1.Rules analysis of acupoint selection on perimenopausal syndrome treated with acupuncture and moxibustion.

With the keywords of "acupuncture" "acupuncture and moxibustion" "perimenopausal syndrome" "rules of acupoint selection", the clinical literature in recent 10 years regarding perimenopausal syndrome treated with acupuncture and moxibustion was searched from China National Knowledge Infrastructure(CNKI). One hundred and ninety-seven relevant articles were acquired,and after excluding, total 67 articles were in accord with research criterion. With method of artificial count, acupoints selected in the inclusive articles were counted. Frequencies of main acupoints, matching acupoints,meridial distributions and locations of the acupoints selected were calculated,and then rules on main acupoints, matching acupoints, meridial distributions and acupoints locations were analyzed. The result indicates that the acupoints with higher selection frequency on perimenopausal syndrome treated with acupuncture and moxibustion are Sanyinjiao(SP 6), Baihui(GV 20),Taichong(LR 3) Shenshu(BL 23), Guanyuan(CV 4), Shenmen(HT 7),Taixi(KI 3), Zusanli(ST 36), Pishu(BL 20) and Ganshu(BL 18), and the meridians are mainly concentrated on governor vessel, bladder meridian, kidney meridian,stomach meridian, conception vessel and spleen meridian,and the distribution of acupoints are mostly concentrated on the 4 limbs and the back and waist.

3.Networked health information on diabetes: layer analysis

Objective Networked health information on diabetes updated by some websites was evaluated to provide evidence for its improvement.Methods Twenty-four domestic diabetes-related websites were identified,and primary index (media index,health information index and diabetes information index) and secondary index (practicability index and novelty index) were evaluated by using hierarchal analysis method.Results Of three primary index,compared with media and health information index,diabetes information index showed higher scores.Moreover,health information and diabetes information had strong causality.For the secondary index,practicability and novelty got relatively lower scores.The website Diabetic Club and Health for All got the highest (3.2) and the slowest scores (1.7),respectively.Conclusion At present,domestic diabetes-related websites may be far from satisfaction and need improvement in technology and human resources and more inputs.

5.Progress of surgical approach to hysterectomy

Nowadays,with the introduction of the concept of minimally invasive surgery,laparoscopic surgery which occupied the mainstream operation method,vaginal surgery,and abdominal surgery were three main ways of gynecological surgery.Hysterectomy operation method as the routine operation of gynecology was changed as well.Laparoscopic hysterectomy has replaced transvaginal hysterectomy and transabdominal hysterectomy gradually.Historical development,operation indication,and advantages and disadvantages of hysterectomy are summarized in this article.
6.Effects of lentiviral delivery in different brain regions on transfection of rat dentate gyrus neurons

Objective To explore the effect of lentiviral delivery green fluorescent protein in different brain regions on the transfection of dentate gyrus neurons.Methods Lenti-pSyn-EGFP was stereotaxic injected into dentate gyrus (DG),Cornu Ammonis 1 (CAl) of hippocampus and medial entorhinal cortex (EC) of rats.All animals were perfused with 4% paraformaldehyde.Their brains were cut into 30 μm sections which were performed with Nissl fluorescent staining.Results The resuts showed that the GFP positive cells in DG area of DG,CA1,EC,DG-EC groups were (18.0± 1.0),(17.0±0.6),(17.3±0.6),(18.3±0.6) respectively,and there was no statistical difference among each group(P>0.05).Conclusion Lentivirus delivery in EC and CA1 both can transfect DG neurons.This is a useful method to transfect dentate gyrus neurons instead of injecting lentivirus into DG directly and avoid the lesion of DG.
7.Construction of recombinant adenovirus vector pAV.Ex1 d-Hiwi using Gateway technology

