1.Analysis of X chromosome mosaicism in patients with premature ovarian failure by fluorescent in-situ hybridization

Objective To investigate whether low level 45, X/46, XX mosaicism presents in women with premature ovarian failure (POF) and the sensitivity and specificity of fluorescent in situ hybridization (FISH) for detecting X chromosome mosaicism. Methods Karyotypes of peripheral lymphocyte in 18 patients with POF and 9 normal controls were analyzed and the orange signals during FISH using X chromosome enumeration probes (CEPX) were counted. Results Single signals of X chromosome (45, X) were found in 7.6% of the total counted cells, which was significantly greater than that in the controls, in spite of the normal 46, XX karyotypes by routine analyses in all POF patients. Conclusions This study indicated that some POF patients may attribute to low level 45, X/46, XX mosaicism. FISH is more sensitive than the routine chromosome analyses in detecting low level X chromosome monosomy.
2.Evaluation of clinical application of approaches in cervical cancer screening

Objective To evaluate the function of Hybrid capture 2 (HC2) human papillomavirus (HPV) test,liquid-based cytology test (LCT),HPV serial test,LCT serial test and HPV-LCT parallel test for cervical high-grade lesions screening and to guide in selecting the scheme of cervical cancer screening.Methods HPV test,LCT,and colpescopic biopsy were performed in 545 women treated in Centrale Hospital of Klamayi from Sep.2009 and Dec.2011.The pathology results were the final diagnosis to evaluate the value of each approach for screening cervical cancer.Results Of 545 patients, 10 had cervical cancer.41 had cervical intraepithelial neoplasia CIN3,69 had CIN2,and 120 had CIN 1.Positive rate of high risk HPV was 64.22 %.The sensitivity,specificity,accuracy,positive pre-value,and negative pre-value of HPV test for detecting high-grade cervical lesions were 95.83 %,84.71%,88.48 %,30.68 %,99.12 %,respectively.The values of LCT were 84.20 %,89.53 %,85.53 %,36.69 %,91.76 %,respectively.The values of LCT serial test were 81.03 %,95.96 %,78.33 %,40.69 %,92.92 %,respectively.The values of HPV serial test were 81.03 %,95.26 %,78.33 %,42.15 %,99.52 %,respectively.The values of HPV-LCT parallel test were 99.20 %,81.53 %,90.03 %,32.09 %,99.33 %,respectively.Conclusion HPV-LCT parallel test can significantly improve the detection rate of cervical cancer screening.
3.Effects of hypoxia and NO on the expression of HIF-1?, VEGF and iNOS in colon cancer cells SW480

AIM: To observe the expression of HIF-1? mRNA, HIF-1?, VEGF and iNOS proteins and to investigate their relationship in h ypoxia-treated SW480 cells. METHODS: HIF-1?, VEGF and iNOS proteins were measured by immuno cytochemistry. Western-blot was used to detect HIF-1? protein. HIF-1? mRNA wa s measured by in situ hybridization. RESULTS: Under hypoxic condition, SW480 cells expressed proteins of HIF-1?, VEGF and iNOS more strongly than that under normoxia condition. How e ver, under hypoxia condition, these three proteins expressed weakly or negativel y when the cells treated with genistein, the inhibitor of HIF-1?. Expressions o f HIF-1? and VEGF proteins in cultured SW480 cells under hypoxic condition were completely or partially inhibited by the addition of SNP but the expression of iNOS was unaffected. Another NO donor NOC5, however, induced the expression of t hese three proteins. L-NAME, a non-specific inhibitor of NOS, inhibited the expr ession of HIF-1?, VEGF and iNOS. The levels of HIF-1? mRNA changed slightly i n different oxygen condition or addition of genistein, NO donor or iNOS inhibitor . CONCLUSIONS: Hypoxia induces the expression of HIF-1?, therefor e upregulates the production of VEGF and iNOS. During hypoxia, SNP inhibits but N OC5 promotes HIF-1? expression, indicating that different NO donor acts on the cells through different mechanisms.
4.Experimental study on the distribution of 99Tc m-MDP in the repair of femoral bone injury in rabbits

