1. Chemical constituents of alkaloids part of Cephalotaxus hainanensis

Objective To study the alkaloids constituents from Cephalotaxus hainanensis. Methods The column chromatography Sephadex LH-20 and ODS were used to isolate and purify the compounds of alkaloids part of C. hainanensis. The chemical structures were identified on basis of physicochemical properties and spectral data. Results Thirteen compounds were isolated and identified as cephalotaxine (1), isocephalotaxinone (2), cephalotaxinone (3), cephalezomine G (4), (2R,3S,4S,5S)-2,3-dihydroxycephalotaxane (5), acetylcephalotaxine (6), deoxyharringtonine (7), (R)-fortunine (8), (S)-fortunine (9), isomer of 3-epi-schellhammericine (10), 1,4 (PrINH)2-anthraquinone (11), 2-ethyl-n-hexyl benzoate (12), and β-sitosterol (13). Conclusion Compounds 3-6 and 10-13 are isolated from this plant for the first time.
2.Effect of antisense CK13 genes on human nasopharyngeal carcinoma HNE1 cell transplantation tumor

[ABSTRACT]OBJECTIVEThe influnence of observation on the antisense cytokeratin 13 (CK13) gene in nasopharyngeal carcinoma HNE1 cell transplantation tumor radiation sensitivity.METHODSHNE1 cell lines can be divided into control group: the control group (HNE1 cell) and lentivirus (transfection slow virus empty carrier) group and experimental group: HNE1-anti-CK13a (transfection antisense CK13a slow virus) and HNE1-anti-CK13b (transfection antisense slow virus CK13b) four groups, set up a corresponding animal model, after radiotherapy by flow cytometry, immunohistochemistry, PCR, Tunel method and Westernblotting detection.RESULTSThe cell cyclede tection of plasmid transfection slow virus group compared with control group after radiotherapy G2/M phase of the block were significantly prolonged; Immunohistochemical results showed emigration tumor CK13 expression decreased in the experimental group; Tunel method to detect apoptosis necrosis rate found that the experimental group,apoptosis rate significantly decreased; Western blotting detection caspase3 apoptosis markers. PCR to detect CDC25mRNA level decreased obviously.CONCLUSIONAntisense CK13 gene by regulating the cell cycle and apoptosis can reduce HNE1 transplantation tumor radiotherapy of nasopharyngeal carcinoma (NPC) sensitivity, and with the caspase 3 apoptotic pathways and CDC25 signaling pathways.
3.Effect of LFA-1 Deficiency on Differentiation of Mice Na?ve T Cells into Th17 Cells in vitro

BacKground:Lymphocyte function-associated antigen-1( LFA-1 ) pIays a cruciaI roIe in the pathogenesis of infIammatory boweI disease by reguIating the activation and function of T ceIIs. Aims:To investigate the effect of LFA-1 deficiency( LFA-1-/-)on differentiation of mice na?ve T ceIIs into Th17 ceIIs in vitro. Methods:LFA-1-/- mice were breeded and the progeny genome DNA was extracted from the taiIs for genotyping by PCR. CD4+CD62L+ na?ve T ceIIs were separated from spIenic mononucIear ceIIs of LFA-1-/- progeny mice and wiId type ( WT ) C57BL/6J mice, respectiveIy,by magnetic-activated ceII sorting( MACS),and then the purity of separated ceIIs was determined. Na?ve T ceIIs obtained were cuItured in different inducing systems[ transforming growth factor-β( TGF-β),TGF-β + interIeukin-6 (IL-6),and TGF-β + IL-6 + IL-23]in vitro for Th17 ceII differentiation;ceIIs in each inducing system were coIIected for anaIyzing the ratio of Th17 ceIIs by fIow cytometry,and the expressions of Th17-specific transcription factor ROR-γt and Th17-specific marker IL-17A were measured by qRT-PCR and ELISA methods. Results:AII fifteen progeny mice were identified as LFA-1-/- genotype. Purity of CD4+CD62L+ na?ve T ceIIs separated by MACS was above 95%. Th17 ceIIs couId be induced by Iow-dose TGF-βcombined with IL-6,and the differentiation ratio was increased obviousIy when IL-23 was added. In inducing system containing TGF-β,IL-6 and IL-23,na?ve T ceIIs from LFA-1-/- mice produced more Th17 ceIIs than those from WT mice(17. 2% ± 1. 4% vs. 5. 7% ± 0. 2%,P<0. 001),expressions of ROR-γt mRNA and IL-17A mRNA were up-reguIated(P<0. 001),and IL-17A concentration in ceII cuIture supernatant was increased(P<0. 01). Conclusions:Deficiency of LFA-1 promotes the differentiation of mice na?ve T ceIIs into Th17 ceIIs in vitro.

5.Quality Standard Study for Gegenbao Soft Capsule

Objective To study the quality standard of Gegenbao Soft Capsule.Methods The qualitative identification of puerarin and daidzein was carried out by TLC,and the contents of puerarin and daidzein were determined by HPLC.Results Puerarin and daidzein could be identified by TLC with high specificity.The linearity was from 0.214 to 1.42 ? g for puerarin and from 0.033 to 0.221 ? g for daidzein by HPLC,and the average recovery was 101.5 % for puerarin and 96.6 % for daidzein with RSD 1.6 % and 1.8 % respectively(n=6).Conclusion This method is simple,accurate and reliable for the quality control of Gegenbao Soft Capsule.
6.Research and design of mobile medical service platform based on cloud computing

Objective:To design a mobile health service platform based on cloud computing so as to reform the current medical problems including serious waste of medical resources, low level of information sharing and poor feel of medical treatment and so on.Methods: On the basic of analyzing cloud computing and relative theories, a solution of mobile medical cloud based on Google App Engine(GAE) was proposed.Results: A mobile medical health service platform including overall business process and deployment of development environment based on GAE has been designed and the traditional medical service mode has been converted.Conclusion: The exploration and attempt that constructe mobile medical health service platform by combining cloud computing with medical services can promote the conversion of medical mode and provide a effective solution for meeting the requirements of medical service quality and service efficiency for the masses.

8.The influences of volatile anesthetic agents on rat liver mitochondrial electron transfer chain

Rat liver mitochondria were exposed to various conentrations of halothane,enflurane, isoflurane and sevoflurane. Electron transfer rates from NADH and succinate to cytochrome C were measured by scanning dual wavelength spectrophotometer. Statistical analysis of the data suggested that halothane at clinical or higher than clinical concentrations markedly inhibited activities of NADH-Cyt,C reductase.in contrast,no decrease occurred in the activities of NADH dehydrogenase,NADH-coenzyme Q reductase and enzymatical system of succinate chain. Enflurane,isoflurane and sevoflurane had little effect on enzymatical system of mitochondrial electron transfer chain. These data indicate that halothane interfere with utilization of NADH-linked substrate by blocking electon transport from NADH to cytochrome C and it is probable that the locus of action is at Q binding protein(Qpn) or complex of Qpn and ubiquinone.