Objective To obtain the Hiwi gene encoding full-length and construct human Hiwi adenoviral vectors carrying green luorescence protein (GFP), and to establish foundation for a further study on Hiwi function and mechanism of inducing leukemia stem cell differentiation and apoptosis.Methods All coding areas of human Hiwi gene full length were amplified using method of overlapping extension PCR technology, and the full length coding aeras were inserted into the vector of Flag-IRES-hrGFP carrying GFP with Gateway technology to construct pDown-Hiwi-3 × flag-IRES-hrGFP. The cloning vector pDown-Hiwi-3 × flag-IRES-hrGFP and expression vector pAV. Des1d were used for homologous recombination reaction to obtain recombinant adenovirus vector pAV.Ex1d-Hiwi-3× flag-IRES-hrGFP.The positive clones were selected by PCR to extract the recombinant adenovirus plasmid and to pack into recombinant Ad-Hiwi-3 × flag-IRES-hrGFP adenovirus. Results The human recombinant Hiwi was successfully cloned and the recombinant adenovirus vector pAV.Ex1d-Hiwi-3×flag-IRES-hrGFP was found to be successfully constructed via restriction enzyme digestion and sequencing methods. The adenovirus vector pAV.Ex1d-Hiwi-3×flag-IRES-hrGFP was transfected into the HEK293A cells using lipofectamine mediated gene transfection method. Under fluorescence microscope, the transfected cells with green fluorescence could be observed.Conclusion The expression plasmid of adenovirus vector pAV.Ex1d-Hiwi-3 × flag-IRES-hrGFP is successfully constructed and it can express in HEK293A cells.
8.High-performance capillary electrophoresis method for ferulic acid assay in serum of rats treated with Buyang Huanwu Decoction

Object To develop a high-performance capillary electrophoresis method to determine ferulic acid concentrations in serum of rats treated with Buyang Huanwu Decoction (BHD). Methods Capillary zone electrophoresis was applied for ferulic acid assay, quantitative determination was based on internal standard and detection was carried on by direct UV. The electrolyte buffer was composed of 25 mmol/L borax-methanol (85∶15). Capillary electrophoresis was performed using a 52 cm (30 cm to detector)?50 ?m fused-silica capillary tube. Separation voltage was 20 kV, sampling time was 3 s, detection wavelength was 320 nm, and the temperature was 30 ℃. Results Ferulic acid was successfully separated within 6 min, the recoveries were 97.2% in serum and 103.28% in BHD, respectively. RSD were 3.73% and 0.91% (n=3), respectively. Conclusion This method can supply reference for the determination of ferulic acid in serum samples and BHD.
10.Influence of miR-21 on radiation sensitivity of cervical cancer HeLa cells

Objective To research the relationship between miR-21 and radiation sensitivity of cervical cancer HeLa cells,and to elucidate the possible action mechanism of miR-21 in apoptosis and autophagy of cells.Methods The HeLa cells were divided into mock groups (transfected with miR-21 NC,mimics,inhibitor NC or inhibitor)and 4 Gy irradiation groups (transfected with miR-2 1 NC, mimics,inhibitor NC or inhibitor with irradiation). The miR-2 1 expression was detected by real-time PCR;colony-forming assay was used to detect the changes of radiation sensitivity;the apoptosis and autophagy were analyzed by FACS assay;the target gene of miR-2 1 was predicted by bioinformatics methods;Dual-luciferase Reporter Assay was used to demonstrate the binding of miR-2 1 and targets;the expression of beclin1 protein was measured by Western blotting method. Results After 4 Gy radiation,compared with mock group,the expression of miR-21 was increased remarkably (P<0.05). Colony-forming assay showed that there were no changes of radiation sensitivity of HeLa cells after overexpressing or inhibiting miR-21 compared with miR-21 NC or miR-21 inhibitor NC groups(P>0.05).Compared with miR-21 NC+4 Gy group,the apoptotic rate of HeLa cells in miR-21 mimics+4 Gy group was increased (P<0.05).After transfection with miR-21 inhibitor,compared with miR-21 inhibitor NC group,the apoptotic rate of HeLa cells in duced by radiation was decreased (P<0.05 ). Compared with miR-21 NC group, the autophagy rate was increased after transfection with miR-21 mimics (P<0.05).After transfection with miR-21 mimics and radiation, the expression level of beclin1 protein was increased (P<0.05 ) compared with miR-21 NC+4 Gy group. Conclusion Beclin1 is the target gene of miR-21 in HeLa cells. Overexpression of miR-21 can increase the radiation-induced apoptosis and autophagy,but has on influence in radiation sensitivity of HeLa cells.
