Objective To explore the predictive value of QT interval dynamicity for sudden death in patients with idiopathic dilated cardiomyopathy ( DCM ). Methods Fifty-five patients with DCM ( DCM group) and 27 healthy subjects (Control group, Con) were enrolled. Investigations included history collection, clinical examination, echocardiography, electrocardiogram and 24 h ambulatory electrocardiogram. Following indexes were determined: left ventricle end diastolic dimension ( LVEDD), left ventricle ejection fraction (LVEF), QT dispersion (QTd), SDNN, the slope of QT/RR plots of the linear regression, ventricular premature beats (VPB) and non-sustained ventricular tachycardia (NSVT). Primary end point for patients with DCM was all cause death. Results LVEDD, QTd, VPB/24 h, NSVT/24 h,QTe/RR slope and QTp/RR slope were significantly higher while LVEF and SDNN were significantly lower in DCM group than in Con group (all P<0.05). LVEDD, LVEF, QTd, SDNN, QTe/RR slope and QTp/RR slope were significantly different among DCM sudden death group, DCM non sudden death group and Con group (P < 0. 05 ). LVEF, SDNN, QTe/RR slope and QTp/RR slope were significantly different between DCM sudden death and non sudden death group (P < 0. 05). LVEF, QTd, VPB/24 h, QTe/RR slope and QTp/RR slope were significantly different between DCM with NSVT and DCM without NSVT group (P <0. 05). The sudden death rate of DCM patients with QTe/RR slope ≥ 0. 210 was significantly higher than DCM patients with QTe/RR slope < 0. 210 (54. 5% vs. 21.1%, P < 0. 05 ). Sudden death rate of QTp/RR slope≥0. 190 was also higher than those <0. 190 (52. 2% vs. 21.9%, P <0. 05). The sudden death rate of DCM patients with both LVEF≤35% and NSVT + was 62. 5%. Combining QTe/RR≥0. 210 with NSVT + or LVEF ≤ 35%, the sudden death rates were 62. 5% or 66.7%. Combining QTp/RR ≥0. 190 with NSVT + or LVEF≤35%, the sudden death rates were 66. 7% or 61.5%. Combining QTe/RR ≥0. 210 or QTp/RR ≥0. 190 with NVST + and LVEF≤35%, the sudden death rates were 77. 8% or 70. 0%. Conclusions High QT/RR slope is a risk factor for sudden death of DCM patients. QT/RR slope is a useful predictor for sudden death in DCM patients either independently or combined with NSVT or LVEF.

Objective To explore the effect of hypergravity on morphology and osteogenesis function of preosteoblast MC3T3-E1 ceils. Methods The cultured MC3T3-E1 cells under hypergravity by different loading forces were divided into five groups, including control group,5 g group,10 g group,15 g group and 20 g group. The experimental groups were loaded for 30 min each time in 3 successive days, and the control group with no g-value was synchronously exposed to the same surrounding. The morphology of cytoskeletal protein was observed by phalIoidin staining,The alkaline phosphatase (ALP) content was examined by ALP activity assay kit,the gene expression of ALP,collagen Ⅰ (Col Ⅰ),osteocalcin (OC),runt-related transcription factors (Runx2) was measured by real-time quantitative PCR,and the protein expression of Col Ⅰ and OC was tested by Western blotting. Results Under the condition of hypergravity, cell body of osteoblast became thinner, but its surface area increased significantly;with the structure of skeletal arrangement becoming loose, actin microfilament structure reduced so that the orderly arrangement of actin-like dispersion lowered. The gene expressions of related indicators of osteogenic differentiation including ALP,Col][,OC,Runx2 were significantly up-regulated,which was the same as Col Ⅰ protein and OC protein after hypergravity loading.A very minute quantity of small red-orange nodules was found in the control group, while the cells in experimental groups after hypergravity loading obviously formed various sizes of red-orange nodules. Conclusions Under hypergravity, changes in osteoblast morphology can be triggered by rearrangements of skeletal structure.Furthermore, osteoblast maturation and differentiation can be stimulated effectively by up-regulating differentiation-related gene and protein expressions.

Aneurysmal subarachnoid hemorrhage(aSAH),primarily caused by the rupture of intracranial aneurysms with bleeding into the subarachnoid space,is an acute neurological disease associated with high disability and mortality.Brain injury after aSAH results from a combination of injury mechanisms, with early brain injury(EBI)occurring within 72 hours post-onset,laying the foundation for subsequent pathophysiological changes in the brain and poor prognosis of patients.Among them, the brain immunoinflammatory response, involving the interaction of various immune cells and active substances, plays a significant role in post-aSAH EBI,and is related to delayed brain injury and long-term prognosis.Systemic inflammatory response following aSAH can also affect the prognosis and outcome of patients. This review summarizes the role of local and systemic immune inflammatory responses in the occurrence and progression of aSAH,as well as the research progress on related inflammatory biomarkers and therapeutic prospects, aiming to provide a theoretical reference for new treatment for aSAH.

Objective: The current study was designed to investigate the specific killing effects against homo-leukemia hybrid cell induced by BCR-ABL multiple epitope fusion protein and explore a powerful chronic myeloid leukemia(CML) immunotherapeutic strategy. Method: A multiple epitope antigen gene of BCR-ABL fusion antigen was used to immunize C57BL/6 mice and the spleen cells were cultured as effective cells. Specific lytic activities of target cells were detected by standard MTT assay. Results: Only the CTLs (cytotoxic lymphocytes) of mice immunized with the fusion protein can lyse the target cells but not the control cells (P<0.01 vs P>0.05). Conclusion: The multiple epitope BCR-ABL fusion antigen elicited antigen-specific, CTL-mediated anti-leukemia immune responses which provided a basis for further study on the developing specific T cell immunotherapy of CML in vivo.
