1.Comparison of ocular biological measurements between type 1 and type 2 diabetic patients with proliferative diabetic retinopathy

Objective To explore the difference of ocular biological measurements between type 1 and type 2 diabetic patients with proliferative diabetic retinopathy.Methods A total of 66 type 2 diabetic patients (67 eyes) with PDR (aged 37-77 years old),10 type 1 diabetic patients (19 eyes) with PDR (aged 18-57 years old) and 35 idiopathic macular hole patients (35 eyes,aged 21-74 years old) from September,2014 to July,2016 in our hospital were retrospectively analyzed,and the differences of axial length,corneal endothelial cell and age among three groups were compared.Results The average axial length of type 1 diabetic patients with PDR was (21.74 ± 0.68) mm,type 2 diabetic patients with PDR was (23.06 ± 0.65) mm,idiopathic macular hole patients was (23.39 ± 0.65) mm,the difference was statistically significant (P < 0.001).There was no statistical difference in average corneal endothelial cells number among three groups (P =0.103),but there was statistical difference in the age (P < 0.001).Conclusion The axial length in patients with type 2 and type 1 diabetic patients with PDR are less than the normal,while type 1 diabetic patient is shorter than type 2 diabetic patient,and the onset time of type 1 diabetic patients with PDR is earlier than that of type 2 diabetic patients with PDR.
2.Silent information regulator 1 regulates the human alveolar epithelial A549 cell apoptosis induced by cigarette smoke extract

Objective To investigate the effect of silent information regulator 1 (SIRT1)on cigarette smoke extract (CSE)-induced apoptosis of human alveolar epithelial cell (AECs).Methods The expression levels of SIRT1 protein were examined by Western Blotting in A549 cells that were treated with CSE at different concentrations.The impairment models of A549 cells induced by CSE were established.The concentration of CSE was 20.0% and the treatment time of CSE was 24 hours.A549 cells were pretreated with 20 μmol/L resveratrol(Res)or 5 mmol/L nicotinamide (NAM) for 2 h before CSE treatment.The protein levels of SIRT1,Bax and Bcl2 were further explored by Western blotting.The proportion of apoptotic A549 cells was measured using MTT.Results The expression of SIRT1 was reduced after treatment with CSE in a dose-dependent manner(P<0.05).Compared with the control group,the group of CSE treatment with 2.5%,5.0%,10.0%,20.0%and 40.0% CSE for 24h showed that the expression of SIRT1 in A549 cells was decreased by 25.6%(P<0.05),35.2%(P<0.05),38.7% (P<0.05),57.9% (P<0.05)and 64.0%(all P<0.05)separately; that A549 cell viability was decreased in a dose-dependent manner; and that A549 cell viability was decreased by 10.2%(t=2.035,P<0.05),18.4%(t=4.269,P<0.05),27.7% (t=5.963,P<0.05),59.0%(t=21.140,P<0.05)and 88.1%(t=58.827,P<0.05)separately.CSE plus 20 μmol/L Res pretreatment reversed the expression levels of SIRT1,Bax and Bcl2 in A549 cells (all P<0.05)and reduced the apoptosis of A549 cells.The effects of CSE on inhibiting SIRT1 pathways were aggravated by NAM (an inhibitor of SIRT1) in the A549 cells (P< 0.05).Conclusions SIRT1 plays important role in regulating the apoptosis of human alveolar epithelial A549cell induced by CSE.SIRT1 may inhibit apoptosis by up-regulating Bcl2 expression and downregulating Bax expression,which has a protective effect on A549 cells apoptosis induced by CSE.
3.Survey and analysis of dental medical students' concerns before and after their clinical internship

