1.Relationship between plasma TRAIL before 20 weeks′gestation and pregnancy induced hypertension

Objective To assess the relationship between maternal plasma tumor necrosis factor-related apoptosis-inducing ligand ( TRAIL ) before 20 weeks′gestation and hypertensive disorder complicating pregnancy (HDCP);and to evaluate the predictive value of plasma TRAIL for HDCP.Methods A 2-phase screening/validation study was designed.In the screening phase , a nested, case-controlled study was performed , the plasma samples collected before 20 weeks′gestation from 20 women who later developed HDCP and 20 age-and gestation week-matched controls were tested in prospective screening test for protein expression profiling during pregnancy and HDCP.Plasma samples were analyzed by a human protein microarray technology designed to detect 507 proteins simultaneously.Differently expressed proteins′functional annotation and clustering were performed by using of Database for Annotation , Visualization and Integrated Discovery ( DAVID) and Gene Ontology ( GO) database.The TRAIL level of plasma samples obtained before 20 weeks′gestation from 53 women who later developed HDCP and 106 similarly matched controls were further validated by ELISA and 62 clinical risk factors were investigated.Logistic regression and ROC analysis were used to evaluate the relationship between TRAIL and HDCP and its predictive value for HDCP.Results In protein microarray analysis , 23 proteins expressed differently before 20 weeks′gestation between the two groups.Further validation results showed that TRAIL levels in HDCP patients were lower significantly (45.7 ±13.1) pg/ml than those in healthy pregnant controls (51.2 ±14.7)pg/ml, P=0.021.Multiple factor logistic regression analysis of 159 pregnancies showed that three features were finally entering the logistic model, they were:anemia (OR=4.87, 95% CI 1.05-24.26), pre-pregnancy BMI (OR=1.72, 95% CI 1.35 -2.19) and TRAIL (OR=0.96, 95% CI 0.92 -0.99).The predictive accuracy of logistic model was 81.8%.The model significantly increases the predictive value (AUC=0.81, 95%CI 0.73-0.87) compared to TRAIL as independent predictor (AUC=0.59, 95%CI 0.51-0.67).Conclusions Totally 23 proteins were expressed differentially before 20 weeks′gestation in plasma of women who later developed HDCP , confirming that HDCP is a heterogeneous disease with different biological changes.The data suggests that plasma TRAIL levels relate with the development of HDCP and its combination with pre-pregnancy BMI and anemia have a high predictive value for HDCP before 20 weeks′gestation.

AIM To study the phenols from Plantaginis Semen.METHODS The 65% and 95% ethanol extracts of Plantaginis Semen were isolated and purified by macroporous resin,silica,ODS,Sephadex and preparative column,then the structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Nine compounds were isolated and identified as (+)-(7R,7'R,8S,8'S)-neo-olivil (1),erythro-guaiacylglycerol-β-ferulic acid ether (2),eriodictyol (3),luteolin (4),chrysoeriol (5),hydroxytyrosol (6),4-(3,4-dihydroxyphenyl)-(E)-3-buten-2-one (7),ferulic acid (8),5,7-dihydroxychromone (9).CONCLUSION Compounds 2-3,6-7 and 9 are isolated from genus Plantago for the first time,compound 5 is first obtained from this plant.
3.Auxiliary value of CT scanning in differentiating acute glioma stroke from simple cerebral hemorrhage

Objective To summarize the auxiliary value of CT scanning in differentiating acute glioma stroke from simple cerebral hemorrhage. Methods The CT data of 12 patients with acute glioma stroke diagnosed by operation and pathological examination were analyzed retrospectively.Results CT scan presented high density hematoma with clear borderline in 12 cases, solid tumor mass in 9 cases, no clear tumor foci in 3 cases, and perifocal edema in 10 cases. Conclusion The combination with the clinical history, CT manifestations and enhanced CT scan can facilitate the diagnosis of acute glioma stroke from simple cerebral hemorrhage.
4.Effects of Bushen Huoxue Recipe on the expressions of TGF-β1, TGF-βRⅡ and Smad2/3 in granulosa cells in mice with premature ovarian failure

