1.Secondary structure and physico-chemical property of fusion proteins: Analysis with bioinformatics network resource

AIM: To analyze secondary structure and physico-chemical property of the fusion protein with bioinformatics network resource, and explore the expression of a secretory anti-osteoblastic carcinoma single-chain bi-functional antibody gene. METHODS: ①The single-chain variable fragment (ScFv) antibody gene and interleukin-2 (IL-2) gene were subcloned into corresponding restriction sites of retrovirus expression vector PLxSN. Mediated by liposome, the recombinant plasmid pL(ScFv-IL-2)SN was packaged with PA317 and selected in G418 to obtain the positive clones, which were able to produce stable retrovirus, and then osteosarcoma (OSC) cells were infected by the recombinant retrovirus, terming OSC/ScFv-IL-2. The virus titer was detected by using NIH3T3.②The transfected OSC9901 cells by ScFv-IL-2 gene were identified by polymerase chain reaction (PCR), reversed transcription-PCR and Western blotting. After the fusion protein was constructed, DNAssist and ANTHEPROT V5 softwares were used to analyze the amino acid sequence, the secondary structure, and the physico-chemical property of fusion protein. RESULTS: ①After the restriction enzyme and PCR identification, the pL(ScFv-IL-2)SN as a fusion protein expression vector, was constructed successfully, and high titer C26 cells were obtained; the expression of recombinant protein was confirmed by Western blotting.②On the fusion genes, the DNA sequence was analyzed with DNAssist nucleic acid sequence analysis software, and their secondary structure and physico-chemical property were analyzed with ANTHEPROT V5. CONCLUSION: The property of fusion protein can be analyzed and forecasted by means of bioinformatics network resources, and the approach may provide evidences for investigating single-chain bi-functional antibody gene.

Meniere's disease (MD), a kind of common disease of otology, is based on the endolymphatic hydrops. The clinical features of MD are intermittent episodes of vertigo, fluctuating sensorineural hearing loss, tinnitus and ear fullness. With the in-depth exploration of the disease, the diagnosis and treatment of MD has made a series of research results. In this paper, the related literature and research reports in recent years were reviewed.

3.Caries-inhibiting effect of topical fluorides for 5 to 6-years old children in two urban districts of Xi’an

Objective:
To explore the effect of a preschool-based fluoride application on early childhood caries (ECC) prevention among 5 to 6-years old children in two urban districts of Xi’an.
Methods:
This cross-sectional study was conducted by using a two-stage cluster method. A total of 564 children aged 5 to 6-years old from eight preschools were recruited for this study. Each participant was assessed dental caries and debris index. Time and materical for topical fluorine application was recorded. Parents/guardians completed a structured self-administered questionnaire, which included demographic, family and social factors (educational level of parents,household monthly income, etc). Children’s oral health related behaviors (age to start brushing, brushing tooth frequency, parents supports in child’s teeth brushing, and whether use fluoride toothpaste); dietary habit (frequency of dessert, desert before sleep, dental discomfort within 12 months, visits to dentist within 12 months). The status of dental caries was shown in the form of the mean dmft and the prevalence of ECC.
Results:
The prevalence of ECC and mean dmft of children was 63.8% and (3.00±3.33) respectively. Among the 294 children aged 5-6 years old, the average intervention time was (4.83±1.00), 33.7% of the children only applied fluorine vanish, 66.3% of the children applied fluorine vanish and fluorine foam. The prevalence of ECC was 53.5%, 63.1% and 68.1% in the children who applied fluorine vanish, fluorine vanish and fluorinated foam, no fluoride, respectively. The mean dmft was (2.45±3.42)(2.83±3.04) and (3.32±3.47), respectively, with statistically significant differences(χ2/F=6.77, 7.08, P<0.05). According to binary Logistic regression equation analysis, the application of fluorine vanish was negatively associated with the prevealence of ECC(OR<1.00). High frequency of dessert intake, low degree of oral hygiene and late start of brushing teeth was positively associated with deciduous teeth caries(OR>1.00). Negative binomial regression analysis of dmft was independent of local fluoride intervention(P>0.05).
Conclusion
The effect of preschool-based fluoride intervention to prevent ECC was limited. It is necessary to improve the effect of intervention for children, carry out targeted health education,encourage parents to intervene eating and brushing behavior of children as early as possible.
4. Effects of microRNA-34a on regulating silent information regulator 1 and influence of the factor on myocardial damage of rats with severe burns at early stage

