1.Analysis of thyroid functional abnormalities in primary Sj(o)gren's syndrome

Objective To explore the clinical characteristics of thyroid disease in patients with primary Sj(o)gren's syndrome (pSS).Methods A total of 81 pSS patients were retrospectively investigated for thyroid dysfunction and the presence of anti-thyroid peroxidase antibody (TPOAb),antithyroglobulin antibody (TGAb).Sex,age,erythrocyte sedimentation rate (ESR),C-reactive protein (CRP),white blood cell count,and serum immunoglobulins were also analyzed.Results Thirty-one patients were identified as having thyroid dysfunction,including 3 with hyperthyroidism,18 with hypothyroidism,and 10 with subclinical hypothyroidism.There were no significant differences in the incidences of some clinical manifestations between patients with or without thyroid dysfunction,such as dry mouth,dry eyes,joint pain,Raynaud's phenomenon,palpable purpura,peripheral neuropathy,interstitial lung disease,liver involvement,and renal tubular acidosis.Differences were not statistically significant for ESR and CRP between two groups.Also,the titers of anti-nuclear antibody,anti-SSA antibody,antiSSB antibodies,and rheumatoid factor between two groups showed no significant differences.However,white blood cell count [(5.65 ± 0.49 vs 7.22 ± 0.48) × 109/L,P<0.05] and hematoglobin [(110.50 ± 3.64 vs 122.80 ± 3.02) g/L,P<0.05],as well as the titers of TGAb [(587.30 ± 204.30 vs 58.76 ± 13.78) U/ml,P<0.05] and TPOAb [(213.70 ± 39.64 vs 29.57 ± 8.49) IU/ml,P < 0.05] were significantly different between the two groups.Conclusions Thyroid diseases are frequently associated with pSS.Therefore,extended screening for existing thyroid diseases during the routine assessment of pSS patients is recommended.
3.Additive effects of the variants in the peroxisome proliferator activated receptor γ2 and uncoupling protein 2 genes on type 2 diabetes

Objective To investigate the additive effects of uncoupling protein 2(UCP2) gene 3′-untranslated region(3′-UTR) 45-base pair insertion/deletion( I/D) variation and peroxisome proliferator activated receptor( PPAR)γ2 gene Pro12Ala variation on type 2 diabetes(T2DM) in Chinese Han popula-tion.Methods The UCP2 gene 3′-UTR I/D variation and PPARγ2 gene Pro12Ala variation were exam-ined by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP) in 490 type 2 dia-betes subjects and in 585 control subjects .The additive effects of the two gene mutations were analyzed . Results ⑴The frequency of PPARγ2 gene Pro12Ala variation in type 2 diabetes was not significantly dif-ferent from that in control subjects.(χ2 =0.058, P =0.809).In T2DM group, A12 allele carriers had larger waist circumference than Pro12Pro genotype carriers;Glucose stimulated insulin secretion by oral glu-cose tolerance test (OGTT) was significantly lower in carriers of the Ala12Ala or Pro12Ala genotype com-pared with Pro12Pro genotype.( Z =2.222, P =0.026; Z =2.051, P =0.040; Z =2.079, P =0.038 ) .⑵There wasn't significant difference among 3 genotypes with 3′-UTR I/D variation in UCP2 gene (χ2 =2.311 , P =0.315 ) .In control group , Glucose stimulated insulin secretion by oral glucose tolerance test ( OGTT) was significantly higher in carriers of the I/D or I/I genotype compared with D/D genotype ( Z =1.997 , P =0.046 ) .⑶The genotype carriers with Pro/Ala +Del/Del were the greatest relation to T2DM (OR=1.22, 95%CI 1.078~1.386).Conclusion Though the UCP2 gene mutation alone or the PPARγ2 gene mutation alone is not associated with T 2DM, the possible additive effects of the two micro genes increase the occurring of T 2DM.
5.Influence of Nimodipine on [Ca~(2+)]i Levels in Red Blood Cell in Newborn Infants with Hypoxic-Ischemic Encephalopathy and Its Clinical Significance

Objective To explore change of RBC[Ca2+]i levels in neonates with hypoxic-ischemic encephalopathy(HIE)and the influence of nimodipine on RBC[Ca2+]i and its clinical significance.Methods Fifty-eight neonates with moderate and severe HIE were randomly divided into 2 groups including routine treatment group(n=28)and nimodipine group(n=30),and 20 healthy full-term neonates were selected as healthy control group.Based on the routine treatment,nimodipine[2 mg,0.5-1.0 ?g/(kg?min)] was given intravenously in the nimodipine group for 7-10 days.Blood samples were collected before and after treatment for 72 hours and 10-14 days,respectively.The levels of RBC[Ca2+]i were measured by Fura-2 pentakis(acetoxymethyl)ester[Fura-2/AM].The results were analyzed by SPSS 10.0 software.Results 1.The levels of RBC[Ca2+]i in neonates with HIE were significantly higher than those in healthy control group[(2.83?0.36)mmol/L vs(2.15?0.18)mmol/L,P

