2.Effect of 3-n-butylphthalide on expression of matrix metalloproteinase andβ-APP in rats

Objective To study the effect of 3-n-butylphthalide on expression of MMP-2 ,MMP-9 andβ-APP in rats following chronic cerebral hypoperfusion at molecular level .Methods Seventy-two male Wistar rats were randomly divided into sham operation group ,ischemic group ,and bu-tylphthalide group (24 in each group) .Rats in ischemic group and butylphthalide group were di-vided into postischemic 7 ,14 ,30 ,60 d subgroups (6 in each group) .Expressions of β-APP ,MMP-2 , MMP-9 in each group were detected by Western blot .Results The β-APP expression level was significantly higher in postischemic 14 d ,30 d ,60 d subgroups than in postischemic 7 d subgroup (P<0 .05 ,P<0 .01) ,and in postischemic 30 d ,60 d subgroups than in postischemic 14 d sub-group (P< 0 .01) .The β-APP expression level differed greatly in 30 d and 60 d butylphthalide subgroups (P<0 .01) .The MMP-2 expression level was significantly higher in postischemic 60 d subgroup than in postischemic 30 d subgroup (P<0 .05 ,P<0 .01) .Theβ-APP and MMP-2 ex-pression levels were significantly higher in ischemic group and butylphthalide group than in sham operation group (P<0 .05 ,P<0 .01) ,and significantly lower in butylphthalide group than in is-chemic group (P< 0 .01) .Conclusion MMP-2 and β-APP tend to change similarly and are in-volved in blood brain barrier destruction and amyloid deposition .Butylphthalide intervenes in am-yloid deposition by downregulating the expression of MMP-2 andβ-APP .

5.Construction and ldentification of Cell Division Cycle 2 Promoter Reporter Gene Vector

Objective To construct the luciferase reporter gene vector of cell division cycle 2 (Cdc2) gene promoter and determine its transcriptional activity. Methods Primers were designed based on human Cdc2 promoter sequence from UCSC software. Then Cdc2 promoter from human genome DNA was replicated. After pGL3-Basic vector and Cdc2 promoter were digested with restriction enzymes SacⅠand XhoⅠseparately, Cdc2 promoter was inserted into pGL3-Basic vector. The recombinant plasmid named pGL3-Cdc2-promoter was transiently co-transfected into U2OS cells with control vector pRL-SV40, and then the activity of dual luciferase was detected. Results pGL3-Cdc2-promoter was constructed successfully. The restriction analysis and sequencing proved the entirely correct sequencing results. The luciferase activity was higher in pGL3-Cdc2-promoter/pRL-SV40 group than that of pGL3-Basic/pRL-SV40 group (1.591 5±0.199 8 vs 0.049 9±0.010 4). Conclusion pGL3-Cdc2-promoter can be transcribed and activated in U2OS cells. This study provided an important basis for screening and evaluation of anticancer drugs.
7.The efficacy and safety of 2 cycles' high-dose dexamethasone treatment adult primary immune thrombocytopenia

Objective To investigate the efficacy and safety of a schedule of 2 cycles' high-dose dexamethasone (HD-DXM) as an initial therapy in adults immune thrombocytopenia (ITP), and compare with conventional dose prednisone therapy. Method A total of 59 newly diagnosed ITP patients were divided into 2 groups randomly. In 30 patients ( Dexamethasone group), oral HD-DXM was administered at 40 mg/d for 4 consecutive days, repeated one week later, and then failed to maintain. In the remaining 29and then gradually tapered. Results For short-term efficacy, after 1 and 2 weeks of treatment, the response rate in Dexamethasone group was significantly higher than that in Prednisone group (50. 0% vs 24. 1%, P <0. 01; 73.3% vs 55.2%, P <0. 05 ), while 3 weeks later, there was no remarkable difference between the two groups(83.3% vs 68.9%, P > 0. 05 ), though the response rate in Dexamethasone group remained higher. For long-term effect, at the end of the 2nd and 3rd months of follow-up, the relapse rate in Dexamethasone group was significantly lower than that in Prednisone group(24. 0% vs 40. 0%, P < 0. 05;32.0% vs 65. 0%, P < 0. 01 ), while at the end of the 1st month of follow-up, there was no significant difference( 16. 0% vs 20. 0%, P >0.05 ). In addition, it's well tolerated and no complications such as severe infection or Cushing syndrome were complained in Dexamethasone group. Conclusion HD-DXM possesses an advantage over conventional dose prednisone therapy in efficacy and safety.

Objective To explore effects of heat stress response on neutrophil apoptosis and respiratory burst function. MethodsNeutrophils from each of 18 healthy volunteers were divided into 3 parts,and one part was served as control group,and the other two parts were induced by heat shock or cadmium chloride for heat stress response and named as heat shock group and cadmium chloride group.The neutrophils were incubated in culture medium.At 0,2,3,4,and 6 h following heat stress induction,the heat shock protein(HSP70) expression and the respiratory burst were detected in the neutrophils respectively by using PCR technique and flow cytometer.Level of apoptosis was observed by immuno-fluorescence and flow cytometer DNA ploid at 24 h after heat stress induction.
