2.The association between subclinical hyperthyroidism and early neurological deterioration in patients with acute cerebral infarction

Objective To detect the association between subclinical hyperthyroidism and early neurological deterioration (END) in patients with acute cerebral infarction.Methods Totally 139 patients diagnosed with acute ischemic stroke were prospectively enrolled between March 2015 and September 2016.The early neurological deterioration was defined as 7-day in-hospital increase (vs.that at admission) in the NIHSS score of ≥2 points without any systemic cause.Multivariable Logistic regression analysis was conducted to evaluate the relationship between subclinical hyperthyroidism and END.Subclinical hyperthyroidism is defined as TSH < 0.44 mU/L,with normal FT3 and FT4.Multivariable Logistic regression analysis was conducted to evaluate the relationship between subclinical hyperthyroidism and END.Results 42 (30.2%) patients with END among 139 with subclinical hyperthyroidism were observed.As compared with patients without END,patients with END showed older age[(71.2±5.7) years vs.(68.1±6.3) years,t=2.695,P=0.008],higher prevalence of subclinical hyperthyroidism (28.6% vs.9.3%,x2 =8.506,P=0.006),and higher levels of C-reactive protein[5.6 (1.0,11.0) mg/L vs.2.5 (1.0,5.0) mg/L,Z=2.125,P=0.034],homocysteine[(15.3±5.8) mmol/L vs.(13.0±4.8) mmol/L,t=2.395,P=0.018]and fasting bloodglucose[(8.1 ± 2.2) mmol/L vs.(6.3 ± 2.6) mmol/L,t =4.108,P =0.001].Logistic regression analysis indicated that subclinical hyperthyroidism was associated with END after adjustment for potential confounders (OR =3.415,95 % CI:1.148-10.167,P =0.027).Conclusions Subclinical hyperthyroidism is a significant and independent predictor for END in patients with acute cerebral infarction.
3.Clinical application of muliplefoliated flap combined with tendon graft from dorsal foot for repairing soft tissue defects of multi-fingers

Objective To summarize clinical experience of application of muliplefoliated flap combined with tendon graft from dorsal foot for repairing soft tissue defects of multi-fingers. Methods Based on the results of microdissection researches, muliplefoliated flap including medial foot flap, the first metatarsal dorsal flap, lateral foot flap, extensor pollicis brevis of toe and extensor proprius of the fifth toe pedicled by anterior tibial vessels and dorsal pedis vessels as trunk, anterior medial malleolus vessels, the first metatarsal vessels and anterior lateral malleolus vessels as branches respective was designed for repairing soft tissue and tendon defects of multi-fingers. Results In all 12 cases, total survival of flap was 11 cases , 1 case had partial necrosis of distal flap and it was healed by dress changing. Conclusion Transplantation of muliplefoliated flap combined with tendon graft from dorsal foot is a good method to repair soft tissue and tendon defects of multi-fingers.
4.Quality evaluation of red ginseng in different transplating systems.

OBJECTIVETo compare the quality of red ginseng in different transplanting systems, and thus provide the basis for ginseng cultivation and processing.
METHODBased on the Chinese Pharmacopoeia and literature relating to red ginseng, the ten ginsenosides, total ash, acid-insoluble ash, volatile oil, ether extract and total protein of red ginseng in different transplanting systems were studied or determined.
RESULT AND CONCLUSIONThe content of total ash and acid-insoluble ash in red ginseng was less than 5.0%, 0.3%, respectively. The content of three ginsenosides (Rg1, Re, Rb1) was in accordance with the requirements of Chinese Pharmacopoeia version 2010, and the content of ten ginsenosides was significant different.

5.Clinical prospective study on maternal-fetal transmission of human bocavirus.

OBJECTIVETo investigate maternal-fetal transmission at human bocavirus (HBoV).
METHODSIgG antibody to HBoV in serum samples of 316 mothers were determined with ELISA and HBoV DNA was determined with real time PCR in the sera of the mothers and their infants.
RESULTSHBoV-IgG was positive in 40.20 percent (127/316) of the mothers, while it was positive in 29.43 percent (93/316) of the cord blood specimens of the infants. The difference between the two groups was significant (X2=8.12, P less than 0.005); 93 samples of both the mothers and the infants were positive for HBoV-IgG.
CONCLUSIONHBoV-IgG can cross the placenta to the fetuses through placenta. Further study is needed to answer the question whether vertical maternal-fetal transmission occurs.

