1.Pharmacodynamics and safety evaluation of clobetasol propionate liposomes

Objective:To investigate pharmacodynamics and safety of clobetasol propionate(CP)liposomes on guinea pigs with chronic dermatitis-eczema.Methods:Right ears of guinea pigs were repeatedly challenged with a small dose of dinitrochlorobenzene(DNCB)for the establishment of model of chronic dermatitis-eczema.The curative effect of CP liposomes was evaluated by cellular infiltrates in dermis and ear swelling thickening.Guinea pigs were given CP liposomes and CP cream on the right abdominal once a day for eight weeks,and the epidermal cell layers was calculated and compared.Results:CP liposomes could effectively reduce the number of cellular infiltrates in dermis and ear swelling thickening.This preparation for the application of eight weeks did not cause reduction of epidermal cells layers. Conclusion:CP liposomes is more effective on guinea pigs model of chronic dermatitis-eczema with less side effects.
3.Clinical efficacy of ambroxol combined with budesonide aerosol inhalation in treating neonatal pneumonia

Objective To observe the effect of aerosol inhalation of ambroxol combined with budesonide on neonatal pneumonia .Methods 66 infants with pneumonia treated were enrolled in this study.The patients were divided into observation group ( n =33 ) and control group ( n =33 ) according to the random number method.The treatment group were treated with ambroxol combined with budesonide and the control group were treated with budesonide.The two groups of infant lung function, clinical outcomes and blood flow were compared.Results The total effective rate (93.94%) in the observation group was significantly higher than that in the control group (45.45%), there was significant difference between the two groups (P<0.05).After treatment, FEV1 and PEF in the two groups were significantly higher than those before treatment, the observation group [(95.21 ± 7.69)%, (93.21 ±7.89)%] were significantly higher than those of the control group[(87.03 ±7.23)%, (84.55 ±7.45)%], the differences between the two groups were statistically significant (P<0.05).The OI of the two groups was significantly higher than that of the control group and the OI of the observation group was significantly higher than that of the control group.PaCO2 of the two groups decreased significantly and the PaCO2 of the observation group was significantly lower than that of the control group, the difference was statistically significant ( P<0.05 ).Conclusion The combination of ambroxol and budesonide is effective in the treatment of neonatal pneumonia.It can effectively improve the infant pulmonary function, shorten the treatment time and improve the hemodynamics of infants .
4.The relationship between the apolipoprotein M-sphingosine-1-phosphate axis and atherosclerosis

Apolipoprotein M-sphingosine-1-phosphate axis ( apoM-S1P axis ) signaling pathway consists of apolipoproteinM (apoM), sphingosine-1-phosphate (S1P) and sphingosine-1-phosphate receptor (S1PR).Plasma apoM belongs to lipocalinsuperfamily members , and is mainly associated to high density lipoprotein( HDL), whereas HDL-cholesterol correlates inversely with cardiovascular risk .The ability of apoM to bind S1P is due to a lipophilic binding pocket within the lipocalin structure of the apoM molecule . S1P, a bioactive mediator of phospholipid metabolism , predominantly abound in HDL among all lipoproteins.S1P can not only be used as intracellular second messengers , but also as intercellular signal molecules, activating of G protein-coupled receptors (S1PR) to mediate various physiological functions.It′s clear that apoM protects human beings from atherosclerosis .Furthermore, recent studies showed that S1P has a significant impact on atherosclerosis , and ApoM-S1P axis may play a important role in the pathogenesis or progression of atherosclerosis .
5.Clinical research of small dose doxepin combined with psychological behavior intervention in the treatment of insomnia

Objective To investigate the effect of small dose of doxepin combined with psychological behav-ior intervention in the treatment of insomnia.Methods 21 patients with insomnia were selected as the study sub-jects,took doxepin 25mg before going to bed at every night,to the research object and psychological behavior interven-tion by hand,continuous treatment for 8 weeks.Used the method of questionnaire survey before and after two periods of time,assessed by using pittsburgh Sleep Quality mder(PSQI)scale,analysed the correlation between sleep quality and sleep beliefs attitudes.Results After treatment with 8 weeks,subjected sleep quality,sleep difficulties,sleep time,sleep efficiency,sleep disturbance,hypnotic drug use,and daytime functional scores were lower than before treat-ment (t =17.34,16.65,10.54,13.37,11.65,7.66,11.57,all P <0.05 ),the difference of the score of PSQI between after treatment[(5.80 ±0.97)points]and before treatment[(13.34 ±2.28)points]was significant(t =21.27,P <0.01).Conclusion Small dose of doxipin combined with psychological behavior intervention in treating insomnia can improve the sleep quality of patients.
7.Coordinate repression of angiostatin and Fas gene on human colon carcinoma

Objective To investigate the coordinate repression of angiostatin(AS)and Fas gene on human colon carcinoma LOVO.Methods Plasmid pcDNA3-AS and pcDNA3-Fas were constructed,and AS,Fas,AS and Fas gene were transfected to human colon carcinoma LOVO cells by liposome method.The expressions of target protein were detected by Western blot.The effects of transfection of AS and Fas gene on the growth of human colon carcinoma cells were detected by MTr methods.AS,Fas,AS and Fas gene were transfected to the human colon carcinoma subcutaneously implanted in nude mice by direct injection into the tumor.The tumor sizes were detected after 14 days of the first gene transfection.Results The effect of gene transfeetion in LOVO cell after twelve hours,24 h,48 h and 72 h were observed and compared with control group,co-transfection of Fas and AS gene group and Fas group significantly inhibited the growth of human colon carcinoma LOVO line cells(P<0.01).Fourteen days after plasmid transfection,the tumor volume in group Fas and group co-transfection were signifieandy smaller than that of control,AS and Fas group(P<0.05).The tumor volume in AS group and Fas group were significantly smaller than that of control(P<0.05).Conclusion Angiostatin and Fas gene have coordinate repression effect on human colon carcinoma in vivo.

9.Gene Therapy of Mice Melanoma by HSV-tk/ACV System

This is the first report on gene therapy of mice melanoma by HSV - tk/ACV system. The sensitivity to ACV of genetically modified B16 cells (B16LNTK) was much higher than that of the parental cells. The sensitivity to ACV of B16 cells was increased when they were co - cultured with B16LNTK cells with various ratios. which showed the exist of the by stander effects. The tumor volume of B16LNTK (0.25cm3) is 94% less than that of B16 in C57BL/6 mice after 20-days ACV treatment. (P
10.Clinical analysis of 82 patients with myasthenic crisis

Objective To evaluate the precipitating factor,mortality,complications of myasthenia gravis crisis and risk factors of prolonged intubation.Methods Totally 82 case records of patients who had been adimitted for having 87 episodes of myasthenia crisis over a period of 15 years from January 1985 to February 2000 and their age, gender, precipitating factors, complications of intubation, and prolonged intubation factors were retrospectively reviewed. Results In 75% of myasthenic crisis patients having precipitating factors, infection was the most often served as the precipitating factors, and then the wound, fatigue, manopause and delivery came the next, 24% of precipitating factors were related to medication. Nine were identified with independent predictors of prolonged intubation. Among the 82 myasthenia crisis patients, 12 died, with a mortality of 14%.Conclusion The prevention and treatment for intubation complications may be the best way for further shortening intubation time and reducing the mortality.
