Objective To observe hematoxylin's cytocidal and apoptosis-inducing effects on human urinary cancer cell-T24,and its cytocidal mechanism to the target cell. Methods Target cells were incubated in the medium 1640 for 24h,which contained hematoxylin in dosage of zero(blank),12.5,25,50,100,200μg/ml,respectively;under inverted microscopy to observe target cells'morphologic change, and then harvest them;by trypan blue tmpochrome method to determine hematoxylin's cytocidal activity to the target cells;by flow cytomelry to detect the effects of hematoxylin in its different levels on target cells'apoptosis. Results The control group(without hematoxylin)showed their target cells in a fusiform adherent growth,plump,close-arranged,and with a good transparence. With the addition and increment of hematoxylin, target cells turned round, not adherent, pyknotic, with a bad transparence,as well as chromatin condensation, the cells clumped.Cell death rate of control group was(2.63±0.29)%,with the increased dosage of hematoxylin the cell death rate of test groups was(10.00±4.82)%,(21.88±3.42)%,(76.41±4.82)%,(92.27±6.54)%,and(96.34±8.70)%respectively.Flow cytometry showed cell apoptosis rate in control group was 0.47%(occurred spontaneously),but hematoxylin in dose of 50μg/ml made the apoptosis rate increased markedly,to 43.1 8%,dead cell rate 48.47%,and survival cell rate 8.35%.With the increased hematoxylin dose, cell apoptosis rate decreased gradually, while dead cell rate increased. Conclusion Hematoxylin can inhibit the target cell by two routes:to induce apoptose or kill it.In a lower dose it is able to induce target cell to apeptose;hematoxylin in a dose over 100μg/ml can directly kill the target cell.Making this trial for checking the cell's morphologic changes benefits determining the optimal dosage level and optimal acting duration for the apoptosis induction.

Objective Calcium-sensing receptor(CaSR)belongs to the family C of G-protein coupled receptors. This study was carried out to observe the influence and mechanism of CaSR on anoxia/reoxygenation(A/R)-induced cardiomyocytes apoptosis. Methods The model of A/R injury was established through anoxia for 2 hours and reoxygenation for 24 hours in cultured cardiomyocytes of neonatal rats.Cardiomyocytes were randomly divided into three groups:control group,A/R group and GdCl3 group(300μmol/L GdCl3 was added to the culture medium at the beginning of reoxygenation).Apoptosis of cardiomyocytes was assessed by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL).The expression of CaSR,cysteine-requiring aspartate protease(caspase)-3,9 and cytochrom c (Cytc)were analyzed by Western blot. Results The TUNEL showed that cardiomyocytes apoptosis was 17%±3% in A/R group, and was higher than that in the control group. At the same time, expression of CaSR in A/R group was markedly increased in response to control group.Compared with A/R group,GdCl3,a specific activator of CaSR,further enhanced cardiomyocytes apoptosis to (28±4)% and decreased the ability of cardiomyocytes to (51.2±6.8)%,along with an increment in CaSR,caspase-3,caspase-9 and Cytc expressions. Conclusion CaSR is involved in anoxia/reoxygenation-induced apoptosis of neonatal rat cardiomyocytes through Cytc-caspase-3 pathway.

