2.Effects of oral biofilm on guided bone regeneration in dental implant

BACKGROUND: Heal-all oral biofilm is a material utilized in repairing oral mucosa and soft tissues defects and characterized by degradation, easily preparation, long preserved duration, convenient transportation and good ossification, which has been widely used in dental implant as guided bone regeneration materials.OBJECTIVE: To check the clinical effective of Heal-all oral biofllm on guided bone regeneration in dental implant.METHODS: A total of 72 patients with bone defects in the implantation area were selected as subjects, who were divided into control group and experimental group at random. Bone defects around implants were repaired by guided bone regeneration technique with BME-10X medical collagen membrane and Heal-all oral biofilm respectively. X-ray and clinical examination were taken at 1 and 3 months after implantation. The amount of new.formed bone tissue was evaluated when stage Ⅱ operation was performed.RESULTS AND CONCLUSION: In stage Ⅱ operation, osseointegration was formed between implants and bone tissue in all 72 patients. The average rate of bone formation was 92% in the experimental group while 91% in the control group. All implants were successfully repaired with implant denture. Occlusal function was restored successfully with all 72 implants during the follow-up period of 3-24 months after restoration. As an alternative option of BME-10X medical collagen membrane, Heal-all oral biofilm can be used in guided bone formation clinically.
3.Roles of HER-2 gene amplification and protein expression in gastric carcinoma with neuroendocrine differentiation

Objective To evaluate the gene amplification and protein expression of HER-2 in gastric carcinoma with or without neuroendocrine differentiation (NED), and to explore the difference in HER-2 gene profile between these two neo?plasms. Methods Cases of gastric carcinoma with NED (n=70) and cases of gastric carcinoma without NED (n=150) were retrospectively reviewed. Gene amplification and protein expression of HER-2 genes in gastric carcinoma with or without NED were detected by combination of immunohistochemical method (IHC) and double color silver dye for in situ hybridiza?tion (DISH). Prognosis of gastric cancer patients with NED were predicted using Kaplan-Meiers survival analysis. Results Expression rates of HER-2 in gastric cancer with and without NED are 20.00%and 21.33%respectively. Amplification of HER-2 in gastric cancer with and without NED are 8.57%and 14.67%respectively. Gene amplification and protein expres?sion of HER-2 between gastric cancer with NED and without NED showed no statistical difference. Chromosome 17 multi-body positively correlated with gene HER-2 amplification in Gastric carcinoma with NED. Postoperative survival period in patients of gastric carcinoma with NED and HER-2 amplification was shorter than that in patients of gastric carcinoma with?out NED but with HER-2 amplification. Gastric carcinoma with or without NED, HER-2 gene amplification, lymph node me?tastasis and operation method obviously correlate with prognosis of gastric carcinoma patients (P<0.05). Conclusion The gastric cancer with NED is a special type of gastric cancer, there was no difference of gene amplification and protein expres?sion of HER-2 gene between gastric carcinoma with NED and without NED. Poor prognosis would be expected in gastric can?cer patients with NED and HER-2 amplification.
4.Investigation on cancer related fatigues in oral cancer patients

Objective To investigate the cancer-related fatigues in oral cancer patients.Methods One hundred oral cancer patients were involved in the survey with self-designed general information questionnaire,revised Piper fatigue correction scale(RPFS).Results Seventy six cases(76.00%)had different degree of fatigue.The total score of RPFS was(5.51+1.23)points. In the descending order of scores,the dimensions were body fatigue,emotional fatigue,behavioral fatigue and cognitive fatigue. Conclusions Oral cancer patients have cancer-related fatigue commonly,with body fatigue the most intense and emotional fatigue at a higher level.Therefore,medical staff should ensure patients intake of enough nutrients in order to reduce the body fatigue and meanwhile should instruct patients to handle their bad moods correctly so as to relieve their metal fatigue.
5.The clinical pathological characteristics of tumor-associated macrophages in esophageal squamous cell carcinoma

Objective To detect the expression of tumor‐associated macrophages(TAM) in esophageal squamous cell carci‐noma ,and to study the clinical pathological characteristics of T AM in the esophageal squamous cell carcinoma .Methods Patients with esophageal squamous cell carcinoma who accepted operation were chosen as study subjects ,and tissue samples of esophagus were collected ,including 90 squamous cell carcinoma tissues ,20 paracancerous atypical hyperplasia and 20 normal mucosa tissues , and the expression of CD206 ,MCP‐1 were detected by immunohistochemisty .Results The positive expression of CD206 was sig‐nificantly increased in tissues of esophageal squamous cell carcinoma (P<0 .01) ,and it was positively correlated with clinical stage , invasion depth and lymph node metastasis in esophageal squamous cell carcinoma (P<0 .05) .The expression of MCP‐1 was signifi‐cantly increased in esophageal squamous carcinoma tissues (P<0 .05) ,and its positive expression was closely correlated with depth of invasion and lymph node metastasis in esophageal squamous cell carcinoma (P< 0 .05) .There was a positive relation between ATM infiltration quantity and the expression of MCP‐1(r=0 .617 ,P<0 .05) .Conclusion The positive expression of TAM was up‐regulated in esophageal squamous cell carcinoma tissues ,and its number was positively correlated with clinical stage ,invasion depth and lymph node metastasis .
6.Clinical analysis of mycoplasma pneumoniae pneumonia with 23SrRNA A2063G gene mutation in 24 cases