ObjectiveTo survey and analyze the changes of dental medical students' concerns before and after their internship in order to provide practical evidences for intern orientation and junior students education.MethodsAnonymous questionnaires were designed with single choice and multiple-choices and were accomplished by 175 dental medical students of different grades to investigate their' concerns before and after the clinical internship and choices for future employment.Results 47% freshmen,43% junior,45% graduates were willing to work in dental restoration field,30%sophomore and 39% senior were willing to work in orthodontics field.Before the internship,the most concem of 97% students was the teaching quality of the practice hospital; 67% students worried that patients may unsatisfy with the treatment.After the internship,the quality of the practice hospital was considered most important by 79% students; 51% students worried that internship may influence the postgraduate entrance exam.ConclusionTeachers should strengthen their self-cultivation and cope with the key concerns before the practice based on the investigation and survey after the practice.Meanwhile teacher should provide employment guidance for students based on students' interests and practice skill ability,encourage them to broaden their horizon and pay attention to the long-term interests in an aim to pave the way for future employment.
4.Effect of postoperative analgesia with difference methods on immunity in patients after thoracic tumour surgery

Objective To investigate the effect of postoperative analgesia with difference methods on immunity in patients after thoracic tumour surgery. Methods Forty ASA Ⅰ-Ⅱ patients aged 35-65 years old undergoing thoracic tumour surgery were randomized to receive either postoperative patient- controlled intravenous analgesia (PCIA) (group Ⅰ, 20 cases) or patient-controlled epidural analgesia (PCEA) (group E, 20 cases) for 48 h. Medicine compatibility in group Ⅰ: sulfentanyl 1μg/ml, tropisetron 0.05 mg/ml, the PCIA pump was set up to deliver a 5 ml bolus dose with a 15-min lockout interval and background infusion at 2 ml/h. Epidual catheter was placed at T4-5interspace before induction of anesthesia in group E. The PCEA solution contained 2 mg/ml ropivacaine. The PCEA pump was set up to deliver a 2 ml bolus dose with a 15-min lockout interval and background infusion at 2 ml/h after a loading dose of 0.33% ropivacame 6 ml. The VAS score, Ramsay sedation score and complications were reeorded. Blood samples were taken before induction (baseline) and at 2 h and 1st, 3rd and 7th day after surgery for determination of plasma concentrations of cortisol, interleukin 2 (IL-2) and the level of natural killer (NK) cells and eytokine-induced killer (CIK) cells. Results There was no significant difference in VAS score at 2 h after operation between two groups [(1.8±0.3) scores in group Ⅰ and (1.8±0.5)scores in group E].Ramsay sedation score at Ist, 3rd and 7th day after operation in group E were significantly lower than those in group Ⅰ (P<0.05), The plasma concentration of cortisol at 2 h and Ist, 3rd, 7th day after operation in group Ewere significantly lower than those in group Ⅰ (P<0.05), the levels of IL-2, NK cells and CIK cells in group E were significantly higher than those in group Ⅰ (P<0.05). Conclusions The efficacy of postoperative PCEA in improving immunity after thoracic tumour surgery is better than that of postoperative PCIA.
5.Progress and application of phosphoproteomics in the proteomics of liver pathological and physiological state.

The phosphorylation is one of most common protein post-translational modifications. The protein phosphorylation plays important roles in the life through the reversible process of phosphorylation and dephosphorylation by kinases and phosphatases. Systematical analysis of the phosphorylation state of proteins would greatly help to reveal the mystery of the life. Recently, with the development of mass spectrometer, bioinformatics sortwares and enrichment methods of phosphopeptides, phosphorylation stduy of orgnism proteins by mass spectrometer has become mature gradually. Liver is one of the most important metabolic and immune organs. In-depth study of protein phosphorylation in liver is of great importance to reveal its function. And booming phosphoproteomics has been applied into the study of liver, which has deepened the knowledge of molecular mechnism of its physiology and pathology states. Here, we review the recent progress on the research and development of phosphoproteomics and their application in liver proteomics study.

6.Molecular Cloning, Recombinant Expression and Functional Characterization of the Soluble Tumor Necrosis Factor-related Apoptosis-inducing Ligand for the Macaca mulatta.

