1.Comparative Study of SFE-CO_2 and Ultrasonic in Extracting the Water Decocted Radix Salviae Miltiorrhiza

Objective To study the different extraction methods for the content of tanshinoneⅡA in water decocted Radix Salviae Miltiorrhiza, in order to develop comprehensive utilization of Salvia miltiorrhiza. Method Tanshinone ⅡA was extracted by supercritical carbon dioxide fluid and compared with ultrasonic extraction. Result When the extraction pressure was 35 MPa, extraction temperature was 40 ℃, extraction time was 2 h and entrainer (ethanol) was 100 %, the yields of the tanshinone ⅡA was up to 3.87 mg, while the ultrasonic extraction was 2.89 mg. Conclusion TanshinoneⅡA extracted by supercritical carbon dioxide fluid was better than ultrasonic extraction and had higher purity. The comprehensive utilization of Salvia miltiorrhiza is available.

3.Effect of Ligustrazine on Expression of Induced Nitricoxide Synthase with Cisplatin-induced Ototoxicosis

Objective To observe the expression of induced nitricoxide synthase(iNOS)in ligustrazine against cisplatin-induced ototoxicity.Methods Fifty white guinea pigs with normal hearing were randomly divided into the control group,ligustrazine group and traditional Chinese drug(TCD)group.The alternations of hearing function were evaluated by the examination of auditory brainstem response(ABR).Some specimens were perfused by AgNO3 and observed the damage of bristle cell.And some were sent for immunohistochemistry to detect the expression of iNOS.Results Compared with the control group,the threshold of ABR in ligustrazine group and TCD group were significantly different.There was a significant difference between the ligustrazine group and TCD group.There was no expression of iNOS in Cortis',spiral ganglion cell and stria vascularis in normal cochlear(control group).In the ligustrazine group,the expression became stronger in spiral ganglion cell,stria vascularis,spiral ligament and Cortis'.INOS also expressed in TCD group.Conclusion Ligustrazine can antagonize cisplatin-induced ototoxicity by inhibiting the expression of iNOS.
4.Effects of ganglioside on cell apoptosis of rats with acute cerebral ischemia-reperfusion injury

BACKGROUND: Cell apoptosis and expression of related apoptotic gene are found in rats with acute cerebral ischemia-reperfusion injury.OBJECTIVE: To study the effects of ganglioside on cell apoptosis in rats with acute cerebral ischemia-reperfusion injury.DESIGN: Randomized control animal experiment.SETTING: Department of Neurology, China-Japan Friendship Hospital of Jilin University.MATERIALS: The experiment was conducted in the Animal Laboratory of China-Japan Friendship Hospital of Jilin University at April 2002.Forty-eight healthy male Wistar rats aged 3-4 months with the body mass of (220±50) g were selected and randomly divided into ischemia-reperfusion group and ischemia-reperfusion + administration group (intraperitoneal injection of ganglioside GM-1 at 30 minutes before ischemia) with 24 rats in each group, and each group was subdivided into three groups according to the reperfusion time: 3-hour, 6-hour and 24-hour with 8 rats in each time-point.METHODS: ①Rat models of cerebral ischemia-reperfusion were established. ②Diphenylamine method was adopted to detect changes of DNA splitting rate in brain tissues at 3 hours,6 hours and 24 hours after cerebral ischemia.100 mg of cerebral cortex was made into 10% homogenate by adding into 0.9 mL of splitting fluid,which was then put in the centrifuge tube for repeated freezing and melting. The supernatant and deposit were collected.DNA splitting rate = supernatant absorption/(supernatant absorption + deposit absorption). ③Immunohistochemical method was used to study the expression of protein kinase Cδ (PKCδ) as well as changes of ganglioside after administration. MAIN OUTCOME MEASURES: Changes of DNA splitting rate in cerebral cortex of rats as well as intensity of PKCδ expression.RESULTS: One rat in the normal saline control group died for exceeding anesthetization at the 6th hour of reperfusion, and 2 rats died at the 24th hour of reperfusion, which were supplemented respectively. With the time of reperfusion increasing, changes of DNA splitting rate significantly increased, which peaked at the 24th hour. The expression of PKCδ peaked at the 6th hour of reperfusion and gradually decreased. The DNA splitting rate and PKCδ expression were remarkably decreased at corresponding time-points in the ganglioside group.CONCLUSION: Ganglioside can inhibit cell apoptosis and reduce PKCδ expression after cerebral ischemia-reperfusion to protect cerebral ischemia-reperfusion injury.
5.Applications of global left ventricular myocardial strain in detection of anthracycline-induced cardiotoxicity in breast cancer survivors with postoperative chemotherapy

