1.Phylogenetic Analyses of HPV53 and Prediction of B and T Cell Epitopes

To construct phylogenetic trees based on HPV53 full length sequences, and predict the physical and chemical parameters, secondary structure, B and T cell epitopes of HPV53 proteins(E1, E2, E4, E6, E7, L1, and L2). The full-length sequences of HPV53 variants were retrieved from the National Center for Biotechnology Information(NCBI), and a phylogenetic tree was constructed to delineate variant lineages. The physical and chemical parameters of HPV53 proteins were analyzed by ProtParam. The secondary structure of proteins was analyzed using PSIPRED and SOPMA. The B and T cell epitopes for HPV53 proteins were predicted by the IEDB analysis server and the ABCpred server, respectively. Then, to select the potential dominant B and T cell epitopes, more parameters including flexibility, hydrophilicity, surface accessibility, antigenicity of predicted B and T cell epitopes were further predicted by bioinformatic methods such as VaxiJen. Finally, for homology analysis, the potential dominant B and T cell epitopes were compared with the 13 high-risk HPV subtypes using NCBI BLAST tool. A total of 54 full-length HPV53 sequences were retrieved from the NCBI database, with 48 entries remaining after deduplication. These 48 HPV53 isolates from different countries/regions were clustered into three main evolutionary branches labeled as lineages A, B, and C. The physicochemical properties of three different HPV53 variants(representing A, B, and C lineages, respectively) were similar. The secondary structure of the E1, E6, and E7 proteins was predominantly α-helices, while E2, E4, L1, and L2 predominantly exhibited random coils. After prediction and screening, a total of 6 potential B-cell epitopes and 9 potential T-cell epitopes were identified on HPV53 proteins. Among these epitopes, B cell epitopes TTPIRPPPPPRPWAPT in E4 region, CYRCQHPLTPEEKQLH in E6 region, and T cell epitopes SGVHSYEEIPMQ in L2 region showed high homologous to HPV56(all > 90%). Bioinformatics analysis and prediction revealed that HPV53 isolates could be clustered into three main evolutionary branches labeled as A, B, and C. These branches exhibited similar physicochemical properties, with minor differences in their secondary structure. Moreover, HPV53 viral proteins contained both B-cell and T-cell antigenic epitopes. These results lay the foundation for further research on vaccines and drugs based on HPV53-related peptides.
2.Relative Analysis between Drug Sensitivity and Escherichia Coli Induced Enzymes in Childhood Diarrhea

Objective To investigate the distribution of pathogenic bacteria,drug sensitivity,and the relationship between drug sensitivity and Escherichia coli(E.coli) induced enzymes in childhood diarrhea in the last 2 years in Chongqing area,so as to provide important evidence for pediatric clinical therapy.Methods Thirty-one E.coli induced enzymes,extended spectrum ?-laetamases(ESBLs),cephalosporinase(AmpC)detected in different phenotype methods,and drug sensitivity was measured in paper strip method,and the specimens were collected from children′s hospital affiliated to chongqing university of medical sciences from Jan.2005 to Dec.2006 were determined.Among the total,there were 18 enteropathogeic E.coli(EPEC) strains,8 enterotoxigenic E.coli(ETEC) strains and 5 enteroinvasive E.coli(EIEC) strains.In addition,drug resistance tests by paper strip included chloramphenicol(CHL),amikacin(AMK),gentamicin(GEN),norfloxacin(NOF),ciproflocacin(CIP),cefazolin(CEZ),cefoperazone(CPZ),ceftriaxone(CRO),ceftazidime(CAZ),cefotacime(CTX),cefepime(FEP),imipenem(IPM).SPSS 12.0 software was used to analyze the data.Results Three point two percent of the 31 E.coli were drug resistant to IPM,and 35.5%,38.7% to NOF,CIP individually,but more than 60% to AMK,GEN,even more than 67.7% towards cephalosporin(except ceftazidime and cefepime);the gross enzyme-produced rate was 87.1%,rate of single ESBLs,AmpC,and induction of both enzymes simultaneously presented 64.5%,6.5%,16.1% respectively;and there was marked difference in drug resistance when bacteria that produced single AmpC versus bacteria that produced single ESBLs or that produced both ESBLs and AmpC(Pa﹤0.05).Conclusions The relationships among enzyme′s quantity,sort and bacterial resistance are different.These data show E.coli infected by bacterial diarrhea children in Chongqing due to a high rate of induced enzymes,and their drug resistance vary according to the state of induced enzymes.
3.Implementation of Real-time Bidirectional Communication Function on Siemens StreamLab Laboratory Automation System

