1.Effects of P-selectin gene knockout on the biological characteristics of mice

Objective To investigate the effects of P-selectin gene knockout on the biological characteristics of mice. Methods P?selectin knockout (PKO) mice and C57BL/6 mice were housed in SPF environment. To make sure that the mice were pure PKO mice by gene identification. To determine the general condition, the activity status, reproduction of P?selectin knockout mice were observed and tested. HE staining was used to observed the histological morphology and struc?tures of 6?week?old and 18?week?old P?selectin knockoutout mice. Results PKO mice and C57 mice have similar multiplica?tion, there was no significant difference in reproduction and blood routine test. The stuctures of liver, spleen, lung and kid?ney were normal. Conclusions P?selectin knockout has no significant effects on reproduction, blood routine test and major organs. This research provides animal model theoretical basis for further study on the effects of P?selection in diseases.
2.Testing of anisotropic modulus and fluid characteristic of Chinese femur and tibia.

The tension moduls and compression modulus of Chinese tibia and femur of male in three different directions, namely axial, circumferential and radial directions, were tested in our experiments. The measured compression moduli are 5.8 +/- 0.34, 3.4 +/- 0.17, 2.1 +/- 0.18 GPa respectively for femur, and 6.1 +/- 0.36, 3.5 +/- 0.17, 2.1 +/- 0.18 GPa respectively for tibia; the measured tension moduli are 15.6 +/- 1.21, 9.8 +/- 0.73, 5.4 +/- 0.52 GPa respectively for femur, and 16.7 +/- 1.09, 10.3 +/- 0.70, 5.6 +/- 0.49 GPa respectively for tibia. The tension and compression ultimate strengths of femur and tibia were tested, too. The measured compression ultimate strengths are 150 +/- 9.3, 111 +/- 6.5, 110 +/- 7.1 MPa respectively for femur, and 191 +/- 12.3, 140 +/- 6.3, 123 +/- 8.3 MPa respectively for tibia; the measured tension ultimate strengths are 115 +/- 9.6, 82 +/- 6.8, 78 +/- 7.3 MPa respectively for femur, and 143 +/- 8.7, 110 +/- 8.3, 97 +/- 6.4 MPa respectively for tibia. Furthermore, the stress-relaxation experiments were conducted. The relation between loading and time during the experiments were obtained by the method of curve fitting. They meet the logarithm function: F = A * B(log t); A is the initial load, and B is the fitting material parameter. For femur, B is equal to 0.985; for tibia, B is equal to 0.987.

3.Effect of P-selectin Deletion on Intestinal Tumorigenesis in 15,24-week-old ApcMin/+ Mice

[Objective] To investigate the effect of P-selectin on the intestinal tumorigenesis in the 15,24-week-old ApcMin/+ mice.[Methods] Male ApcMin/+ mice were mated with female P-selectin knockout (P-sel-/-) mice to generate AMin/+Ps-/-mice.The diameters and numbers of the intestinal tumors in the intestines of ApcMin/+ and AM/+ps-/-mice were measured using an inverted microscope.The tumor volumes were measured using the following equation:volume =0.52 × (length × width2).[Results] There were no significant difference between the volume and number of intestinal tumor in 15-week-old ApcMin/+ mice and 15-week-old A+ P-mice.The volume and number of intestinal tumor were significantly increased in 24-week-old A+P-mice compared to 24-weekold ApcMin/+ mice.P-selectin deletion significantly prolonged the survival time of ApcMin/+ mice.[Conclusions] P-selectin deletion promotes the intestinal tumorigenesis in the 24-week-old ApcMin/+ mice.
4. Establishment and analysis of predictive models of postoperative complications in elderly patients undergoing hip arthroplasty

Objective:
To establish a preoperative evaluation model and evaluate the accuracy of postoperative complications by analyzing the risk factors of postoperative complications in hip arthroplasty in elderly patients, and to provide a basis for grading nursing.
Methods:
From January 2012 to August 2018, 148 patients with elderly patients undergoing hip arthroplasty admitted to People′s hospital of Wanning were included in the study. Among them, 23 patients with postoperative complications were included as complications and 125 patients without complications served as a control group. Univariate analysis of the factors associated with complications, statistically significant indicators were included in the multivariate logistic regression model, and the risk factors for postoperative complications in hip arthroplasty were analyzed. Based on the results of the regression analysis, a preoperative evaluation model was established. The ROC curve and the Hosmer-Lemeshow test were used to evaluate the model prediction performance.

