1.Therapeutic Effects Analysis of Combined Therapy on 40 Lubar Intervertebral Disc Protrusion Patients with Magnetic Resonance Imaging

Objective To observe the changes of combined therapy on disci intervertebrales and their surrounding tissues and thus to determine its therapeutic effects on lubar intervertebral disc protrusion with magnafic resonance imaging. Methods After combined therapy including physical agents, drugs, traction, manipulation and remedial exercises had been adopted to treat 40 iubax intervertebral disc protrusion patients for 30 days, morphologic changes of disci intervertebrales and their surrounding tissues of these patients were observed by magnetic resonance imaging. Results The effective rate of combined therapy on lubar intervertebral disc protrusion was 82.5%, there were no obvious changes of thickness, salient degree of disci intervertebrales, anteroposterior diameter, transverse diameter and latero-cryptae extent of vertebral canal after the treatment. Conclusion Though the combined therapy can not change the morphous of lobar intervertebral disc protrusion, it may efficiently relief the clinical symptoms of patients with lobar intervertebral disc protrusion.
3.An experimental study on the expression of heme oxygenase-2 mRNA in Hirschsprung's disease.

In order to investigate the relationship between the expression of heme oxygenase-2 (HO-2) mRNA and the pathogenesis of Hirschsprung's disease (HD), total ribonucleic acid (RNA) was extracted in the aganglionic and ganglionic segments of colon respectively from 15 cases of HD. The single-stranded cDNA of HO-2 was synthesized and further amplified by reverse transcription-polymerase chain reaction (RT-PCR). The expression of HO-2 mRNA was normal in ganglionic segments, but absent in aganglionic segments. It is concluded that the absence of HO-2 mRNA expression may be an important mechanism responsible for HD.

5.Changes of nucleus pulposus after in vitro culture of rabbit whole intervertebral disc and spinal motion segment.

OBJECTIVETo compare the changes of nucleus pulposus after in vitro culture of rabbit whole intervertebral disc and spinal motion segment.
METHODSTwenty-one New Zealand white rabbits which were randomly divided into organ group with 8 rabbits and segment group with 13 rabbits. Fifty intervertebral discs and 50 spinal motion segments were harvested respectively under aseptic conditions from two groups. These specimens were maintained in organ culture with hyperosmotic media (410 mOsm/kg), then 10 discs of the two groups were observed respectively by HE staining, immunohistochemistry of collagen type III, proteoglycan content and cells viability of nucleus pulposus before culture and at 3, 7, 14, 21 days after culture.
RESULTSHE staining showed the intervertebral disc tissue structure remained intact after culture of 21 days organ group and 14 days segment group,but there was severely degenerated of 21 days segment group. The intensity value of type II collagen immunohistochemical staining in the nucleus pulposus were not changed significantly between 21 days organ group and 14 days segment group (P > 0.05), but the staining of segment group at 21 days became shallower, there was significant difference compared with before each time points and organ group at 21 days (P < 0.05). PAS/AB staining of proteoglycan of nucleus pulposus showed that there were not decrease of tinting strength of two groups within 7 days, but the strength weakened slightly of two groups at 14 days, and the tinting strength became weaker at 21 days segment group, the change is more obvious than the organ group. The intensity value of fluorescence staining of nucleus pulposus cells was not changed significantly within 7 days of two groups (P > 0.05), the intensity value decreased slightly at 21 days organ group and 14 days segment group, but there were no significant difference compared with before time points (P > 0.05) however at 21 days segment group the intensity decreased as cells viability of nucleus pulposus decreased,and there was a significant difference compared with before each time points and organ group at 21 days (P < 0.05).
CONCLUSIONIt is not obviously degenerated of the discs of organ group cultured within 21 days and segment group cultured within 14 days, but there was significant degeneration of the intervertebral disc of segment group after cultured 21 days, so the rabbit spinal motion segment can be used on research about the biomechanics of intervertebral disc as a vitro experimental model within 14 days.