Human tumor necrosis factor-related apoptosis-inducing ligand (hTRAIL) might be developed as a novel anti-tumor drug due to its selective cytotoxicity in tumor cells. The predicted Macaca mulatta TRAIL (mmTRAIL) is highly homologous to hTRAIL in nucleotide acid as well as amino acid sequence, suggesting that mmTRAIL might induce apoptosis of human cancer cells. However, the cytotoxicity of mmTRAIL in human cancer cells has not been investigated. In this paper, it is reported that the gene encoding mmTRAIL has been cloned by using reverse-transcriptase polymerase chain reaction (RT-PCR) from monkey peripheral blood mononuclear cells (PBMCs) in our laboratory. Subsequently, an expression plasmid was constructed by inserting mmTRAIL gene into pQE30 plasmid. After induction by addition of Isopropyl β-D-1-Thiogalactopyranoside (IPTG), mmTRAIL was expressed. MmTRAIL was recovered from supernatant of sonicated bacteria by Ni-NTA agarose affinity chromatography. SDS-PAGE and gel filtration chromatography demonstrated that mmTRAIL forms trimer in solution. In vitro assays indicated that mmTRAIL was cytotoxic to human COLO205 tumor cells but not to normal cells at low concentration of nanomole. In addition, antitumor effect of mmTRAIL was evaluated in mice bearing human COLO205 tumor xenografts. Intratumorally injected mmTRAIL significantly inhibited growth of tumor grafts. These results suggested that mmTRAIL was valuable as candidate drug for cancer-targeted therapy.

7.Multi-slice Spiral CT Study of Liver Graft Regeneration after Living Donor Liver Transplantation

Objective To study the effect of liver transplantation using right liver lobe Ⅴ and Ⅷ segments with middle hepatic vein(MHV) and without main MHV on the regeneration of liver transplantation. Methods 25 pair of donors and receptors of liver transplantation were divided in to two groups:with M HV group (group A, n= 14) and without main MHV (group B, n=11). The volumes of Ⅴ and Ⅷ segments of liver were measured in donors presurgicaUy,half month and 3 month in receptors postsurgically, the rates of regeneration of the Ⅴ and Ⅷ segment were calculated half month and 3 months postsurgically and compared between two groups. Results In group A,the regenerate rates of the Ⅴ,Ⅷ segment were 0.360±0.043 and 0.853±0.059 half month after surgery,0.253±0.043 and 0.708±0.059 three months after surgery respectively. In group B, the regenerate rates of the Ⅴ,Ⅷ segment were 0.306±0.049 and 0.815±0.066, respectively half month after graft, 0.161±0.049 and 0.627±0.066, respectively three months after surgery. There were no statistically significant differences in regenerate rates of the Ⅴ, Ⅷ segment of graft be-tween the two groups (P= 0.685 ,P>0.05 and P= 0.738, P>0.05). Conclusion The right lobe living donor liver transplantation, maintaining the MHV or reconstruct MHV tributaries without MHV has similar effects on the regeneration of Ⅴ,Ⅷ segment of graft.
8.Expressions of natural killer cells, interleukin-27 and CXC chemokine ligand 10 in triple-negative breast cancer and their clinical significances

Objective:To investigate the expressions of natural killer (NK) cells, interleukin-27 (IL-27) and CXC chemokine ligand 10 (CXCL10) in triple-negative breast cancer patients and non-triple-negative breast cancer patients and their clinical significances.Methods:Forty-eight cases of triple-negative breast cancer, 50 cases of non-triple-negative breast cancer and 50 cases of benign breast diseases diagnosed by pathology in the People's Hospital of Xinjiang Uygur Autonomous Region from January 2012 to June 2016 were selected. The expression of NK cells, IL-27 and CXCL10 in the paraffin sections of tissues of the patients after surgery were detected by immunohistochemistry, and the relationships between them and clinical features and prognosis of the patients were analyzed.Results:The positive expression rates of NK cells in triple-negative breast cancer, non-triple-negative breast cancer and benign breast diseases tissues were 81.25% (39/48), 64.00% (32/50) and 26.00% (13/50), respectively, and the difference was statistically significant ( χ2 = 28.229, P < 0.01); the positive expression rates of IL-27 in the three groups were 85.42% (41/48), 76.00% (35/50) and 16.00% (10/50), and the difference was statistically significant ( χ2 = 47.439, P < 0.01); the positive expression rates of CXCL10 in the three groups were 72.92% (35/48), 62.00% (31/50) and 18.00% (9/50), and the difference was statistically significant ( χ2 = 33.422, P < 0.01). In the triple-negative breast cancer group, the positive rate of NK cells was higher in patients with lymph node metastasis ( χ2 = 7.134, P = 0.008), the positive rate of IL-27 was higher in patients with tumor long-axis diameter > 2 cm ( P = 0.034), and the positive rate of CXCL10 was higher in patients with late tumor stage and lymph node metastasis ( χ2 = 8.574, P = 0.003; χ2 = 4.789, P = 0.049). In the non-triple-negative breast cancer group, the positive rate of IL-27 was higher in patients with tumor long-axis diameter > 2 cm ( χ2 = 6.711, P = 0.010), and the positive rate of CXCL10 was higher in patients with high tumor histological grade ( χ2 = 7.621, P = 0.006). The overall survival difference between NK cells-positive and NK cells-negative triple-negative breast cancer patients was statistically significant ( P = 0.047), and the prognosis of NK cells-positive patients was worse. Conclusions:The expressions of NK cells, IL-27 and CXCL10 are related to the occurrence and development of breast cancer. NK cells can be used as a potential prognostic indicator and a clinical treatment target for triple-negative breast cancer patients.
9.Study of craniofacial tissues of northeastern females with bimaxillary protrusion in non-surgical treatment

