1.Improved methods of establishing rabbit VX2 hepatocarcinoma models
Chinese Journal of Tissue Engineering Research 2015;(27):4361-4364
BACKGROUND:Rabbit VX2 hepatocarcinoma models are characterized by expanded and infiltrative growth, abundant blood supply, similar pathological and imaging features to human hepatocelular carcinoma, so this model is ideal for imaging experiments. OBJECTIVE: To establish rabbit VX2 hepatocarcinoma models and compare two different laparotomy methods in establishing rabbit VX2 hepatocarcinoma models. METHODS:Thirty New Zealand rabbits were randomly divided into two groups: traditional laparotomy (tumor particles were directly embedded into the liver with tweezers) and improved laparotomy (tumor particles were injected into the liver through a 15G syringe needle). The growth features of the tumor established in two different ways were compared by ultrasound. RESULTS AND CONCLUSION:The tumor formation rate was 100% in the two groups; the single rate was respectively 50% and 90%; the ectopic implantation rate was 50% and 10%, there were significant differences between two groups (P < 0.05). The mortality rate within 2 weeks was 33% and 0 in the two groups, with significant differences (P < 0.05). The improved laparotomy method is feasible for modeling rabbit VX2 hepatocarcinoma, due to minimal injury, short operation time, high success rate and stable features.
2.Research of RBP1 like protein--a novel autoantigen in RA
Chinese Journal of Immunology 2017;33(7):976-978
Objective:Rheumatoid arthritis(RA) is a systemic autoimmune disease, but its etiology is unclear. Therefore,it is urgent to search antigens which is relate to induce this disease, however, there are few reports about the new self-antigens at home and abroad. In this study,we investigate new autoantigen through cloning. Methods: We used antigens in synovium of patients with rheumatoid arthritis as probe, extracted mRNA from synoviocyte, performed cDNA library immunological screening and sequence, and analyzed the conformation and performed Northern blotting. Results: We found that new autoantigen had 36.5% homology with retinoblastoma binding protein 1(RBP1)and it was found that these special protein structure such as AT-rich interaction domain, chromo domain,A+T-hook and TAF homology domain. Therefore,it will be named RBP1-like Protein(Rbik).Rbik RNA is not only expressed in the synovial membrane, but also in the heart, small intestine, muscle and other organs. Conclusion: We report that the discovery of the new autoantibodies to RBP1-like Protein(Rbik)may provide a new possible index and target in diagnosis and therapy.
3.Correlational on tibial intercondylar hyperplasia with the area of abnormal signalanterior cruciate ligament in patients with knee osteoarthritis.
China Journal of Orthopaedics and Traumatology 2013;26(12):1002-1004
OBJECTIVESTo investigate whether tibial intercondylar hyperplasia can cause anterior cruciate ligament (ACL) injuries.
METHODSFrom February 2009 to October 2012, 58 cases (including 70 knees) with knee osteoarthritis (KOA) without trauma history were selected from the out-patient department of orthopaedics in Guang'anmen Hospital attached to China Academy of Chinese Medical Science. All cases were tested by MRI scanning, then vertical height of the medial and lateral intercondylar spine in coronal position and the area of abnormal signal of ACL in sagittal position were tested from patients. Correlation between the height of intercondylar spine and the area of abnormal signal of ACL were analyzed.
RESULTSFives cases of 58 cases( 70 knees) were excluded from episome in knee joint, and other 65 knees were in accordance with the inclusion criteria. Among them, there were 10 males and 48 females aged from 39 to 79 years old with an average of 61 years old. The vertical height of the medial intercondylar spine was (10.02 +/- 1.46) mm and lateral was (8.92 +/- 1.69) mm. The area of abnormal signal ACL was (318.42 +/- 130.10) mm2. There was positive correlation between the height of medial intercondylar spine and the area of abnormal signal of ACL (r=0.06, P=0.00). But there was no correlation between lateral intercondylar spine and the area of abnormal signal of ACL (P=0.10).
CONCLUSIONThe height of medial intercondylar spine and the area of abnormal signal of ACL is positively correlated. The clinical significance is in study, while the clinical meaning still need further study.

Chinese Journal of Tissue Engineering Research 2013;(41):7284-7289
BACKGROUND:The skin lesions, pathological and immunological characteristics of Nc/Nga mice are consistent with human atopic dermatitis, and as an atopic dermatitis animal model, it has great research value. But there are no reports on the physiological and biochemical parameters of Nc/Nga mice in China. OBJECTIVE:To observe the growth and reproduction and blood physiological and biochemical parameters of atopic dermatitis model of NC/Nga mice METHODS:The data of reproductive performance of NC/Nga mice from the first to the third generation was analyzed, including mean litter size, weaning rate, pregnancy rate and generation interval. The body mass of 60 Nc/Nga mice with 1-56 days old was measured, 30 mice of female and male, then the growth curve was draw. The blood samples from the infraorbital vessels were col ected to detect the blood physiological and biochemical parameters in the mice. RESULTS AND CONCLUSION:The mean delivery interval of Nc/Nga mice was (25.8±3.1) days, mean litter size was (7.5±2.5) mice, mean weaning rate was (97.2±1.2)%, the mean pregnancy rate was (97.0±1.4)%, and there was no significant difference among the three generations of these mice (P>0.05). The body mass of Nc/Nga mice was increased with the time increasing of days, the body mass of the mice was maximal within 1-2 weeks post-weaning, and there was no significant difference in body mass between males and females at 6 weeks post-weaning (P>0.05). Comparison of the blood physiological and biochemical parameters of mice at the same age between males and females showed that the levels of red blood cells and hemoglobin in female mice were significantly higher than those in the male mice;the platelet count in the male mice was significantly higher than that in the female mice (P<0.05);the triglycerides and albumin levels in the female mice were higher than those in the male mice (P<0.05). The results indicate that the gender and age may influence the blood physiological and biochemical parameters of Nc/Nga mice.