Objective:
To explore the effects of microRNA-34a on regulating silent information regulator 1 (SIRT1) and influence of SIRT1 on myocardial damage of rats with severe burns at early stage.
Methods:
(1) Twenty-four Sprague-Dawley (SD) rats were divided into sham injury (SI) group, simple burns (SB) group and SIRT1 agonist (SA) group according to the random number table (the same grouping method below), with 8 rats in each group. Rats in groups SB and SA were inflicted with 30% total body surface area full-thickness scald (hereinafter referred to as burns) on the back, and rats in group SI were sham injuried on the back. Immediately after injury, rats in groups SI and SB were intraperitoneally injected with normal saline of 50 mL/kg, and rats in group SA were intraperitoneally injected with normal saline of 50 mL/kg and 1 mg/mL resveratrol of 50 mg/kg. At 6 h post injury, abdominal aortic blood was collected to make serum and myocardial tissue of rats was collected. (2) Myocardial cells of twelve neonatal SD rats were collected and divided into microRNA-34a mimic control (MMC) group, microRNA-34a mimic (MM) group, microRNA-34a inhibitor control (MIC) group, and microRNA-34a inhibitor (MI) group, which were respectively transfected with gene sequences of mimic control, mimic, inhibitor control, and inhibitor of microRNA-34a. The microRNA-34a expression level and protein expression level of SIRT1 in myocardial cells were respectively detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blotting. Another batch of myocardial cells were divided into microRNA-34a inhibitor control+ burn serum (MCB) group, microRNA-34a inhibitor+ burn serum (MB) group, and microRNA-34a inhibitor+ burn serum + EX527 (MBE) group. Myocardial cells in group MCB were transfected with gene sequence of inhibitor control, and myocardial cells in the later groups were transfected with gene sequence of inhibitor of microRNA-34a. After transfection of 48 h, myocardial cells in group MBE were cultured in Dulbecco′s modified Eagle′s medium (DMEM) solution for 6 hours, with serum in group SB of volume fraction of 10% and final amount-of-substance concentration of 1 mol/L, and myocardial cells in the other 2 groups were cultured in DMEM solution with serum from rats of group SB of volume fraction of 10%. The protein expression levels of myocardial cells of SIRT1, cleaved-caspase-3, and Bax were detected by Western blotting. (3) Myocardial tissue from (1) was collected to detect expression levels of microRNA-34a and mRNA of SIRT1 in groups SI and SB by real-time fluorescence quantitative RT-PCR. Morphology of myocardial tissue of rats in groups SI, SB, and SA was observed with biological image navigator. The mRNA expression levels of interleukin 1β (IL-1β) and tumor necrosis factor (TNF-α) of rats in groups SI, SB, and SA were detected by real-time fluorescence quantitative RT-PCR. The expression levels of cleaved-caspase-3, and Bax of myocardial tissue of rats in groups SI, SB, and SA were detected by Western blotting. Data were processed with one-way analysis of variance and least-significant difference test.
Results:
(1) After transfection of 48 h, the expression level of microRNA-34a of myocardial cells in group MM was 4.67±0.92, significantly higher than 1.03±0.04 in group MMC (

OBJECTIVE:
To carry out genetic and metabolite analysis for an infant with cerebral creatine deficiency syndrome type 2 (CCDS2).
METHODS:
Clinical data of the child was collected. Whole-exome sequencing was carried out to identify potential variants by next generation sequencing. Candidate variants were confirmed by Sanger sequencing. Metabolites were determined by tandem mass spectrometry and magnetic resonance spectroscopy. Treatment was carried out following the diagnosis and genetic counseling for the affected family.
RESULTS:
Two novel heterozygous variants (c.289delC and c.392-1G>C) of the GAMT gene were identified in the proband, which were respectively inherited from her father and mother. In silico analysis suggested both variants to be pathogenic. Creatine (Cr) level of the child was very low, and cerebral guanidinoacetate (GAA) level was slightly increased. But both had recovered to normal in two weeks, and cerebral Cr level was significantly improved after two months. Intellectual and motor development of the child were significantly improved.
CONCLUSION
The child was diagnosed with CCDS type 2, for which pathogenic variants of the GAMT gene may be accountable. Treatment has attained a satisfactory effect for the patient.
6.Research progress in the application of hybrid cell membrane biomimetic nano-drug delivery system in cancer treatment

