Objective To prevent the prevalence of drug resistance viruses,we analyzed drug resistance mutations of HIV-1 among HIV-1 infected individuals in seven provinces of China. Methods We acquired 718 HIV-1 infected patients' treatment and compliance from questionnaires, amplified HIV-1 pol genes by RT-PCR and nest-PCR,sequenced RT and protease regions and run clustal with the subtype B consensus amino acid sequence in Stanford HIV drug resistance sequence database. Results The viral load in treated patients was obviously lower than that in untreated patients(P

Objective To explore the bioactivity and stability of the antifungal substance produced by Streptomyces NG-715 as well as to establish the assay for biological activity detection. Method Take the antifungal substance as experimental materials, and test its minimum inhibitory concentration and minimum bactericidal concentration on four fungi strains including Saccharomyces cerevisiae, Penicillium citrinum, Aspergillus niger, Rhizopus sp. Aspergillus niger was used as indicator strain to measure the biological activity and stability of the antifungal substance. Results The results showed that the MIC of the antifungal substance on four fungi strains including Saccharomyces cerevisiae,Penicillium citrinum,Aspergillus niger,Rhizopus sp were 1.25, 2.5, 3.75 and 3.75μg/mL, respectively. The MBC of the antifungal substance on four fungi strains were 2.5, 10, 17.5 and 17.5μg/mL, respectively. Linearity regress equation of the antifungal substance in Aspergillus niger was y=26.963 x-27.6,R 2=0.9991. The antifungal substance was pH-stable, heat-stable but ultravio1 et-sensible. Conclusion The results from this study will porvide useful information for the further extraction and analysis on the bioactive compound.

OBJECTIVETo investigate the clinical effects of double-plating internal fixation for the treatment of medial tibial plateau fractures with fracture-dislocation type through combined anterior and inverted "L" posteromedial knee approaches.
METHODSFrom February 2010 to September 2013, 17 patients with medial tibial plateau fractures with fracture-dislocation type were treated by double-plating internal fixation at our department. Anterior knee approach was adopted for medial mass reduction and fixation, while the inverted "L" posteromedial approach was used for posteromedial mass. There were 11 males and 6 females, with an average age of 42 years old (ranged, 21 to 65 years old). The efficacy were investigated by comparing pre- and post-operative radiographic data, and by evaluating clinical symptoms using the HSS score system.
RESULTSAll the patients were followed with a mean time of 18 months (ranged, 11 to 25 months). The tibial-plateau varus angle immediately after operation and at the latest follow-up were respectively (87.8 ± 4.2)° and (88.2 ± 4.6)°, and there was no statistical difference (P = 0.458). The posterior slope angle immediately after operation and at the latest follow-up were respectively (10.1 ± 3.7)° and (10.3 ± 4.1)°, and there was no statistical difference (P = 0.512). The knee joint HSS score was 86.7 ± 6.1. No patient had complications such as infection, skin necrosis, internal fixation loosening or loss of fracture reduction.
CONCLUSIONDoubl-plating internal fixation through combined approaches is an ideal operation method for medial tibial plateau fractures with fracture-dislocation type, because it can provide clear surgical field, satisfactory reduction, reliable fixation, and chance for early functional exercise.

Objective To investigate the relatonahip between TNF-α and ventricular arrhythmias after acute myocardial infarction(AMI)and its mechanism.Method Both the clinical and animal experiments were done.(1)Clinical experiment:Eighty patients with AMI were included in Union Hospital,Tongji Medical College,Huazhong University of Science and Techology, from May 2005 to November 2006 according to the WHO diagnostic criteria.Co-infection of diseases such as severe upper respiratory infection, lung infection, high fever, cancer,et al were excluded. The relationship between the levels of TNF-α and arrhythmias were observed at different times after AMI.A straight line correlation, analysis Was done.(2)Animal experiment:Different concentrations of TNF-αwere added to isohted rat hearts for observing the arrhythrnia effects. The effect of TNF-α on intracellular Ca2+ concentration was detected by laser confocal technique. All data were analyzed by SNK-q test using SPSS 13.0 sofeware prograrn.Results(1)The plasma levels of TNF-α were significantly associated with the Lown class of PVC after AMI and they were higher in AMI of anterior wall[(46.41±10.34)pg/ml]than other positions [(28.25±6.35)pg/ml,P<0.05].2)The frequency of ventricular arrhythmias was interrelated with the concentralions of TNF-α.Using etanercept beforehand,TNF-α induced a slight increase of intracellular Ca2+ intensity (P<0.05).Conclusions There was a relationship between TNF-αlevels and ventricular arrhythmisa in patients with AMI.Animal experiments confirmed the isolated heart perfusion with TNF-α induced ventricular arrhytrnias.Expression of TNF-α after AMI was related with the occurrence of ventricular arrhythrnias. The effect might be associated with the increased inuaeellular Ca2+ intensity caused by TNF-α.

AIM: To study the culture and characteristics of mouse adult bone marrow-derived pluripotential mesenchymal stem cells and its potential to differentiate into insulin secretion cells. METHODS: Cells were plated on 60% DMEM-LG and 40% MCDB-201 medium supplemented with 2% fetal calf serum and 10 ?g/L PDGF-BB, 10 ?g/L EGF and 1?10~6 U/L LIF. The proliferation rate, phenotype and oct-4 mRNA were tested. After it was plated on serum-free medium DMEM/F12 with GLP-1 and nicotinamide, the nkx2.2 ngn3, pdx-1 and insulin 2 mRNA were tested. RESULTS: The cells were round with large nucleus and scant cytoplasma. They were CD13~+, CD44~-, CD45~- and MHCⅡ~-. Oct-4 mRNA were present. The nkx2.2 pdx-1 and insulin 2 mRNA were presented in cells plated on the inducing medium at 14 days. CONCLUSION: The adult bone marrow-derived pluripotent stem cells were cultured and they has the possibilities to be induced into insulin-secreting cells.
