1.Comparison of clinical effects of different kinds of gingival retraction technique in treatment of anterior teeth restoration

Objective To compare the clinical effects of four kinds of gingival retraction technique in treat-ment of anterior teeth restoration.Methods 169 patients for 352 teeth with tooth body and tooth dentition defect by anterior crown bridge repair were chosen and divided into 4 group by patients'order,including A group(88cases)with single track method,B group(91cases)with single line for two times method,C group(89cases)with double line method and D group(94cases)with double line for two times method.And the fixed restoration satisfaction rate of anterior teeth of both groups was compared.Results The satisfaction rates of gums,shoulder,impression,model and restoration of A group were 73.9%,70.1%,69.3%,65.9%,61.4% respectively,which of B group were 83.5%, 80.2%,80.2%,80.2%,76.9%;which of C group were 84.3%,80.9%,78.7%,77.5%,76.4%;which of D group were 94.6%,93.6%,92.6%,92.6%,90.4%.There were no significant differences in the fixed restoration satisfaction rate of anterior teeth between B group and C group(χ2 =2.43,1.17,2.85,3.04,2.30,all P >0.05). The fixed restoration satisfaction rate of anterior teeth of D group were significantly better than A,B and C groups (χ2 =14.90,16.39,15.83,19.87,21.38,all P <0.05 ).Conclusion Double line for two times method in the treatment of anterior teeth restoration can efficiently avoid ree gingival damage in the situation with abutment shoulder preparation,improve the quality of preparation,impression and model,and are helpful to higher the fitness of the neck of anterior teeth fixed restorations.
2. Screening for simulation peptide specifically binding to the first and the second extra-cellular domain of CCR5 and its therapeutic effect on mice with autoimmune encephalomyelitis

Objective: To screen for simulation peptide binding specifically to the first and the second extra-cellular domain of CC chemokine receptor 5 (CCR5), and to observe their therapeutic effect on mice with experimental autoimmune encephalomyelitis (EAE). Methods: Phage display peptide library was used to screen for peptide sequence binding specifically to CCR5; ELISA was used to identify its binding activity and analyze its DNA sequence. The simulation peptide was synthesized and was injected into abdominal cavity of the EAE mice. Spinal cord tissues were obtained and the pathologic changes were studied by H-E staining in EAE control group and simulation peptide group. Results: Twenty phage clones were randomly chosen for identification and ELISA showed that there were eighteen clones had a strong binding activity with CCR5. The positive clones were sequenced and four peptides of high frequency were obtained: STFTTTL, TPIPQLL, SLPLPKP, and QTSSAAL. Mean clinical score of mice in the EAE model group was 3 and that of the simulation peptide group was 1. H-E staining found that the spinal cord tissues in EAE model group had great number of inflammatory cells and evident demyelination changes; the simulation peptide group had less inflammatory cells and no demyelination changes. The four short peptides had an effect of suppressing and delaying the development of EAE, with the average inhibition rate being 43% (P<0.05). Conclusion: The screened CCR5 simulation peptide has evident inhibitory effect against EAE, indicating that CCR5 may play an important role in the course of multiple sclerosis (MS).

4.Analysis of peblA gene sequence and construction of eukaryotic expression recombinant plasmid of peblA gene of Campylobacter jejuni Pen:19

Objective As the genome sequence of Campylobacter jejuni is a hypervariable sequence, the peblA gene sequence of Campylobacter jejuni Pen∶19 was sequenced and the peblA gene conservative was identified. To construct the eukaryotic expression recombinant plasmid of the peblA gene of Campylobacter jejuni Pen∶19 . Methods Total DNA of Campylobacter jejuni Pen∶2, Pen∶8, Pen∶19 and Pen∶21 as templates respectively, peblA gene DNA with KpnⅠ and EcoRⅠ sites was amplified by PCR using the same primer and the PCR product of Pen∶19 was sequenced. Because most close association with Guillain-Barre syndrome, the PCR product of Pen∶19 was selected as target gene and cloned into pcDNA3.1(+) and constructed the recombinant plasmid that was identified by endonuclease digestion and PCR and confirmed by sequencing. The recombinant plasmid was then transfected into mammalian cell COS-7 for expression. The transient expression was investigated by RT-PCR. The expression of peblA gene in the culture supernatant of positive clones was analyzed by ELISA. Results The peblA gene sequence of Campylobacter jejuni Pen∶2 and Pen∶19 was identical. The target gene was amplified from COS-7 cells transfected with recombinant plasmids by RT-PCR. PEBl protein could express in the culture supernatant in the transfected COS-7 cells by ELISA. Conclusion The peblA gene of Campylobacter jejuni was conservative. The recombinant plasmid was constructed and expressed in COS-7 cells successfully. The results obtained lay the foundation for research on development of Campylobacter jejuni DNA vaccine.
5.Effect of Edaravone Combined with Plantula Ixeridis Sonchifoliae on Relieving Neurologic Impairment Symptoms of Patients with Acute Cerebral Infarction

