Objective To investigate the effects of high concentration of semen samples in detection by fully automated sperm quality analyzer for sperm concentration, progressive motility(PR),non-progressive motility(NP)and percentage of immotile sperm(IM).Methods The semen samples from the patients who visited the reproductive clinic in our hospital from February 2023 to May 2023 were analyzed using the fully automated sperm quality analyzer.A total of 155 semen samples with initial sperm concentration ≥100× 106/mL and semen volume ≥1.5 mL were set as the high concentration semen group(group A).The semen samples with self seminal plasma dilution group(group B)and 37 ℃ normal saline dilution group(group C)were prepared with their own seminal plasma and normal saline at 1∶2 dilution ratio respectively. The sperm concentration of group A was detected by the manual method as the control. The sperm concentration and motility parameters of each group were detected. The differences of sperm concentration and the values of PR,NP and IM among the group A,B and C were analyzed. The correlations between sperm concentration and PR,NP and IM were also assessed. Results The sperm concentration of group A was significantly lower than that of the control group and groups B and C(all P<0.05).There was no statistically significant difference for the results between group B and group C(P>0.05).The PR in group C was significantly lower than that in groups A and B(all P<0.05),and there was no significant difference of PR between group A and group B(P>0.05).The values of NP in group A was significantly higher than that in groups B and C(all P<0.05),and there was no significant difference in NP between group B and group C(P>0.05).The IM in group A was significantly lower than that in groups B and C(all P<0.05),and there was no statistical difference of IM values between group B ang group C(P>0.05).Conclusion The direct analysis of undiluted semen samples with high sperm concentration may lead to lower results of sperm concentration, increased percentage of NP sperm and decreased percentage of IM sperm. The dilution of semen samples may improve the accuracy of the detec-tion results. The dilution with 37 ℃ normal saline can lead to decrease of the percentage of PR sperm. The self seminal plasma should be recommended as the first choice of diluent for the semen samples with high sperm concentration.

Journal of Public Health and Preventive Medicine 2022;33(2):61-64
Objectives To investigate the sanitary condition of the secondary water supply for passenger trains in the administration area, to evaluate the influencing factors of the secondary water supply system for passenger trains, and to put forward suggestions for improving the water quality of the secondary water supply for passenger trains. Methods The water quality of secondary water supply of 48 trains of 24 pairs of CRH type passenger trains and 160 trains of 10 pairs of type 22 passenger trains and 30 pairs of type 25 passenger trains in Lanzhou Railway Bureau in 2019 was analyzed and compared. Results The qualification rate of 208 samples tested was 73.08%. The qualification rate of secondary water supply quality of type 22 and 25 passenger trains was 62.50% and 66.67%, respectively, and the difference was not statistically significant(χ2=0.23 ,P>0.05). The qualification rate of secondary water supply of CRH passenger train was 97.92%, which was statistically significantly different compared with that of type 22 and 25 passenger trains(χ2=18.40, 18.15, P<0.05). The qualification rate of secondary water supply quality of type 22 and 25 passenger trains in different seats was 65.00%, 67.50%, 62.50% and 67.50%, respectively, and the difference was not statistically significant (χ2=0.056, 0.054, 0.056, 0.22, 0, and 0.22, P>0.05). The total number of bacterial colonies, total coliform, turbidity and iron in the secondary water supply of type 22 and 25 passenger trains did not meet the standard, while only one water sample for the secondary water supply of CRH passenger trains did not meet the standard. Conclusion The quality of secondary water supply of CRH passenger train is obviously better than that of type 22 and 25 passenger trains. Changing the position of defecation port and water injection pipe of type 22 and 25 passenger trains, selecting non direct train defecation collection system, or adding appropriate amount of chlorine disinfectant into the water supply hose can significantly improve the qualification rate of bacteriological indexes of secondary water supply of passenger trains.

Objective To investigate the protective effect and molecular mechanism of bloodletting acupuncture at Jing-well points on acute high-altitude hypoxia brain injury through regulating PI3K/AKT/mTOR signaling pathway-mediated mitochondrial autophagy, and to provide an effective target and theoretical basis for the clinical use of bloodletting acupuncture at Jing-well points to prevent and treat acute high-altitude hypoxia brain injury. Methods Male SD rats were randomly divided into Control group(n=15)and experimental group, and the experimental group was divided into Model group(n=15),Bloodletting Acupuncture at Jing-well Points of hand group(BAJP group,n=15),Bloodletting Acupuncture at Non-Acupoint group(BANA group,n=15).The low pressure oxygen chamber was depressurized to 6000 m altitude, and the rats in each experimental group were treated with low-pressure hypoxia for 72 h to replicate the acute high-altitude hypoxia brain injury rat model. The rats in the BAJP group were bled according to the order of"Shaoshang"(LU11),"Shangyang"(LI1),"Zhongchong"(PC9),"Guanchong"(SJ1),"Shaochong"(HT9),"Shaoze"(SI1),once a day for 7 days. The rats in the BANA group were bled by cutting the tail tip daily, and the amount of blood bled was 15-20 μL in both groups. The expression levels of PI3K,AKT and mTOR in hippocampal tissues of rats were detected by Western blot;AKT and mTOR mRNA expression levels in hippocampal tissues of rats were detected by PCR.Results Compared with the Control group, the number of degenerative necrotic vertebral cells in CA1 area of hippocampal tissue, swelling of mitochondria, appearance of autophagosomes, and increase of apoptosis in hippocampal tissue of Acute High-altitude Hypoxia(AHH)rats were significantly increased; After bloodletting acupuncture at Jing-well points of hand treatment, various brain injury manifestations in AHH rats were alleviated;Bloodletting acupuncture at non-acupoint had no significant ameliorating effect on AHH rats′ brain injury.Western blot detected a significant decrease in the phosphorylation levels of PI3K,AKT,and mTOR in the hippocampal tissues of AHH rats compared to Control group rats(P<0.01),and the phosphorylation levels of the three molecules were further decreased after bloodletting acupuncture at Jing-well points of hand treatment(P<0.01),and bloodletting acupuncture at non-acupoint treatment did not have significantly affect on the phosphorylation levels of these molecules(P>0.05),and AKT,mTOR mRNA expression levels further demonstrated the above trend. Conclusion Bloodletting acupuncture at Jing-well points of hand can play a protective role against acute high-altitude hypoxia brain injury with points specificity, and the mechanism may be related to the inhibition of PI3K/AKT/mTOR pathway to promote the elevated level of mitochondrial autophagy, improve mitochondrial physiology, and enhance the body′s ability to resist apoptosis and hypoxia.
