1.Protective effect of taurine on diaphragm against adriamycin cytotoxicity

AIM To study protective effect of taurine on diaphragm against adriamycim cytotoxity in rabbits.METHODS The rabbits were divided randomly into three groups.①Control group:The animals were administered NS 2 ml*kg-1 for five days. ②Adriamycin+NS group:The animals were administered NS 2ml*kg-1 for five days and then administered adriamycin 10 mg*kg-1.③Adriamycin+taurine group:The animals were administered taurine 100 mg*kg-1 for five days and then administered adriamycin 10 mg*kg-1.After 24 h,three groups animals were anaesthetized.The transdiaphragmatic pressure(Pdi) and diaphragm evoked potential(DEP) were measured.SOD and MDA were tested.the ultrastructure of diaphragm cells was investigated by eletronmicroscopy.RESULTS The Pdi and amplitude of DEP were lowered by adriamycin(P<0.01).The levels of MDA was increased and the activity of SOD was decreased in diaphragm(P<0.05).The disorder of myofibrills,swelling of mitochondria with broken cristase can be observed by electronmicroscopy.The above changes were inhibited by taurine.CONCLUSIONS The taurine could scavenge the toxic radicals generated by adriamycin and protect the diaphragm myocytes.

Thyroid-stimulating hormone receptor(TSHR)is an important autoantigen whose cleavage and shedding can induce autoimmunity.The TSHR is the main functional receptor of the thyroid.The shedding of the ? subunit plays an important role in the forming of TSAb and balancing between TSAb and TBAb.Research on the cleavage and shedding of the TSHR extracellualar domain helps to gain a deep insight into the structure and function of TSHR,illuminate the pathogenesis of autoimmune thyroid disease and find out a new therapy.The structure of TSHR,the mechanism and the significance of its cleavage and shedding are reviewed in this article.
5.Effects of oxidative stress induced by high glucose in endothelial cells

Objective To explore the effects of oxidative stress in porcine iliac endothelial cells(PIECs) induced by high glucose. Methods After being intervened by high glucose for some time, dihydroethidium (DHE) or dihydrorhodamine 123 (DHR123) was used as a reactive oxygen species(ROS) capture. The mean fluorescent intensity(MFI) of above probes which were the products of intracellular oxidation was detected by fluorescent microscopy and flow cytometry, and the level of ROS was thus measured. With lucigenin as chemiluminescence agent, luminescence changes were detected by chemiluminescence analyzer after addition of NADPH to observe the effect of high glucose on activity of NADPH oxidase(NOX). Results Intracellular MFI was markedly elevated with the concentration of high glucose and time of exposure to high glucose. Objective To investigate the clinical characteristics,pathogen distribution and outcome of lower respiratory tract fungal infection in patients with mechanical ventilation in emergency intensive care unit(EICU). Methods The clinical data of patients with lower respiratory tract fungal infection treated with mechanical ventilation in EICU were retrospectively analysed. Results Eighty(64.5%) of the 124 patients with mechanical ventilation experienced lower respiratory tract infection,among whom 21(26.2%) were complicated with fungal infection.The predominant pathogen was Candida(Candida albicans,n=11;Candida tropicalis,n=5;Candida smooth,n=3;Candida parapsilosis,n=2).Compared with patients with non-fungal infection(n=59),those with fungal infection(n=21) had higher APACHE II score,longer mechanical ventilation time and longer ICU stay duration(P
7.Study on the effectiveness and safety of ginseng polysaccharide adjuvant GP chemotherapy in the treatment of non small cell lung cancer

