Objective To investigate the effects of RDP1258 on survival of rat cardiac allograft. Methods RDP1258 was synthesized and the model of rat heart abdominal transplantation was established. Animals were divided into four groups. Group 1 received no immunosuppression. Group 2 received CsA alone. Group 3 received RDP1258 alone. Group 4 received RDP1258 and subtherapeutic CsA. In all cases RDP1258 was administrated intraperitoneally and CsA was gavaged. Light and electron microscopic examinations were taken . Transplanted hearts were monitored daily by direct palpation. Results The purity of synthesized RDP1258 was over 95 % and the molecular weight was in accord with theoretical value. The histology and the ultrastructure changed little in grafts in group 3 and group 4. Survival of rat cardiac allograft was significantly prolonged in group 4. Conclusions RDP1258 can suppress acute rejection. Perioperative administration of RDP1258 in combination with CsA can significantly prolong survival of rat cardiac allograft.

Objective To evaluate the feasibility of using stroke volume variation (SVV) as the left ventricular preload to draw the cardiac function curve.Methods Twenty-seven patients of both sexes,aged 18-64 yr,with body mass index of 18-30 kg/m2,of American Society of Anesthesiologists physical status Ⅱ (New York Heart Association Ⅱ),with abnormal cardiac function,scheduled for elective offpany coronary artery byp grafting,were enrolled in this study.Twenty-five patients,aged 18-64 yr,with body mass index of 18-30 kg/m2,of American Society of Anesthesiologists physical status Ⅰ (New York Heart Association Ⅰ),with normal cardiac function,scheduled for elective non-cardiac surgery,were also enrolled in this study.SVV and SV were monitored by using a FloTrac/VigileoTM system.The patients were in the supine position from the end of anesthesia induction to the beginning of surgery.After the hemodynamics was kept stable for 5 min,SVV and SV were recorded.6％ hydroxyethyl starch 130/0.4 500 ml was intravenously infused over 20 min.SVV and SV were recorded after volume expansion.Spearman rank-order correlation was used to analyze the relationship between SVV and SV.The quadratic regression analysis was used to draw the SVV-SV curve,and the SVV-SV curve was compared with the Frank-Starling curve.Results Compared with the value before volume expansion,SVV and HR were significantly decreased,and SV was increased after volume expansion in the patients with normal cardiac function,and SVV was decreased after volume expansion in the patients with abnormal cardiac function (P＜0.01).SVV was negatively correlated with SV in the patients with normal cardiac function,and r=-0.467 (P＜0.05).SVV was negatively correlated with SV in the patients with abnormal cardiac function,and r=-0.378 (P＜0.05).The mirror symmetry was found between the SVV-SV curve in the patients with normal cardiac function and the normal Frank-Starling curve,and the general trend was close.The symmetry was not detected between the SVV-SV curve in the patients with abnormal cardiac function and the Frank-Starling curve in the patients with decreased myocardial contractility,and the general trend was not close.Conclusion For the patients with normal cardiac function,SVV can be used as the left ventricular preload to draw the cardiac function curve.

OBJECTIVE To investigate hand hygiene of healthcare workers in burn ward in our hospital and provide scientific basis for the effective prevention and control of hospital infection in burn ward.METHODS Selected randomly 30 healthcare workers in burn ward and sampled from hands for bacterial culture.RESULTS The average colony of hand pathogens of the 30 healthcare workers was 105.42 CFU/cm2,the maximam was 1093 CFU/cm2.Among the hand pathogens,40.91% were Bacillus subtilis,36.36% were micrococcus and 13.64% were Staphylococcus aureus.CONCLUSIONS Hand hygiene of healthcare workers are not optimistic in burn ward.Strengthening the education of hand hygiene and raising the executable rate of hand hygiene are important measures to prevent and control the hospital infection in burn ward.