Objective To analyse the clinical manifestations of mycoplasma pneumoniae pneumonia(MPP) with 23SrRNA A2063G gene mutation,and improve the ability of diagnosis and treatment of patient infected with MPP.Methods MP-DNA was detected by fluorescent quantitative real-time PCR in sputum specimens from 36 children with MPP,then we detected the drug resistance gene mutation sites by nest-PCR and DNA sequencing,on this basis we classified into two groups of macrolide-resistant MP and macrolide-sensitive MP,and compared the clinical manifestations,laboratory findings,chest imagings and treatment between two groups.Results Of these 36 cases of MPP,24 cases had macrolide-resistant gene mutation with an A2063G transition in domain V of the 23SrRNA,12 cases had no macrolide-resistant gene mutation.Compared to macrolide-sensitive MP group,macrolide-resistant MP group had longer hospitalization duration,longer total cough period,longer total febrile period,longer fever duration after macrolide therapy,longer course of disease,and had higher white blood cells counts and CRP.In the macrolide-resistant MP group,the temperature subsided within 5 days after macrolide treatment alone of 12 cases,3 cases needed switch to fluoroquinolones therapy,10 cases combined with glucocorticoids and 6 cases combined with intravenous immunoglobulin,all 24 patients had good outcomes.While in macrolide-sensitive MP group,the temperature susided between 12 hours to 3 days after macrolide treatment of 8 cases.Conclusions Compared to patients infected by macrolide-sensitive MP,those mycoplasma pneumoniae pneumonia patients with 23SrRNA A2063G gene mutation have longer hospitalization duration,longer total cough period,longer total febrile period,longer fever duration after macrolide therapy,longer course of disease,and have higher white blood cells counts and CRP.Some macrolide-resistant MPP patients have good response to macrolide antibiotics treatment,while the severe cases need combined with glucocorticoids and immunoglobulin,or should change antibiotics.
7.Sodium thiosulfate attenuates high phosphorous induced vascular calcification in rats with 5/6 nephrectomy

Objective To observe the expression of Klotho and Na+/Pi cotransporter in high phosphorous-induced rats with 5/6 nephrectomy and its relationship with vascular calcification,as well as to investigate the effect of early intervention by sodium thiosulfate (STS) on the progression of vascular calcification.Methods Either 5/6 nephrectomy (n=21) or sham operation (n=14) was conducted on 35 Sprague Dawley rats,who were then fed with high phosphorus (HP) diet or normal phosphorus (NP) diet for 16 weeks respectively.The rats were divided into 5 groups as follows:(1) remnant kidney rats receiving HP diet (NHP,n=7),(2) remnant kidney rats receiving NP diet (NNP,n=7),(3) sham operation rats receiving NP diet (SNP,n=7),(4) sham operation rats receiving HP diet (SHP,n=7),(5) remnant kidney rats receiving HP diet with STS (THP,n=7).The treatment group was given STS intraperitoneally three times a week for 16 weeks.At the end of the 16th week,rats tail artery blood pressures were tested,serum creatinine (Scr),calcium (Ca2+),phosphorus (P3+),FGF23,iPTH and urine protein were examined.Throacic aorta and kidney were then removed.Vascular calcification was confirmed by Von kossa staining.Klotho and Pit-1 expression in aortas were determined by immunohistochemistry.Renal lesion was determined by PASM-Masson staining.Renal Klotho and NaPi-2a mRNA were determined by real time RT-PCR.Results After 16 weeks,Scr,p3+,FGF23,iPTH,uric protein and blood pressure were significantly higher in NHP than those in SNP rats (all P < 0.05).PASM-Masson staining revealed typical renal pathology of chronic renal failure in NHP group.With the treatment of STS,THP rats showed significant decrease in Scr,t3+,FGF23,iPTH,uric protein and blood pressure by comparison with NHP group (all P < 0.05).Significant vascular calcification was found in NHP group while NNP and SHP group occasionally had vascular calcification;THP group had marked alleviation of vascular calcification.The aorta and renal expression of Klotho decreased remarkably while expression of Pit-1 and NaPi-2a increased significantly in NHP compared with SNP group (all P < 0.05).Accordingly,the aorta and renal expression of Klotho increased and Pit-1 and NaPi-2a decreased significantly in THP compared with NHP group (P < 0.05).Conclusions The early intervention of sodium thiosulfate might regulate Klotho and Na +/Pi cotransporter expression in both aorth and kidney,decreasing serum phosphate,delaying progression of vascular calcification and improving renal function.
8.Role of anterior thalamic nucleus in trigeminal nerve electrostimulation-induced reduction of seizures and hippocampal damage

