4.Investigation of properties of the resveratrol-beta-cyclodextrin inclusion complex by drug-protein interaction spectra and molecular modeling.

The supramolecular inclusion properties of beta-cyclodextrin (beta-CD) and resveratrol (Res) were investigated using drug-protein interaction spectroscopy method. The differences between the results of interaction spectroscopy method and the results of classical method were compared. The total energy of the stable inclusion of cyclodextrin-resveratrol was calculated by Gaussian theory calculation. The stable inclusions in the process of interaction between resveratrol/inclusion complex and bovine lactgoferrin (BLF) were studied by molecular modeling. The results showed that the interaction spectroscopy method could explain the property of the inclusion in a more sensitive manner, it also interpreted the conveying mechanism of BLF binding with inclusion complex. The molecular modeling result showed consistent results with Gaussian theory calculation; both of the two methods obtained the stable configuration of beta-CD-Res inclusion. The relevant result provided an experimental consequence for the pharmacology research of beta-cyclodextrin-resveratrol inclusion complex as well as offering a new reference to the future research of supramolecular inclusion compound.

6.The complications of traumatic optic neuropathy

Objective:To investigate the complications of traumatic optic neuropathy and to call the surgeons' attention to precaution. Method:Retrospectively analysis of 122 cases patients with traumatic optic neuropathy and 3 cases were analyzed in detail including 1 case with purulent meningitis, another 1 case with internal carotid artery pseudoaneurysm and the other 1 case with internal carotid artery cavernous sinus fistula. Result:Most of the patients had the complications of orbital fracture, maxillofacial fracture, ocular and craniocerebral injury. A few of patients had other injuries all over the body. The case with purulent meningitis was cured with antibiotics. The case with internal carotid artery pseudoaneurysm was cured with neurosurgery. The visual acuity of the both cases were improved. The case with internal carotid artery cavernous sinus fistula died of severe hemorrhea. Conclusion: The patient with traumatic optic neuropathy has the possibility of severe cranial disorders,orbital fracture, maxillofacial fracture and injuries of viscera or limbs. It should be paid mare attention and treated accordingly.
7.Behavior assessment and magnetic resonance imaging examination on premature rats with periventricular leukomalacia induced by intracerebral injection of 3-nitropropionic acid

Objective To establish the model for periventricular leukomalacia(PVL) by intracerebral injection of 3-nitropropionic acid (3-NPA) and explore realistic model for concerned studies and investigate the diagnostic method with magnetic resonance imaging (MRI) examination. MethodsSprague-Dawley rat pups of both sexes at the age of postnatal day 5 (P5) were randomized and divided into two groups: NPA group and PBS group , and they were injected the same volume of 3-NPA and PBS with a tip located at the corpus callosum above the left ventricle by stereotaxis instrument, respectively. One day (P6), two days (P7), three days (P8) and nine days (P14) after injection, the injections were transcardially perfused and brains were collected. Then sections of brains were undertaken HE staining; growth and the time of opening eyes of the rats in the two different groups were observed and compared. Neurobehaviorai activity and memory tests were performed on postnatal day 29 (P29) and day 30 (P30). MRI examination was performed on postnatal day 30 (P30). ResultsMore weight increase and slower opening eye time were found in the NPA group compared with PBS group (P < 0.05). In the NPA group, sub-cortical and periventricular white matter rarefaction were observed by HE staining on P6, P7 and P8, significant lateral ventricle enlargement was found on P14, while the same changes were not found in the PBS group, and no histological changes in gray matter were noted in both groups. The outcomes of neurobehavioral tests of NPA group were much more abnormal compared with the PBS group (P < 0.05). MRI examination disclosed the signal changes of brain tissue is worse in the NPA group than that of the PBS group in muscle strength of limbs, autocinesis, capability and white matter. ConclusionsThe model for PVL induced by intracerebral injection of 3-NPA is characterized by damage to the periventricular white matter. The model can stimulate the pathologic change factually in vivo. The neurobehavioral movement is consistent with.clinical symptom. It can be used as a model to investigate some related disease. MRI examination is a feasible diagnostic method to show anatomic changes of white matter injury of the brain.
8.Effect of Overexpression of NAD+-dependent Malic Enzyme on Anaerobic Mixed Acid Fermentation of E.coli FMJ39

In order to investigate the effect of malic enzyme on anaerobic mixed acid fermentation,the gene encoding NAD+-dependent malic enzyme was amplified from E.coli DH5? genome by using PCR and cloned into vector pTrc99a to give an expression vector pTrc99a-sfcA.Malic enzyme was overproduced by E.coli FMJ39(ldh,pfl)harboring pTrc99a-sfcA under the condition of IPTG induction.The results of anaerobic fermentation indicated overexpression of malic enzyme would have influence on the pathway of formate,acetate and succinate of FMJ39.The concentration of formate and acetate was 17.58% and 15.27% higher than FMJ39,the succinate concentration was reduced by 26.87%.There was no obvious citrate concentration change.The experimental results also indicated that high concentration of L-Thr and L-Ser would induce the Tdc operon to convert pyruvate into formate and acetate even when pfl gene encoding the pyruvate formate lyase was inactivated.
9.Tuina Intervention and Sleep and Emotional Disorders due to Chronic Fatigue Syndrome

Objective:To study the tuina effect on sleep disorder and emotional problems of chronic fatigue syndrome (CFS) cases.Method:Thirty CFS cases were treated in the treatment group,by applying one-thumb pushing,rolling and pressing-kneading manipulations along the Governor Vessel,Bladder and Yangming Meridians.The intended major points include Fengfu (GV 16),Yaoyangguan (GV 3),Xinshu (BL 15),Pishu (BL 20),Ganshu (BL 18),Hegu (LI 4) and Taixi (KI 3).The treatment was done once every other day;10 times constitute a course of treatment.Another 30 healthy cases were selected in the control group,without any intervention.The sleep quality and mental state in the two groups were evaluated by PSQI,HAMA and HAMD.In addition,the pre-treatment and post-treatment score changes were observed.Results:Before treatment,the total score of PSQI,sleep quality,time of falling asleep,sleep efficiency,sleep disorder,daytime functional impairment,and scores of HAMD and HAMA in treatment group were all substantially higher than the control group (P<0.01).After 10 tuina treatments,the total score of PSQI,time of falling asleep,sleep disorder and daytime functional impairment as well as scores of HAMD and HAMA in treatment group were significantly changed,compared with before treatment,P<0.05.Conclusion:Those with CFS may present with sleep disorder and mental or psychological abnormality,tuina can improve their sleep quality and adjust their mental conditions.
10.The application of array comparative genomic hybridization for the detection of chromosomal defects in newborns

Objective To study the application of the array comparative genomic hybridization (Array-CGH) for the detection of chromosomal disorders in newborns.Method The Array-CGH technique was used to analyze the whole genome of the patients who were suspected of chromosomal disease in neonatal ward of our hospital from January to December in 2014,and further verification in genomic unbalanced ectopia was carried out by FISH (fluorescence in situ hybridization,FISH).Result Among 514 patients,104 were found carrying chromosomal abnormalities with a detection rate of 20.2%.The most common chromosomal disease is the Down syndrome syndrome (24 cases),followed by the chubby Willy and Angel syndrome(17 cases),while the Wolf-Hirschhorn syndrome in 5 cases,Williams syndrome in 5 cases and the Criduchat syndrone in 5 cases.The results of FISH were consistent with Array-CGH.Conclusion The technique of Array-CGH can be used to scan the whole genome of children with unknown disease.As a high-throughput and rapid research method,this technique has important clinical significance in the screening of chromosomal diseases.
