1.Isolation and Characterization of a ?-tubulin Gene from Trichoderma Harzianum

Based on a ?-tubulin gene EST sequence from Trichoderma harzianum mycelium cDNA library,a 1.74kb ?-tubulin gene coding region,together with a 1.5kb 5'-flanking region and a 1.0kb 3'-flanking region was cloned from T.harzianum by IPCR methods.The gene encodes a 446 amino acid polypeptide,which shows a high degree of homology with other fungal ?-tubulins.A three-dimensional model of the T.harzianum ?-tubulin was built based on the crystal structure of bovine tubulin using homology modeling method.Tertiary protein structure of the ?-tubulin should provide a basis for understanding a significant body of research on tubulin's properties and interactions with antimicrotubule agents.

3.A Rapid Method for Generation of Homologous Recombinant Fragments—Fusion PCR

Fusion PCR, which employs chimeric primers to generate PCR products with complementary ends in its amplifications, is a rapid and flexible method in joining different DNA fragments. This method can assemble DNA fragments without the treatment of restriction endonucleases and T4 DNA ligase. It offered a shortcut for the construction of homologous recombinant fragments. Through assembling three recombinant fragments successfully, fusion PCR procedure was improved and manipulation essentials of fusion PCR were described in detail. The results indicated that the improved procedure can assemble three or four fragments simultaneously, and the length of each fusion product is above 4.5 kb. The result recombinants were proposed to be use in further experiments, which structures had been confirmed by sequencing.
4.Clinical diagnosis and treatment in 230 cases of cryptorchidism

Objective To evaluate the diagnosis and treatment of cryptorchidism and its complications,and to emphasize the appropriate surgical treatment age of cryptorchidism. Methods The data of diagnosis and treatment of 230 cases of cryptorchidism (a total of 299 undescended testicles) were reviewed.Their ages varied from 1 to 59 years and the mean age was (9.5?9.0)years.88 cases had cryptorchidism on the left side;73 cases on the right side;and 69 cases on both sides.Cryptorchidism was complicated by hypospadias in 11 cases and by latent penis in 2 cases. Results In these series only 25 cases (10.9%) underwent surgical treatment within 2 years of age.Surgical operations were performed in these cases (289 sides) and the locations of the undescended testicles were determined.Of them 273 ones underwent primary descending and fixation,15 ones underwent resection and histopathological examinations for mahgnant or shrinked tendency 1 was absent during the detection,and they were followed up for 4 months to 5 years with the success rate being 98.9 %.Of these cases 260 ones(90.0%) were external abdominal;25 ones (8.7%) were intra-abdominal;4 ones were missing (1.4%).For external abdominal testicles the positive ratio of detections by B-ultrasonography was 86.3%(126/146) and that of physical examination was 63.0% (92/146),which showed statistically significant difference ( P

8.Epidemiological investigation of an outbreak of P1-Ⅱ Mycoplasma pneumoniae infection in a school

Abstract
On October 13 2023, the Beijing Economic and Technological Development Area Center for Disease Control and Prevention recAAAAAeived a report from a school indicating an outbreak of fever among students in Class 2, Grade 4. An on-site epidemiological investigation was immediately conducted. A total of 14 cases were reported, all of whom were students, with an attack rate of 34.15%. The onset of illness was concentrated between October 6 and 15, with the main clinical symptoms being fever, cough and fatigue. There were no cases of hospitalization, severe illness or death. Seven cases tested positive for Mycoplasma pneumoniae P1-Ⅱ. Based on the on-site investigation and laboratory test results, it was an outbreak of influenza-like illness caused by Mycoplasma pneumoniae infection. The likely cause of the spread was cases infecting other classmates through respiratory droplets while attending class. The outbreak occurred only in one class and did not spread to neighboring classes. Schools should actively carry out health education to raise awareness among parents for reporting infectious diseases, and strengthen morning and afternoon screenings to improve the sensitivity of identification.

There is currently no effective treatment for PD, and dyskinesia symptoms affect the quality of life of patients with PD. 10.Nonspecific apoptosis induced by long, double-stranded RNA in human osteosarcoma cells

Objective To construct the plasmid that can produce long,double-stranded RNA and identify nonspecific apoptosis induced by long, double-stranded RNA in human osteosarcoma cells. Methods Plasmid pCI-neo-dsRNA was constructed by two promoters driven DNA sequence based on nontoxic, completely exoteric gene EGFP from the head and the end simultaneously to synthesize sense and antisense RNAs. Human osteosarcoma cells MG63 were transfected with pCI-neo-dsRNA and 48 h later the transfected cells were assessed by Apoptosis Detection System, FCMand MTT method. Results Plasmid pCI-neo-dsRNA was constructed successfully, MTT measurement showed that the absorbance of cells decreased gradually with the increase of plasmid pCI-neo-dsRNA dose(0.05 ?g, 0.10 ?g, 0.15 ?g, 0.20 ?g,0.25 ?g,0.30 ?g). MG3 transfected with pCI-neo-dsEGFP presented a great amount of irregular green fluorescence that indicated cell apoptosis. As the quantity of dsRNA increased, the percent of apoptosis cells also raised. While the dose of pCI-neo-dsEGF was 3.0 ?g, the apoptotic results of pCI-neo-dsEGF in MG3 were similar to that of 5-FU. Conclusion Long, double- stranded RNA could trigger human osteosarcoma cell apoptosis and the further research on using long, double-stranded RNA in osteosarcoma gene therapy can be done based on this theory.
