1.Effects of intense pulsed light and radiofrequency radiation on the content of collagen in BALB/c mouse skin

ObjectiveTo investigate the effects of intense pulsed light(IPL) and radiofrequency radiation on the appearance,histopathological manifestation,dermal depth and collagen content in BALB/c mouse skin.MethodsThe back of BALB/c mice was irradiated with intense pulsed light(IPL group,n =9),radiofrequency (RF group,N =9),IPL and radiofrequency (IPL+RF group,n =9) respectively,for 4 sessions at 1-week intervals followed by 2 sessions at 2-week intervals.Three mice remaining untreated served as the control group.The appearance of skin on the back of mice was observed at different time points after irradiation.Some mice were sacrificed and skin specimens were obtained from the back at week 4,8 and 12 after the first irradiation.Hematoxylin and eosin staining was performed to observe the histopathological manifestation and measure the dermal depth of mouse skin,and Masson staining to detect the expression of collagen fiber in mouse skin.ResultsNo local adverse effects were observed in the back skin of mice after irradiation with IPL or radiofrequency.Since the 8th week after the first irradiation,the dermal layer had appeared to be thickened,with an increase in the content of collagen fiber and extraeellular matrix,in the IPL group,RF group and IPL+ RF group compared witb the control group,and the increase was more obvious in the IPL+RF group than in the IPL group and RF group (all P < 0.05 ).Increased collagen fiber was densely arranged in irradiated mice.There was no apparent thickening of skin epidermis after irradiation.ConclusionsBoth IPL and radiofrequency can stimulate an increase in dermal collagen production in mice,and a synergistic effect exists between IPL and radiofrequency in the biostimulation of collagen synthesis.
2.Methodological evaluation on ELISA for detecting concentration of apo A-V in human plasma

Objective To establish an ELISA to detect the concentration of apo A-V in human plasma and evaluate the sensitivity and stability of the method.Methods The standard curve of recombinant human apo A-V, the lowest detectable concentration, the day-to-day variation and between-plate-within-day variation, the interference of other non-specialized apolipoprotein were examined, and the reference range was established by detecting the plasma concentration of apo A-V in 505 healthy subjects.Results The linear range of the established method was 5.86 - 187.5 ng/ml with lowest detectable concentration of 2.34 ng/ml. The interferences for apo A-V detection due to Lp(a), apo AI and apo B was never found in our study. The day-to-day variation and between-plate-within-day variation were less than 10% respectively.Conclusion An ELISA for detecting the concentration of apo A-V in human plasma was established and the method can serve as a useful tool for research on mechanism and function of apo A-V in TG metabolism.
3.Effect of Depression on the Incidence of Osteoporosis in Patients with Maintenance Peritoneal Dialysis

Objective To investigate the effect of depression on the incidence of osteoporosis in patients with maintenance peritoneal dialysis (MPD).Methods We enrolled 80 MPD patients,who underwent peritoneal dialysis in our dialysis center.The clinical data and biochemical parameters of all patients were collected,bone mineral density was detected and the Self-rating depression scale was used to evaluate depression state.Results Among the 80 enrolled MPD patients,34 patients had osteoporosis (42.5%),48 patients had the presence of depression (60%).Logistic regression analysis showed that age,dialysis age,gender,diabetes history and depression state were the risk factors for osteoporosis in MPD patients,the depression state was negatively correlated with BMD of lumbar and femoral neck (r=-0.347,r=0.426,P<0.05).Conclusion Depression state may be a risk factor for osteoporosis in clinic,it is of great significance to focus on the psychological state of MPD patients in the prevention and treatment of osteoporosis.

5.Application of spectral karyotyping in diagnosis and prenatal diagnosis of the marker chromosome

Objective To determine the value of spectral karyotyping(SKY)in identification of the marker chromosome.Methods Selected six cases that could not be identified in clinic were studied,using samples of peripheral blood from four cases,and samples of amonic fluid and fetal cord blood for prenatal diagnosis in two cases were investigated.All cases were analyzed with the routine SKY method.and the results with the SKY View software.The SKY results were identified by using fluorescence in situ hybridization(FISH).And C-banding technique was used to help diagnose the heterochromatin.Results SKY wag successfully performed on all of 6 cases.The origin of all marker chromosomes was identified by SKY.Except case No.4,the others were confirmed by FISH.It helped determine the pregnancy outcome in two cases of prenatal diagnosis:one case of genetic marker chromosome continued the pregnancy,and another case of de novo marker chromosome was terminated of the pregnancy.Conclusion SKY may be a vahable tool to diagnose the marker chromosome with rapidness,direct-viewing and sensitiveness.It can be used to assess the prognosis and the pregnancy outcome.
6.Expression Diversity of Quorum-sensing-Related Genes in Pseudomonas aeruginosa

