Recently,increasing cancer researches focus on antibody-drug conjugates(ADCs) which can improve the anti-tumor potency with less adverse effect while benefiting patients in the future. However,safety evaluation of ADCs is a big challenge because of complex components as well as in experience in preclinical studies. In this review,the authors reviewed the mode of action,hazard risks,and toxicity observed in preclinical/clinical studies of ADCs,summarized the preclinical studies of Adcetris(brentuximab vedotin)and Kadcyla(ado-trastuzumab emtansine),and suggested a better strategy of ADCs in preclinical safety evaluation.
6.Study on Intervention of Serum 3-deoxyglucosone byBu-Shen Hua-TanFang in Rats with Diabetes Mellitus and Cognitive Impairment

This paper was aimed to study the influence ofBu-Shen Hua-TanFang (BSHTF) on the learning and memory function of diabetes mellitus (DM) rats. A total of 50 3-month-old male Wistar rats were randomlydivided into the control group, model group and 3 BSHTF groups (i.e., high-, middle-, and low-dose). A total of 35 mg·kg-1 streptozotocin (STZ) was intraperitoneally injected into the model and BSHT group. The same volume of citric acid-sodium citrate buffer solution was intraperitoneally injected into the control group. The intragastric administration of BSHTF at the dose of 20, 10, and 5 g·kg-1 was given to the BSHTF groups, respectively. The behavior and content of serum 3-deoxyglucosone (3-DG) were detected among DM rats after 30 days. The results showed that the levels of serum glucose in the model group and the BSHTF groups were obviously higher than that of the control group (P < 0.01). The swimming time to platform of rats in the model group was obviously prolonged compared to the control group with significant difference from the second day on (P < 0.01). The required time in different BSHTF intervention groups was obviously decreased compared with the model group from the 4th day (P< 0.05). The level of serum 3-DG in rats of the model group was obviously higher than that of the control group (P < 0.01). The level of serum 3-DG in rats of the middle-dose BSHTF group was obviously decreased compared with the model group (P < 0.05). It was concluded that BSHTF was able to improve the cognitive impairment among DM rats by reducing the levels of serum glucose and serum 3-DG.
7.Studies on Pharmacokinetics and Bioequivalence of Valaciclovir Hydrochloride Tablets

OBJECTIVE: To study pharmacokinetics of valaciclovir hydrochloride tablets and to evaluate the bioequivalence of test and reference tablets. METHODS: In a two-period cross-over design test, 18 healthy volunteers were given a single oral dose of 600 mg test valaciclovir hydrochloride and 600 mg reference formulations respectively. Plasma concentrations of acyclovir were determined by HPLC method. The pharmacokinetic parameters of the two formulations were calculated and analyzed statistically. RESULTS: The main pharmacokinetic parameters of test vs. reference valacyclovir hydrochloride tablets were as follows:tmax(1.69?0.25)h vs. (1.72?0.26)h, Cmax(3.34?0.58) ?g?mL-1 vs. (3.40?0.49)?g?mL-1, t1/2(2.73?0.31)h vs. (2.97?0.33)h, AUC0~14(11.22?2.21) ?g?h?mL-1 vs. (11.12?1.90)?g?h?mL-1, AUC0~∞(11.76?2.15) ?g?h?mL-1 vs. (11.61?1.86)?g?h?mL-1, respectively. As compared with reference formulation, the relative bioavailability of test formulation was (101.06?11.72)%. CONCLUSION: The two preparations are bioequivalent.
9.Effect of Shexiang Baoxin pill on coronary vasodilation by analysis of coronary angiography.

OBJECTIVETo evaluate the effect of Shexiang Baoxin Pill (SBP) on coronary vasodilation by analysis of coronary angiography (CAG).
METHODSA consecutive cohort of 300 patients who underwent CAG between January 2013 and July 2013 were recruited and randomly assigned to 2 groups before operation. Patients in the SBP group sublingually took SBP, while those in the control group sublingually took placebos. All patients repeatedly underwent CAG 5 min after administration. The vascular diameter was calculated by quantitative angiography analysis method. The diameter of the left anterior descending coronary artery was measured in patients whose coronary arteries had no stenosis. The narrowest vascular diameter was measured in patients whose coronary arteries had stenosis. The heart rate, blood pressure, and the vascular diameter were compared between before and after administration in the two groups.
RESULTSIn the two groups, there was no significant difference in changes of heart rate, systolic pressure, or diastolic pressure between before and after administration (all P > 0.05). There were 64 patients with normal CAG in the two groups, 30 in the control group and 34 in the SBP group. CAG showed there were 236 patients with stenotic coronary artery, 110 in the control group and 126 in the SBP group. The vascular diameter was obviously larger in patients in the SBP group with normal or abnormal CAG after administration (all P < 0.01). It was also obviously larger than that of the control group after administration (P < 0.05, P < 0.01).
CONCLUSIONSBP could dilate both normal coronary artery and lesioned coronary arteries, but did not lead to fastened heart rate and decreased blood pressure.

10.Meta-analysis about time of indwelling urinary catheter after and before general anesthesia

Objective In order to know the influence of different time of indwelling urinary catheteron postoperative comfortation of patients with general anesthesia. Methods Six ease-control studies re-lated the influence of different time of indwelling urinary catheter on postoperative eomfortation of patientswith general anesthesia had analyzed by Meta-analysis method. Results According to the results ofMeta-analysis from the 6 documents, the postoperative comfortable level of patients with indwelling urinarycatheter before general anesthesia was better than patients with indwelling urinary catheter after generalanesthesia, OR=0.12(0.05~0.30). Conclusions Indwelling urinary catheter before general anesthesia can prevent postoperative uncomfortation, which is a kind of proper method.
