1.Influence of indapamide on pharmacokinetics of telmisartan in male and female rats

AIM To study the influence of indapamide(Ind) on pharmacokinetics of telmisartan(Tel) and observe the difference between male and female rats. METHODS Wistar rats were divided into Tel and Tel+Ind groups, each group containing 8 female and 8 male rats, and were ig administered a single dose of either Tel 3.6 mg·kg-1 or Tel 3.6 mg·kg-1+Ind 0.135 mg·kg-1, respectively. Blood samples were collected at intervals over 96 h after administration. The Tel concentrations in plasma were determined by high performance liquid chromatography with fluorescence detector. The Tel concentration-time curves were simulated by 3p97 software and the pharmacokinetic parameters were calculated. RESULTS Whatever in female or male rats, there were no significant differences in the main pharmacokinetic parameters of Tel between Tel and Tel+Ind groups. However, females had higher values for area under the concentration-time curve and maximum plasma concentration than males, but lower values for total clearance in both Tel and Tel+Ind groups. CONCLUSION Ind has no significant influences on the pharmacokinetics of Tel. However, pharmacokinetics of Tel is significant different between male and female rats.
2.The Value of CT in Diagnosing and Following-up Gastrointestinal Stromal Tumor

Objective To analyze the value of CT in diagnosing and following-up gastrointestinal stromal tumors(GIST).Methods 117patients with CT examination and histological diagnosis of GIST were retrospectively reviewed.Results 117 patients had 118 GIST.Thelocations of primary tumor were as follows: stomach(n=56),small intestine(n=40),esophagus(n=6),colorectum(n=6),mesentery(n=3),retroperitoneum(n=2) and indefinite origin(n=5).Size of tumor was 1.7~18 cm.Tumors(5 cm) often demonstrated lobulated shape with central low attenuation area and heterogeneous enhancement.There were 17 tumors with bubble,13 with scattered calcifications,12 with metastases of liver.With following-up,23 of 37 were developed local recurrence or hepatic metastases.Conclusion CT is aneffective and useful imaging modality in the diagnosis and follow-up gastrointestinal stromal tumors.
3.Radiofrequency Catheter Ablation of Idiopathic Ventricular Tachycardia in Children

Objective To investigate the value of radiofrequency catheter ablation(RFCA) on idiopathic ventricular tachycardia(IVT) in children.Methods The treatment efficiency of 35 patients accepting RFCA from March 2000 to February 2006 was analyzed,which was divided into control group and study group,randomly.The target site of IVT was localized by routine mapping in control group,and study group was localized by routine mapping combining doppler tissue imaging(DTI) on the basic treatment.Results Numbers of discharge,X-ray exposure time in study group were shorter than that in control group.There were no significance of radiofrequency energy,achievement rates and relapse rates between two groups.Conclusions RFCA is a safe and effective method on IVT.Children are localized combined with DTI shall decrease numbers of discharge and X-ray exposure time.
4.Chemical constituents from Solanum rostratum.

Ten compounds were isolated from the aerial part of Solanum rostratum by means of various chromatographic techniques such as silica gel, Sephadex LH-20, ODS and preparative HPLC. Their structures were identified as dioscin (1), hypoglaucin H (2), hyperin (3), isoquercitrin (4), isorhamnetin-3-O-beta-D-galactopyranoside (5), kaempferol-3-O-beta-D-glucoside (6), smilaxchinoside A (7), 26-O-beta-D-glucopyranosyl-3beta, 20alpha,26-triol-25 (R) -delta5,22-dienofurostan-3-O-alpha-L-rhamnopyranosyl (1 --> 2) -[ alpha-L-rhamnopyranosyl (1 --> 4)] -beta-D-glucopyranoside (8), beta-sitosterol (9), and daucosterol (10), on the basis of physicochemical properties and spectroscopic data analysis. Among them ,compounds 7 and 8 were isolated from the genus Solanum for the first time, and the remaining compounds were obtained from this plant for the first time.

5.Pharmacokinetic effect of combined administration on spinosin and ferulic acid in monarch drug Ziziphi Spinosae Semen kernel.

To study the pharmacokinetic effect of different combined administration with monarch drug Ziziphi Spinosae Semen on its main components in rats. Sprague-Dawley (SD) rats were randomly divided into Ziziphi Spinosae Semen group, Ziziphi Spinosae Semen-Fructus Schisandrae Chinensis group, Ziziphi spinosae Semen-Salviae Miltiorrhize Radix et Rhizoma group and Zaoren Ansheng prescription group. After oral administration, HPLC was eluted with the mobile phase of acetonitrle-0.03% phosphate acid water in a gradient mode. The detection wavelength was 280 nm. The pharmacokinetic parameters of spinosin and ferulic acid were calculated by DAS 2. 0 software. Compared with Ziziphi Spinosae Semen group, Ziziphi Spinosae Semen-Fructus Schisandrae Chinensis group, Ziziphi Spinosae Semen-Salviae miltiorrhizae Radix et Rhizoma group showed a lower maximum plasma concentration (C(max)) and area under curve (AUC(0-t)) for spinosin and ferulic acid but higher clearance speed (CL/F); whereas the Zaoren Ansheng prescription group showed higher maximum plasma concentration (C(max)) and area under curve (AUC(0-t)) for spinosin and ferulic acid but lower clearance speed (CL/F). Compared with Ziziphi Spinosae Semen group, prescription group showed slower metabolism of spinosin and ferulic

7.Improved preparation of buffy-coat-derived platelet concentrates and factors affecting platelet recovery

