1.An ABCC8 gene mutation leading to the onset of KATP-congenital hyperinsulinism in a Chinese boy and his pedigree analysis

ABCC8,KCNJ11,and GLUD1 gene mutations were investigated in a male patient with congenital hyperinsulinism and his parents were also investigated.A 1484 G>A mutation was found in the 10th exon of ABCC8 gene in the patient,which leads to amino acid substitution at the 495 residue of the sulphonylurea receptor SUR1 protein.The patient's father also carried the same heterozygous inactive mutation,while the genotype of the mother was normal,indicating that the gene mutation of the patient was paternally inherited.According to that mutation,it is deduced that the patient may suffer from the focal type of congenital hyperinsulinism.
2.Management moved forward and process control of detail problem in disinfection supply room

Objective To establish the management moved forward mechanism of detail problem in disinfection supply room,so as to achieve the purpose of the process control.Methods Management moved forward mechanism was established,such as finding problem,recording detail problem,discussing details problem,and tracing back to the effect in disinfection supply room.Deviation situation,efficiency targets of job quality,clinic satisfaction were compared before and after the management mechanism implemented.Results Compared before and after the management moved forward mechanism implemented in disinfection supply room,decontamination area deviation were cases (78 vs 12),qualified cleaning were ( 175 vs 290 cases),satisfaction survey scores were (94.26 ± 1.22 vs 98.46 ± 1.67),the difference was statistically significance ( P < 0.05 ).Conclusions Management moved forward mechanism established in disinfection supply room can ensure the process coritrol on the details.
3.Observation on corrosion reaction and disinfection effect of two kinds of chlorine disinfectants to surgical instruments

Objective To observe the corrosion reaction and disinfection effect of the two different chlorine disinfectants toward the surgical instruments, to prevent the corrosion of instruments and the professional infection risk of cleaning staffs during washing the infectious disease-contaminated medical instruments manually in the primary hospitals. Methods The surgical instruments were soaked in the diluted chlorine effervescent tablets and the diluted chlorine disinfection powder, respectively. The corrosion of the two disinfectants to the equipment at different times was observed. The bloody surgical instruments were soaked in the diluted chlorine effervescent tablets and the chlorine disinfection powder for 4 hrs, respectively. The instruments' surfaces status was observed,and the bacterial reproduction status after soaking the instruments in the diluted liquid for 48 hrs was monitored.Results The diluted liquid of effervescent chlorine tablets had corrosion reaction after only 5- 10 minutes,but the diluted liquid of chlorine disinfection powder had no corrosion reaction after 48 hours. The dilution liquid of effervescent tablets and the dilution liquid of disinfection powder had a significant statistic difference in the rate of instruments corrosion. The bloody instruments' surface had difference after immersed for 4 hrs,and the pass rates of bacteria monitor was 100% after 48 hrs of immersion. Conclusions For the primary hospitals without cleansing and disinfection equipments,using the chlorine disinfection powder to soak the medical instruments before cleaning the infectious disease-contaminated medical instruments manually not only has no equipment corrosion but also achieves the disinfection effect.
4.Evaluation on the detail of quality control method during the implementation of the new CSSD guideline in district level hospitals

Objective To study the detail of quality control method during the implementing the new CSSD guideline in the district level hospitals. Methods The quality control was divided into 5 steps:Wrote the hospital' s CSSD quality system documents and comply them into a booklet. Set up reasonable quality control positions and staff and designed relative record files. Record the problems in the dynamic patrol handbook in time. Summary and discuss the problem by period. Made flow charts to improve the quality control details. All the above 5 steps formed the frame of the detail quality control. Results It had distinctly difference for inspection on quality efficiency ( Lumen clean, 175 vs 290, χ2 = 25.29, P < 0. 01 ), and thescores of nursing inspection of( satisfaction ,94.26 ± 1.22 vs 98.46 ± 1.67 ,t =42. 10,P <0. 05 ), after the detail quality control.Conclusions It can solve the anti-guideline problems, improve work quality and efficiency and low down cost for the implementation of the CSSD guideline of the district level hospital by detail quality control method.
5.Establishment and evaluation of method to collect rat gingival crevicular fluid by using absorbent paper points

Objective To establish and evaluate the method to collect the rat gingival crevicular fluid (GCF)by using absorbent paper points, and to lay foundation for analysis on GCF.Methods 20 healthy male rats were selected and randomly divided into GCF group and saliva group.The GCF of the right upper molar gingival trough of the rats in GCF group and the saliva of the rats in saliva group were collected by using 1 5# absorbent paper points.The SDS-PAGE analysis and abundance detection were applied to analyze the protein bands of the samples in two groups.Results The SDS-PAGE analysis identified the proteins at 77 000,66 000,55 000,51 000,and 28 000,especially 66 000 in GCF group.While saliva group had lower brightness protein bands at 66 000,60 000, and 48 000.The data of protein abundance of 66 000 between two groups had statistically significant difference (P<0.05).Conclusion The number and types of the protein bands are different between GCF Group and saliva group,so using 15# absorbent paper points can collect the rat GCF successfully.
6.Bcl-2 Antisense Oligodeoxynucleotides Increase Apoptosis of Lung Cancer Cells Induced by Radiation

Objective To study the effects of Bcl-2 antisense oligodeoxynucleotides(ASODN)on the apoptosis of lung cancer cells induced by radiation in vitro.Methods NCI-H446 lung cancer cell strains were divided into 5 groups:control simple radiation,lipofectin plus radiation,nonsense sqnence radiation and ASODN plus radiation.The cells cultured in five groups were collected at 6h,12h,24h,48h and 72h,with Wright-Giemsa stain,morphology analysis for which was done;the mRNA expression for p53、bcl-2 and PTEN gene was examined by RT-PCR half quantivity and DNA-ploid of the cells in five groups was detected by flow cyfometric method.Results Cell proliferation is obviously restrained and conformation is changed too with the shape crimpled and adherence function decreased obviously after irradiated for 10 Gy dose by the linac;p53 and PTEN expression clearly increased for the combination of Bcl-2 ASODN and bcl-2 mRNA expression clearly decreased.The apoptosis rate after 72 hours among control,pure radiation,lipofectin+radiation,nonsense+radiation and ASODN +radiation grouop is 0.14?0.09,13.17?2.47,11.84?1.76,13.72?1.4,21.26?2.97 respectively,the difference between ASODN combined with radiation grouop and other 4 groups are significant(P

10.Influence factors and quality control of bloodstatned surgical instruments clena effect

Objective To compare the effect of two kinds of soaking methods on bloodstained surgical insturments,and analyze influence factors for instruments clean effect,control key link,improve quality and clean surgical instruments washing efficiency,and to reduce the consumption.Methods Points on three analysis phase:analysis of the cause and gory surgical instruments clean effect.Factors validation phase:on the basis of the moisturizing method is verified.Improve phase:for key factors,establishing countermeasure.Results Oxidation rate of instruments in water group was 19.60%,and 2.30% in disinfection group,the difference between two groups was significant ( x2=26.04,P <0.01 ).Re-washing rate of oval forceps was 23.7% and consumption rate was 15.0% before quality control and those were 0.7% and 0 after quality control,the difference was significant ( x2=74.25,48.69,respectively; P < 0.01 ).Conclusions Sterilization supply &distribution control two keys could improve the quality and washing efficiency of surgical instruments,and reduce the consumption.
