1.Clinical profile and progress of serpiginous choroiditis
Chinese Journal of Ocular Fundus Diseases 2017;33(1):100-103
Serpiginous choroiditis (SC) is infrequent, chronic and posterior uveitis displaying a geographic pattern of choroiditis easy to recur. Studies reveal that the active lesions of inflammatory processes are mainly localized to the choriocapillaris and retinal pigment epithelium cells. SC may manifest with variable features, although a creeping pattern of choroiditis, extending from the juxtapapillary area, with grayish yellow discoloration. Fundus fluorescein angiography, indocyanine green angiography, fundus auto-fluorescence and optical coherence tomography are helpful to diagnose atypical SC. In addition, these image examinations can evaluate the activity and progression of lesion, and detect any complication that might occur. SC is mainly distinguished from multifocal SC related with tuberculosis or virus and etc. Pathogenesis is unclear, an organ-specific autoimmune inflammation or infection seems likely to be the underlying process. It is mainly using glucocorticoid with immunosuppressant therapy at present. Timely and effectively control inflammation can effectively prevent vision loss, choroidal neovascularization and choroidal scar in SC patients.

Objective: To evaluate effects of tRA on activity and content of UCP in rat brown adipose tissue mitochondria. Methods: The rats were divided into tRA treatment and control groups using 3H GTP binded UCP in mitochondria. Radioactivity was measured by liquid aglimmer instrument, the dissociation constant( K d)and maximal binding( B max )was determined from Scathard plot. Results: In tRA treatment rat, B max increased by 1.48 fold, K d remained unchanged, rat body weight loss and brown fat weight decreased, white fat became rosiness. Conclusion: tRA can increase concentration of UCP in brown adipose tissue mitochondria, but do not change its activity,it also can increase fat consumption, produce UCP from white adipose tissue. [

Chimeric antigen receptor-T cell (CAR-T) therapy is one of the effective novel immunotherapy for malignancies, and it has a great effect on leukemia and lymphoma. More researches are needed to improve the understanding of CAR-T therapy and promote the development of this technology in order to treat hematological malignancies from a novel perspective.

Objective To study the localization of specific allergen of Dermatophagoides pteronyssinus. \ Methods\ Through optical microscope, the specific allergens of D.pteronyssinus were observed in paraffin sections using D.pteronyssinus\|specific IgE antibodies from the patient sera. \ Results and Conclusion \ The digestive system was found occupying large parts of body cavity of D.pteronyssinus by HE staining, while the specific allergens of D.pteronyssinus were mostly occurred in the midgut tissue, gut contents, cuticle and reproductive system in the immunostained sections. The results also showed that many parts of D. pteronyssinus were recognized by the specific IgE antibodies obtained from allergic individuals to D.pteronyssinus, which provided a theoretic base for further study of isolation and purification of the specific allergen.

A yeast strain Rhodotorula sp.NJ-298,which was isolated from antarctic sea-ice,could produce carotenoids pigment, and the total production was 1986?g?g~(-1).UV-visible spectrum and LC/MS analysis indicated astaxanthin was the major component of the carotenoids, and HPLC analysis showed its content accounted for 81.9 % of total carotenoids productoin.Maximum astaxanthin production 6506 ?g?L~(-1) was obtained when the yeast was grown at steady-state phase.Optimum temperature for the strain grow was at 8℃,and sodium acetate and peptone were the best carbon and nitrogen sources for the astaxanthin production. Moreover, the pigment had strong activity of scavenging free radicals, and the IC_(50) was 0.40 ?g?mL~(-1) by DPPH assay.

Objective Classification of non-small cell lung lymph (NSCLC) node (N) is one of the key factors influencing treatment, however, the cilinical noninvasive and invasive approaches to N classification have their limitations. This study aimed to investigate the risk factors of lymph node metastasis of peripheral lungadenocarcinoma by using CT and PET / CT scans. Methods Retrospective analysis had been done on a total of 248 patients who underwent surgical resection from February 2010 to November 2015 in our hospital. All of them underwent chest CT and 80 patients underwent PET/CT examination.Univariate analysis was applied in the relation of lymph node metastasis to gender, age, smoking situation, CEA, SUV, cancer size, pathological variants, and the degree of differentiation.Multivariable logistic regression analysiss were performed in the prediction of risk factors for lymph node metastasis.ResultsSeventy-four patients (29.8%) had regional lymph node metastases.Univariate analysis showed that lymph node metastasis was related to the serum CEA level, degree of differentiation, SUVmax, tumor size, lobulation/spiculation, pleural retraction, mediastinal or hilar lymphadenopathy (P<0.05).In the multivariable analysis of risk factors, including serum CEA, SUVmax and CT features, for predicting lymph node metastasis, the most important and significantly independent risk factors identified were SUVmax, CEA level, mediastinal or hilar lymphadenopathy, cavitation/bubble-likelucency and pleural retraction (P<0.05).Conclusion The lymph node metastasis is associated with SUVmax of primary tumor, serum CEA level, mediastinal or hilar lymphadenopathy, cavitation/bubble-likelucency and pleural retraction. The combination of radiographic features and serum CEA can help to predict more accurately the risk of lymph node metastasis in patients with peripheral lung adenocarcinoma.

Objective To identify and investigate B(A)02 allele in a patient. Methods Serological tests were performed with standard serological methods in a patient with B(A)02 allele. DNA sequences of all seven exons and exon-intron boundaries of ABO gene were analyzed by polymerase chain reaction (PCR), direct DNA sequencing and sequencing after gene cloning. In order to analyze the allele, PyMOL software was used to establish 3D model of Glycosyltransferases B (GTB). Results The serological results showed the characteristics of B(A) phenotype. DNA analysis revealed that ABO gene of the individual was heterozygous of B(A)02/O01 allele. 700C>G mutation was identified in B101 allele, which resulted in the amino acid substitution P234A in GTB. Through the analysis of the 3D structure of GTB, it was speculated that the P234A replacement affected the intermolecular forces of the 234 amino acid and Met-266, thus changed the conformation of the donor-binding pocket of GTB,that made GTB capable of recognizing and tranferring the GalNac to the H antigen, which can lead to the formation of the weak A antigen on membrane of red blood cells. Conclusion The P234A replacement can affect the spatial conformation of the specific recognition region conformed by Met-266 and Ala-268 residues, which leads to the antigenicity change of the ABO blood group.
