1.The Antifungal Mechanism of Bioactive Metabolites Produced by Penicillum sp. TS67

The antifungal mechanism of the TS67(Penicillum sp.) producing bioactive metabolites was investigated in this paper. The test indicated that mycelia growth, spores forming and spores germination of Bipolaris maydis and Fusarium oxysporum can be inhibitied by metabolites, after 50% fermentation broth acting 120 hours, the inhibition rate of hyphal growth can reach about 77.78%, 70.30%, the inhibition rate of spores forming also arrived at 58.8%, 73.5%; under 50% fermentation broth acting 12 hours, the inhibition rate of spores germination were up to 78.3%, 62.0%, the mycelia under activity metabolites conditoins also showed surface nodulation and growing point expanding irregularly, protoplasmic condesing, etc. Based on all these results, we may draw a conclusion that activity metabolites maybe target at fungal cell wall.

3.Clinical observation of cervus blood granule combined with GP in the treatment of advanced non-small cell ;lung cancer after chemotherapy

Objective To discuss the efficacy of GP combined with cervus blood granule in treating advanced non -small cell lung cancer (NSCLC ) after chemotherapy.Methods Eighty patients with advanced NSCLC were randomly divided into two groups by digital table method.In the combined group,forty patients were trea-ted by GP combined with cervus blood granule.In the control group,forty patients were treated by GP alone.All the patients were tested routine blood in the seventh and tenth day.The value of platelet was recorded,the incidence of thrombocytopenia between two groups was compared.Results At the seventh day of chemotherapy,in the control group,thrombocytopenia Ⅰ found in 5 cases,2 cases of thrombocytopeniaⅡ,1 case of thrombocytopeniaⅢ,thrombo-cytopenia Ⅳ in 1 case.In the combined group,thrombocytopenia Ⅰ occurred in 3 cases,1 case of thrombocytopeniaⅡ,no grade 3 or 4 thrombocytopenia,there was no statistically significant difference between the two groups(Z=-1.259,P=0.208).At the tenth day of chemotherapy,in the control group,thrombocytopenia Ⅰ found in 6 cases, 3 cases of thrombocytopenia Ⅱ,1 case of thrombocytopenia Ⅲ,thrombocytopenia Ⅳ in 1 case.In the combined group,4 cases occurred thrombocytopenia Ⅰ,thrombocytopenia Ⅱ in 1 case,no grade 3 or 4 thrombocytopenia.The difference between the two groups was statistically significant(Z=-1.966,P=0.049).Conclusion Cervus blood granule combined with GP can effectively reduce the classification of thrombocytopenia caused by chemotherapy.To some extent,it can also prevent the happening of the thrombocytopenia.
4.Rats hyperuricemia model established by lipid emulsion simulating irregular of diet.

Due to the irregular of diet and overfeeding greasy and surfeit flavor closely associated with hyperuricemia disease, the lipid emulsion containing high cholesterol was used to model. To obtain a more stable and sustained animal model for the efficacy evaluation of traditional Chinese herbs, we observed the influence on the serum uric acid of rat induced by the lipid emulsion compared with high purine diet. 36 SD male rats were randomized to the normal control group, high purine diet group and lipid emulsion group respectively. The general behavior, body weight and daily food intake of rats were observed. The orbital blood was taken to separate into the serum and 24 hours urine was collected. The serum indexes such as UA, BUN, Cr, ALT, AST, TC, TG, LDL-c were determined every 2 weeks, and XOD, ADA enzyme activity were determined at the 4th week. The urine indexes such as UA, Cr and Cua/Ccr were determined at the 4th week. After stopping modeling, the serum UA were determined two weeks and four weeks later respectively. At the 2nd week, the body weight and daily food intake of rats in the lipid emulsion group reduced significantly, and the level of serum UA, BUN, Cr, TC, LDL-c, ATL, AST raised significantly meanwhile TG reduced. At the 4th week, the serum UA in high purine diet group did not raise, and the serum XOD raised obviously while ADA did not; the serum UA in lipid emulsion group was higher significantly, and the serum XOD and ADA raised while Cua/Ccr reduced obviously. At the 6th weeks, the serum UA in both the high purine diet group and lipid emulsion group raised obviously. After stopping modeling, the serum UA in lipid emulsion group still maintained a high level at the 2nd week and back to the normal level at the 4th week. Compared with high purine diet, the hyperuricemia model induced by lipid emulsion forms earlierand more stable. It maybe has great value to study the pharmacodynamics of traditional Chinese medicine treatment to hyperuricemia disease. Its mechanism may be related to increasing XOD and ADA enzyme activity which can promote uric acid synthesis, meanwhile inhibiting of uric acid excretion.

5.Application of lift and squeeze technique in phacoemulsification of hypermature cataract

AlM: To evaluate the effectiveness and security of lift and squeeze technique in phacoemulsification of hypermature cataract.
METHODS: From June 2010 to June 2013, totally 156 eyes with hypermature cataract, which received phacoemulsification in our hospital, were enrolled. Lift and squeeze technique was used to chop the nucleus, and 1g/L Trypan blue was used for capsulorhexis. Average time of phaco complication, corneal edema and visual outcome were recorded.
RESULTS: The best-corrected visual acuity ( BCVA ) was 0. 1-0. 4 in 15 eyes (9. 6%), 0. 5-0. 7 in 82 (52. 6%) eyes, and 0. 8-1. 0 in 59 ( 37. 8%) eyes at 3mo after surgery. The phaco time was 25-56s (average 42±10s), the maximum phaco power was 30%. Posterior capsular rupture and vitreous loss happened in 2 eyes (1. 3%), and the lOLs were implanted in the sulcus. Corneal edema classified at grade Ⅰ were seen in 12 eyes (7. 7%), and 5 eyes (3. 2%) at gradeⅡ, no eye at gradeⅢ and gradeⅣ. The mean endothelial cell loss was 8. 7%at 3mo.
CONCLUSlON: Crystalline lens capsule staining with Trypan blue increase the success rate of intact continuous curvilinear capsulorhexis ( CCC ) . The lift and squeeze technique reduces the stress on the zonules and capsule, and decreases the phaco time and phaco power.

