OBJECTIVE To investigate the quinolone-resistance mechanisms of multi-drug-resistant Klebsiella pneumoniae(MDRKP).METHODS Seven kinds of chromosome and plasmid mediated quinolone-resistance genes were analyzed by PCR and verified by DNA sequencing in 25 strains of MDRKP.RESULTS In 25 strains of MDRKP,the positive rate of genes of gyrA,aac(6′)-Ⅰb-Cr,qnrA1,qnrB4-like,qnrS1,mdfA,and qepA were 76.0%,36.0%,8.0%,8.0%,12.0%,100.0%,and 0,respectively.CONCLUSIONS The mutation of gyrA gene is the main cause of the resistance of quinolone in the 25 strains of MDRKP.

Objective To evaluate the anti-osteoporosis effect of the mixtures of epimedin C and icariin monomers with invariant molarity on zebrafish osteoporosis model. Methods The zebrafishes, fertilized 4 d ago, were exposed in 11 groups of nutrient solutions with prednisolone (25 μmol/L) as well as epimedin C and icariin (15 μmol/L) of various contents. The ratio of epimedin C and icariin in the 11groups were as follows: A (10:0), B (9:1), C (8:2), D (7:3), E (6:4), F (5:5), G (4:6), H (3:7), I (2:8), J (1:9), and K (0:10). Meanwhile, a negative control group with prednisolone (25 μmol/L) was prepare as S. The selected zebrafish fetus was put into the 24-hole culture plate, and ensure every 5 zebrafishes in 1 hole and 2 holes as a group. They were placed in incubator at 28.5 °C, and the daily changes of fluid were investigated until they were put to death on day 8 and then fixed. After dyeing with alizarin red, the segmental venter of zebrafish skulls was observed and quantitative analysis of dyed area was conducted. Results Compared with the negative control group S, the integrated optical density (IOD) values of cranial dyed area in all groups increased significantly (P < 0.05); Compared with group S, the IOD value of cranial dyed area in mixtures of epimedium monomers increased significantly (P < 0.05). The mixtures of epimedium monomers were all effective in facilitating zebrafish cranial mineralization and preventing prednisolone from inducing osteoporosis. According to mixtures of A-K groups, zebrafish cranial mineralization gradually decreased with gradually reduced content of epimedin C, with significant difference among groups (P < 0.05). Conclusion The higher the content of epimedin C in mixtures with invariant molarity is, the more active the anti-osteoporosis effect of epimedinC to zebrafish osteoporosis model is.

Liver perfusion technique has been used in drug study for many decades. Liver perfusion has outstanding advantages over other techniques, such as isolated hepatocytes, hepatic cell lines, and hepatocyte membrane vesicles.It is an ideal experimental model used in drug metabolism, drug-drug interactions, and pharmacokinetic studies. Liver perfusion technique, its advantages or disadvantages, and its extensive applications have been reviewed.

Objective To establish a purified model of rat retinal ganglion cells (RGCs) cultured by serum-free medium,and provide a good cell model to investigate the damage of RGCs in glaucoma,retinal ischemia,and degenerative retinopathy. Methods Two monoclonal antibodies,anti-rat SIRP (OX-41) against rat macrophage and antibody against rat Thy-1 (OX-7),were used to purify and characterize RGCs from 1-3-day old Sprague-Dawley (SD) rats by means of two-step filtration.Purified RGCs were cultured in serum-free neurobasal medium containing B27 and ciliary neurotrophic factor (CNTF) meeting the neuronal cell's special requirements.Photomicrographs illustration,immunfluorescence staining of Thy-1,calcein-acetoxymethyl ester (calcein-AM) fluorescence images were used to observe and identify cultured retinal cells and purified RGCs. Results Among the primary cultured rat retinal cells,91% were retinal neurons.Protuberances of RGCs were seen after cultured for 24 hours.At the4th to 8th day,many cells had uniform configuration,large body,and long protuberances. At the 14th day,over 60% cells maintained viability.Immunoflurescence staining of Thy-1 showed the purity of RGCs was about 90%. The results of calcein-AM staining,which stained the living cells only,showed large cell body of RGCs and most of RGCs had a protuberance whose length was twice longer than the diameter of the cells. Conclusion RGCs cultured by serum-free medium has uniform size,good configuration,and high purity,which is adapt to the research of damage of RGCs caused by various factors and to evaluate the protective effects of neuroprotective agents.

