Objective To observe the effects of aerosol imiquimod on the level of Interleukin-12 (IL-12) and interleukin-13 (IL-13) in bronchial alveolar lavage fluid(BALF) and serum immunoglobulin E(IgE) level in mouse asthma models.Methods Thirty BALB/c mice were divided into 3 groups, control group, asthma group and imiquimod group, ten in each group.The asthma group and imiquimod group were established by sensitization and challenge with ovalbumin (OVA) ,imiquimod group was interfered with aerosol imiquimod 30 min before challenge.Serum in each group was collected by excising eyeball 24 h after the last challenge.The level of IgE in serum was measured by ELISA.Bronchoalveolar lavage fluid (BALF) was performed, and total cell counts,cell differential were examined.The level of IL-12 and IL-13 in BALF was measured by ELISA and total cell counts, cell differential were examined in BALF.The inflammation changes of lung was observed by HE staining.Results (1) By hematoxylin-eosin (HE) staining, the result of asthma mice were small airway wall thickening ,secretion increased, epithelial cells fall off, arranging disorderly, and submucosal had plenty of inflammatory cells infiltration, mainly with lymphocytes.But imquimod group attenuated the airway inflammation of asthmatic mice.(2) Total cell counts and all kinds of inflammatory cells of asthma group were increased greatly compared with normal group;Eosinophil and lymphocyte cells were much lower in imquimod group than those in asthma group (P ＜0.05).(3) The level of IL-13 in BALF and the level of IgE in serum in imiquimod group was obviously higher than that in control group,IL-13 level were positively correlated with IgE level(r =0.774 ,P ＜ 0.05), but imiquimod group was greatly decreased compared with control group and increased in comparison with that in group A;The level of IL-12 in asthma group was reduced and the level of IgE was increased compared with normal group.IL-12 level in imiquimod group was evidently higher and IgE level was lower than that in asthma group.IL-12 level negatively correlated with IgE level(r =-0.802, P ＜ 0.05).Conclusion Imiquimod aerosol inhalation may up-regulate the level of Th1 chemokine IL-12,down-regulates the level of Th2 chemokine IL-13 and decrease the level of IgE in serum so that it may suppress antigen-induced airway inflammation.

BACKGROUND:Previous studies have confirmed the presence of bis-(3'-5')-cyclic dimeric guanosine
monophosphate signaling pathway in Streptococcus mutans, which construct the streptococcus mutans gcp gene knockout strains.
OBJECTIVE:To compare the gene expression differences between Streptococcus mutans wild strains and gcp mutant strains, and to screen the biofilm-related genes from them for the fol ow-up study.
METHODS:The total RNA of two kinds of strains were extracted and stained with cy3 and cy5 respectively after reverse transcription. The gene chip was scanned after hybridization and the differential gene were obtained
through the data analysis. The different expression genes were verified by real-time PCR.
RESULTS AND CONCLUSION:Differential genes were mainly relative about glucose metabolism and biofilm formation. We selected two genes for real-time PCR verification. The PCR results were consistent with the
microarray results. After Streptococcus mutans gcp gene knockout, the gene expressions of gcp mutant strains were upregulated and the gene expressions of phosphotransferase system were downregulated, this result
suggested that two different genes were related with the c-di-GMP signal pathway downstream.

ObjectiveTo survey and analyze the changes of dental medical students' concerns before and after their internship in order to provide practical evidences for intern orientation and junior students education.MethodsAnonymous questionnaires were designed with single choice and multiple-choices and were accomplished by 175 dental medical students of different grades to investigate their' concerns before and after the clinical internship and choices for future employment.Results 47％ freshmen,43％ junior,45％ graduates were willing to work in dental restoration field,30％sophomore and 39％ senior were willing to work in orthodontics field.Before the internship,the most concem of 97％ students was the teaching quality of the practice hospital; 67％ students worried that patients may unsatisfy with the treatment.After the internship,the quality of the practice hospital was considered most important by 79％ students; 51％ students worried that internship may influence the postgraduate entrance exam.ConclusionTeachers should strengthen their self-cultivation and cope with the key concerns before the practice based on the investigation and survey after the practice.Meanwhile teacher should provide employment guidance for students based on students' interests and practice skill ability,encourage them to broaden their horizon and pay attention to the long-term interests in an aim to pave the way for future employment.