Objective To implement a real-time bidirectional communication function on Siemens StreamLab laboratory automation system, Methods Firstly, the system selecting sample was changed from the default settings and then the communication character string sent by the StreamLab was utilized by the laboratory information system to build up a real-time bidirectional control of the samples assay. Results New functions such as real-time signing in and real-time detection information getting of specimens could be performed by the StreamLab automatically.Conclusion The real-time bidirectional communication between laboratory automation system and laboratory information system can optimize the working flow and improve the work efficiency.

5.Remote Control System Construction in Core Medical Laboratory

Objective To implement a remote monitor system in a core medical laboratory.Methods Firstly,the Hub of StreamLab laboratory automation system was connected to a wireless router to build a wireless network,and several equipments were also integrated into the wireless network using wireless network cards.Secondly,VNC Server was installed into the control PCs of StreamLab LAS,CA7000 Coagulation Analyzer,DPC IMMULITE 2000 SMS and its chemiluminescence photometer,and remote control function of Dimension RxL Max was activated,then some control software such as VNC Viewer,Q/Remote and PC Anywhere were setup in two agency control computers.Finally,PCAnywhere and VNC Viewer were setup in the remote control computers in the Internet and Intranet,respectively.Results A remote control system with three functions was built successfully in a core medical laboratory.Conclusion The remote control system makes the daily operation and remote maintenance of the equipments easy.
6.Clinical analysis of orbital implantation and postoperative complications

Objective To discuss the cause, managment and prevention of the postoperative complications of orbital implantation by clinical case analysis, and to provide reference for the future clinical work. Methods 175 cases of orbital implantation from Aug. 2011 to Aug. 2013 were retrospectively analyzed in our department. The postoperative complications occurrence, related symptomatic treatment, and the reasons of complications were analyzed. Results Complications occurred in 175 cases of 9 cases(5. 14%),including 4 cases of the con-junctival cyst (2.29%),3 cases of orbital implants exposure(1.71%),1 case of orbital chronic infection(0.57%)and 1 case of subcon-junctival tissue atrophy(0. 57%). All of them had obtained good curative through cyst excision or orbital implanting again after the active treatment. There were no complication occurred again. Conclusion The postoperative complications should be positively analyzed, and early positive precaution could reduce most of the complications.

9.Effect Evaluation of Clinical Pharmacists in Continued Service for Patients with Ischemic Stroke

Objective:To evaluate the effect of clinical pharmacists in the continued therapy of stroke patients .Methods:Totally 146 cases were selected and randomly divided into the observation group and the control group according to the standard matching meth -od.The observation group was treated with the continued therapy .The control group was given one-month telephone follow-up after the discharge.The differences in the drug compliance (Morisky scale), systolic blood pressure (SBP), diastolic blood pressure (DBP), glycosylated hemoglobin (HbA1C), serum total cholesterol ( TC), low density lipoprotein ( LDL-C), health behaviors ( HPLPⅡscale) and relapse rate were compared between the two groups in the 3rd, 6th and 12th month after the discharge.Results:The differ-ences in the above indices were statistically significant between the two groups ( P <0.05) at the various time points , and those were more significant with time extending (P<0.01).The difference of the relapse rate in the 3rd, 6th and 12th month after the discharge between the two groups were significant .Conclusion: Clinical pharmacists can improve the drug compliance and health behaviors of stroke patients and reduce the relapse rate .It is worthy to be recommended in clinics .