AIM: To examine the effects of cigarette smo ke extract (CSE) and lipo polysaccharide (LPS) on the production of transforming growth factor-? 1 (TGF- ? 1 ) mRNA and protein in cultured human embryonic lung fibroblasts (HELF). METHODS: The cultured HELF were incubated with CSE, LPS or CSE in com bination with LP S for 24 hours at 37℃, respectively. The total RNA was extracted from the cells . The expression levels of TGF-? 1 mRNA were evaluated by reverse transcription - p olymerase chain reaction (RT-PCR). The mRNA levels of TGF-? 1 were corrected by GAPDH transcripts, and TGF-? 1 protein levels were determined by immunocytoch emistry. RESULTS: CSE stimulated the TGF-? 1 mRNA expression i n HELF at lower concentr ati ons (1∶50 and 1∶25)( P 0.05). LPS enhanced TGF-? 1 mRNA levels in HELF at a ll three doses (0.1 mg/L, 1 mg/L, and 10 mg/L)( P

Objective To determine the intestinal absorption characters of zingerone in rats.Methods The concentration of zingerone in the intestine perfusate was determined by HPLC.The absorption kinetics were obtained by the in-situ perfusion method in rats.Results The absorption rate constants(Ka) of zingerone were 0.0137,0.0135 and 0.0139 min-1 at the concentration of 25.0,50.0 and 100.0 ?g?mL-1,respectively;Ka of zingerone in the duodenum,jejunum,ileum and colon were 0.0070,0.0031,0.0032 and 0.0026 min-1,respectively.Conclusion Concentration of zingerone has no significant effect on its absorption kinetics.The best absorption segment of the intestine was duodenum,ileum,jejunum and colon by turns.The intestinal absorption of zingerone has the first-order kinetic characters with passive diffusion mechanism.

Objective: In order to observe the therapeutic effect of acupuncture in reducing muscular tension of the limb in apoplectic hemiplegia. Methods:Yin-meridian electroacupuncture was used to treat 37 cases of high muscular tension of limbs in apoplectic hemiplegia. Results:The total effective rate was 83.3% in reducing muscular tension and was 86.5% in increasing the index in the activity of daily life (ADL). Conclusion: The findings show that yin- meridian electro- acupuncture has a good effect in reducing muscular tension and improving the activity of daily living and is beneficial to rehabilitation of paralyzed limb.

Objective To investigate the effect of position ventilation(PPV) on oxygenation,the airway sputum drainage and hemodynamic of acute respiratory distress comprehensive syndrome(ARDS).Methods From June 2015 to April 2016,13 cases with moderate or severe ARDS were researched.The changes of PaO2,PaCO2,SpO2,PaO2/FiO2 and airway sputum drainage were observed as well as HR,MAP,CVP,CO,CI,SVV,etc.Results Totally 56 PPV were taken.Compared with pre-treatment of PPV,the number of PaO2 and PaO2/FiO2 increased from (74.7±13.18) mm Hg and 168.65±22.12 to (92.13±17.82) mm Hg and 208.80±24.04(P<0.05) respectively;SpO2 and airway sputum drainage increased from (95.32±2.76)% and (7.84±1.68)mL to (97.04±1.84)% and (13.25±3.55)mL(P=0.000).However,there were no significant differences in HR,MAP,CVP,CO,CI and SVV(P>0.05).Conclusion PPV can effectively improve oxygenation in patients with ARDS and promote airway sputum drainage,and hemodynamic effects are not obvious.However,nursing intervention still should be strengthened.
