1.Endoscropic assist in rat orotracheal intubation

Objective To investigate endoscropic assist in rat orotracheal intubation.Methods 40 rats were randomly divided into two groups.The procedure time, total time, intubation frequency,the success rate of the first intubation and the survival rate in 24h were compared between the conventional group and the endoscropic group.Results The total time in two groups were nearly the same.But the procedure time and frequency of intubation were significantly less in the endoscropic group than those in the conventional group.The success rate of the first intubation in the endoscropic group was obviously higher than that of the conventional group.There was no obviously difference between the survival rate after extubation in 24h between groups.Conclusion Endoscropic assist is superior to the conventional method and worth promoting.
2.CHANGES OF CYTOSKELETAL GENES EXPRESSION IN CORTICOSPINAL NEURONS AFTER SPINAL CORD SEMITRANSECTION INJURY IN RAT

Objective To investigate the pattern and the role of cytoskeletal genes expression during nerve regeneration. Methods We examined the changes of expression of ? tubulin(? Tub)and three neurofilament(NF)subunits mRNA in rat corticospinal neurons of sensorimotor cortex on the 1st,3rd,5th,7th,10th,14th,21st,28th and 56th days after spinal cord semitransection injury by using in situ hybridization. Results Levels of ? Tub,NF L,NF M and NF H mRNA were dramatically downregulated within large sized pyramidal neurons in layer V of the sensorimotor cortex of side lateral after spinal cord injury.The level of ? Tub mRNA was decreased as early as 1 day postinjury,whereas the levels of NF subunits mRNA were not decreased until later time(on the 3rd days postinjury),and both of them did not return to control levels on the 56th days following the lesion. Conclusion ? Tub gene expression if inhibited in CNS neurons postinjury. The axotomy signeal and the continuance downregulation of NF mRNA expressioon following spinal cord injury may contribute to the ineffective regeneration response of CNS neurons.
3.ALTERED EXPRESSION OF NEUROFILAMENT GENES IN RAT SENSORY NEURONS OF THE DORSAL ROOT GANGLION AFTER SCIATIC NERVE CRUSH

Objective In order to explore the mechanisms that regulate the expression of genes coding for cytoskeletal proteins during nerve regeneration. Methods In the present study, in situ hybridization was used to examine the changes of light(NF L),medium(NF M)and heavy (NF H)neurofilament protein subunits mRNA in L 4-6 dosal root ganglion (DRG) sensory neurons during nerve regeneration following a unilateral crush of the sciatic nerve. Results The hybridization signals of each neurofilament subunit mRNA were dramatic decrees in DRG sensory neurons post axotomy by light microscope.The signals of NF\|L and NF\|M mRNA were located in cytoplasm of neurons,whereas NF H mRNA was found in both nucleus and cytoplasm of neurons. Conclusion There are the different mechanisms of regulation of neurofilament subunit genes expression,the reduced neurofilament gene expression may represent a general response to axonal injury and plays an important role in effective nerve regeneration.
4.Correlational Analysis of Diastolic Heart Failure and Bone Mineral Density in the Elderly

Objective To study the relationship between the severity of diastolic heart failure(DHF)and bone mineral density in the elderly. Methods Totally 80 elderly patients aged over 80 years who were tested as normal for cardiac diastolic function by Doppler tissue imaging(DTI) were selected and divided into four groups by the e/a ratio,i.e.,the normal group(n=18),the DHF 1 group(0.8≤e/a<1,n=25),the DHF 2 group (0.6≤e/a<0.8,n=22),and the DHF 3 group(e/a<0.6,n=15). And the other 20 healthy people by physical examination were set as the normal control group.All subjects underwent bone mineral density(BMD)measurement(including femoral neck,total femoral hip and lumbar vertebra 1?4) by dual energy X?ray absorptiometry. Results Bone mineral density(BMD)was significantly decreased(P<0.05)in DHF groups(DHF 1,DHF 2,and DHF 3). Bone mineral density significantly decreased along with the severity of DHF. Bone mineral density was positively correlated with the e/a ratio in the elderly with DHF(r=0.75,P<0.01). Conclusion The severity of diastolic heart failure is closely related to bone mineral density in the elderly. The severity of diastolic heart failure could predict osteoporosis.
5.Efficacy of Capparis Spinosa extracts in the treatment of scleroderma:an experimental study in animal models

Objective To observe the effect of ethanol extract and ethyl acetate extract of Capparis Spinosa on the thickness of dermis,synthesis of collagen type Ⅰ,type Ⅲ,and expression of transforming growth factor-β1 in mouse models of scleroderma.Methods Mouse models of scleroderma were established through local injection of bleomycin on the back once a day for 4 weeks.After confirmation of model establishment,72 mouse models were equally and randomly divided into three groups.Two groups received topical treatment with ethanol extract of Capparis Spinosa and ethyl acetate extract of Capparis Spinosa,respectively,no treatment was given to the rest of the control group.After 2-,4-,6-week treatment,8 mice were sacrificed and tissue samples were obtained from the back,and subiected to the measurement of dermal thickness by HE staining,as well as to the analysis of expression of collagen type Ⅰ,collagen type Ⅲ and transforming growth factor-β1 by immunohistochemical staining.Results On week 2,4,6,the thickness of dermis was 23.22,24.94,19.97 μm respectively in mice treated with ethanol extract of Capparis Spinosa,27.66.26.15,22.13 μm respectively in those treated with ethyl acetate extract of Capparis Spinosa.Compared with the mouse models without treatment,the thickness of dermis significantly decreased(F=12.99,P<0.01),the expression of collagen type Ⅰ(F=7.47,P<0.01)and transforming growth factor-β1(F=11.76,P<0.01)were also inhibited in those receiving treatment.However,the expression of collagen type Ⅲ was not affected obviously by the treatment.Conclusion The ethanol extract and ethyl acetate extract of Capparis Spinosa have the effect against skin fibrosis.
6.Myringotomy with Grommet Insertion for Treatment of Tympanic Membrane