Objective To discuss the treatment effect of ginseng polysaccharide injection auxiliary GP chemotherapy for patients with non-small cell lung cancer.Methods 130 cases of patients with non-small cell lung cancer from June 2011 to June 2013 were randomly divided into two groups, 65 cases in each group.The control group was given gemcitabine and cisplatin treatment.The observation group were treated with ginseng polysaccharide injection on the basis of the control group.After 3 months of continuous therapy, the therapeutic effect, T-lymphocyte subsets, adverse events, quality of life scores of the two groups were compared, and survival in two groups of patients were also compared after 1 year follow-up.Results The total efficiency and clinical benefit rate in observation group were 58.5%, significantly better than the control group ( 41.5%) , the difference was statistically significant (χ2 =3.72, P<0.05);CD3, CD4 and CD4 /CD8 in observation group were significantly higher than the control group, the differences were statistically significant (t =5.735, 7.885, 7.326, P<0.05), whereas CD8 was significantly lower than the control group, the difference was statistically significant (t=2.442, P<0.05).The main adverse reactions of the two groups of patients during treatment were myelosuppression (WBC and platelet decreasing and anemia ) and gastrointestinal reaction.Score of life quality in observation group was ( 71.5 ±12.9 ) after treatment, significantly higher than the control group (64.2 ±12.2), the difference was statistically significant (t=3.315, P<0.05).1-year survival rate in the observation group were 63.1%, and 52.3% in the control group.Conclusion Ginseng Polysaccharide injection auxiliary GP chemotherapy for non-small cell lung cancer has significantly efficacy, enhances immunity, reduces side effects, better quality of life.
8.Clinical analysis of 6 patients with cardiac surgery under cardiopulmonary bypass during pregnancy

Objective To evaluate the optimal management of cardiac surgery during pregnancy,in the second or third trimester,and the maternal and fetal outcomes in pregnant patients after surgery.Methods Six pregnant women with heart diseases were identified,who underwent cardiac surgery with cardiopulmonary bypass.Surgery was performed using cardiopulmonary bypass at mild hypothermia/normothermia,with pulsatile perfusion.Natural progesterone(20 mg)was added in priming solution,monitoring uterine contractions and heart rate changes.The mode of surgeries included mitral valve replacement+tricuspid valve plastic in three cases,aortic and mitral valve replacement+tricuspid valve plastic in one,left ventricular myxomatomy in one,left ventricular septal myectomy+artificial aortic valvular vegetations dissection+tricuspid valve plastic in one.Results The cardiopulmonary bypass and cross clamp time averaged 169 minutes(range,96~419 minutes)and 113 minutes(range,56~296 minutes),respectively.Five patients were alive.One maternal and fetal death occurred 6 h after surgery caused by heart failure and pulmonary edema.The other 5 patients terminated pregnancies after heart surgeries,one underwent with cesarean section 3 d after surgery in second trimester,two of whom with induction of labor,and two full-term labor(one with cesarean section,one with normal delivery).Two newborns were alive with no malformation.Conclusion Cardiopulmonary bypass can be used safely with satisfactory maternal and fetal outcomes in pregnant patients with heart disease undergoing cardiac surgery.

Objective: Effects of Shenmai injection and aminophylline alone or in combination on transdiaphragmatic pressure and electrical activity of diaphragm were observed, so as to evaluate their effecfs on diaphragmatic fatigue. Methods: The diaphramatic fatique (DiF) model was estab- lished by stimulating bilateral phrenic nerves for 40 minutes. The animals were allocated to three groups: Shenmai group (2mL/kg, N=7), aminophylline group (20mg/kg, N=7) and combi- nation group (Shenmai injection 2mL/kg+aminophylline 20mg/kg, N=7) The transdi- aphragmatic pressure (Pdi) and diaphragmatic electromyogram (EMGdi) were recorded before and after treatment. EMGdi was analysed by computer and then the high/low frequency ratio (H/L) and the central frequency (Fc) were caculated. The diaphragm evoked potential (DEP) was record- ed simutaneously. 10.A genetic transformation study of introducing wild Gastrodia elata DNA into Solanum tuberosum by soaking seedling method

Objective Using the soaking seedling method to introduce wild Gastrodia elata DNA into potato plantlets and analyze gastrodin of transformed Solanum tuberosum.Methods After the tuber grown up,the solution of gastrodin was extracted from the potatoes which were transformed by wild G.(elata) DNA.The transformed plants were scaned by ultraviolet.PCR was used to analyze GAFP gene by SDS-PAGE.TLC was used to analyze the gastrodin of transformed S.tuberosum.Results(1)The 21 transformed plants had obvious absorption peak at 220 nm in 200 transformed S.tuberosunr.(2)The fragment of wild G.elata GAFP gene was found by PCR from 5 transformed plants.(3)The protein expressions in transgenic S.tuberosum were obviously different from the normal S.tuberosum;a band was detected in transgenic S.tuberosum and wild G.elata,but wasn't detected in normal S.tuberosum.(4)The expression of the gastrodin was detected by TLC from three transformed plants. Conclusion It is feasible to use the soaking seedling method to introduce exogenous DNA into plants.
