1.Expression of early growth response factor 1 and tissue factor in rats with carotid artery injury

BACKGROUND: DNA enzyme targeting early growth response factor 1 (Egr-1) mRNA (ED5) can inhibit expression of downstream target genes by specificaly inhibiting expression of early growth response factor 1. OBJECTIVE:To investigate the effects of ED5 on the expression of plasma tissue factor after vascular baloon injury in rats and the mechanism of inhibiting neointimal hyperplasia. METHODS: Intimal injury models of the left carotid artery were made in rats. Then, ED5, MgCl2 and FuGene6 were injected into the injured vascular segment. RESULTS AND CONCLUSION:At days 3, 7, 14, 21, the expression levels of Egr-1 and tissue factor in plasma were significantly down-regulated in the ED5 transfection group compared with the MgCl2 and FuGene6 groups (P < 0.01); and neointimal hyperplasia was significantly inhibited by ED5 at days 7, 14 and 21 after modeling (P < 0.01). ED5 may inhibit neointimal hyperplasia folowing baloon injury of rat common carotid artery through down-regulation of tissue factors.
2.The Application of CR Imaging System in Conventional Pantomography Equipment

Objective To introduce a simple and practical digital equipment for photographic children's oral cavity.Methods Using the existing panoura ULTRA pantomography dental equipment,its axletree pole of imaging carrier bracket and the position of the handle of film box were improved,as a result,the traditional intensifying screen film was changed into the IP board carrying digital X-ray imaging to take pictures and using FCR5000R to read the IP board,and then the acquired images were dealt with through terminal processor,and printed with FUJIFILM DRY PIX7000 laser camera to print.Consequently,the clear digital tooth image with high quality were got.Results 200 children were examined randomly with the improved pantomography dental equipment.The excellent image guality was up to 98%.Conclusion The improved pantomography dental equipment is of advantages,and it should be of a promising future in the field of oral cavity clinic,scientific research and teaching.
3.Improvement of Image Carrier Bracket of Orthopantomograph Dental Machine

Objective To introduce a kind of simple and practical digital dental photographic equipment. Methods PanouraULTRA Orthopantomograph dental equipment was adopted to improve its axletree pole of imaging carrier bracket and the position of the handle of film box, so as to make it possible to change the traditional screen film into the IP board carrying digital X-ray imaging to take pictures. By reading the IP board with FCR5000R read implement, dealing with the acquired images through terminal processor, and then using FUJIFILM DRY PIX7000 laser camera to print, a highly clear digital tooth image with high quality would be got. Results Taking Orthopantomograph photos with the improved imaging carrier bracket, successful photos were over 98% of the total. Conclusion This applied machine is characterized by several advantages as follows: appropriate design, convenient operation, low cost, energy-saving and environment protection; high quality of imaging and low quantity of radiation. It can be combined with PACS, which is convenient for distant consultation. This equipment has a promising future in the fields of clinical oral cavity, scientific research and teaching.
4.Investigation about preoperative gastrointestinal cancer patients' quality of life and the related factors

Objective In order to know preoperative gastrointestinal cancer patients' quality of life and the related factors. Methods A cross-sectional study was conducted in 356 preoperative gastrointestinal cancer patients. The data were analyzed using descriptive, linear correlation and multiple regression analysis. Results The total number of valid return was 323, and the valid usable return rate was 90.7%.The preoperative gastrointestinal cancer patients' quality of life reached the middle level and varied with gender, age, residence, income and psychological state. Emotional function, physical function and fatigue were influencing factors of total quality of life. Conclusions Anxiety and emotional function were the main influencing factors of preoperative gastrointestinal cancer patients' quality of life.

6.Research and application of patient satisfaction information system

Objective To establish and apply the statistical information system of patient satisfaction, improve statistical work practice of patient satisfaction. Methods Developed the patient satisfaction research statistic information system which was generated by authority, security settings, the correction and collection of the patient questionnaire, multi-dimension statistics and a copy of data and statistics form through the content and data demand of nursing job satisfaction questionnaire. The system would count up the satisfaction from the whole hospital, every department, every dimension and every item as well as the unsatisfaction item and developed the statistic comparison chart automatically. Results After the formal operation for 6 months, the input time data of patient satisfaction statistic information system decreased from 2.0 days to 0.5 days, the statistic time decreased from 2.0 days to 0.5 days, which saved 6 statistic workers and 3 computers. Conclusions The patient satisfaction statistic information system improves the efficiency and accuracy of data collection dramatically, avoid the waste of human and material resources, achieve the function that data can develop the charts automatically and multi-dimension statistic, which provide the data support for the nursery management work.
7.T type calciumchannel activated by brain injury induces neural stem cell proliferation

ABSTRACT:Objective To investigate the mechanism and induction of T type calcium channel on neural stem cells after brain injury .Methods Adult mice brain injury model was established and divided into control ,sham operation ,surgery ,and surgery+mebefradil groups .Neural stem cells were separated from the subventricular zone (SVZ) and identified .Moreover ,we analyzed the results using MTT and neural stem cell sphere counting after adding different doses of mibefradil in culture medium ,respectively .Then we calculated the half maximal inhibitory concentration (IC50) of mibefradil on neural stem cells .Finally ,we analyzed the expression of Cav3 .2 in SVZ and the protein expressions of Cav3 .2 ,Cyclin A and caspase‐3 in neural stem cells by Western blot .Results In vivo , neural stem cell proliferation was increased in surgery group compared with that in control and sham‐operation groups .The proliferation of neural stem cells in surgery + mibefradil groups was significantly decreased compared with that in surgery group after mibefradil‐induced inhibition of T type calcium channel protein . In vitro , the formation of neural stem cell sphere was significantly inhibited after adding mibefradil .The cell growth ratio was significantly decreased when the concentration of mibefradil was above 5μmol/L .A values in 5 μmol/L ,10 μmol/L and 20μmol/L groups were significantly lower than those in control group (P<0 .05) .IC50 was 8 .93μmol/L .The protein expressions of Cyclin A and Cav3 .2 were inhibited while that of caspase‐3 was increased after mibefradil treatment .Conclusion Neural stem cell proliferation was enhanced by activating T type calcium channel after brain injury .
8.Inhibitory effect of EGCG on apoptosis of Langerhans cells after UVB irradiation