Objective To study the effect of atrial natriuretic peptide(ANP)deletion on melanoma growth. Methods A subcutaneous xenotransplanted melanoma model was established in ANP knockout mice and C57BL/6J mice. The tumor volume was measured at the sixth day after establishment of the subcutaneous transplantation. Tumor cell proliferation and tumor-induced angiogenesis were detected by immunohistochemical staining. Results The volume and weight of xenotransplanted melanoma were significantly decreased in the ANP knockout mice compared with those in the C57BL/6J mice. The proliferative tumor cells and microvessel density were significantly decreased in the tumor tissues of ANP knockout mice compared with those in the C57BL/6J mice. Conclusions ANP deletion significantly suppresses xenotransplanted melanoma growth through inhibiting the tumor cell proliferation and angiogenesis.
6.Effect of atrial natriuretic peptide ( ANP) on pulmonary melanoma metastasis in ANP knockout mice and C57BL/6J mice

Objective To investigate the role of atrial natriuretic peptide (ANP) in lung metastasis of melanoma. Methods Melanoma B16F10 cells were intravenously injected into ANP knockout mice and C57BL/6J mice. After three weeks, the mice were sacrificed, and the lungs were removed, embedded, and examined by pathology using HE staining. The numbers of metastatic foci on the lung surface and micrometastatic foci in the lung tissues were counted. Results The ANP knockout mice displayed fewer metastatic foci on the lung surface and less micrometastatic foci in the lung tissues of ANP knockout mice than in the C57BL/6J mice. Conclusions ANP deletion significantly suppresses the metastasis of melanoma in the lung of mice.
7.Development of a functional mandibular model.

This study was conducted to establish a functional mandibular biomechanical model for use in the follow-up biomechanical study of the integrated and fractured mandible. The integrated and dry human mandible was used, and the corresponding maxilla and cranial base was duplicated by resin and plaster. 2-mm silicon rubber was used for simulation of the temporomandibular joint (TMJ) disc. A simulated TMJ and physiological mandibular model was developed by four pairs of muscular loadings (Masseter= 180 N, Temporalis = 190 N, Medial Pterygoideus = 120 N, Lateral Pterygoideus = 40 N) in each muscular center, and the functional loading corresponding with physiological condition was reflected and simulated more realistically when compared with that of the previously reported mandibular models which were developed by occlusion loading or by only one pair of muscles loading. In summary, we have established a functional mandibular model which can be used to analyze the biomechanical behavior in various functional conditions.

8.Schisantherin B Improves the Pathological Manifestations of Mice Caused by Behavior Desperation in Different Ages-Depression with Cognitive Impairment

Depression is a major mood disorder. Abnormal expression of glial glutamate transporter-1 (GLT-1) is associated with depression. Schisantherin B (STB) is one bioactive of lignans isolated from Schisandra chinensis (Turcz.) Baill which has been commonly used as a traditional herbal medicine for thousands of years. This paper was designed to investigate the effects of STB on depressive mice induced by forced swimming test (FST). Additionally, we also assessed the impairment of FST on cognitive function in mice with different ages. FST and open field test (OFT) were used for assessing depressive symptoms, and Y-maze was used for evaluating cognition processes. Our study showed that STB acting as an antidepressant, which increased GLT-1 levels by promoting PI3K/AKT/mTOR pathway. Although the damage is reversible, short-term learning and memory impairment caused by FST test is more serious in the aged mice, and STB also exerts cognition improvement ability in the meanwhile. Our findings suggested that STB might be a promising therapeutic agent of depression by regulating the GLT-1 restoration as well as activating PI3K/AKT/mTOR pathway.

9.Effects and mechanism ofandrographolide on diabetic nephropathy mice

To investigate the effects and mechanism of andrographolide(Andro)on type II diabetic nephropathy in Leprdb/dbmice,levels of urinary protein,serum creatinine and blood urea nitrogen after 24 h of Andro(1 mg/kg,ip)treatment on Leprdb/dbmice were measured by kit.The changes on renal pathology and sugar levels in diabetic mice treated by Andro were observed by HE staining and PAS staining.The expression of fibronectin (FN)and NOX-4 protein were analyzed by immunofluorescence.Results showed that levels of urinary protein, serum creatinine and blood urea nitrogen in Leprdb / dbmice after Andro treatment for 24 h were significantly lower than those of the control group. Kidney pathologic change was inhibited, and FN and NOX-4 expression were markedly decrease in the Andro group.In conclusion,Andro can significantly reduce the diabetic nephropathy in Leprdb/dbmice with spontaneous type II diabetes mellitus.