AIM To explore the effects of Bushen Huoxue Recipe (Fluoritum,Psoraleae Fructus,Cuscutae Semen,etc.) on the protein expressions of transforming growth factor (TGF)-β1,TGF-βRⅡ and Smad2/3 in follicle wall granulosa cells in mice with autoimmune premature ovarian failure (POF).METHODS Balb/c female mice were subcutaneously injected with mouse zona pellucida 3 at multiple points to establish autoimmune POF model.POF mice were divided into model group,positive group (progynova),low-,middle-and high-dose of Bushen Huoxue Recipe groups.After intervention for 30 days,ovarian tissue was stained by hematoxylin-eosin (HE),and the protein expressions of TGF-β1,TGF-βRⅡ and Smad2/3 in follicle wall granulosa cells and ovarian tissue were detected by immunohistochemistry and Western blot,respectively.RESULTS Compared with the model group,the number of mature follicles in the Bushen Huoxue Recipe groups and the positive group was obviously increased,together with the decreased number of atretic follicles.The protein expressions of TGF-β1,TGF-βRⅡ and Smad2/3 in follicle wall granulosa cells and ovarian tissue in the Bushen Huoxue Recipe groups and the positive group were much higher than those in the model group (P < 0.05).CONCLUSION Bushen Huoxue Recipe can improve ovarian function by up-regulating the protein expressions of TGF-β1,TGF-βRⅡ and Smad2/3 in granulosa cells.
5.Evaluation of Ion Release from Four Dental Sealants

The purpose of this study was to determine the ion release from four commercially available pit-and-fissure sealants [3M Clinpro,3M ConciseTM,BeautiSealant (BS),and GI FX-Ⅱ)].With each brand,18 specimens were prepared.Their fluoride release in de-ionized water was measured by fluoride electrode,while the release of silicate (Si),aluminum (A1),sodium (Na),calcium (Ca),strontium (Sr),and phosphorus (P) was measured by inductively coupled plasma atomic emission spectroscopy (ICP-AES) on days 1,3,7,14,21,and 28.The result showed that fluoride was not released from 3M ConciseTM.GI FX-Ⅱ displayed the largest fluoride release,which,however,dropped rapidly on day 3.3M Clinpro exhibited less fluoride release than GI FX-Ⅱ and BS did.At any time during the 28-day experimental period,GI FX-Ⅱ released more Na than the other sealants (P<0.001).BS ranked the second in Na release,and a small amount of Na ions was released from the 3M Clinpro and 3M ConciseTM samples.A1 ions were only detected from BS and GI FX-Ⅱ,but not from the 3M Clinpro and 3M ConciseTM.Additionally GI FX-Ⅱ had the largest Si release among the four brands at any time during the experimental period (P<0.001).Ca ions were detected from 3M Clinpro and 3M ConciseTM,but not from GI FX-Ⅱ.BS released more Sr than the other sealants at any time during the experimental period (P<0.001).All the samples released similar amounts of P continuously during these 28 days.In conclusion,based on the type and the amount of ion release,BS is the best pit-and-fissure sealant among the four brands.
6.To re-evaluate the clinical classification criteria of liver failure.

OBJECTIVETo study the clinical feature and more reasonable diagnostic typing criteria for patients with liver failure.
METHODS13/21 cases of ALF, SALF with no past liver disease, 49/72 cases of with chronic hepatitis, and 23/73 cases ALF, SALF with liver cirrhosis, were analyzed respectively.
RESULTS1 ALF patients (1). There exist significant statistic differences in ALB, ALT, CHE in three ALF groups.(2). It had statistic differences in those patients with hepatic encephalopathy.(3). The prognosis of the patients with chronic hepatitis group (42.85 percent) was best than that of chronic cirrhosis (26.09 percent) and no past liver disease (15.38 percent). (2) In SALF patients (1). There exist significant statistic differences in ALB, GLO, ALT, AST, BDIL, GLU and CHE in three SALF groups.(2). It had statistic differences in those patients with hepatic encephalopathy in three SALF groups.(3). The prognosis of the patients with chronic hepatitis group (51.39 percent) was best than that of chronic cirrhosis (36.85 percent) and no past liver disease (33.33 percent).
CONCLUSIONThere are different clinic feature and prognosis in three ALF or SALF groups, so we suggest that it were clinic practicability and science in classify of liver failure at present.

