2.Expression and construction of an adeno-associated virus vector containing dual genes-GDNF and TH

Objective Rat glial cell derived neurotrophic factor(GDNF) gene and tyrosine hydroxylase(TH) gene were widely used in gene therapy of Parkinson's Disease(PD).The present study was designed to investigate whether the two target genes GDNF and TH could express simultaneously in one adeno-associated virus(AAV)vector and to discuss the probability of using it in gene therapy of PD.Methods RNA of GDNF and TH in HEK293 packaging cells were examined by reverse transcription-polymerase chain reaction(RT-PCR).Expressions of GDNF and TH in infected bone marrow stromal cells(BMSCs) and rat brain coronal sections were examined with immunocytochemistry and immunohistochemistry assay.Results RNA of GDNF and TH were detected in HEK293 packaging cells.Expressions of this two genes were observed in both bone marrow stromal cells and brain sections of rats.Theexpression efficiency of AAV-LacZ was about 50% in HEK293 packaging cells and approximately 15% in infected BMSCs and the expressions of TH and GDNF on the AAV-GDNF/TH virus injected points both had significant differences(P
3.Application and biocompatibility of a new absorbable magnesium alloy stent in blood vessels

BACKGROUND: In vivo animal experiments have found that magnesium al oy stents can be completely degraded in a slow, orderly and non-toxic manner fol owing the vascular remodeling. OBJECTIVE: To investigate the applied effect and biocompatibility of a new type of magnesium al oy stent in blood vessels. METHODS: Forty-two crossbreed dogs were implanted with absorbable magnesium al oy stents via the coronary artery or left femoral artery. At days 1, 3, 5 and weeks 1, 2, 3, 4 after implantation, coronary or femoral artery angiography, inflammatory factor detection and morphological analysis targeting intimal hyperplasia were performed. RESULTS AND CONCLUSION: At 1 day after implantation, the stents had the complete shape and were ful y extended; at 3 days after implantation, the stents were degraded partial y; at 1 week after implantation, the stents were degraded completely. Within 1-3 weeks after implantation, the levels of tumor necrosis factor α, interleukin-6 and interleukin-8 continued to decrease. At 2 and 3 weeks after implantation, the target vessels presented with mild intimal hyperplasia. As time went on, the hyperplasia area increased gradual y, and the percentage of intimal hyperplasia increased gradual y. Blood magnesium concentration remained unchanged before and after stent implantation. The results show that the new absorbable magnesium al oy stent can be completely degraded within 1 week after stent implantation and has good biocompatibility.

It can perform high-throughput detection and data processing with multiple param eters, potentially enabling the identification and quantification of all sm all m etabolites in a biological system . It can be used to provide com prehensive inform ation on the toxicity effects, toxi-cological mechanisms and biom arkers, sensitively finding the unusual m etabolic changes caused by poi-son. This article mainly review s application of m etabonom ics in toxicological studies of abused drugs, pesticides, poisonous plants and poisonous anim als, and also illustrates the new direction of forensic toxi-cology research.
7.Effect of Nasal Inhalation of Compound Acorus Liquid and Oxygen on the Recovery of Nervous Function in Cerebral Infarction Patients

It can perform high-throughput detection and data processing with multiple parameters, potentially enabling the identification and quantification of all small metabolites in a biological system. It can be used to provide comprehensive information on the toxicity effects, toxicological mechanisms and biomarkers, sensitively finding the unusual metabolic changes caused by poison. This article mainly reviews application of metabonomics in toxicological studies of abused drugs, pesticides, poisonous plants and poisonous animals, and also illustrates the new direction of forensic toxicology research.

9.Relationship between leptin level and glycometabolism in pregnancy with abnormal glucose challenge test

Objective To investigate the relationship between serum leptin(LP) level and glycometabolism in pregnancy with abnormal glucose challenge test(GCT) and the corresponding outcome of pregnancy. Methods Serum LP level,fasting serum insulin(FINS) and fasting blood glucose(FBG) levels were measured by radioimmunoassay in 23 patients with abnormal GCT but normal oral glucose tolerance test(OGTT),20 with impaired glucose tolerance(IGT),20 with gestational diabetes mellitus(GDM)(studied groups above) and 30 normal pregnant women(control group).LP,FINS,FBG, insulin resistance index(HOMA-IR) and ?-cell function index(HOMA-IS) were compared among the groups and the outcomes of pregnancy were observed. Results ①There were significant differences in LP,FINS,FBG,HOMA-IR and HOMA-IS between the studied groups and the control group(P0.05).③There were signi-ficant differences in the gestational weeks of delivery,cesarean section rate and neonatal weight between the studied groups and the control group(P0.05). Conclusion Both insulin secretion and insulin resistance are elevated in pregnancy with abnormal glycometabolism,and are closely correlated to the increased serum level of LP.There existed abnormal glycometabolism in patients with abnormal GCT but normal OGTT,which is correlated to the outcome of pregnancy.
10.Clinical and pathological analysis of hepatitis B virus-associated glomerulonephritis in 20 cases

Objective To understand the clinical and pathological features of hepatitis B virus-associated glomerulonephritis(HBV-GN).Methods Twenty children with HBV-GN were analysed including 17 cases of nephritic syndrome (4 cases of simple nephrosis, 13 cases of nephritic nephrosis),2 cases of isolated proteinuria, 1 case of nephritic syndrome according to the clinical classification.Results On the basis of pathology of kidney biopsy, there were 15 cases of membranous glomerulonephritis(MN),3 cases of manbranoproliferative glomerulonephritis(MPGN),2 cases of mesangial proliferative glomerulonephritis(MsPGN).Eight of 20 cases were treated with recombinated human alpha-interfeon. The average follow-up time was 5.7 years. As the result, complete remission clinically was on 5 cases,partial remission on 2 cases and ineffectiveness on one case. Besides, serum HBsAg, HBeAg were converted to negative on 3 cases , HBeAg disappeared and anti-HBe appeared on one case.Conclusion MN is common in childhood with HBV-GN.It also indicates the prognosis of the associated nephropathies is quite favorable.