Objective To evaluate the efficacy of myringotomy with grommet insertion in the treatment of tympanic membrane atelectasis .Methods From July 2011 to July 2013 ,62 patient with tympanic membrane atelecta-sis were treated with myringotomy with grommet insertion in our hospital ,we evaluated 54 patients(56 ears)whose follow -up period was 1 year or more .Of the 54 patients included in this study ,28 (30 ears))were female and 26(26 ears)were male .The age of the patients ranged from 8 to 54 ,with a mean age of 25 .8 ± 6 .6 years old ,and 18 pa-tients (32% ) were under the age of 16 .The air -bone gap(ABG)was compared at pre -operation and 1 year after post-operation .Results The average ABG of pre-operation and post -operation ,including all patients ,were 27 . 07 ± 5 .03 dB and 15 .52 ± 5 .73 dB ,respectively (P<0 .05) ,and the difference was statistically significant .All the 3 groups ,the ABG of pre- and post-operation ,the difference of each group was statistically significant .Conclusion Myringotomy with grommet insertion was effective in treating tympanic membrane atelectasis .
7.Separation of soybean isoflavone glucosides from Semen Sojae Praeparatum by preparative reversed-phase high performance liquid chromatography

Objective:To search for a rapid and efficient method for the isolation of 3 kinds of isoflavone glucosides from the ethanol extract of Semen Sojae Praeparatum.Methods: The crude isoflavones were extracted by 75% aqueous ethanol after removing the oil by Soxhlet extraction.Then 1300 macroporous resin,water and different concentrations of aqueous ethanol(10%,20%,30%,40%,50%,70%,and 95%) were used to separate and elute the crude isoflavones.The fraction eluted by 40% aqueous ethanol was subjected to preparative HPLC analysis.The chromatographic conditions: A YWG C18(10.0 mm?200 mm,i.d.10 ?m) column was used;the mobile phase consisted of acetonitrile-water-acetic acid at volume ratio of 25751(V/V/V) and a flow rate of 3.0 ml/min;the injection volume was 750 ?l;and the detection wave length was set at 260 nm.Results: Daidzin,glycitin and genistin were rapidly separated with the purities over 99% as determined by external standard HPLC.Conclusion: This technique is simple and suitable for the isolation of daidzin,glycitin and genistin from Semen Sojae Praeparatum.
8.Detection of qacE△1-sul1 Gene in Pseudomonas aeruginosa Isolated from Burned Patients and Its Clinical Significance

OBJECTIVE To investigate the disinfectant-resistant gene qacE△1-sul1 in Pseudomonas aeruginosa isolated from burned patients. METHODS GNS-448 and K-B tests were performed to detect the susceptibility of 19 kinds of antimicrobial agents against these strains. Genotype was analyzed by polymerase chain reaction(PCR) and verified by DNA sequencing. RESULTS The 32 strains isolated were all resistant to ampicillin,cefuroxime,cefoxitin,SMZ/TPM. The sensitive rates to amikacin and gentamicin were 68.0%,and 46.9%,respectively,the resistant rates to imipenem and meropenem were 68.8% and 59.4%,respectively. The positive rate of gene qacE△1-sul1 was 50.0%. CONCLUSIONS The resistance of P. aeruginosa isolated from burned patients is a serious issue.There is high positive percentage of qacE△1-sul1 gene in P. aeruginosa isolated from burned patients.
9.Comparison of sufentanil and oxycodone hydrochloride injection for general anesthesia in breast cancer patients

Objective To explore the efficacy and the safety of oxycodone hydrochloride injection in the induction and maintenance of general anesthesia in breast cancer patients.Methods A total of 200 female patients,aged 25-65 years,ASA physical status Ⅰ or Ⅱ,scheduled for elective breast-conservative surgery,were randomly divided into 2 groups (n=100 each): oxycodone hydrochloride group (group O) and sufentanil group (group S).Oxycodone 0.2 mg/kg (group O) or sufentanil 0.2 μg/kg (group S) with propofol 2 mg/kg and cisatracurium 0.15 mg/kg were administered intravenously for general anesthesia induction.Propofol target-controlled infusion combined with remifentanil were used for maintenance of general anesthesia during the operation.Anesthesia time,awake time,extubation time,total consumption of propofol and remifentanil were recorded.The adverse events and VAS scores after surgery were observed.Results There was no significant difference between groups in anesthesia time,awake time,extubation time,consumption of propofol and remifentanil.Compared with group S,group O had lower VAS score at 2 h after surgery (P<0.05),but with no significant difference at other time points.There was no significant difference at the incidence of moderate pain between group.The incidence of dizziness was 18% in both groups.The incidence of nausea was 11 (11%) in group S and 9 (9%) in group O with no significant difference.Conclusion Taken together,oxycodone hydrochloride used for general anesthesia in breast cancer patient is practicable.
10.Study on reducing blood exposure through protection measures

Objective To evaluate the effect of occupation al protection measures on reducing blood exposure. Methods A s urvey was carried out to investigate medical staff in Shanghai hospitals. Sing le-factor and multi-fa ctor analysis measures were used. Results The more protection m eas ures adopted, such as gloves using, occupational training and strict rules and r egulations, the less occupational exposure. The resul ts also showed that there were statistical difference. Conclusions It is important for medical staff to strength occupational protection in order to avoid acqu iring hospital infection.