7.Nosocomial infection in extremely preterm infants

Objective:To study risk factors and clinical outcomes of nosocomial infection (NI) in extremely premature infants (EPIs).Methods:From January 2017 to December 2021, EPIs admitted to NICU of our hospital were retrospectively studied. The infants were assigned into NI group and non-NI group. The NI group were further assigned into survival group and death group. Single factor analysis was conducted using chi-square test, t-test and non-parametric test. Multiple logistic regression was used to analyze risk factors of NI and death.Results:A total of 115 cases were enrolled, including 67 (58.3%) in NI group, 54 in survival group and 12 in death group. One case gave up treatment. 48 cases were in non-NI group. 81 episodes of nosocomial infections occurred in NI group and the case infection rate was 70.4% (81/115). 48 cases (41.7%) had late-onset sepsis, 14 (12.2%) neonatal necrotizing enterocolitis, 13 (11.3%) pneumonia, 1 (0.9%) urinary tract infection and 1 (0.9%) thrush. Hemodynamic significant patent ductus arteriosus (hsPDA) and long duration of parenteral nutrition were independent risk factors for NI in EPIs ( P<0.05). The incidence of bronchopulmonary dysplasia (BPD) (100.0% vs. 70.8%) and mortality (17.9% vs. 2.1%) in NI group was higher than non-NI group ( P<0.05). Septic shock was an independent risk factor for the death due to NI. Conclusions:The incidence of NI in EPIs is high. hsPDA and long duration of parenteral nutrition are independent risk factors for NI in EPIs. EPIs with NI have high incidence of BPD and mortality and septic shock is an independent risk factor for death.
8.Application of Maslow's need hierarchy theory in postoperative nursing of patients with cervical spondylopathy

Objective To observe the effect of Maslow' s need hierarchy theory on the postoperative care of patients with cervical spondylosis operation.Methods Totals of 70 cases of cervical spondylosis of the intended line operation were divided into the experimental group and the control group,with 35 cases in each group.Control group received routine nursing model,while experimental group received nursing model implementation of care and related health education according to the Maslow' s need hierarchy theory.After implementation,the treatment and nursing effect of two groups was compared.Results In experimental group,the postoperative hospital stay [(6.2 ± 1.1) d vs (6.9 ± 1.5) d] and the incidence of anxiety (11.43% vs 31.43%) were significandy lower than that in control group (t =2.226,x2 =4.16;P < 0.05).And in experimental group,functional exercise knowledge [(96.3 ± 1.6) % vs (89.8 ± 2.7) %] and nursing job satisfaction [(96.7 ± 1.5) % vs (94.1 ± 2.8) %] was significantly higher than that in control group,and the difference was statistically significant(t =12.253,4.842 ; P < 0.01).Conclusions Maslow' s need hierarchy theory as a guide to carry out treatment and care can improve the quality of medical care and nursing satisfaction,with the benefit of modern nursing development,is a suitable good nursing mode for cervical spondylosis operation patients.
9.Significance of high-risk human papillomavirus DNA in atypical squamous cells, cannot exclude high-grade squamous intraepithelial lesion.

OBJECTIVETo explore significance of high-risk human papillomavirus (HR-HPV) testing in atypical squamous cells, cannot exclude high grade squamous intraepithelial lesion (ASC-H).
METHODSPresence of HR-HPV DNA was examined in 45 patients with ASC-H using hybrid capture II (HC-II) test. Colposcopic examination and biopsy were taken all results were evaluated.
RESULTSOverall, 33 of 45 (73.3%) ASC-H cases were biopsy proven cervical intraepithelial lesion (CIN). 36 of 45 ASC-H cases were HPV-DNA positive, including 19 cases of HSIL and over lesion; whereas no HSIL or over was found in 9 HR-HPV negative cases. Sensitivity and negativity predictive value of HR-HPV in ASC-H with HSIL and over lesion were both 100%.
CONCLUSIONSASC-H strongly predicts the presence of HSIL, HR-HPV may serve as a predict select whether a patient with ASC-H should take colposcopic examination immediately, patients with positive HR-HPV should undergo immediate colposcopic examination, while negative HR-HPV is an excellent predictor of the absence of HSIL.

10.Expression of adenosine receptors in human retinal pigment epithelium cells in vitro.

BACKGROUNDAdenosine receptors (ADORs) have been reported to play a role in experimental myopia. This study aimed to determine the distribution of ADORs in human retinal pigment epithelium (RPE) cells cultured in vitro.
METHODSHuman RPE cells (cell line D407) were cultured in vitro. ADOR mRNA in RPE was detected by reverse transcription polymerase chain reaction. ADOR protein expression in RPE was confirmed by Western blotting analysis of cell lysates. Confocal fluorescence microscopy was used to study the subcellular distribution of ADORs.
RESULTSAll four subtypes of ADORs mRNA and protein were expressed in human RPE. This was confirmed by Western blotting analysis. The ADOR subtypes were differently distributed within the cells. ADORA1 was expressed in nucleus, perinucleus and cytoplasm of RPE. ADORA2A was concentrated mainly in one side of the perinucleus and cytoplasm of RPE. ADORA2B was strongly expressed in the nucleus, perinucleus and the cytoplasm, and ADORA3 was expressed weakly in the cytoplasm of RPE.
CONCLUSIONSADORs are expressed in human RPE. The different distribution at the subcellular level suggests different functions of ADOR subtypes.