Objective To observe the effect of movement exercise combined with electroaeupuneture on the expression of Nestin in the hippocampus dentate gyrus (DG) after cerebral ischemia-repeffusion.Methods Fifty- four Wistar rats were used and randomly divided into a control group (Group A),an exercise training group (Group B),and an exercise training combined with electroacupuncture group (Group C).The middle cerebral arteries (MCA) of all the rats were occluded for 1 h,followed by reperfusion for 7,14 and 21 davs.Immunohistochemistry method was used to detect the expression of Nestin in the hippoeampus dentate gyrus.Results The number of Nes- tin-positive cells peaked in DG in all groups on the 7th day after cerebral isehemia-reperfusion.The number of Nes- tin-positive cells in DG ipsilateral to the ischemia-reperfusion lesion were significantly more than those in the opposite side at various time points (P

Objective To explore the clinical significance of the blood lactic dehydrogenase(LDH), ?_2-microglobulin(?_2-MG),D-dimer measuring in the diagnosis and treatment of Non-Hodgkin lymphoma. Methods In 40 cases with NHL,LDH was measured by L-P continuous monitoring method,?_2-MG was measured by luminescent immunoassay,D-dimer was measured by immunoturbidimettic assay.Results The levels of the blood LDH,?_2-MG and D-dimer in patients with NHL were higher than those of in the controls(P 0.05).Con- clusion The levels of blood LDH,?_2-MG,D-dimer can be taken as an auxiliary clinical index to diagnose, classify the phase,evaluate the effectiveness of treatment and prognosis in the NHL patients,and have impor- tant clinical significance.
9.Study on effect of four traditional Chinese medicines distributed along lung meridian on TLR2 and NF-κB expressions in mice with lung heat syndrome.

OBJECTIVETo investigate the effect of four traditional Chinese medicines distributed along lung meridian, namely Ephedrae Hebra, Zingiberis Rhizoma, Scutellariae Radix and Mori Cortex, on TLR2 and NF-κB expressions in mice with lung heat syndrome, in order to study the intervention effect of the four traditional Chinese medicines (TCMs) on the lung heat syndrome.
METHODOne hundred KM mice were randomly divided into the normal control group, the model group, the Ephedrae Hebra group, the Zingiberis Rhizoma group, the Scutellariae Radix group and the Mori Cortex group (20, 10 g x kg(-1)), nasally dripped with streptococcus pneumoniae to establish the mouse lung heat syndrome model, and then administered with different TCMs. The expressions of TLR2, NF-κB p65 proteins in lung tissues were analyzed by the immunohistochemical method. The expressions of TLR2, NF-κB p65 mRNA were measured by real time PCR.
RESULTCompared with the normal control group, the expressions of TLR2 and NF-κB p65 proteins in lung tissues in the model group were higher (P < 0.01), and the expressions of TLR2 and NF-κB p65 mRNA in lung tissues were up-regulated (P < 0.01 or P < 0.05). Compared with the model group, Ephedrae Hebra high and low dose groups, the Zingiberis Rhizoma low dose group and the Scutellariae Radix high dose group showed decreased expression of TLR2 protein (P < 0.05 or P < 0.01); Ephedrae Hebra high and low dose groups, the Zingiberis Rhizoma low dose group, Scutellariae Radix high and low dose groups and Mori Cortex high and low dose groups showed reduced expression of NF-κB p65 protein (P < 0.05 or P < 0.01). Ephedrae Hebra high and low dose groups, Zingiberis Rhizoma high and low dose groups, Scutellariae Radix high dose group and Mori Cortex high dose group showed down-regulated expression of TLR2 mRNA (P < 0.01 or P < 0.05).
CONCLUSIONEphedrae Hebra, Zingiberis Rhizoma, Scutellariae Radix and Mori Cortex can induce the TLR2/NF-κB inflammatory signal pathways by down-regulating the expressions of TLR2 and NF-κB p65 in protein and mRNA, so as to alleviate the lung tissue injury in mice with lung heat syndrome.

10.Small interfering RNA mediated silencing of cytochrome P450 3A4 gene

Objective To investigate the inhibitory effect of the cytochrome P450 3A4(CYP3A4) gene expression and function in CHL-3A4 cells lines by vector-ecpressed small hairpin interfering RNA(shRNA).Methods The shRNA expression vectors targeting CYP 3A4 gene(CYP3A4Ⅰ,CYP3A4Ⅱ,CYP3A4Ⅲ) were designed and constructed.The inhibitory effect of shRNA was detected by Western blot and RT-PCR analysis.The inhibitory effect of cytotoxic of cyclophosphamide using shRNA was measured by MTT assay.Results CYP3A4Ⅲ shRNA expressing vector significantly reduced the CYP3A4 mRNA(70%) and protein expression levels(75%) of the CYP3A4 gene in CHL3A4 cells suppression of CYP3A4 gene expression by CYP3A4Ⅲ largely reversed cyclophosphamide cytotoxicity(75%) in CHL-3A4 cells.Conclusion Vector-based RNAi could suppress CYP3A4 gene expression and function,and the use of RNAi to inhibit CYP3A4 gene expression in mammalian cells was a promising new tool for the study of cytochrome P450 gene function.