Objective To assess the clinical significance of three-dimensional speckle tracking imaging (3D-STI) in the evaluation of left ventricular myocardial function in breast cancer survivors with postoperative anthracycline-based chemotherapy.Methods A total of 51 breast cancer survivors with postoperative anthracycline-based chemotherapy were recruited and 31 female healthy volunteers as the controls.Conventional echocardiography and 3D-STI derived parameters [including global longitudinal strain (GLS) and global area strain (GAS)] were measured before and at 3 and 6 anthracycline-based chemotherapeutic cycles.The receiver operating characteristics (ROC) curve was constructed to determine optimal sensitivity and specificity of 3ID-STI derived parameters for the prediction of future cardiotoxicity.Results In comparison with the controls and baseline cases,GLS and GAS deteriorated significantly (P < 0.05 for both).No statistical difference in GCS and conventional parameters showed before and after the initiation of chemotherapy (P <0.05,respectively).The ROC curves showed that GAS as the best 3D-STI predictor of patients who develop cardiotoxicity during the follow-up.The area under ROC(AUC) of GAS was 0.894,and its optimal cut-off value was-28.4%,with a specificity of 88.0% and a sensitivity of 82.9%.AUC of GLS was 0.802,and its optimal cut-off value was-14.3%,with a specificity of 62.0% and a sensitivity of 85.4%.Conclusions Early decreases in GAS and GLS based on 3D-STI may allow the prediction of subsequent cardiotoxic development accurately in breast cancer survivors with postoperative anthracycline-based chemotherapy.
6.Effects of neostigmine on the transdiaphragmatic pressure and diaphragm evoked potential in rabbit

Experiments were performed on spontaneously breathing rabbits anesthetized with urethan. The diaphragmatic fatigue (DiF) was reproduced by the phrenic nerve stimulation (30 Hz, 10 V,0. 2 ms). The transdiaphragmatic pressure (Pdi) and diaphragm evoked potential (DEP) were measured before injury and after injury 30, 60, 90, and 120 min. Pdimax and the amplitude of DEP obviously decreased and the latency of DEP obviously delayed after injury 30min. The amplitude of Pdi and DEP were quickly promoted by introvenous injection of neostig-mine after DiF. The latency of DEP was partially recovered by introvenous injection of neostig-mine. It is concluded that neostigmine can improve the generation of the fatigued diaphragm. and promote its recovery.
7.Effect of clinical nursing path with GTI on health education to patients with complex retinal detachment

Objective To study the effect of clinical nursing path with graph and text instructions (GTI) on health education to patients with complex retinal detachment.Methods Toally 76 inpatients with complex retinal detachment were enrolled in the study:38 of them hosptalized from March to August 2015 were put in the control group received routine treatment and the others hosptalized from September 2015 to March 2016 as the experiment group treated with GTI.The clinical outcomes were compared.Result The patients treated with GTI were better in the knowledge of the complex retinal detachment and moreover,they felt significantly more satisfied compared to that of the control group (P<0.05).Conclusion It is beneficial to use GTI in the patients with complex retinal detachment.GTI is effective for improving their knowledge on the disease,and increasing their satisfaction with the clinical nursing.
8.Clinical comparison of using sevoflurane and ketamine in infants' removing stitches after cleft lip surgery

