1.Determination of Thimerosal of Chondroitin Sulfate Eye Drops in Different Packing Materials by Volt-am-pere Polarography Method

Objective:To establish a volt-ampere polarography method for the determination of thimerosal in chondroitin sulfate eye drops, and study the adsorption of thimerosal in different packing materials. Methods:The hanging mercury drop electrode ( HM-DE) in multi-functional electrode was used and the sample solution was added to potassium chloride-hydrochloric acid buffer solution. Nitrogen deoxygenation time was 200 s, the enrichment time was 60 s, and the equilibrium time was 5 s. The potentiometric scan was performed using a two standard addition method. Results:Thimerosal had a good linear relationship within the range of 0. 01-1. 0 mg· ml-1(r=0. 9990). The detection limit was 1. 78 μg·ml-1 and the average recovery was 99. 1% (RSD=2. 7%, n=9). Conclu-sion:The method is simple and accurate, and suitable for the determination of thimerosal in chondroitin sulfate eye drops. Low-density polyethylene and polypropylene both show obvious adsorption of thimerosal,so chondroitin sulfate eye drops should not be packed in the above packing materials.

3.Analysis on staphylococcus xylosus inducing postoperative nosocomial infection

Objective To investigate the pathogens inducing a posto perative nosocomial infection. Methods Specimens was collected fro m exudates or air for bacteria culture and identification. Re sults The postoperative infection was induced by staphylococcus x ylosus. Conclusions The relevant factors affecting the po stoperative nosocomial infection include incomplete sterilization of operative r oom and operative tools. Thus strict control measures must be put into effect.
5.Effect of pseudolaric acid B on proliferation of human LiBr cell line in vitro

Object To study the inhibitory effects and mechanism of pseudolaric acid B (PAB) on the growth of LiBr cell line. Methods PAB in different concentrations was added into the medium in which LiBr cells were cultured. The changes of cell morphology were observed by phase contrast microscope. Cytotoxicity of PAB was detected by MTT. The number of apoptotic cell was calculated by double fluorescent staining. The level of p21 WAF1 protein was measured with immunocytochemistry. Results The growth of LiBr cell line was remarkably inhibited by PAB. The IC 50 values of PAB for LiBr cells was 2.5?10 -5 mol/L. The inhibitory rate and the apoptotic cell rate were related with the medicine concentration. The level of p21 WAF1 protein in LiBr cells treated with PAB was obviously increased. Conclusion PAB can effectively inhibit the proliferation of LiBr cell line.
6. Isolation and preparation of leonurine from Leonurus japonicus by high speed countercurrent chromatography

Objective: To establish a simple and effective method for the isolation of leonurine from Leonurus japonicus by high speed countercurrent chromatography (HSCCC). Methods: The extraction conditions of leonurine were optimized by one-factor experimental design. After comparing several different solvent systems, the two phase system of ethyl acetate-n-butanol-water (3∶2∶5) was finally chosen as operating solvent of HSCCC for the separation of leonurine, in which the lower phase was determined as the mobile phase and the upper phase as stationary. The detection of eluates was performed with an ultraviolet detector at 277 nm. The rotation speed was adjusted at 850 r/min, and the flow rate was 2.2 mL/min. Results: Leonurine was successively isolated from n-butanol fraction by HSCCC, and the above established method was also successively applied to the crude extact of L. japonicus. Finally, 68 mg of leonurine with purity about 96.2% could be obtained from 2.48 g crude extract of L. japonicus in a single injection. Conclusion: The described approaches actively promote efficient preparation strategy to obtain high purity of leonurine.
7.A auditory-P300 research on male heroin addicts in convalescence and drug craving state