18 fungal strains including N.coenophialum,N.lolii, N.huerfanum、N.chisosum、N.aotearoae、N.sp.and 8 varieties of grass seeds belonging to Festuca arundinacea and Lolium perenne have been studied.With amplification of IS1～IS3 and F1～R1 of genomic DNA, the primers Tub-2-F～Tub-2-R from Tubulin-2 gene and F3～R3 from NC25 gene have been designed.A PCR method to detect N.coenophialum and N.lolii was established, and also a nested-PCR method to detect N.coenophialum and N.lolii in single seed was established.These PCR detection methods are strongly special and much credible and rapid-speeded.

BACKGROUND:Large numbers of experimental data have confirmed that bone marrow mesenchymal stem cel s have a positive therapeutic effect on cerebral ischemia-reperfusion injury, but there are few reports about intravenous administration of bone marrow mesenchymal stem cel conditioned medium in the treatment of stroke.
OBJECTIVE:To investigate the effects of the conditioned medium of rat bone marrow mesenchymal stem cel s on the recovery of neurological function in rats after cerebral ischemia-reperfusion injury.
METHODS:The bone marrow mesenchymal stem cel s were isolated from rat bone marrow. When cel s at passage 2 or 3 reached 90%confluence, the original culture medium was removed. Then the cel s were cultured in serum-free DMEM for 18 hours. After that, the culture solution was col ected as the conditioned medium of rat bone marrow mesenchymal stem cel s. Adult rats were subjected to 2 hours of right middle cerebral artery occlusion. Ischemia-reperfusion injury rats were randomly assigned to three groups:control group, simple culture medium group and conditioned medium group, and respectively given injection of normal saline, DMEM, conditioned medium (10 mL/kg) via the tail vein at 2, 24, 48 hours after operation.
RESULTS AND CONCLUSION:There was no difference in the behavioral tests among the three groups at postoperative 2 hours (P>0.05). Compared with the control and simple culture medium group, neurological impairment was significantly improved in the conditioned medium group at postoperative 1, 3, 5 days (P<0.05), but there was no significant difference between the control and simple culture medium groups. At postoperative 5 days, brain edema was significantly eased in the conditioned medium group in comparison to the control and simple culture medium groups (P<0.05), and there was also no difference between the latter two groups (P>0.05). These results suggest that rat bone marrow mesenchymal stem cel s-conditioned medium via intravenous administration can significantly ease brain edema and improve the neurologic function after cerebral ischemia-reperfusion injury.

Objective:To investigate the effects of estradiol on ~(60)Co?-ray induced apoptosis of bone marrow hematopoietic cells of mice,and to discuss the related anti-irradiation mechanism.Methods:KM mice were randomly divided into 3 groups(15 mice/each group):control group(without radiation),pure radiation group and estradiol+radiation group(ER group).The pure radiation group was irradiated by 4.0 Gy?-ray at a dose rate of 1.15Gy/min;the ER group was administered with 0.1 mg estradiol(IM)at 10 days before 4.0 Gy?-ray radiation;and the control group received no special treatment.The apoptotic DNA segments of bone marrow hematopoietic cells were analyzed by DNA agarose gel electrophoresis;flow cytometry was used to examine the apoptosis rate of cells and expression of Fas and Bcl-2 at 4 h,8 h,and 12 h after irradiation.Results:Eight hours after radiation,the apoptotic DNA segments were obviously increased and apoptotic DNA ladder appeared,which was not seen in the other 2 groups.The apoptosis rate of bone marrow hematopoietic cells in ER group was significantly lower than that in the pure radiation group at 4,8,and 12 h after irradiation(P

AIMTo prepare the sustained-release nitrendipine microspheres with a solid dispersed structure in liquid system.

METHODSThe sustained-release nitrendipine microspheres with a solid dispersed structure was prepared in liquid system by combining spherical crystallization technique and solvent deposition method in one step. The resultant microspheres were evaluated for the recovery, micromeritc properties, incorporation efficiency. The factors of effect on the formation and the release rate of microspheres were also investigated.