OBJECTIVETo evaluate clinical effect and safety of floating needle therapy and duloxetine in treating patients with persistent somatoform pain disorder (PSPD).
METHODSTotally 108 PSPD patients were randomly assigned to the floating needle treatment group, the duloxetine treatment group, and the placebo treatment group, 36 in each group. Patients in the floating needle treatment group received floating needle therapy and placebo. Those in the duloxetine treatment group received duloxetine and simulated floating needle therapy. Those in the placebo treatment group received the placebo and simulated floating needle therapy. All treatment lasted for six weeks. Efficacy and adverse reactions were evaluated using Simple McGill pain scale (SF-MPQ) and Treatment Emergent Symptom Scale (TESS) before treatment and immediately after treatment, as well as at the end of 1st, 2nd, 4th, and 6th week of treatment, respectively. Hamilton Depression Scale (HAMD, 17 items), Hamilton Anxiety Scale (HAMA) were assessed before treatment and at the end of 1st, 2nd, 4th, and 6th week of treatment, respectively. Patients in the floating needle treatment group and the duloxetine treatment group with the total reducing score rate of SF-MPQ in Pain Rating index (PRI) ≥ 50% after 6 weeks' treatment were involved in the follow-up study.
RESULTS(1) Compared with the same group before treatment, SF-MPQ score, HAMD score and HAMA total scores all decreased in all the three groups at the end of 1st, 2nd, 4th, and 6th week of treatment (P < 0.05, P < 0.01). Besides , each item of SF-MPQ significantly decreased immediately after treatment in the floating needle treatment group (P < 0.01). Compared with the placebo treatment group, SF-MPQ, HAMD, and HAMA total score in the floating needle treatment group significantly decreased after 1, 2, 4, and 6 weeks of treatment (P < 0.05, P < 0.01). SF-MPQ score, HAMD score and HAMA total score in the duloxetine treatment group also significantly decreased after 2, 4, and 6 weeks of treatment (P < 0.05, P < 0.01). (2) There were 3 patients (8.3%) who had adverse reactions in the floating needle treatment group, 17 (50.0%) in the duloxetine treatment group, and 7 (21.2%) in the placebo treatment group. Compared with the placebo treatment group, the incidence of adverse reaction increased in the duloxetine treatment group (χ² = 6.04, P < 0.05). Besides, it was higher in the duloxetine treatment group than in the floating needle treatment group (χ² = 14.9, P < 0.05). (3) There were 19 patients in the floating needle treatment group and 17 patients in the duloxetine treatment group involved in the follow-up study. Compared with 6 weeks after treatment, no significant difference was observed at 3 and 6 months after treatment in the score of SF-MPQ, HAMD, and HAMA in the floating needle treatment group and the duloxetine treatment group. No significant difference was observed between the two groups (P > 0.05). There were 5 patients (29.4%) who had adverse reactions in the duloxetine treatment group, and no adverse reactions were observed in the floating needle treatment group. The adverse reaction rate was significantly different between the two groups (χ² = 4.26, P < 0.05).
CONCLUSIONSFloating needle therapy and duloxetine were effective in treatment of patients with PSPD. However, floating needle therapy could relieve pain more rapidly than duloxetine, with obviously less adverse reactions.

Journal of Korean Academy of Community Health Nursing 2015;26(4):321-329
PURPOSE: This study aimed to explore the prevalence of bullying and to examine the effect of bullying on psychological well-being including depression, self-esteem, and academic major satisfaction among nursing students during clinical training. METHODS: Three hundreds one nursing students who were recruited from three universities in D City were assessed with self-report questionnaires of bullying experience and psychological well-being. Data analyses were performed using the SPSS 21.0 program, which included one-way ANOVA, independent t-test, Pearson's correlation, and multiple linear regression analyses. RESULTS: More than three quarters of the participants experienced bullying during their clinical training, and their experience of being bullied was a significant predictor of psychological well-being even after controlling for perceived academic performance, relationship between nurses and students, teachers' or nurses' help to deal with bullying, and religion. CONCLUSION: Bullying was an issue among nursing students during clinical placement. Bullying experience yielded negative psychological outcomes associated with high depression, low self-esteem, and low academic major satisfaction. Practical guidelines are required in nursing education to protect students from the possible harm of bullying in clinical settings during training.

Objective To study the effects of sensitized dendritic cells in the treatment of bladder tumor and further discuss the mechanism of this immunotherapy. Methods 44 female F344 rats, which irrigated N-methyl-N-nitrosourea into bladders every other week for a total of five doses, were induced to bladder tumor. They were treated subcutaneously with either PBS, unsensitized DC, freeze thawing supernatant of tumor cells, or sensitized DC respectively every week for a total of four times. In the fifteenth week, their bladders were weighted and harvested for observation by naked eye and microscope, their blood was harvested for examination CTL by FCM. Results The weight of bladders in sensitized DC group was lower than those in PBS group, unsensitized DC group and freeze thawing supernatant of bladder tumor cells group (P<0.05). The stages of bladder tumor in sensitized DC group were statistically descended compared with those in PBS group (P <0.05). The CD+3 T cells in sensitized DC group was lower than those in PBS group, unsensitized DC group and freeze thawing supernatant of bladder tumor cells group (P <0.05). The CD+3 CD+8 CD+28 T cells in sensitized DC group was higher than those in PBS group, unsensitized DC group and freeze thawing supematant of bladder tumor cells group(P <0.001). Conclusion Sensitized DC injecting subcutaneously can reduce the stages of F344 rats' bladder tumor, Unsensitized DC injecting subcutaneously has not effect in the treatment of bladder tumor;, while the effect of freeze thawing supematant of tumor cells injecting subcutaneously is not well. The mechanism of sensitized DC in the treatment of blader tumor is that DC plays an immunol killing role by presenting antigen, stimulating CTL.