Objective:To study the craniofacial characters of non-surgical treatment of the female patients with bimaxillary protrusion in northeastern region of China.Methods:80 patients with bimaxillary protrusion in northeastern region of China received orthodontic treatment were divided into two groups,the ANB angles of the first group were no more than 4.7?,the second one exceeds 4.7?.Cephalograms were analyzed in all the subjects.Compared with the normals in Beijing.Results: Most of the female patients with bimaxillary protrusion had shorter anterior cranial bases,longer posterior cranial bases,longer and more prognathic maxillas,longer mandible lengths,higher anterior and anterior upper facial heights,obvious procumbency,thicker mental thickness,procumbent soft tissue profile with a low lip line.Different groups had the features as follows:the main abnormal factors were the maxilla in sagittal aspect of the first group.They had the suitable facial planes.The second group's abnormal factors were the mandible,they had the divergent facial plane.Conclusion:The non-surgical treatment′s patients of bimaxillary protrusion have unique character.The results suggest it is of critical importance to adjust the position of jaws to benefit the improvement of facial features.

Objective To observe the expression of Klotho and Na+/Pi cotransporter in high phosphorous-induced rats with 5/6 nephrectomy and its relationship with vascular calcification,as well as to investigate the effect of early intervention by sodium thiosulfate (STS) on the progression of vascular calcification.Methods Either 5/6 nephrectomy (n=21) or sham operation (n=14) was conducted on 35 Sprague Dawley rats,who were then fed with high phosphorus (HP) diet or normal phosphorus (NP) diet for 16 weeks respectively.The rats were divided into 5 groups as follows:(1) remnant kidney rats receiving HP diet (NHP,n=7),(2) remnant kidney rats receiving NP diet (NNP,n=7),(3) sham operation rats receiving NP diet (SNP,n=7),(4) sham operation rats receiving HP diet (SHP,n=7),(5) remnant kidney rats receiving HP diet with STS (THP,n=7).The treatment group was given STS intraperitoneally three times a week for 16 weeks.At the end of the 16th week,rats tail artery blood pressures were tested,serum creatinine (Scr),calcium (Ca2+),phosphorus (P3+),FGF23,iPTH and urine protein were examined.Throacic aorta and kidney were then removed.Vascular calcification was confirmed by Von kossa staining.Klotho and Pit-1 expression in aortas were determined by immunohistochemistry.Renal lesion was determined by PASM-Masson staining.Renal Klotho and NaPi-2a mRNA were determined by real time RT-PCR.Results After 16 weeks,Scr,p3+,FGF23,iPTH,uric protein and blood pressure were significantly higher in NHP than those in SNP rats (all P ＜ 0.05).PASM-Masson staining revealed typical renal pathology of chronic renal failure in NHP group.With the treatment of STS,THP rats showed significant decrease in Scr,t3+,FGF23,iPTH,uric protein and blood pressure by comparison with NHP group (all P ＜ 0.05).Significant vascular calcification was found in NHP group while NNP and SHP group occasionally had vascular calcification;THP group had marked alleviation of vascular calcification.The aorta and renal expression of Klotho decreased remarkably while expression of Pit-1 and NaPi-2a increased significantly in NHP compared with SNP group (all P ＜ 0.05).Accordingly,the aorta and renal expression of Klotho increased and Pit-1 and NaPi-2a decreased significantly in THP compared with NHP group (P ＜ 0.05).Conclusions The early intervention of sodium thiosulfate might regulate Klotho and Na +/Pi cotransporter expression in both aorth and kidney,decreasing serum phosphate,delaying progression of vascular calcification and improving renal function.

The present article aimed at importance of self-efficacy assessment in behavioral change ,self-management capaci-ty ,empowerment capacity and active response in patients with diabetes mellitus (DM) ,introduced research progress of dia-betes self-efficacy assessment scale in China and overseas ,and influencing factors affecting self-efficacy of DM patients , such as baseline data ,well-awareness of blood glucose control ,method and content of health education and social support etc .,in order to promote research and development of Chinese self-efficacy assessment tool .