Objective:To investigate the distribution of 99Tc m-methylene diphosphonate (MDP) at different stages of bone injury repair. Methods:A total of 30 rabbit models of femur injury were established by the method of electric drill and perforation of femur. According to the different stages of bone injury repair (at 1, 2 and 3 week), rabbits were divided into group A, B and C ( n=10 each group). Femoral SPECT/CT imaging was performed on the last day of different stages of bone injury repair to obtain radioactivity counts in the region of interest (ROI) on the test side and control side and to calculate target/background ratio (T/B). The light intensity of 3 groups was analyzed by phosphor screen imaging and the distribution of 99Tc m-MDP in bone cells was observed by autoradiography. One-way analysis of variance and paired t test were used to analyze the data. Results:The T/B values of group A, B and C were 1.16±0.14, 1.39±0.23 and 1.18±0.10, respectively ( F=5.83, P<0.01). There were significant differences of the maximum radiation count between the test side (50.00±12.45, 59.50±12.83 and 55.10±9.26) and the control side (43.20±9.57, 50.00±12.30 and 44.30± 6.50) in group A, B and C ( t values: 3.24, 2.28 and 5.77, all P<0.05). There were significant differences in the light intensity of bone specimens in group A, B and C by phosphor screen imaging (37 324.67±6 481.50, 60 950.33±9 781.72 and 43 905.00±4 957.92; F=8.25, P=0.02). 99Tc m-MDP were deposited in both intracellular and extracellular during different stages of bone repair in osteocytes and osteoblasts under autoradiography. Conclusion:At different stages of bone injury repair, the concentration of 99Tc m-MDP is significantly distributed, suggesting that there are other ways of concentration mechanism of 99Tc m-MDP in bone tissue besides the chemical adsorption with hydroxyapatite.
5.Photochemical Effect of the Photosensitizer Hemoporphyrin Derivative on MCF-7 Cell Line and Umbilical Cord Blood Hematopoietic Cells

To study the effect of hemoporphyrin derivative(HPD) combined with laser irradiation on human breast tumor cell line MCF-7 and normal human umbilical cord blood-derived hematopoietic cells by using MTT assay. The results showed that HPD plus laser irradiation was more efficient for killing MCF-7 cells than normal human umbilical cord blood-derived hematopoietic cells. The photochemical effect with laser irradiation were higher than with light irradiation and it's effect on MCF-7 cells was higher gradually with the increase of HPD concentration.
6.Changes in antioxidant capacity of the guinea pig exposed to noise and the protective effect of alpha-lipoic acid against acoustic trauma.

The study was aimed at exploring the effect of noise on total antioxidant capacity (TAC) in serum, nitric oxide (NO) level in the cochlea and the protective action of alpha-lipoic acid against noise-induced hearing loss (NIHL). Sixty guinea pigs (350-400 g) were divided randomly into three groups (control group, noise+saline group and noise+alpha-lipoic acid group). Serum and cochlear tissue were treated immediately after noise exposure (4-kHz octave band, 115 dB SPL 5 h) to determine the level of TAC and NO, respectively. Auditory brainstem responses (ABRs) were measured before and immediately after exposure. The threshold of hearing in the control group was relatively stable, while the hearing threshold in the noise+saline group was significantly higher than those in the noise+alpha-lipoic acid group (P<0.05). TAC level of the noise+saline group was significantly lower than that of the control group P<0.05 . TAC level of the noise+alpha-lipoic acid group was significantly higher than that of the noise+saline group P<0.05 , while there was no significant difference in the levels between the noise+alpha-lipoic acid group and the control group (P>0.05). The NO level of the cochlear tissue in the noise+saline group was significantly higher than that of the control group (P<0.05). Cochlear NO level in the noise+alpha-lipoic acid group was significantly lower than that of the noise+saline group (P<0.05), while there was no significant difference in cochlear NO levels between the noise+alpha-lipoic acid group and the control group (P>0.05). The results obtained indicate that noise exposure causes a decrease in serum TAC and an increase in NO in cochlea. alpha-Lipoid acid exerts a protective effect against hearing loss in acoustic trauma through its antioxidant effects.

7.Applied anatomy of the path of retroperitoneal laparoscopy surgery.

