1.Expression of Bax and Bcl-2 gene in rat testis tissues induced by testicular denervation.

OBJECTIVETo detect the changes of the expression of Bax and Bcl-2 gene in the denervated testis, and to explore the possible mechanisms underlying the apoptosis of germ cells induced by testicular denervation at the genetic translation level.
METHODSEighteen mature SD rats (350-375 g) were equally divided into 3 groups: a sham operation group( SO) , a superior spermatic nerve group (SSN) and an inferior spermatic nerve group (ISN) , and the latter two received bilateral surgical removal of the superior spermatic nerve and the inferior spermatic nerve, respectively. The animals were killed I month after the operation. ISH SP-method was used to detect the expression of Bax and Bcl-2 protein.
RESULTSSignificant up-regulation of Bax protein was detected in both the treatment groups 1 month after surgery( P <0. 05) , and the level of Bcl-2 protein remained unchanged.
CONCLUSIONBax gene is involved in the apoptosis of germ cells induced by testicular denervation.

2.Effect of anti-CD49d monoclonal antibody on mobilization of CD34 positive cell into peripheral blood in mice.

To study the mobilization of peripheral blood stem cells (PBSC) with anti-CD49d monoclonal antibody and try to find a new method for mobilization of PBSC, anti-CD49d McAb, rhG-CSF and combination of anti-CD49d McAb with rhG-CSF were administered subcutaneously to mice separately, the count of white blood cells (WBC) and percentage of CD34(+) cells in peripheral blood of donor mice were dynamically observed, CD34 positive cells obtained by above methods were transfused to recipient mice. The results showed that the count of WBC and percentage of CD34(+) cells in peripheral blood of donor mice elevated significantly after the administration of anti-CD49d McAb, rhG-CSF or combination of anti-CD49d McAb with rhG-CSF. The most effective method for mobilization is the combination of rhG-CSF with anti-CD49d McAb. Reconstitution of hematopoiesis was successful in all group recipient mice after transplantation. Most rapid hematopoietic recovery was observed in recipient mice by rhG-CSF plus anti-CD49d McAb for mabilization. In conclusion, anti-CD49d McAb is effective and synergistic with rhG-CSF in mobilization of CD34 positive cells from bone marrow into peripheral blood.

3.Genetic Polymorphisms of 17 Y-STR Loci in Nantong Han Population

Objective To analyze the genetic phenotypes of Nantong Han population and evaluate the application value of 17 Y-STR loci in Nantong population.Methods The peripheral blood samples were collected from 343 unrelated Nantong Han males and the genomic DNA were extracted by Chelex-100 method.Genotyping was performed using the AmpF(l)STR YfilerTM Kit.The results were compared with other 12 Han populations,including Anhui,Jiangsu,Jiangxi,Shandong,Shanghai,Zhejiang (1),Lanzhou,Nanyang,Luzhou,Mudanjiang,Shanxi and Zhejiang (2),and 9 minority populations (Mongol,Xibe,Tibetan in Lhasa,Tibetan in Qinghai,Kazak,Uighur,Manchu,Paiwan in Taiwan and Tujia).Results A total of 327 different haplotypes were found in 17 Y-STR loci in Nantong Han population.The haplotype diversity (HD) was 0.999 7.The R,value between Nantong Han and other Chinese populations ranged from-0.000 6 to 0.263 5.The multidimensional scaling results showed that Nantong Han population had no significant differences between most of the Han populations,but had significant differences between most of Chinese minority populations.Conclusion Seventeen Y-STR loci can be a powerful tool for forensic application because of its high polymorphism in Nantong Han population.
4.Blood component quality sampling: the use of characteristic curve scheme

