mechanism and Chemical Basis of the Effect of Bacteria on Urinary Stone Formation

There are two populations of bacteria to affect the formation of urinary stones in humanity. The first one can promote the formation of urinary stone by increasing urinary pH, decreasing concentration of urinary inhibitors , and damaging the protective urothelial glycosaminoglycan layer. The second inhibit the formation of urinary stones. These bacteria ( mainly the intestinal oxalate degrading bacteria such as Oxalobacter formigenes, lactic acid bacteria, Enterococcus faecalis etc) can decrease urinary oxalate concentration by regulating exogenous oxalate. The problems faced and the developing direction were also indicated.

Objective To explore the clinical value and the difference of bone mineral density(BMD) in every vertebra of lumbar spine in middle old-aged Chinese female with quantitative computed tomography (QCT).Methods The BMD in every vertebra of lumbar spine was investigated by QCT in 725 healthy females aged from 35 to 98 years. They were divided into groups every five years.The BMD in vertebra of lumbar spine (L 1~L 4) were measured by QCT and the groups tested for significant differences,using SPSS10.0 software for the analysis.Results Lumbar spine (L 1~L 4) BMD declined with aging. 9.The effects of extracorporeal shock waves on the lumbar spinal cord

Objective To investigate whether or not extracorporeal shock wave (ESW) treatment is safe for treating low back pain caused by lumbar facet joint degeneration by observing its effects on the lumbar segment of the spinal cord.Methods Five adult dogs were used for this study.ESWs were focused on their right L_(4~5) facet joints. The energy output was set at 40% of the maximum output,and a total of 1000 shocks were delivered at a frequency of 120/min.Before and 3 days after the treatment,the somatosensory evoked potentials (SEPs) induced by stimulating the tibial nerve were recorded from the animals' scalps,and the motor function of their hind limbs was evaluated with Tarlov's scale.The animals were then sacrificed and the L_(4~5) segments of their spinal cords were taken for histopatho- logical observation,with the T_(12) segments taken as controls.Results Three days after ESW treatment,the left and the right hind limbs scored 5 and 4 on the Tarlov scale.The amplitude of SEPs decreased and their latency increased. Histopathological observation showed mild swelling of the L_(4~5) segment of the spinal cord with congested veins on the surface.Erythrocyte and leukocyte exudation as well as myelin sheath swelling in the spinal cord were also observed un- der the microscope,but with no demyelination.Conclusion ESW had some adverse effects on the spinal cord when focused on the facet joint.Further studies of the safety range of ESW energy and distance are needed.
10.The influence of acupuncture on Nestin expression in the cortex of hypoxia-ischemic neonatal rats

Objective To investigate the influence of acupuncture on Nestin expression in the cerebral cor- tex of hypoxia-ischemic neonatal rats.Methods A total of 72 Sprague-Dawley(SD)rats aged 7 days were random- ly divided into three groups:A normal group(n=24),the hypoxic-ischemic encepbalopathy(HIE)group(n=24), and an acupuncture+HIE group(n=24).The rats were sacrificed at time points including 3d,7d,14d and 28d af- ter insult,their brains were removed and sections were made.Any nerve stem cells(NSCs)in the cerebral cortex were detected using immunohistochemical staining of Nestin.Results Nestin-positive cells were found in the cere- bral cortex in the normal neonatal SD rats early after birth.After HI insult,the Nestin-positive cells had increased in the injured areas of the cortex and in contralateral mirror areas,especially around the injured areas.Peak Nestin ex- pression was 3 days after insult.In the acupuncture+HIE group,Nestin positive cells were significantly increased when compared with that in the HIE group,the peak was postponed,and the peak was longer than in the HIE group. The Nestin positive cells decreased,were seldom seen,and were not different among three groups 28 days after the HI insult.Conclusions Endogenous neural stem cells in the cerebral cortex were increased by acupuncture in hy- poxia-ischemic in neonatal rats,and acupuncture made the peak of Nestin expression longer.
