2.Analysis of the difference of three imaging methods of the MRA in detection of cerebral infarction

Objective To compare the clinical value of fluid attenuated inversion recovery imaging (FLAIR),magnetic resonance diffusion-weighted imaging(DWI) and magnetic resonance angiography(MRA) in diagnosis of early cerebral infarction.Methods 70 cerebral infarction patients were chosen.They underwent FLAIR,DWI and MRA examination.Results 70 patients with early cerebral infarction mostly were multiple lessions accouting for 60%.Supper actue period had 22 cases and acute period had 48 cases.DWI had the highest detection rate of cerebral infarction,particularly in the hyperacute period the detection rate reached 100%,which was significantly higher than the FLAIR and MRA(x2 =12.4,P < 0.05).Conclusion Especially in the early diagnosis of hyperacute cerebral infarction,DWI is superior to FLAIR and MRA.FLAIR showed better on the cortex and periventricular lesions,MRA can check out the blood vessel lesions in the infarction area.

5.Changes and Clinical Significance of Serum GM-CSF, IL-6 and TNF-α Levels in Patients with Rheumatoid Arthritis

Objective:To investigate the changes and clinical significances of serum levels of granulocyte macrophage colony-stimulating factor (GM-CSF),interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) levels in patients with rheumatoid arthritis (RA).Methods:126 cases of patients with RA in our hospital from June 2013 to December 2016 were selected as RA group and divided into two subgroups as active RA group (69 cases) and remission RA group (57 cases) according to the severity of the disease.55 healthy cases who receive physical check in our hospital at same time were selected as the control group.The serum GM-CSF,IL-6 TNF-α levels were detected by enzyme linked immunosorbent assay (ELISA) and compared between different groups,the correlation of serum GM-CSF,IL-6 TNF-α levels with DAS28 were analyzed.Results:The serum GM-CSF,IL-6 TNF-α levels of RA group were significantly higher than those of the remission RA group and control group (P<0.01),and the serum GM-CSF,IL-6 TNF-α levels of remission RA group were also significantly higher than those of the control group (P<0.01).Positive correlation between serum GM-CSF,IL-6 and TNF-α levels were found(P<.0.01).The level of DAS28 was significantly correlated with the levels of serum GM-CSF,IL-6 and TNF-α (r=0.473,0.584,0.675,all P<0.01).Conclusion:The levels of serum GM-CSF,IL-6 and TNF-α were up-regulated in patients with RA and positively correlated with the activity of RA,which might be of important clinical significance in the early diagnosis,disease evaluation and prognosis of RA.
6.Detection and significance of phosphatidylinositol 3-kinase in adipose tissue of polycystic ovary syndrome patients with insulin resistance

Objective To investigate the expression of phosphatidylinositol 3-kinase(PI-3K)in adipose tissue of polycystic ovary syndrome patients(PCOS),and explore molecular mechanisms of insulin resistance(IR)in PCOS.Methods Samples from patients with PCOS with IR(n=19),PCOS without IR (n=10)and controls(n=15)were collected.Serum fasting insulin(FIN)and fasting plasma glucose (FPG)were measure& Insulin resistance index was calculated using homeostasis model assessment (HOMA)to analyze the relationship between these markers and IR.Western blot technique was used to detect the PI-3K p85 subunit.Gene expression of PI-3K p85 subunit was detected by reverse transcription polymerase chain reaction(RT-PCR)method.Kinase activity was detected by immunoprecipitation,thin- layer chromatography and gamma scintillation counting.Results(1)The levels of FIN[(25.2?3.8) mU/L]and HOMA-IR(1.6?0.3)in PCOS with IR were significantly higher than those in PCOS without IR[(13.4 +3.8)mU/L,0.9?0.3]and controls[(9.5 +2.6)mU/L,0.5?0.3;all P<0.05).(2) There was no significant difference in the protein(0.65?0.10)and gene expression(0.92?0.12)of PI-3 K p85 subunit in PCOS with IR compared with PCOS without IR(0.72?0.10,1.01?0.10)and control groups(0.73?0.14,1.00?0.12;P>0.05).(3)PI-3K activity in PCOS with IR(81%)and PCOS without IR(89%)was significantly decreased(P<0.01,P<0.05)and negatively correlated with HOMA- IR(r=-0.69,P<0.01;r=-0.62,P<0.05).Conclusions No significant difference in the protein and gene expression of PI-3K p85 subunit in PCOS with IR is found.The decreased PI-3K activity may lead to IR of PCOS.