【Objective】 To explore the optimal sampling scheme for functional indexes of blood components, so as to ensure the accuracy of sampling. 【Methods】 The operation characteristic curves of 5 sampling schemes as (4, 1), (6, 1), (8, 2), (10, 2) and (12, 3) were drawn based on the characteristic curve theory of sampling inspection, the discriminability of those 5 sampling schemes was compared, and the risk of misjudgment and omission of sampling schemes was analyzed. 【Results】 The discriminability of 5 sampling schemes from high to low were (10, 2), (6, 1), (12, 3), (8, 2) and (4, 1), ; the operation characteristic curves of scheme (10, 2) were in basic concordance with (6, 1), (12, 3) and (8, 2) as the percent defective was <0.25. The risk of misjudgment of the 5 sampling schemes was low, and the risk of omission was high in scheme (4, 1)(0.738, 3), and low in (10, 2) and (6, 1) (0.525 6 and 0.533 9, respectively). 【Conclusion】 On the premise of strictly controlling the process of blood collection and supply and ensuring the quality of blood preparations, the sampling scheme (4, 1) can effectively monitor the stability of the whole process. However, for blood preparations with high nonconforming rate, sampling scheme (6, 1) or (10, 2) was recommended as the sampling amount should be elevated appropriately.
5.Clinical Significance of Serum Midkine in Children with Henoch-Schonlein Purpura.

OBJECTIVETo investigate the clinical significance of serum midkine (MK) in children with henoch-schnlein purpura(HSP).
METHODSOne hundred and six children with HSP admitted in our hospital from March 2013 to March 2016 were enrolled in HSP group, but then 80 healthy volunteers were used as control. MK, interleukin-2 (IL-2), IL-4, IL-6, IL-10, tumor necrosis factor (TNF), interferon γ (IFN-γ) and IL-17 in peripheral blood were measured by ELISA, biochemical indicators including white blood cell count (WBC), platelet (Plt), C-reactive protein (CRP), erythrocyte sedimentation rate (ESR), D-dimer, immunoglobulin A (IgA), IgE, IgG, IgM and other clinical data were recorded and analyzed.
RESULTSAmong 106 cases of henoch-schonlein purpura, 42 patients combined with renal failure (isolated hematuria in 4 cases, 17 cases of isolated proteinuria, hematuria and proteinuria in 21 cases). Midkine level in children with HSP was significantly higher than that in the control group [291.70(248.50-396.41) pg/ml vs 217.30(198.98-243.65) pg/ml)](P<0.05); the MK level in group of HSP with nephritis was higher than that in group of HSP without nephritis [326.58(266.58-459.25) pg/ml vs 280.72(233.67-384.36) pg/ml] (P<0.05). IL-4, IL-6, IL-17, TNF and IFN-γ levels in children with HSP were higher than those in the control group(P<0.05), but IL-10 level was lower than that in control group. IL-2 level was not significantly different between 2 groups. Cytokines levels in HSP nephritis group and HSP without renal involvement were not significantly different. Pearson correlation analysis showed that midkine level positively correlated with IL-4, IL-6, IL-17, IgA and IgE (P<0.05). By ROC curve analysis, the AUC of midkine was 0.902, 95% CI 0.841-0.963 (P<0.001), threshold 295.50 pg/ml. The sensitivity and specificity of midkine for predicting Henoch-Schonlein purpura nephritis were 80.60% and 88.30% respecitvely.
CONCLUSIONPlasma MK plays an important role in the development of Henoch-Schonlein purpura and Henoch-Schonlein purpura nephritis. Plasma MK can be used as an effective indicator for early diagnosis and prediction of HSP and renal damage.
6.Relationship between levels of Annexin A1, lipoprotein-associated phospholipase A2 and Bax and human carotid atherosclerotic plaques