Objective To observe the damage to Langerhans cells induced by UVB irradiation,and to evaluate photoprotective effect of these cells from UVB irradiation by epigallocatechin-3-gallate (EGCG).Methods Biopsy specimens were obtained from normal adult foreskin,and epidermal cells were isolated.Density gradient centrifugation and magnetic cell sorting were used simultaneously to purify Langerhans cells from the cell suspension.These cells were then divided into three groups,control (no ir- radiation or EGCG treatment),UVB (irradiation) and EGCG (irradiation+ECCG treatment) groups. The cells in the UVB and EGCG group were irradiated by UVB (30 mJ/cm~2).After the irradiation,the U- VB group was incubated with RPMI-1640 containing 10% bovine serum for 4 hours,while the EGCG group with the same medium containing 200?g/mL of EGCG for 4 hours.Another four hours after the treatment, the cells were collected for the detection of apoptosis rate by propidium iodide staining and flow cytometry. Results Exposure to UVB (30 mJ/cm~2) significantly increased the apoptotic rate of Langerhans cells.The apoptotic rate in EGCG group was significantly lower than that in the UVB group,but was higher than that in the control group.Conclusion Rate of apoptosis of Langerhans cells could be increased by UVB irradia- tion,while EGCG could prevent the increase of apoptosis.
9.Case-control study on three antithrombotic agents for the prevention of venous thromboembolism after unilateral total knee arthroplasty.

OBJECTIVETo evaluate the efficacy and safety of three antithrombotic agents on venous thromboembolism (VTE) after unilateral total knee arthroplasty.
METHODSFrom November 2011 to March 2014, 149 patients undergoing unilateral total knee arthroplasty for knee osteoarthritis were reviewed. Among them, there were 66 males and 83 females, ranging in age from 48 to 76 years old. All the cases were randomly divided into three groups including Aspirin group, low-molecular-weight heparin (LMWH) group, and rivaroxaban group, according to antithrombotic agents. Deep vein thrombosis (DVT), pulmonary embolism (PE) and bleeding complication (including wound ecchymosis, hematoma and other local complications, gastrointestinal, cardiovascular, urinary hemorrhage and other major bleeding events) of antithrombotic agents were observed and analyzed statistically at the 6 week, 8 week, and 12 week after operation.
RESULTSAmong patients who received Aspirin (48 cases), 4 patients had DVT, in 1 patient had PE, and 2 patients had bleeding complication. Among 54 patients in low-molecular-weight heparin group, 3 patients had DVT, 1 patient had PE, and 3 patients had bleeding complication. While among those patients received the rivaroxaban (47 cases), 3 patients had DVT, 0 patient had PE, and 11 patients had bleeding complication. There were no statistically differences among three groups on DVT, and PE (P>0.05). The incidence of bleeding complication in rivaroxaban group was higher than the other two antithrombotic agents, and the difference among the three groups was statistically significant (P<0.05).
CONCLUSIONAspirin, low-molecular-weight heparin, and rivaroxaban could effectively reduce the incidence of VTE after total knee arthroplasty, and their efficacy was similar. Rivaroxaban has a higher incidence of bleeding complication and further clinical trials are required to be conducted to assess the safety of rivaroxaban in clinical.

10.Expression of pSTAT5 in pancreatic carcinoma cells and effect of growth hormone on pSTAT5

Objective To investigate the expression of pSTAT5 in 7 pancreatic carcinoma cell lines,and the change of expression of pSTAT5 in pancreatic carcinoma cells SW1990 after growth hormone (GH) treatment, and explore its molecular mechanism. Methods Human pancreatic carcinoma cell lines (SW1990, Cap-1, Colo, Mia, AsPc, P3, PANC1) were cultured in vitro, and Western blotting was used to detect the expression of pSTAT5 in these cell lines. SW1990 in exponential growth phase was collected and nude Balb/c mice were inoculated with SW1990 cells. When tumors became palpable after inoculation, mice (normal saline group). 1 h, 2 h and 24 h after the last dose of GH treatment, the mice were sacrificed.Western blotting was used to detect the expression of pSTAT5 in SW1990 and inoculation tumor cells after GH injection. Results Positive expression of pSTAT5 was observed in all human pancreatic carcinoma cell lines (SW1990, Cap-1, Colo, Mia, Aspc, P3, PANC1). 5 minutes after GH (50 ng/ml) stimulation, the expression of pSTAT5 in SW1990 was 0.57 ±0.05, which was significantly increased; and it reached 0.64 ±0.04 at 10 minutes, then decreased to 0.39 ±0.03 at 15 minutes, however, it remained higher than that in the control group at 1 h (0.33 ± 0.02 vs 0.25 ± 0.06), and its expression at 2 h was 0.26 ± 0.03 and returned to the normal level. The expression of pSTAT5 in xenograft was not significantly changed. Conclusions GH could rapidly up-regulate the expression of pSTAT5 in SW1990 but the effect lasted for a relatively short period. GH had no significant effect on the expression of pSTAT5 in xenograft.