Objective To investigate the association of interleukin-6 (IL-6) with depression using meta analysis. Methods A systematic literature search was conducted to examine depression and interleukin-6 in Chinese patients us-ing“depression”and“interleukin-6”in Databases including PubMed, CBM, VIP, CNKI and WanFang Data. A me-ta-analysis was performed to analyze correlation between depression and interleukin-6 using RevMan 5.2.8 software. Results A total of 9 studies were recruited, involving 432 depression patients and 277 healthy controls. The results of meta-analysis showed that concentration of IL-6 was higher in depression patients than in healthy controls(SMD=1.48, 95%CI:0.78~2.18,P<0.05). Subgroup analysis in the first-episode subgroup and the non first-episode subgroup showed that concentration of interleukin-6 was higher in depression patients than that in controls [the first-episode subgroup (SMD=1.94,95%CI:0.07~3.81,P<0.05), the non first-episode subgroup (SMD=1.26,95%CI:0.60~1.93,P<0.05)]. Conclusion There is a good correlation between depression and the serum IL-6 concentration. Due to the small number of studies and heterogeneity in different studies, the present meta-analysis has not yet confirmed the quantitative correla-tion between IL-6 and depression.

Chinese Journal of Clinical Oncology 2014;(17):1089-1093
Objective:To investigate the relationship between cancer-related fatigue and cortisol in cancer patients and elucidate the underlying mechanism. Methods:A total of 80 cancer cases were divided into two groups:fatigue group (50 cases with cancer-related fatigue) and non-fatigue group (30 cases without fatigue). The scores were evaluated through the Multidimensional Fatigue Symptom Inventory-Short Form (MFSI-SF) and the Fatigue Symptom Inventory (FSI) report. Serum specimens were examined through electrochemiluminesence immunoassay and enzyme-linked immunosorbent assay. Serum cholesterol was examined through the CHOD-PAP method, and serum total protein and albumin were determined via the Biuret method. Agarose gel electrophoresis was conducted to determine alpha 2 globulin ratio and to calculate serum alpha 2 globulin concentration. Results: The cortisol level in the fatigue group was significantly lower than that in the non-fatigue group[(119.68±5.34) nmol/L vs. (163.45± 31.49) nmol/L, P<0.05], and the adrenocorticotropic hormone (ACTH) level in the fatigue group was significantly higher than that in the non-fatigue group [(104.50 ± 17.15) ng/L vs. (51.43±13.24) ng/L, P<0.05]. Cortisol negatively correlated with MFSI-SF (r=-0.867, P<0.001) but positively correlated with ACTH (r=0.809, P<0.001). Furthermore, cortisol negatively correlated with FSI (r=-0.747, P<0.001) but positively correlated with ACTH (r=0.70, P<0.001). The levels of serum cholesterol [(1.25±0.70) mmol/L vs. (3.28±0.73) mmol/L, P<0.05], albumin[(18.24 ± 7.03) g/L vs. (37.40 ± 8.05) g/L, P<0.05], and alpha-2 globulin [(2.25±1.07) g/L vs. (5.36±1.09) g/L, P<0.05]were significantly lower in the fatigue group than in the non-fatigue group. Conclusion:The patients with cancer-related fatigue exhibited increased MFSI-SF score, decreased serum cortisol level, and enhanced ACTH level. The low serum cortisol levels caused a disorder in the serum ACTH and cancer-related fatigue of malignant tumor patients. The mechanism underlying the reduction in serum cortisol level correlated with the insufficient amounts of serum cholesterol, the composite material of cortisols, and of serum albumin, particularly alpha-2 globulin, the carrier protein of serum cholesterol.

Objective To study the expression and clinical significance of matrix metalloproteinase 2 (MMP-2) in chinese patients with bladder cancer. Methods The relevant original articles published in journals from 1998 to 2014 were selected. The ratio of the case group and the control group (OR value) regarded as an effect index. Then the original data were analyzed using Meta analysis software RevMan 5.0, the combined OR values and 95%confidence interval were calculated, and the forest map was draw. Results 13 studies involving 692 patients that met the inclusion criteria were included in the Meta analysis. The OR values of bladder cancer tissue group and normal bladder tissue group, superficial bladder cancer group and invasive bladder cancer group, and low grade bladder cancer group and senior bladder cancer group were 45.83, 0.22 and 0.31, respectively. The positive rates of MMP-2 in bladder cancer tissue group, invasive bladder cancer group and advanced bladder cancer group were significantly higher than those in normal bladder tissue group, superficial bladder cancer group and low grade bladder cancer group (all P< 0.05). Conclusions The high expression of MMP-2 has a certain effect in bladder cancer. The detection of MMP-2 contributes to the understanding of the biology of bladder cancer, and provides information for clinical treatment and prognosis.