Objective To explore the auditory P300 potentials in rehabilitated male patient of heroin addicts (MPHD),as well as the change of P300 amplitude in the drug craving state.Methods Rehabilitated MPHD patients (n =20) and healthy controls (n =20) underwent standard auditory P300 test.Only the patient group completed auditory P300 test after viewing neutral video and drug craving video respectively.Results Rehabilitated MPHD patients showed reduction of P300 amplitude (Cz:(2.72 ± 1.90) μV vs (3.99 ± 1.74) μV ; Pz:(4.48 ± 1.87) μV vs (5.61 ± 1.39) μV ; P < 0.05) than that of healthy controls in standard auditory P300 test.Patients who had been successfully induced by drug craving video demonstrated higher P300 amplitude (Pz:(5.00± 1.68) μV vs (4.19 ± 1.62) μV; P<0.05) after viewing drug craving video than that of viewing neutral video.Conclusion MPHD patients have cognitive impairment in their rehabilitation period;and P300 index can partially reflect the attention change in patients' drug craving state.
8.Effects of acupuncture plus language training on language function and cerebral blood flow in patients with motor aphasia after ischemic stroke

Objective: To observe the effects of acupuncture at the Governor Vessel acupoints plus language training on the language function and cerebral blood flow in patients with motor aphasia after ischemic stroke. Methods: Eighty-six patients were randomized into a control group and an observation group, with 43 cases in each group. Conventional symptomatic treatment was offered to both groups. Besides, the control group received language training, while the observation group received language training plus additional acupuncture at the Governor Vessel acupoints. Before and after treatment, the aphasia battery of Chinese (ABC) and Chinese functional communication profile (CFCP) were tested, and the mean velocity (Vm) and resistance index (RI) of the left middle cerebral artery (MCA) were detected. Results: The total effective rate was higher in the observation group than in the control group (P<0.05). After treatment, the observation group gained higher scores in oral expression, comprehension, repeating, naming objects, reading, and writing, as well as the general score of ABC (all P<0.05), higher than those in the control group (all P<0.05). The CFCP score increased in both groups after intervention, showing significant intra-group differences (both P<0.05), and the CFCP score was higher in the observation group than in the control group (P<0.05). After treatment, Vm of the left side MCA increased in the control group (P<0.05), while no significant change was observed in RI (P>0.05); in the observation group, Vm of the left side MCA increased and RI decreased significantly compared with the baseline (both P<0.05), and were markedly different from those in the control group (both P<0.05). Conclusion: Acupuncture at the Governor Vessel acupoints plus language training can produce valid efficacy in treating motor aphasia after ischemic stroke; it can notably improve the language function, everyday oral communication ability, and increase cerebral perfusion of the patients.
9.Influence of rehabilitation training on heart rate of stroke patients in the early stage

ObjectiveTo observe the influence of rehabilitation training on heart rate of stroke patients in the early stage.MethodsThe heart rate (HR) of 30 patients within one week after the onset of stroke was evaluated by FUKUDA DS 880A teleelectrocardiograph and the rating of perceived exertion (RPE) was collected during practicing bridging, rolling and moving the arms.ResultsThe HR of 30 patients was only increased 7.57, 6.23 and 6.57 beats per minute during practicing bridging, rolling and moving the arms. RPE of all patients was less than 11.ConclusionAppropriate rehabilitation training is safety and unable to increase the loading of heart in the early stage after the onset of stroke.
10.Granulosa cells model transfected by ENO1 overexpression adenovirus vector and the effect of ENO1 overexpression on progesterone secretion.

OBJECTIVETo construct primary cultured granulosa cells model of Zi Gooses tansfected by alpha-enolase (ENO1) overexpression adenovirus vector, and to detect the effect of ENO1 overexpression of granulose cells on progesterone secretion.
METHODSGranulosa cells were infected with Ad-CMV-ENO1 in gradient multiplicity of infection(MOI) levels:100, 250, 350 and 400 pfu/cell. Twenty four hours and 48 h after infection, green fluorescent protein (GDP) was respectively detected by fluorescence inverted microscopy. The effect of ENO1 overexpression of granulose cells on progesterone secretion was detected by the step double antibody sandwich enzyme-linked immunosorbent assay (ELISA).
RESULTSThe optimal infection rate (100%) was achieved when MOI was 800 pfu/cell,48h after infection. Real time RT-PCR and Western blot showed that the level of mRNA and protein expression ENO1 were increased significantly after infection (P < 0.01); The granulosa cells progesterone secretion of Ad-CMV-ENO1 group increased signigicantly (P < 0.01).
CONCLUSIONENO1 overexpression could make the primary culture follicle granulosa cells in vitro improve progesterone secretion.