Objective:To study the feasibility of using sevoflurane induction in infants' removing stitches after cleft lip surgery. Methods:60 infants after cleft lip surgery were randomly divided into three groups:group K (ketamine group,n=20),group S (sevoflurane group,n=20) and group SN (sevoflurane and nitrous oxide group,n=20). Group K were given intramuscular ketamine 5 mg/kg,midazolam 0.05 mg/kg and atropine 0.01 mg/kg. Group S were induced with inhalation of 8% sevoflurane under 6 L/min oxygen. Group SN were induced with inhalation of 8% sevoflurane under 4 L/min nitrous oxide and 2 L/min oxygen. After induction,anesthesia was continued with inhalation of sevoflurane under 3 L/min oxygen for 2 min before starting removing stitches. HR and SpO2 were monitored regularly during operation. The induction time,recovery time,occurrence of head moving,complication such as respiratory depression and increased secretion were recorded. Results:Induction and recover time in group S and group SN were similar,but faster than that of group K. Head moving in group S and group SN were less than that in group K. There happened glossoptosis and increased secretion in all the three groups,but no differences were found significantly. Conclusion:Inhaled induction of sevoflurane has more rapid induction and recover compared with intramuscular ketamine,and can be used safely in infants' removing stitches after cleft lip surgery. Additional inhalation of nitrous oxide can not shorten infants' induction and recovery time than sevoflurane inhalation alone.
9.Clinical application of arthroscopic arthrolysis for the treatment of elbow stiffness

Objective To evaluate the clinical results of arthroscopic arthrolysis for the treatment of elbow stiffness.Methods Totally 34 cases with elbow stiffness caused by trauma who needed to undergo arthroscopic arthrolysis were selected from General Hospital of Shenyang Military Area Command from October 2014 to February 2016.Among them,3 cases were lost to follow-up,and the other 31 cases were included in the study.All the patients were performed arthroscopic arthrolysis.Removed the hyperplasia of osteophyte and girdle,cleaned the articular cavity,restored the smooth of articular surface,and released the joint capsular and ligament around the elbow joint during the operation.Manipulation release was used coordinate with analgesia and rehabilitation therapy after operation.Mayo scores of elbow were used for function evaluation and range of motion(ROM) was collected.Results Patients were followed-up from 9 months to 12 months,(10.4±1.5) months on average.Before the operation,the mean maximal flexion angle of the elbow was (87.6±5.9)°,the extension angle was (35.5±6.4)°,the pronation angle was (75.9±9.6)°,the rotational angle was (67.2±7.4)°,and it was (125.9±6.5)°,(5.0±1.9)°,(82.0±9.1)°,(81.0±7.0)°respectively after operation,and the differences before and after opreation were statistically significant (P<0.05).The Mayo scores was (61.9±7.6) preoperatively and (88.8±4.7) postoperatively,and the difference was statistically significant(P<0.05).Conclusion Arthroscopic arthrolysis has the advantages of clear operative field,minimal invasion and rapid recovery.It can effectively improve the elbow joint activity and arthroscopic arthrolysis is an effective surgical treatment for elbow stiffness.
10.Oleanolic acid inhibits proliferation of HUVECs, and inhibits migration and tube formation via VEGF pathway.

To investigate the effects of oleanolic acid (OA) on the proliferation, migration and the formation of tube-like structure in human vascular endothelial cells (HUVECs). MTT assay, flat plate scarification, Transwell plates and matrigel-induced tube formation assay were performed to detect the effects of OA on proliferation, migration and tube formation. MTT assay showed that the inhibition rates of HUVECs treated with 60 and 100 microg x mL(-1) of OA for 24 h were 19% and 83% respectively. Treatment of HUVECs significantly inhibited the cell migration in a dose-dependent manner. The vascular indexes of HUVECs treated with 40 and 60 microg x mL(-1) OA were 33% and 20% respectively. Western blotting analysis showed that treatment of the cells with OA significantly attenuated the expression and secretion of VEGF. Additionally, VEGF could in part reverse the effects of OA on migration and tube formation of HUVECs. In conclusion, OA inhibits the proliferation, and VEGF plays an important role in OA induced decreased migration and tube formation of HUVECs.