Objective To observe the effect of edaravone combined with Plantula Ixeridis Sonchifoliae(PIS) on relieving neurologic impairment symptoms of patients with acute cerebral infarction(ACI).Methods Ninety-two ACI patients were equally randomized into the treatment group and the control group.The two groups were given routine western medical treatment such as use of aspirin and neurotrophic drugs,and decreasing intracranial pressure and symptomatic treatment for large-size cerebral ischemia.Additionally,the treatment group received intravenous injection of edaravone injection plus PIS injection,and the control group received PIS injection.Fourteen days constituted one treatment course,and the treatment lasted 2 courses.The score of neurologic impairment was evaluated with European Stroke Scale(ESS),and the score of activities of daily life(ADL) was observed before treatment,and after one and two treatment course(s).The adverse reaction was recorded during the treatment.Results At the end of the first treatment course,the scores of ESS and ADL were improved in both groups(P0.05).At the end of the second treatment course,the scores of ESS and ADL were markedly improved in both groups(P
6.Survey and thought of laboratory medicine undergraduates

Objective To investigate students' view about the laboratory medicine curriculum and teaching reform,and to provide reference for further reform and development. Methods The students specialized in laboratory medicine in School of Medicine,Shanghai Jiaotong University were involved in the survey.Questionnaires were answered and analyzed.The research method of expert consultation was adopted. Results Students had some different opinions for course content,structure and so on,and they expected more clinical practice,more hands operation,and more teachers' guidance.Conclusion To train laboratory technicians,it was necessary to build a reasonable system of professional courses and train practical talents.

The plastic splint PCL,which is introduced in this article,is a new type of medical exter- nal immobilization material.During the course of its development,the burden has been optimality seeked with regressive orthogonality after screening of the material.Its function has attained the international levels during the eighties.This splint may be used not only in the treatment of burns and various orthopedic conditions,but also in the immobilization of fracture and rehabilation treatment of orthopedic patients etc.
8.Effects of esculentoside A on production of IL-1 and TNF by rabbit synovial cells

Objective: To investigate the influence of esculentoside A(EsA) on production of IL-1 and TNF by rabbit synovial cells induced by LPS. Methods: levels of IL-1 and TNF in the supernatant of rabbit synovial cell were determined by examining proliferation of thymic cells and by bioassay L929 cells as target cells, respectively. Results: EsA in 5-40 μg/ml could significantly inhibit the production of IL-1 and TNF from rabbit synovial cells induced by LPS. Conclusion: EsA can inhibit the production of IL-1 and TNF from synovial cells. It suggests that EsA may play a role in improving the rheumatoid arthritis.
9.Study on preparation and oral efficacy of insulin-loaded poly(lactic-co-glycolic acid) nanoparticles.

AIMTo investigate the possibility of poly(lactic-co-glycolic acid) as a carrier for the delivery of macromolecular.
METHODSInsulin-loaded poly (lactic-co-glycolic acid) nanoparticles (INS-PLGA-NPs) was prepared by a double-emulsion solvent evaporation method. The size distribution was examined by photo-correlation spectrometry. The entrapment efficiency was determined by HPLC and important factors that affected the entrapment efficiency were investigated. The loading mechanism of different size nanoparticles was assayed by radioimmunoassay (RIA). INS-PLGA-NPs release behavior in vitro was carried out under sink condition. After oral administration of the nanoparticles to alloxan-induced diabetic rats, its glucose level was determined by glucose oxidize method and the oral pharmacological bioavailability in contrast to s.c. of insulin solution was calculated according to the area over the curve.
RESULTSThe INS-PLGA-NPs was prepared with poloxamer 188 as a emulsifier, the mean diameter was 149.6 nm and the polydispersity index was decreased to 0.09. While the entrapment efficiency was increased to 42.8%. Most of the insulin loaded was adsorbed on the surface of the nanoparticles. The release behavior in vitro showed an initial burst effect followed by a slower rate stage. After oral administration of 10 u.kg-1 INS-PLGA-NPs, the plasma glucose level decreased significantly after 4 h (P < 0.05), 10 h later the glucose level decreased to the lowest (52.4% +/- 10.2%, P < 0.01) and the relative pharmacological bioavailability is (10.3 +/- 0.8)%.
CONCLUSIONPLGA-NPs might be used as a new oral carrier for protein drug delivery systems in the future.

10.Study of production of sesquiterpenes of Aquilaria senensis stimulated by Lasiodiplodia theobromae.

To investigate the mechanism of agarwood formation in Aquilaria sinensis induced by Lasiodiplodia theobromae, the fermentation liquor of L. theobromae was analyzed qualitatively and quantitatively by gas chromatography-mass spectrometry (GC-MS). JAs were detected in the fermentation liquor. The effect of the fermentation liquor on the abundance of sesquiterpenes in the callus of A. sinensis was analyzed by solid phase microextraction-gas chromatography-mass spectrometry (SPME-GC-MS). And the fermentation liquor stimulated alpha-guaiene, alpha-humulene and delta-guaiene biosynthesis in calli. It was inferred that L. theobromae produced JAs, which resulted in a significant increase of sesquiterpenes in A. sinensis.
