Objective Rat glial cell derived neurotrophic factor(GDNF) gene and tyrosine hydroxylase(TH) gene were widely used in gene therapy of Parkinson's Disease(PD).The present study was designed to investigate whether the two target genes GDNF and TH could express simultaneously in one adeno-associated virus(AAV)vector and to discuss the probability of using it in gene therapy of PD.Methods RNA of GDNF and TH in HEK293 packaging cells were examined by reverse transcription-polymerase chain reaction(RT-PCR).Expressions of GDNF and TH in infected bone marrow stromal cells(BMSCs) and rat brain coronal sections were examined with immunocytochemistry and immunohistochemistry assay.Results RNA of GDNF and TH were detected in HEK293 packaging cells.Expressions of this two genes were observed in both bone marrow stromal cells and brain sections of rats.Theexpression efficiency of AAV-LacZ was about 50% in HEK293 packaging cells and approximately 15% in infected BMSCs and the expressions of TH and GDNF on the AAV-GDNF/TH virus injected points both had significant differences(P

Objective To investigate the correlation between plasma N-terminal pro-brain natriuretic peptide (NT-proBNP) and haemodynamics evaluated by right-heart catheterization in patients with chronic thromboembolic pulmonary hypertension (CTEPH).Methods Fifty-three patients with CTEPH were retrospectively reviewed.All patients underwent right-heart catheterization to detect haemodynamics.The level of plasma NT-proBNP was measured by electrochemical luminescence.The indexes were analyzed.Results Pulmonary artery systolic pressure was (84.9 ± 21.6) mmHg (1 mmHg =0.133 kPa).Pulmonary artery diastolic pressure was (31.1 ± 9.7) mmHg.Pulmonary artery pressure was (49.3 ± 13.4) mmHg.Pulmonary vascular resistance was (1 047.4 ± 86.1) dyn ·s/cm5.Pulmonary capillary wedge pressure was (8.16 ± 3.02) mmHg.Right cardiac output was (3.36 ± 1.42) ml.Right cardiac work was (1.97 ± 0.95) kg·m.The level of plasma NT-proBNP in CTEPH was (2 301.5 ± 1 787.3) ng/L.The level of plasma NT-proBNP in CTEPH had positive correlation with pulmonary vascular resistance (r =0.429,P =0.02),and had negative correlation with right cardiac output (r =-0.583,P＜ 0.01) and right cardiac work (r =-0.521,P ＜ 0.01).The level of plasma NT-proBNP in CTEPH had no correlation with pulmonary artery systolic pressure,pulmonary artery diastolic pressure,pulmonary artery pressure,pulmonary capillary wedge pressure (P ＞ 0.05).Conclusion The level of plasma NT-proBNP can be used as a better predictor for evaluating pulmonary vascular resistance and right cardiac function in CTEPH.

Objective To explore the relationship between HPV infection and the incidence of cervical canc-er and precancerous lesions in women.Methods 108 patients with cervical disease who accepted cervical screening were chosen as the research subjects, and they were given colposcopy and pathological examination, hybrid capture 2 generation method for quantitative detection of HPV-DNA content,RLU/CO was used for viral load and HPV-DNA content was more than 1ng/L(RLU/CO>1) can be judged as positive.Results The positive rates of HPV-DNA was 35.00%,67.74%and 95.83%of chronic cervicitis,cervical cancer precursor lesions( CIN) and invasive cervical squamous cell carcinoma,the difference was statistically significant(χ2 =10.73,P<0.05) .The positive rates of HPV DNA in patients with typeⅠCIN,typeⅡCIN,typeⅢCIN were 44.44%,62.50%and 95.00%respective-ly,the difference was statistically significant(χ2 =11.43,P<0.05).HPV-DNA and the positive rate of cervical cancer patients with histological grade,clinical stage,lymph node metastasis,there was no significant correlation(χ2 =1.76,1.44,1.89, all P >0.05 ) .Conclusion HPV -DNA positive rate increased with the severity of cervical lesions in patients,but it had no significant correlation with clinical and pathological parameters,the active prevention of HPV infection of cervical cancer prevention had important significance.

Abstract: Objective To analyze the changes of coagulation indicators in patients with hepatitis B complicated with Clonorchis sinensis (C. Sinensis), and provide reference value for diagnosis, drug using and prognosis monitoring. Methods　The patient samples were collected from the Third Affiliated Hospital of Sun Yat-sen University from June 2018 to February 2022 and divided into six groups. They were 40 healthy patients, 47 patients with simple chronic hepatitis B, 47 patients with post-hepatitis B liver cirrhosis, 40 patients with C. Sinensis mono-infection, 30 patients with chronic hepatitis B patients co-infected with C. Sinensis and 27 patients with post-hepatitis B liver cirrhosis patients coinfected with C. Sinensis. Four coagulation indexes, prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT) and fibrinogen (FIB), were detected and compared among the groups. Results Compared with the healthy group, C. Sinensis mono-infection group had higher level of FIB and delayed PT, APTT; chronic hepatitis B group and chronic hepatitis B patients co-infected with C. Sinensis group had delayed PT, APTT, TT and significant lower FIB, these differences were statistically significant (all P<0.05). Compared with simple chronic hepatitis B group, post-hepatitis B liver cirrhosis and chronic hepatitis B patients co-infected with C. Sinensis had significant delayed PT, APTT, TT and lower FIB (all P<0.05). Compared with the post-hepatitis B liver cirrhosis and chronic hepatitis B patients co-infected with C. Sinensis, post-hepatitis B liver cirrhosis patients coinfected with C. Sinensis group had significant delayed PT, APTT, TT and lower FIB (all P<0.05). Conclusions The coinfection of C. Sinensis will further aggravate the coagulation dysfunction of HBV patients, leading to poor treatment and prognosis. HBV patients will have worse coagulation function in the process to post-hepatitis B liver cirrhosis; Therefore, it is important to pay attention to C. Sinensis co-infection when treating HBV patients, so that further guidance on clinical use and monitoring of prognosis can be provided.