One hundred Pseudomonas aeruginosa quorum-sensing-related genes were selected and their primers were synthesized. The fragments of specific sequences which are related QS genes were amplified by PCR. These verified sequences were inserted into the vector pMD-18T for sequencing. These DNA fragments were dotted onto glass slides to make cDNA microarray. Hybridization was performed with cy3/cy5-dCTP labeled probes. The scanning data of early stationary phase and mid-logarithmic phase indicated that 9 genes were up-regulated and 6 genes were down-regulated. Undergoing the different medicines,we took tobramycin as an example to compare the expression diversity. The results confirm that the QS cDNA chip is useful,and may contribute to better understand the mechanism of quorum-sensing,and can help us find the new targets for restraining the growth of Pseudomonas aeruginosa.

Objective Clinical significance of using ELISA to determine ?-amyloid(A?)_(1-42) antibody levels in the sera of patients with Alzheimer's disease(AD).Methods 96 wells PVC plate was coated with A?_(1-42)peptide.Serum of AD patient was competing with mouse A?_(1-42)monoclonal antibody in this assay.The second antibody was horseradish peroxidase(HRP)conjugated goat anti-mouse IgG.Serum A?_(1-42)antibody levels were determined by ELISA.Results The sensitivity of this assay was about 1 ng/ml.The recovery rate of this test was between 96.5% and 104.7%.The residual A?_(1-42)antibody levels in human serum or horse serum after A?_(1-42)antibody was removed by absorption were less than 1 ng/ml. 9.Protective effect of total flavonoids of epimedium on the kidney in experimental diabetic rats.

OBJECTIVETo investigate the influence of total flavonoids of epimedium (TFE) on the streptozocin (STZ)-induced kidney injury in diabetic rats and discuss the possible mechanism.
METHODSDiabetes was produced by a single injection of streptozocin (40 mg/kg, iv) in male SD rats. The rats were randomly divided into three groups (n = 10): control group, model group and TFE group (100 mg/kg, ig). Animals were sacrificed 12 weeks later. The level of blood glucose, blood urea nitrogen (BUN) and creatinine (Cr) as well as the renal index were determined. Detect the specific biochemical of renal tissue: superoxide dismutase (SOD), malondialdehyde (MDA). Use masson staining to observe the morphology of the renal tissue. Immunohistochemistry was employed to determine the protein levels of transforming growth factor-beta1 (TGF-beta1).
RESULTSCompared to control group, the enhancement of blood glucose, renal index, BUN and Cr was found in model group, which was significantly attenuated by treatment with TFE. Meanwhile, elevated MDA level in renal tissue as well as decreased SOD activities in renal tissue were significantly remitted by TFE. Furthermore, TFE decreased the expression of TGF-beta1.
CONCLUSIONTFE can evidently relieve renal damage in rats with diabetic nephropathy induced by STZ, which might be related to antioxidation and modulating the expression of TGF-beta1 protein.

10.Changes of Streptococcus mutans concentration of plaque during fixed appliance treatment.

OBJECTIVETo evaluate the dynamic changes of Streptococcus mutans concentration of plaque during fixed appliance treatment and the effects of two materials of ligation on Streptococcus mutans concentration.
METHODSTwenty-eight patients undergoing fixed appliance treatment were observed. Ligature wire and elastomeric rings were applied on one side of arches, stochastically. The dynamic changes on the quantity and percentage of Streptococcus mutans were observed before and after fixed appliance bonding.
RESULTSStatistically significant increase of the quantity and percentage of Streptococcus mutans was found after fixed appliance bonding, and the percentage of Streptococcus mutans in the plaque around the brackets ligated with elastomeric rings was more than that of ligature wire at the beginning of fixed appliance bonding, statistically.
CONCLUSIONSThe finding suggested that the caries-associated capability of the plaque increased after bonding and there was greater caries-associated capability of the plague on the teeth when elastomeric rings was used than that of the plague when ligature wire was used at treatment beginning. The ligature wire is recommended in the fixed appliance treatment.