The biomimetic strategy of using the cell membrane-coated nanoparticles can retain the physical and chemical properties of the nanoparticles and show the biological characteristics of the source cell membrane, which can further enhance the role of the nanodrug in tumor treatment. A hybrid cell membrane is the fusion of two or more different types of cell membranes. A hybrid cell membrane can endow nanoparticles with multiple biofunctions derived from the source cells compared with a single cell membrane. Hybrid cell membranes provide a foundation to stimulate extensive research into multifunctional biomimetic nano-drug delivery system (NDDS), which is expected to broaden the application of biomimetic nanotechnology in drug delivery systems. In this review paper, the types of hybrid cell membrane used to construct nano-drug delivery systems, the preparation and characterization methods, and cancer treatment research progress in recent years were reviewed.
7.Calcium-sensing receptor overexpression induces proliferation and differentiation of immature white matter progenitor cells after ischemia in vitro

Objective:To explore the promoted role of calcium-sensing receptor (CaSR) overexpression in proliferation and differentiation of immature white matter progenitor cells after ischemia in vitro. Methods:Periventricular white matter cells from 5-d-old rats were cultured in vitro and divided into control group, oxygen-glucose deprivation (OGD) group, OGD+gadolinium chloride (GdCl 3) group, and OGD+ CaSR silenced group. CaSR expression was agitated by GdCl 3, and CaSR gene expression was inhibited by gene silencing. CaSR mRNA levels 24, 48 and 72 h, and 7 and 14 d after OGD were detected by real-time fluorescence quantification-PCR (RT-qPCR); cell proliferation 48 h after OGD was detected by inverted microscope, and cell differentiation 24, 48, and 72 h, and 7 and 14 d was detected by double immunofluorescence staining. Results:(1) CaSR mRNA expressions: CaSR mRNA expressions in OGD group 48 and 72 h and 7 d after OGD were statistically higher than those in control group ( P<0.05). Forty-eight and 72 h, and 7 and 14 d after OGD, CaSR mRNA expressions in OGD+GDCL 3 group were statistically higher than those in control group and OGD group ( P<0.05); the CaSR mRNA expressions in the OGD+CaSR silenced group were significantly lower than those in the control group ( P<0.05). (2) Cell proliferation and differentiation: 48 h after OGD, the cell sphere diameter of OGD group ([75.26±26.07] μm) was significantly increased as compared with that of control group ([57.96±18.92] μm, P<0.05); the cell sphere diameter of OGD+GdCl 3 group ([91.92±21.82] μm) was significantly increased as compared with that of control group and OGD group ( P<0.05); and the cell sphere diameter of OGD+ CaSR silenced group ([24.09±8.34] μm) was significantly shorter than that of control group and OGD group ( P<0.05). At 48 and 72 h after OGD, the number of O4 +/CaSR + olidoendrocyte precursor cells (OPCs) in OGD group was significantly larger than that in control group, that in OGD+GdCl 3 group was significantly larger than that in control group and OGD group, and that in OGD+ CaSR silenced group was significantly smaller than that in control group and OGD group ( P<0.05). Conclusion:CaSR overexpression could promote the proliferation and differentiation of progenitor cells to OPCs.
8.Effects of psychological intervention based on resourcefulness theory on management of aggression and violence attitude and resilience of emergency nurses