Objective:To study the craniofacial characters of non-surgical treatment of the female patients with bimaxillary protrusion in northeastern region of China.Methods:80 patients with bimaxillary protrusion in northeastern region of China received orthodontic treatment were divided into two groups,the ANB angles of the first group were no more than 4.7?,the second one exceeds 4.7?.Cephalograms were analyzed in all the subjects.Compared with the normals in Beijing.Results: Most of the female patients with bimaxillary protrusion had shorter anterior cranial bases,longer posterior cranial bases,longer and more prognathic maxillas,longer mandible lengths,higher anterior and anterior upper facial heights,obvious procumbency,thicker mental thickness,procumbent soft tissue profile with a low lip line.Different groups had the features as follows:the main abnormal factors were the maxilla in sagittal aspect of the first group.They had the suitable facial planes.The second group's abnormal factors were the mandible,they had the divergent facial plane.Conclusion:The non-surgical treatment′s patients of bimaxillary protrusion have unique character.The results suggest it is of critical importance to adjust the position of jaws to benefit the improvement of facial features.
10.Effects of dexmedetomidine on expression of spinal matrix metalloproteinase-9 in a rat model of neuropathic pain

Objective To evaluate the effects of dexmedetomidine on the expression of spinal matrix metalloproteinase-9 (MMP-9) in a rat model of neuropathic pain (NP).Methods Eighty-one adult male SpragueDawley rats,weighing 190-230 g,were randomly divided into 3 groups (n =27 each) using a random number table:sham operation group (group S); group NP; dexmedetomidine group (group Dex).The animals were anesthetized with intraperitoneal 10% chloral hydrate 350 mg/kg.The right sciatic nerve was exposed and 4 loose ligatures were placed on the sciatic nerve at 1 mm intervals with 4-0 silk thread in NP and Dex groups.In group Dex,dexmedetomidine 50 μg/kg was injected intraperitoneally once a day starting from the end of operation until the animals were sacrificed.The equal volume of normal saline was given instead of dexmedetomidine in S and NP groups.Paw withdrawal threshold to von Frey filament stimulation (MWT) and paw withdrawal latency to thermal stimulation (TWL) were measured at 1 day before operation (To,baseline) and 5,9 and 16 days after operation (T1-3).Nine animals were sacrificed after measurement of pain threshold at T1-3 and their lumbar segments (L4,5) of the spinal cord were removed for detection of MMP-9 expression (by immuno-histochemistry) and tumor necrosis factor-alpha (TNF-α) content (by ELISA).Results Compared with group S,MWT was significantly decreased,TWL was shortened,and the levels of MMP-9 and TNF-α were increased at T1-3 in NP and Dex groups.Compared with NP group,MWT was significantly increased,TWL was prolonged,and the levels of MMP-9 and TNF-α were decreased at T1-3 in Dex group.Conclusion Dexmedetomidine can inhibit up-regulation of MMP-9 expression,and decrease inflammatory responses,thus attenuating NP in rats.