Objective To investigate the correlation between levels of Annexin A1,Bax and lipoprotein-associated phospholipase A2 (Lp-PLA2) and both formation and stability of human carotid atherosclerotic plaques.Methods Forty-five specimens from patients with carotid atherosclerotic plaques (test group,group A),admitted to our hospital from May 2010 to June 2011 and performed carotid endarterectomy (CEA),and 20 specimens from patients with mesenteric artery and 20 healthy subjects (control groups,group B and group C,respectively) were collected in our study.The carotid atherosclerotic plaque specimens were divided into soft plaque group A1 (n=15),mixed plaque group A2 (n=15) and hard plaque group A3 (n=15) according to the results of carotid artery ultrasound.The Lp-PLA2 level in group A and group C was measured by enzyme linked immunosorbent assay (ELISA),and the mRNA and protein expressions of Annexin A1 and Bax in group A and group B were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting.Results The level of Lp-PLA2 in all patients with carotid atherosclerotic plaques was significantly increased as compared with that in group C (P<0.05); that in group A1 was significantly higher than that in group A3 (P<0.05).The mRNA and protein expressions of Annexin A1 and Bax in all patients with carotid atherosclerotic plaques were significantly increased as compared with those in group B (P<0.05); group A1 exhibited lower mRNA and protein expressions of Annexin A1 than group A3 (P<0.05),while group A1 exhibited higher mRNA and protein expressions of Bax than group A3 (P<0.05).Conclusion Annexin A1,Lp-PLA2 and Bax participate in the formation and stability of human carotid atherosclerotic plaques.
7.Role of complement C3 in early predicting pregnancy outcomes of frozen-thawed embryo transfer

Objective To investigate the role of complement C3 in early predicting pregnancy outcomes of frozen-thawed embryo transfer(F-ET).Methods A total of 378 F-ET cycles were prospectively collected and divided into group A(complement C3≤1.05,120 cycles)and group B(complement C3>1.05,258 cycles)based on the best cutoff value of complement C3 for predicting F-ET pregnancy outcomes.The outcomes of the two groups were compared,and the best cutoff value of complement C3 for predicting F-ET spontaneous abortion was analyzed in group B.Results Age was a risk factor for successful F-ET pregnancy(P<0.05),and complement C3 and embryo type were protective factors for successful F-ET pregnancy(P<0.05).The area under the receiver-operating characteristic curve(ROC)of complement C3 for predicting F-ET pregnancy outcome was 0.702,and the best cutoff value was 1.05 g/L,with a clinical pregnancy sensitivity of 87.60%and a specificity of 52.00%.The clinical pregnancy rate and embryo implantation rate in group B were both significantly higher than those in group A(67.05%vs.52.75%,P<0.05).The best cutoff value of complement C3 for predicting spontaneous abortion after F-ET was 1.32 g/L,with an area under the ROC curve of 0.760,a sensitivity of 69.00%,and a specificity of 81.20%.Conclusions Complement C3 is of significance in the early prediction of F-ET pregnancy outcome.When complement C3 exceeds the level of 1.32 g/L,it may lead to an increase in the rate of spontaneous abortion.
8.Analysis of heart rate variability during blood donation

【Objective】 To observe the regulation of autonomic nerves in blood donors during blood donation by heart rate variability analysis and explore the possible mechanism of donation related vasovagal reaction. 【Methods】 Electrocardiogram (ECG) of 90 blood donors was monitored by Fontaine Ⅰlead during the whole process of blood donation, and the 5-min heart rate variation before, during and after blood donation was analyzed. 【Results】 During the whole process of blood donation, the sympathetic HRV index (LF nu) and the sympathetic and vagal balance ability index (LF/HF) increased, whereas the vagal nerve index (pNN50, RMSSD, HFnu) and heart rate variability index (SDNN, Total power) decreased. For baseline heart rate variability of different blood donors (first-time vs. repeated, male vs. female, 18-24 years old vs. ≥25 years old, <400 mL vs. 400 mL) before blood donation, the pNN50, RMSSD and Total power of 18-24 years old blood donors were higher, but other indicators showed no significant difference. There were differences in HRV indexes of different types of blood donors during blood donation compared with before blood donation. The decrease of pNN50 and HFnu and the increase of LF/HF were larger in experienced blood donors than in first-time blood donors. The decrease of RMSSD was larger in male blood donors than in female blood donors; the change of LF/HF was larger in blood donors aged≥25 years than in blood donors aged 18-24 years; other indicators had no significant difference. 【Conclusion】 Blood donation leads to reflex readjustment of the cardiac autonomic tone: the sympathetic nerve is excited while the vagal nerve is suppressed. The cardiac autonomic nerve function of first-time blood donors, female donors and low-age (18-24 years old) donors to blood donation stress is not fully regulated. Donation related vasovagal reaction may be related to the autonomic nerve regulation function of blood donors.