BACKGROUND:Buffy-coat-derived platelet concentrates and plasma-rich platelet concentrates have a high incidence of invalid infusion and adverse reactions.
OBJECTIVE:To observe the improved preparation of buffy-coat-derived platelet concentrates and to analyze the influential factors relevant to platelet recovery.
METHODS:400 mL of blood sample extracted from 126 cases were randomly divided into improved buffy-coat group, buffer-coat group and platelet-rich plasma after 4-6 hours. The 3-step centrifugal method was used for improved preparation of buffy-coat-derived platelet concentrates:step 1, centrifugation at 2 300 r/min for 12 minutes at (22±2)℃ with a deceleration of 5;step 2, centrifugation at 910 r/min for 10 minutes at (22±2)℃;step 3, centrifugation at 2 800 r/min for 12 minutes at (22±2)℃. After centrifugation, the upper layer containing few platelets was removed, and the rest 30 mL platelet suspension was platelet concentrates. Factors affecting platelet recovery were analyzed through literature retrieval.
RESULTS AND CONCLUSION:There was no difference in platelet number among the three groups before preparation of platelet concentrates (P>0.05). A higher rate of platelet recovery was found in the platelet-rich plasma group and improved buffy-coat group compared with the buffy-coat group (P<0.05), but there was no difference between the former two groups (P>0.05). There were less residual red blood cells and white blood cells in the two buffy-coat groups than the platelet-rich plasma group (P<0.05), but there was no difference between the two buffy-coat groups (P>0.05). The recovery rate of prepared platelet concentrates was affected by the whole blood amount, centrifugal speed, centrifugation time and methods. Improved buffy-coat method for preparation of platelet concentrates can be generalized in blood centers or blood stations, because it can reduce residual red blood cells and white blood cells and increase rate of platelet recovery.
8.Asiatic acid induces apoptosis in T98 G human glioblastoma cells by changing autophagy

Aim To investigate the effect of asiatic acid on apoptosis and autophagy in human glioblastoma T98G cells. Methods MTT colorimetry was employed to assay the cellular proliferating activity. The fluores-cence microscope and Hoechst 33258 staining were used to detect the morphological changes. The cell ap-optosis and autophagy were analyzed by flow cytometry with Annexin-V/7-AAD and MDC staining respective-ly. The expressions of associated proteins were detected by Western blot to analyze the mechanism of apoptosis and autophagy. Results MTT assay showed that the growth of T 9 8 G cells was inhibited by asiatic acid ( IC50 =46. 3 μmol · L-1 ) . Annexin V/7-AAD stai-ning and Western blot revealed that asiatic acid in-duced apoptosis in T98 G cells by reducing the expres-sion of Akt, decreasing the mitochondrial membrane potential, and increasing the expression of Caspase-3. MDC staining and Western blot showed that the per-centage of MDC-positive cells was decreased and the expressions of Beclin-1 , LC3-II and Atgs were inhibi-ted by asiatic acid treatment. 5 μmol·L-1 chloroquine was used to up-regulate the expressions of LC3-Ⅱand Beclin-1 . Asiatic acid-inhibited autophagy was blocked and the total apoptotic rate was reduced remarkably. Conclusion Asiatic acid suppresses T98 G cells pro-liferation by inducing apoptosis and inhibiting cell au-tophagy, and the very role of inhibiting autophagy could promote apoptosis to a certain extent.
9.Effect of Tongmai Yangxin Pill on the Serum Hepcidin Level of Patients with Coronary Atherosclerotic Heart Disease and Anemia of Chronic Disease

Objective:To investigate the effect of Tongrnai Yangxin Pill on the serum hepcidin level of patients with Coronary Atherosclerotic Heart Disease and Anemia of Chronic Disease.Methods:Seventy patients with Coronary Atherosclerotic Heart Disease and Anemia of Chronic Disease whose age were above 40 years old were enrolled as the research group and divided into the medication group (group Ⅱ) (n=35) and the nonmedication group (group Ⅲ)(n=35),while 40 CAD patients were enrolled as the normal control group (group Ⅰ).Before and after medication,the Hepc,Hb,etc levels were compared between two groups.Results:Before medication,the levels of Hepc in the two research subgroup were significantly higher than that of the control group (P<0.05).At 8 weeks after treatment,the Hepc level of group Ⅱ was significantly declined,and the level of Hb was increased than those before treatment (P <0.05);the Hepc levels of group Ⅰ and group Ⅲ showed no significant difference (P>0.05),the Hepc levels of group Ⅱ and group Ⅲ were obviously higher than that of the control group (P <0.05),the Hepc levels of group Ⅲ was obviously higher than that of the group Ⅱ(P<0.05),the Hb level of group Ⅲ was obviously lower than those of group Ⅰ and group Ⅱ (P<0.05).Conclusion:Tongmai Yangxin Pill could reduce the level of Hepc and enhance the Hb levels of patients with Coronary Atherosclerotic Heart Disease and Anemia of Chronic Disease.It was useful to the patients with Coronary Atherosclerotic Heart Disease and Anemia of Chronic Disease,especially patients with mild anemia.
10.Study on the Extracting Method and Producing Conditions of Phyllosticta commelimecola Toxin

The crude toxin was extracted from hypha and culture solution of Phyllosticta commelimecola through three different polarity solvent: benzinum, puncificatum ethyl acetate and chloroform. The result indicated that the toxin secreted by Phyllosticta commelimecola not only was in hypha but also in culture solution and the extracting effect of ethyl acetate was the best. The soybean median and PSK media can be respectively used as solid and liquid culture media to produce toxin and grow mycelium. The optimal cultural conditions for producing toxin were temperature 32℃,cultured period 14d, cultured ways shaking of 150r/min.