Objective:To explore the effects of psychological intervention based on resourcefulness theory on the management of aggression and violence attitude and resilience of emergency nurses.Methods:Totally 60 nurses who worked in the Emergency Department of the 904 Hospital, the People's Liberation Army Joint Logistic Support Force from January 2020 to December 2021 were selected by convenience sampling. According to the enrollment time, the nurses enrolled from January to December 2020 were included as the control group, and the nurses enrolled from January to December 2021 were included as the intervention group, with 30 nurses in each group. Nurses in the control group received routine psychological training, while nurses in the intervention group was trained with a psychological intervention program based on resourcefulness theory based on the training provided to the control group. The nurses were surveyed and the intervention effect was evaluated by Resourcefulness Scale, Management of Aggression and Violence Attitude Scale (MAVAS), Medical Professionals Resilience Scale (MeRS), and Nurses' Work Alienation Questionnaire (NWAQ) .Results:After intervention, the scores of Resourcefulness Scale, MAVAS and MeRS of nurses in the intervention group were higher than those in the control group, and the scores of NWAQ were lower than those in the control group, with statistical significance between the two groups ( P<0.05) . Conclusions:Psychological intervention based on resourcefulness theory can enhance the resourcefulness of emergency nurses, improve their management of aggression and violence attitudes, strengthen their deep understanding of work violence and their level of resilience, reduce their sense of alienation at work, and alleviate negative psychological emotions. Therefore, it is worth promoting in clinical practice.
9.Effectiveness and safety of laparoscopic-assisted liposuction for treatment of gynaecomastia

Objective:To explore the effectiveness and safety of laparoscopic-assisted liposuction in the treatment of gynecomastia.Methods:A retrospective study was conducted on 115 male breast development patients from January 2021 to May 2023 at the First Affiliated Hospital of Henan University and Shaoguan Hospital of Southern Medical University. The patients were divided into two groups based on surgical methods: the laparoscopic combined liposuction technique group (observation group) and the traditional areola incision group (control group). The control group consisted of 59 cases, aged between 18 and 52 years (26.2±5.2); There were 56 cases in the observation group, aged between 18 and 55 years (26.5±5.2). The differences in surgical time, intraoperative blood loss, drainage tube removal time, incidence of surgical complications, postoperative drainage volume, pain visual analog scale (VAS), and patient satisfaction were compared between two groups of patients.Results:The intraoperative bleeding volume, drainage tube removal time, and postoperative drainage volume in the observation group were 12.25±2.23, 2.85±0.53, and 80.52±7.53, respectively, all of which were lower than those in the control group (26.53±2.35, 4.22±0.59, 81.25±8.54, respectively), and the differences were statistically significant (all P<0.05).The incidence of sensory abnormalities in the nipple areola area of the observation group was 1.8% (1/56), which was lower than the 10.2% (6/59) of the control group, and the difference was statistically significant ( P<0.05).The postoperative breast shape, nipple shape, and incision score of the observation group were 81.15±18.52, 77.85±22.15, and 72.58±10.56 points, respectively, all higher than the control group's 69.34±18.48, 78.12±21.75, and 60.35±9.35 points, and the differences were statistically significant (all P<0.05). Conclusions:Laparoscopic combined with liposuction technology for the treatment of gynecomastia can reduce intraoperative bleeding and postoperative drainage volume and shorten the time for removing drainage tubes with better safety.
10.Dose analysis of hippocampus in T3,T4nasopharyngeal carcinoma patients treated with intensity modulated radiotherapy

Objective To analyze the exposed dose of hippocampus(HC)of T3,T4nasopharyngeal carcinoma patients treated with intensity modulated radiotherapy(IMRT). Methods The bilateral HCs were delineated and were divided into head(HH),body(HB)and tail(HT)for 62 nasopharyngeal carcinoma patients treated with IMRT.The dose parameters of HC were then analyzed. Results The mean dose of left and right HC was(1 127±704)cGy,(1 173±762)cGy. The mean dose of left HH,HB and HT was(1 732±1029)cGy,(820±632)cGy,(423±366)cGy(P=0.000);while the mean dose of right HH, HB and HT was(1 985±1101)cGy,(837±531)cGy,(432±343)cGy(P=0.000).The exposed dose and the volume exposed in different dose of HH were obviously higher than those of HB and HT.The dose parameters of HH,HB and HT decreased in turn. The involvement of sphenoid sinus,ethmoid sinus and cavernous sinus correlated with high exposed dose of HC. Conclusions The exposed dose of HH,HB and HT was different in nasopharyngeal carcinoma patients treated with IMRT.The exposed dose of HH was the highest,which should be emphasized especially. The involvement of sphenoid sinus,ethmoid sinus and cavernous sinus suggest high exposed dose of HC.