Objective Through the two D-dimer content detection of the fracture patients and healthy con-trols,and analyze the difference between them ,to explore the correlation between fractures were detected two D-dimer and the importance and the patient′s condition.Methods 306 of fracture patients were collected for determination of D-dimer content were detected at the same time ,100 healthy people as control two D-dimer content;analysis of the correlation between the fracture in 306 cases patients with D-dimer content .Results The plasma D-dimer level of pa-tients with two fracture(10.7 ±2.6)mg/L,which was significantly higher than that of healthy subjects (0.3 ±0.1) mg/L(t=11.07,P<0.01);The fracture number,fracture patients plasma D-dimer detection results content is high-er,the more serious condition .Conclusion The plasma D-dimer level was significantly higher in patients with two fractures,coagulation Stata ,fibrinolytic enhancement ,easily lead to venous thrombosis and disseminated intravascular coagulation in patients with more complications such as fracture;fracture,the higher the content of plasma D-dimer, patients with more serious ,there is correlation .

A case of overseas imported quartan malaria was reported in Weihai City. The patient worked in Africa for many years had no blood transfusion history and had not been to malaria endemic regions of China. In approximately half a month af?ter returning from Africa the patient appeared suspected malaria symptoms such as irregular fever sweating and headache. The patient was diagnosed as quartan malaria by a blood test in basic hospital reviewed with a microscope by Weihai Centre for Disease Control and Prevention and checked through the microscopic examination of malaria diagnosis and reference laboratory and PCR amplification by Shandong Institute of Parasitic Diseases. The patient was cured after the treatment with chloroquine/primaquine for 8 days and did not recur in the 3?month following up.

Objective To investigate the effects of zinc fingers and homeoboxes 3 (ZHX3) silence on expressions of smad3, smad4 and RUNX2 in bone marrow mesenchymal stem cells (BMSCs). Methods ZHX3 low expression vector (ZHX3 silent group) was constructed and was transfected to rat BMSCs. Empty vector was transfected into BMSCs and was used as vehicle control group, and wild type BMSCs was used as the control group. The cell transfection rate was measured under a fluorescence microscope, and then the successful transfection was identified. The immunocytochemistry and immu?noblotting methods were used to detect the expression levels of smad3, smad4 and RUNX2. Results (1) Cells with BMSCs phenotype can be obtained by recovery culturing. (2) After transfection, the green fluorescent protein was found in ZHX3 si?lence group and vehicle control group. Blank control group showed no significant fluorescence. The expression level of ZHX 3 was significantly lower in ZHX3 silence group than that of vehicle control group. (3) Results of immunofluorescence asssay showed that the positive expressions of smad3 and smad4 were located in nucleus and cytoplasm, the positive expression of RUNX2 was mainly located in nucleus. Positive cells were observed in three groups. There was no significant difference in fluorescence intensity between the control group and the vehicle control group, but the fluorescence intensity was significant?ly lower in ZHX3 gene silence group than that of two control groups. (4) There were no significant differences in expressions of smad3, smad4 and RUNX2 betweem control group and the vehicle control group, but they were significantly higher than those of ZHX3 silence group(P < 0.05). Conclusion ZHX3 gene silence can delay vitro osteogenesis of BMSCs, which may play a role by the down-regulated expression levels of smad3, smad4 and RUNX2.

Objective To compare different ways to trace bone marrow mesenchymal stem cells (BMSCs) after being transplanted in cerebral ischemia-reperfusion injury. Methods Male SD rats of SPF grade were randomly divided into sham group, model group (ischemia-reperfusion,IR), BrdU tracing group, PKH26 tracing group and GFP tracing group. Fo?cal cerebral ischemia-reperfusion model was established by blocking middle cerebral artery. 24 hours after cerebral isch?emia-reperfusion injury, 10μL BMSCs that were labeled respectively by BrdU, PKH26, GFP were added respectively into BrdU, PKH26 and GFP tracing group while equal volum of normal saline was added into sham group and model group. Mod?el and transplanting cells efficacy was determined by neural behavioral score, TTC staining and brain water content;Neurons were counted using tar violet staining;The number of transplant cells in the transplanting site was assessed by fluorescence microscopy. Results Before transplanting, there was no significant difference among BrdU, PKH26 and GFP group in cell labeled efficacy. By contrast, neural behavioral score, brain infarct volume and brain tissue water content were significantly lower in all three tracing groups than that in model group 4 weeks after transplantation while neuron counts were markedly higher. There was no significant difference of above parameters among the three tracing groups. However, the number of traced transplanting cells in damaging area in GFP group is significantly higher than that in BrdU group and PKH26 group. Conclusion In cerebral ischemia-reperfusion injury, the tracing effect of GFP last longer, therefore it is significantly more effective than BrdU and PKH26.