RESULTSThe recovery of microspheres (280-900 microns) was more than 70% and the bulk density was around 0.7 kg.L-1. The incorporation efficiency always exceeded 95%. The formation of microspheres was mainly affected by the amount of bridging liquid and the emulsifying agents in poor solvent. The release rate of nitrendipine from the microspheres could be controlled as desired by adjusting the ratio of talc to Eudragit RS PO in the formulation.

CONCLUSIONThe presented method was suitable for preparing sustained-release microspheres of a water insoluble drug.

OBJECTIVETo establish a method used for optimization of harvesting time and determine the best time for harvesting Rumex gmelini.

METHODAn HPLC method was applied to determinate the contents of seven active constituents(resveratrol, polydatin, chrysophanol 1-glucoside, nepodin, emodin, chrysophanol and physcion)of R. gmelini at different development stage. The result was analyzed by principal component analysis.

RESULTThe accumulation of active constituents showed a regular pattern.

CONCLUSIONThe best harvesting time of R. gmelini is early July.

A sample pretreatment method combining column clean-up with dispersive liquid-liquid microextraction (CCU-DLLME) for determination of polycyclic aromatic hydrocarbons (PAHs) in oil-field water was proposed. With this method,most organic interferences in matrix were cleaned up,and PAHs were purified, enriched and analyzed by gas chromatography/mass spectrometry directly. The influences on extraction efficiency including the kinds of column packing,weight ratio between column packing and sample, column flow rate,type and volume of extraction solvent, type and volume of disperser solvent and extraction time were investigated, respectively. Finally, 12 g of H103 macroporous resin was selected as column packing,12﹕5 of weight ratio between column packing and sample and 4 BV/h of column flow rate were selected in CCU. The resulting eluate was added with 1.00 mL of acetone (disperser solvent) and 15 μL of carbon tetrachloride (extraction solvent),followed by DLLME for 2 min. Under the optimum conditions,the enrichment factor of PAHs was 730-1579,the limits of detection (S/N=3) were 1.1-5.3 ng/L, the linear range was 0.01-50 μg/L,the RSDs(n=5) were 0.6%-3.4% and the recoveries were 82.6%-104.6%. This method could greatly reduce the influence of organic interferences in matrix, and was fit for the rapid analysis of pollutants in oil-field water especially.

Objective To observe the application of high-speed vitrectomy system in combined surgery of phacoemulsification and vitrectomy .Methods Eighty-one patients (86 eyes) aged over 60 years with degree Ⅱlens opacity and vitreous retinal disorders treated between October 2012 and March 2013 were enrolled .The pa-tients included 43 males (46 eyes) and 38 females (40 eyes), at a mean age of (72.1 ±10.5) years (60-83 years).All the patients received routine ophthalmological examination , A/B mode ultrasound, and intraocular lens degree testing .With a 23G minimally invasive incision , phacoemulsification and intraocular lens implanta-tion were performed after closed the intraocular perfusion .With the viscoelastic agent left in the anterior cham-ber, high-speed vitrectomy was conducted using Resight noncontact wide -angle lens system , finished by filling with silicone oil , C3 F8 , or intraocular infusion fluid .Visual acuity , fundus condition , and postoperative compli-cations of the patients were observed .Results The combined surgery was successfully performed in all the pa-tients, with a mean operation time of (48.0 ±12.6) minutes (35-65 minutes).Among the 86 eyes, vision was improved in 80 eyes (93.0%), unchanged in 5 eyes (5.8%), and deteriorated in 1 eye (1.2%).After the surgery , 10 eyes (11.6%) developed corneal edema , and 25 (29.1%) showed anterior chamber reaction , all mitigated by treatment after 1 week.No other complications were reported .Conclusions For the elderly pa-tients with degree II lens opacity and the need of vitrectomy , the high-speed vitrectomy combined with phacoemul-sification and intraocular lens implantation can rapidly improve eyesight and reduce the incidence of postoperative complications .