Objective To observe the changes of plasma pulmonary surfactant protein-A (SP-A) and serum ferritin (SF) in neonates with mechanical ventilation and its clinical significance in ventilation-induced lung injury (VILI).Methods The study was carried out in 36 neonates with mechanical ventilation and 31 neonates without mechanical ventilation .Plasma SP-A and SF levels in venous blood were mea- sured on 1,24,48,72 hours after mechanical ventilation and 24 hours after mechanical ventilation removal by Western-dot blot and radioimmunoassay (RIA).Plasma SP-A and SF levels of non-mechanical ventilation group were determined at the same time.Results Plasma SP-A levels in neonates on 24,48,72 hours after mechanical ventilation were significantly higher than those of non-mechanical ventilation group[( 2.20? 0.22)vs(1.97?0.29) ?g/L,(2.43?0.28)vs(1.94?0.33) ?g/L,(2.61?0.38)vs(1.80?0.34) ?g/L,respectively P
7.Correlation Between Preoperative Anxiety and Intraoperative Anesthetic Requirements

Objective To investigate the correlation between preoperative anxiety and intraoperative anesthetic requirements,and to determine the proper anxiety evaluation scale.Methods Sixty ASA I patients undergoing knee arthroscopy were enrolled.The anxiety level of the patients was assessed by using the State-Trait Anxiety Inventory(STAI),Zung Self-Rating Anxiety Scale(SAS),and Visual Analogue Scale(VAS) 30 min before the surgery.The low frequency(LF) and low frequency/high frequency(LF/HF) of the heart rate variability were recorded before the surgery.Target controlled infusion(TCI) of propofol was started to provide a blood concentration of 1.5 ?g/ml,and was increased by 0.5 ?g/ml every 30 s until OAA/S value reached 4,which was defined as the point of consciousness loss.BIS value and propofol requirements were recorded.TCI of propofol was maintained for 30 min to keep a BIS value at 50,LF and LF/HF of heart rate variability and maintenance dose of propofol were recorded during this period.Results Propofol requirements at the point of consciousness loss were significantly liner correlated with the SAI,TAI,and Zung scores,as well as LF and LF/HF(r=0.362,0.285,0.315,0.274,0.411,P
8.Effect of Delayed Entry into 2 Different Blood Culture Systems(BacT/Alert 3D and BACTEC 9120) on Culture Positivity

OBJECTIVE To evaluate the effect of delayed entry(0-24 h) into BacT/Alert 3D and BACTEC 9120 and the effect of 2 incubation temperatures(22 ℃ vs 35 ℃) on culture positivity.METHODS We utilized the BacT/Alert system with FA bottles and the BACTEC 9120 system with Plus(Aerobic) bottles.Four clinical bacterial species,Staphylococcus aureus,Escherichia coli,Streptococus pneumoniae,Candida albicans,were used as the test strains.and 5 ml of blood were added to each bottle.Each species was inoculated into the bottles 3 times at an inoculum size of 10 CFU/ml,102 CFU/ml and 108 CFU/ml,respectively.The inoculated bottles were cultured using the respective instruments after they were allowed to stand at room temperature(22 ℃)and 35 ℃ for 0,8,16 and 24 h.Time-to-detection and culture positivity were evaluated.RESULTS The delay in transportation of blood culture bottles stored at room temperature(22 ℃) or 35 ℃ had no effect on the recovery rate for BACTEC 3D at less than 24 h preincubation time and for BacT/Alert 9120 at less than 16h preincubation time.The positivity rate decreased significantly for BacT/Alert 9120 for 24 h of delay.Culture positivity of BacT/Alert 3D was higher than BACTEC 9120 for 24 h of delay.CONCLUSIONS The delayed entry for BacT/Alert 3D should be within 24 h,but the delayed entry for BACTEC 9120 should be within 16 h.

10.The Effects of Asiaticoside on the Growth of Melanoma B16 Cells

Objective To study the effects of asiaticoside on the growth of melanoma B16 cell cultures in vitro. Methods Melanoma B16 cells were subcultured and the inhibition of cellular growth was investigated. The morphology of the cells was observed after inhibition. The induction of apoptosis by asiaticoside was determined by flow cytometry. Results It was found that asiaticoside could significantly inhibit the growth of B16 cell cultures in vitro in a dose-dependent manner. The annexin-v positive cells were increased, along with that cells intaking R123 marked mitochondria were decreased, and PI positive cells increased, which indicated that cellular apoptosis was induced. Conclusion Asiaticoside plays an inhibitory role in the growth of melanoma B16 cells.