OBJECTIVETo discover the anatomical basis of retroperitoneal laparoscopic surgery.
METHODSTwenty Chinese adult cadavers including 12 and 8 woman were anatomized. The relationship between nerves, vessels of the lateral abdominal wall, retroperitoneal structures and the path of retroperitoneal laparoscopic surgery were measured.
RESULTSIn male cadavers, the distance between the iliac crest and lumbar trigonum was 4.02 +/- 1.26 cm on the left, and 3.83 +/- 0.90 cm an the right. The distance between the iliac crest and ilio-hypogastric nerve was 0.82 +/- 0.13 cm on the left and 0.84 +/- 0.08 cm on the right. The distance between the subcostal nerve and XII rib in the posterior axillary line and midaxilla line was 0.79 +/- 0.26 cm, 1.65 +/- 1.12 cm on the left and 0.78 +/- 0.30 cm, 1.59 +/- 1.07 cm on the right respectively. The distance between the ureter of inferior pole of kidney and extra-border of psoas was 2.24 +/- 0.67 cm on the left and 2.19 +/- 0.73 cm on the right. The distance between crossing of the ureter and iliac vessels and extra-border of psoas was 3.91 +/- 0.59 cm on the left and 3.76 +/- 0.53 cm on the right. In female cadavers, the distance between the iliac crest and trigonum lumbale was 3.90 +/- 0.37 cm on the left and 3.87 +/- 0.37 cm on the right. The distance between the iliac crest and ilio-hypogastric nerve was 0.94 +/- 0.06 cm on the left and 0.93 +/- 0.10 cm on the right. The distance between the subcostal nerve and XII rib in the posterior axillary line and midaxilla line was 0.61 +/- 0.14 cm and 1.37 +/- 0.89 cm on the left and 0.64 +/- 0.13 cm, 1.32 +/- 0.95 cm on the right respectively. The distance between the ureter of inferior pole of kidney and extra-border of psoas was 2.00 +/- 0.23 cm on the left and 1.91 +/- 0.13 cm on the right. The distance between crossing of the ureter and iliac vessels and extra-border of psoas was 3.43 +/- 0.31 cm on the left and 3.85 +/- 0.43 cm on the right.
CONCLUSIONSThe top of the lumbar trigonum which is 2 cm above the iliac crest selected as the first point and 2 cm inferior to the XII rib in the posterior axillary line selected as the second point is the anatomical basis for retroperitoneal laparoscopic surgery. It can reduce the opportunity to injury nerves, vessels and retroperitoneal structures while making retroperitoneal laparoscopic surgery easy.

Multidrug resistance (MDR) of cancer cells to anti-tumor drugs remains a major impediment to successful chemotherapy. There has been an increasing interest in the studies of the mechanism and reverse of the MDR. Being a reliable and safe way to reverse MDR, drug delivery systems (DDS) such as micelle, liposome and nanoparticle, represent a promising prospect both in research and application in recent years. On the basis of recent studies, the effect and mechanism of micelles on reversing MDR are reviewed. And it is anticipated that DDS could contribute greatly to reversing MDR in the future.

9.Nursing intervention of acupuncture plus rehabilitation training on patients with hemiplegia after stroke

Objective To study the nursing intervention of acupuncture plus rehabilitation training on patients with hemiplegia after stroke. Methods 60 patients with hemiplegia after stroke from February 2016 to January 2017 were randomly divided into the control group and the experimental group, each with 30 cases. The control group was given routine treatment and nursing measures, the patients in the experimental group were received acupuncture treatment plus rehabilitation training, and given specific nursing intervention measures, the psychological state of patients with attention, eliminating the patient anxiety and anxiety and other negative emotions, explaining to patients patiently, rehabilitation training guide patients. Clinical indicators of two groups of patients were compared and analyzed. Results After treatment, the total score of quality of life of patients in the experimental group was (75.2±11.2) points, the control group quality of life in patients was (64.2±9.3) points, the effective rate of the experimental group (90.00%) was significantly higher than (70.00%) in the control group with statistical difference (P<0.05). Conclusion Acupuncture plus rehabilitation training combined with nursing intervention in clinical application of hemiplegia patients after stroke, could significantly improve the quality of life and life ability of patients with high safety and clinical significance.
10.The efficacy and safety of Danhong injection combined with pantoprazole sodium on patients with acute exacerbation of respiratory failure

Objective To investigate the clinical efficacy and safety of Danhong injection combined with pantoprazole sodium on patients with acute exacerbation of respiratory failure. Methods 104 patients with acute exacerbation of respiratory failure were treated in our hospital from January 2016 to January 2017. They were selected and equally divided into two groups with the method of random number table. The two groups were given the same basic treatment, and the control group was treated with pantoprazole sodium on this basis. The treatment group was treated with Danhong injection on the basis of the control group. The clinical efficacy and adverse reactions of two groups were recorded. Results After 15 days of the treatment, the total effective rate of the treatment group was 94.2%, which was significantly higher than that of the control group 71.2%(P<0.05). There was no significant difference in the incidence of adverse reactions between the control group and the treatment group. Conclusion Danhong injection combined with pantoprazole sodium is effective and safe in the treatment of acute exacerbations of respiratory failure, which is worthy of clinical practice and popularization.