6.Identification of KI polyomavirus in children with lower respiratory tract infections from Zhejiang region of China.

KI polyomavirus, which was firstly discovered in 2007, is a new human polyomavirus belonging to Polyomaviridae and containing circular double-strand genomic DNA. This study was based on identification assay of KI polyomavirus reported. Total 2293 clinical sputum specimens from children under 3-years-old were collected and screened from Wenzhou Medical College affiliated Wenling Hospital, Zhejiang Province. A KI polyomavirus was detected and identified, the positive rate was 0.04%. The sequences of PCR products was identical to that of the viral capsid protein (VP1) gene derived from KI polyomavirus. The results strongly suggested that the KI polyomavirus was found firstly in Chinese children with acute lower respiratory tract infections from Zhejiang region. This study provided new information for further investigation of etiopathogenisis and diagnosis in children with lower respiratory tract infections.

7.Discovery and identification of WU polyomavirus in children from Zhejing region.

WU polyomavirus, which was firstly discovered in 2007, is a new human polyomavirus belonging to Polyomaviridae and containing circular double-stranded genomic DNA. In this study, the 278 clinical sputum specimens from children under 5 years old were collected from Wenzhou Medical College affiliated Wenling First Hospital, Zhejiang Province. Based on identification assay of WU polyomavirus previously reported, a WU polyomavirus was identified from clinical samples successfully, the positive rate was 0.4%. The sequences of PCR products were identical to that of VP2 gene and large T antigen gene derived from WU polyomavirus reported. The above results strongly suggested that the WU polyomavirus isolated was firstly found in Chinese children with acute lower respiratory tract infections. This study provides a firm basis for further research of WU polyomavirus.

8.Expression of recombinant VP2 gene in insect sf9 cells and screening of clinical specimens.

OBJECTIVETo clone and express VP, gene from HBoV, and the expressed VP, protein was as the antigen in order to detect serum from children in Wenling area with lower respiratory tract infections.
METHODSThe VP, gene was recombined with the genome of Baculovirus, which infected the insect cell. The fusion protein with HA tag was applied to confirm the specificity of expressed protein. Furthermore, the recombinant protein was observed using electron microscopy. The 176 serum from children in Wenling area with lower respiratory tract infections was screened using Western blot.
RESULTSThe expressed VP2 protein was more than 60% in total proteins from insect cell, and MWt about 60 x 10(3). The virus-like particle (VLP) was observed using electron microscopy, and size about 20 nm. The 176 serum from children in wenling area with lower respiratory tract infections was screened using Western blot. The HBoV positive rate was 2.28% (4/176).
CONCLUSIONThe VP2 protein from human bocavirus was expressed in insect cell successfully. Through HA tag the VP2 protein was specific, and then the assay using SDS-PAGE with Western blot could detect and screen the antibody in serum from children with lower respiratory tract infections rapidly and accurately.

9.Soluble expression of A/H1N1 influenza virus HA with Drosophila S2 cell line and its bio-activity identification.

OBJECTIVETo express soluble HA of A/H1N1 influenza virus in drosophila S2 cell line and identify its bio-activity.
METHODSHA gene was amplified from A/Shenzhen/71/09 virus strain using RT-PCR, then we constructed pAC5.1-HA expression vector, which was co-transfected into S2 cell with pCoblast vector. After transfection, stable S2 cell was selected through Blasticindin. HA in the supernatant was identified with Western Blot assay and purified with Ni-column. Recombinant HA was immunized into BALB/c mice 3 times, and the Abs titers were evaluated with ELISA.
RESULTSWe successfully cloned HA gene with 1.7 x 10(3) bp of A/Shenzhen/71/09 virus strain and got recombinant pAC5. 1-HA expression vector. Stable S2 cell line was established after transfection and selection, which continuously expressed HA with molecular weight 75 x 10(3) D. After immunization with HA, the Abs titers were 1:1280 and 1: 5120 respectively on 10 d, 30 d.
CONCLUSIONWe expressed soluble HA with good bio-activity, which contributed to research on immune diagnosis, subunit vaccine, and monoclonal Abs for influenza.