7.Biocompatibility of a new ureteral stent material

BACKGROUND:Studies have shown that polyethylene polymer-synthesized ureteral stents have good biocompatibility in vivo, but the side effects are found to induce the body to produce inflammatory, pyrogenic, and al ergic reactions. Therefore, studies have mainly aimed to explore the biocompatibility, toxic and side effects, inflammation of newly developed ureteral stents synthesized by L-lactic acid, glycolide, and barium sulfate. OBJECTIVE:Based on the biocompatibility comparisons on cel toxicity test, inflammatory reaction, al ergic reaction, pyrogen reaction and mortality after implantation between the usage of polyethylene material and L-lactic acid, glycolide, barium sulphate syntheses, to provide a better reference to introduce a new clinical ureteral stent material. METHODS:There were three groups in the experiment:blank control group, new material group, polyethylene group. Cel s were cultured in normal medium, leach liquid of L-lactic acid, glycolide, barium sulphate syntheses and leach liquid of polyethylene, respectively. RESULTS AND CONCLUSION:The toxic effects were significantly enhanced in the urethral epithelial cel s that grew in the leach liquids of two materials compared with the blank control group (P<0.05), but the two kinds of materials had no significant statistical difference in the toxicity test (P>0.05). Hematoxylin-eosin staining results showed that there were obvious inflammatory infiltrations in muscle tissue and the number of neutrophils and eosinophils were increased when the two kinds of materials were implanted in the rats for 2 and 6 weeks (P<0.05), but the inflammatory infiltration and tissue damage in the new material group was less than that in the polyethylene group (P<0.05). ELISA results showed that the serum concentrations of interleukin-10 and interleukin-23 were significantly increased when the two kinds of materials were implanted in the rats for 6 weeks (P<0.05), but the content of inflammatory cytokines in the new material group was less than that in the polyethylene group (P<0.05). In the new material group, there was only one rat that developed al ergic reaction and there was no pyrogen reaction or death;however, in the polyethylene group, there were three rats with presence of al ergic reaction and one rat with pyrogen reaction but no death. These findings indicate that the new ureteral stent material synthesized with L-lactic acid, glycolide and barium sulphate can reduce inflammation reactions, al ergic reactions and pyrogen reactions to the body, and has better biocompatibility than polyethylene composite material.
8.Optimum extraction of polysaccharides from ophiopogon japonicus by response surface methodology

Objective To optimize the extraction of polysaccharides from Ophiopogon japonicus by the use of response surface methodology.Method According to the design principle of Box-Behnken, effects of extraction temperature, extraction time , extraction times and solid-liquid ratio on the extraction yield of Ophiopogon japonicus polysaccharides were studied by response surface methodology,meanwhile regression analysis of these experimental data was used by Design-Expert 8.05b software.Results The optimum extraction conditions from Ophiopogon japonicus were concluded as follows: the extraction temperature was 93℃, extraction time was 2 h, extraction times was 3 times, and solid-liquid ratio was 1:15 g/ml.Conclusion Under these conditions, the extraction yield of Ophiopogon japonicus polysaccharides is up to 5.440%, and the relative error is 7.328%.
9.Risk factors for surgical site infection in patients undergoing cardiac surgery

Objective To analyze risk factors for surgical site infection(SSI) in cardiac surgery patients,so as to provide basis for making control measures.Methods 215 patients who underwent cardiac surgery in a hospital between January 2012 to December 2014 were investigated,univariate analysis and multivariate logistic regression analysis were used to analyze the related risk factors for postoperative SSI.Results The incidence of postoperative SSI in cardiac surgery patients was 16.28% (35/215).Univariate analysis showed that the incidence of postoperative SSI was relatively higher in patients aged>65 years,duration of operation>5 hours,continuous operation,preoperative hospitalization time >7 days,irrational perioperative antimicrobial use,as well as patients with underlying diseases such as hypertension and diabetes mellitus.Multivariate logistic regression analysis indicated that the age,duration of operation,continuous operation,preoperative hospitalization time,irrational perioperative use of antimicrobial agents,and patients with underlying diseases were independent risk factors for SSI in patients who underwent cardiac surgery.Conclusion Incidence of SSI is higher in patients with cardiac surgery,preventive measures should be taken to control risk factors such as duration of operation,so as to reduce the incidence of SSI.
10.Exploration on new role of ancient perilous but key acupoint of renying (ST 9).

The specific functions, stimulating methods and ranges of clinical application of Renying (ST 9) are explored. Because Renying (ST 9) is located in the neck which is a dangerous area, it is perilous if the acupoint is stimulated, so it is mostly forbidden in the past dynasties. In recent years, the main stimulating methods for Renying (ST 9) include acupuncture and press. The keys of acupuncture are to apply correct technique of needle insertion and control the depth and direction of needle. The keys of press are focused on strength and time of press. Renying (ST 9) is effective for vertigo, palpitation, asthma, hemiplegia, aphasia, hysteria, acute attack of pain, hyperplasia of mammary, continuous hiccup, disorder of throat and so on. As long as the acupoint location is accurate, manipulation is careful and stimulating method is correct, not only the safety could be guaranteed, but also the function can be well-played in clinical application.