[ ABSTRACT] AIM: To investigate the role of anterior thalamic nucleus in trigeminal nerve electrostimulation ( TNS)-induced effects on seizures and hippocampal damage.METHODS: The rats were intraperitoneally injected with pilocarpine to induce chronic epilepsy, and then received sham treatment, TNS treatment and TNS treatment after stereotac-tic lesion to the anterior thalamic nucleus.The TNS treatment lasted for 1 month in each group.Another injection of pilo-carpine was conducted to induce seizures, and the severity and duration of seizures were quantitatively evaluated.TUNEL, Fluoro-Jade B ( FJB) and Nissl staining were applied to determine in situ apoptosis, neuronal degeneration and neuron loss in the hippocampal CA1 area respectively.RESULTS:Compared with TNS group, TNS treatment after stereotactic lesion to the anterior thalamic nucleus significantly increased the severity and duration of seizures (P<0.05), and the numbers of TUNEL positive cells, FJB positive cells and lost neurons in the hippocampal CA1 area (P<0.01).CONCLUSION:Anterior thalamic nucleus plays a role in TNS-induced reduction of seizures and hippocampal damage.The mechanisms might be due to the chronically activation of the cortex through anterior thalamic nucleus pathway induced by TNS, resulting in the down-regulation of neuronal excitatory susceptibility.
9.Effects of N-palmitoylethanolamide on anxiety-and depression-like behaviors in mice induced by chronic restraint stress

AIM:To study the effects of N-palmitoylethanolamide ( PEA) on the anxiety-and depression-like behaviors of the mouse model induced by restraint stress , and to explore the possible mechanism of anxiolytic and antide-pressant effects of PEA .METHODS:The mice were intragastrically treated with 2.5, 5 and 10 mg/kg of PEA for 21 con-secutive days once daily .Thirty min after intragastric administration , the mice ( except the normal control group ) were placed in the glass tube to accept 4-h chronic restraint stress for 21 d.After the last administration , the mice were submit-ted to the forced stress test and the open field test (OFT) to observe the effects of PEA on the depression-like behaviors. The cumulative immobility time was recorded during the 4-min interval in the forced swimming test ( FST) or during the 5-min interval in the tail suspension test (TST).The elevated plus maze (EPM) test was used to investigate the effect of PEA on the mouse anxiety-like behaviors , and the water maze method was used to investigate the learning and memory abi-lities, spatial orientation and cognitive function of mice .After the behavior tests , the serum was collected and the hippo-campus was removed . The serum contents of adrenocorticotropic hormone ( ACTH ) , cortisol ( CORT ) and 5-hydroxytryptamine (5-HT) in the hippocampus were detected by ELISA .The changes of acetylcholinesterase ( AChE) ac-tivity in the hippocampal homogenate was measured by spectrophotometry .RESULTS:Compared with model group , in the FST or TST, the immobility time in the mice treated with PEA at 2.5~10 mg/kg and fluoxetine was significantly reduced . In the OFT, the total locomotion distance and total movement time were increased significantly in the mice , but only 10 mg/kg PEA and fluoxetine increased the numbers of rearing .In the EPM test , the percentage of the time spent in open arms, the entries into open arms and the total locomotion distance in 4 arms in the mice were significantly increased .In wa-ter maze test , PEA at 5 and 10 mg/kg and fluoxetine significantly shortened the latency to find the security zone in the mice, and PEA at 10 mg/kg and fluoxetine obviously shorten the swimming distance .Compared with model group , PEA at 10 mg/kg and fluoxetine reduced the mouse serum levels of ACTH and CORT , and the adrenal index , increased the 5-HT content and decreased the AChE activity in the hippocampus .CONCLUSION:PEA produces antagonistic effects on an-xiety-and depression-like behaviors in the mice induced by restraint stress .Its specific mechanism may be related to the re-gulation of the hypothalamic-pituitary-adrenal axis function by increasing the 5-HT level in hippocampus , thus participating in the regulation of central cholinergic system .
10.FGF-21 protects H9c2 cardiomyoblasts against hydrogen peroxide-induced oxidative stress injury.

Fibroblast growth factor-21 (FGF-21) is an important metabolism regulator, however, whether FGF-21 has effects on cardiovascular remains unclear. In this study, H2O2-induced injury in H9c2 cells was used as a cell model, the anti-apoptosis potential and mechanism of FGF-21 against oxidative injury were evaluated by MTT assay, flow cytometry assay and real-time PCR. The results showed that FGF-21 could increase the cell survival of H2O2-induced injury in H9c2 cells and prevent H9c2 cells from oxidative stress-induced apoptosis. Furthermore, FGF-21 can elevate SOD activity and regulate Bcl-2/Bax expression in H9c2 cells. The results suggest that FGF-21 have protective effect against the H2O2-induced apoptosis in H9c2 cells.
