1.Experiment Study on In-vitro Anti-rotavirus Effect of Zhixie Tuire Tablets

Objective To evaluate the pharmacological effect of Zhixie Tuire Tablets(ZTT) on human rotavirus RV709 in host cells in vitro.Methods By improved methods of methyl thiazolyl tetrazolium colorimetry assay(MTT) and cytopathic effect(CPE),the anti-rotavirus effect of ZTT on RV709 was evaluated.Results The half toxic concentration(TC50) of ZTT for MA104 was 0.49699 mg/mL.ZTT neither can directly kill human rotavirus,nor prevent human rotavirus from invasion of host cells.The halft inhibitory concentration of ZTT was 0.00519 mg/mL for RV709.Conclusion In-vitro anti-rotavirus mechanism of ZTT is related to the inhibition of the biological synthesis of rotavirus in host cells.
2.Therapeutic Mechanism of Zhixie Tuire Tablet for Rotavirus-induced Diarrhea

Objective To probe into the therapeutic mechanism of Zhixie Tuire Tablet (ZTT) for rotavirus-induced diarrhea. Methods We established the 4-day old Kunming neonatal mice model by infecting group A G3 type of rotavirus,and then gave the inoculated mice with ZTT at the dose of 0.78 g/kg for three days. Fluorescent quantification PCR method was used to detect the changes of sodium glucose transporter SGLT1 mRNA level and radio-immunity method was used for the examination of PGE2 level in neonatal mice small intestine. Results SGLT1mRNA level was lower in ZTT group than that in the normal group,the difference being significant (P 0.05). 3.High-performance capillary electrophoresis method for ferulic acid assay in serum of rats treated with Buyang Huanwu Decoction

Object To develop a high-performance capillary electrophoresis method to determine ferulic acid concentrations in serum of rats treated with Buyang Huanwu Decoction (BHD). Methods Capillary zone electrophoresis was applied for ferulic acid assay, quantitative determination was based on internal standard and detection was carried on by direct UV. The electrolyte buffer was composed of 25 mmol/L borax-methanol (85∶15). Capillary electrophoresis was performed using a 52 cm (30 cm to detector)?50 ?m fused-silica capillary tube. Separation voltage was 20 kV, sampling time was 3 s, detection wavelength was 320 nm, and the temperature was 30 ℃. Results Ferulic acid was successfully separated within 6 min, the recoveries were 97.2% in serum and 103.28% in BHD, respectively. RSD were 3.73% and 0.91% (n=3), respectively. Conclusion This method can supply reference for the determination of ferulic acid in serum samples and BHD.

5.Effect of Gegen Qinlian Decoction on Rotavirus-inducing Diarrhea in Neonatal Mice

Objective To investigate the anti-rotavirus effect of Gegen Qinlian decoction on rotavirus (RV)-inducing diarrhea neonatal mice model, and provide evidences for pharmacological and pharmacodynamic studies. Methods Neonatal diarrhea mice model of Kunming family infected with RV-Wa was developed. Symptom, diarrhea ratio and severity level, histological changes of neonatal mice’s small intestine and lung was assessed after Gegen Qinlian decoction (0.78 g/kg) treatment. Results Diarrhea ratio, diarrhea severity level, histological changes of neonatal mice’s small intestine and lung were obviously decreased after Gegen Qinlian decoction (0.78 g/kg) treatment. Conclusion Gegen Qinlian decoction has anti-rotavirus effect on RV induced diarrhea.
6.Forensic Application of Objective Assessment on Visual Acuity by ERP

Objective T o explore the com position characteristic of event related potential (E R P ) in dif-ferent visual acuity levels, and to provide theoretical basis for the objective assessm ent of visual acuity. Methods M onocular stim ulus w as perform ed on 16 selected subjects. T he subjects w ere required to look straight at the screen ahead and count the am ount of stim uli from different directions. T he pictures of optotype stim ulus w hich corresponding to three different visual acuity levels w ere show ed in the center of the screen. The ERP results w ere recorded separately. Results (1) The P1 am plitudes of m atch stim uli recorded under the supra-threshold visual acuity level w ere higher than that of m atch stim uli recorded under the threshold and sub-threshold visual acuity levels. T here w as no significant difference betw een the P 1 am plitudes of m atch stim uli recorded under the threshold and sub-threshold visual acuity levels. T he tendency of conflict stim uli w as sim ilar to that of m atch stim uli under three visual acuity levels. (2) In the 340-500 m s post-stim ulus range, P 300 com ponent w as found under supra-threshold and threshold visual acuity levels; no P 300 com ponent w as found in corresponding tim e w indow under sub-threshold visual acuity. T he differences of P 300 am plitudes am ong three visual acuity levels w ere statistically significant. T he am plitudes from high to low w ere the supra-threshold, threshold and sub-threshold visual acuity levels. Conclusion E R P can be a potential new m ethod for the objective assessm ent of visual acuity in forensic m edicine.
7.Effect of Different Attentional Conditions on ERP Detection of Visual Acuity

Objective T o study the effect of different attentional conditions on the event-related potential (E R P ) com ponents generated by the visual inform ation stim uli related to visual acuity, and provide a theoretical reference for clinical forensic visual objective evaluation. Methods W ith visual acuity opto-types as norm al form of visual inform ation stim uli, 15 volunteers as study subjects w ere supposed to ac-count the visual acuity optotypes under the attentional condition of visual stim uli. Furtherm ore, the sub-jects w ere required to listen to the storytelling carefully under the non-attentional condition of visual stim uli, and after the exam ination, they needed to answ er the story-related questions. A ll the E E G re-sults of tw o different attentional conditions from the subjects w ere recorded by 32 channel E R P system . Results U nder tw o attentional conditions, P 1 and P 300 com ponents w ere evoked by the visual acuity opto-types on supra-threshold and threshold visual acuity levels, w hile only P 1 com ponent w ere evoked by the visual acuity optotypes on sub-threshold levels. In the E R P w aveform s evoked by the visual acuity optotypes on supra-threshold, P 1 and P 300 am plitudes under attentional condition w ere larger than that un-der non-attentional condition. Conclusion A ttentional conditions can influence the detection of visual acuity. P 300 com ponent can be used to distinguish the visual acuity levels w ith supra-threshold and sub-threshold under non-attentional condition.

Traditional research ideas of miRNA-target gene-biological function have ignored the contact between the target genes and signaling pathways involved , making the integrity and relevance of miRNA regulatory mechanisms not be fully elucidated .Integrated with systematic and relevant way of thinking , summarization and analysis for the luciferase reporter assay validated miR-205 target genes and their related signaling pathways will pave the way for new research area for miR-205 , and , it will be helpful for breaking through the status quo and exploring the novel research areas for miRNA .
9.Iodine nutritional status of children aged 8-10 after implementation of a new standard of iodized salt in northern area of Jiangsu Province

Objective To investigate and analyze the iodine nutritional status of school-age children six months after implementation of a new standard of iodized salts in northern area of Jiangsu Province,and to provide a basis for control of iodine deficiency disorders.Methods According to the water iodine content,northern area of Jiangsu Province was divided into low,moderate and high iodine areas (the water iodine contents were < 10,l0-150 and > 150 μg/L,respectively).Two towns were selected in each area; one elementary school was selected in each town; and 120 children aged 8-10 were selected in each school.The samples of urine,household salt,and water in each town were collected.Urinary iodine content was tested by arsenic cerium catalytic spectrophotometry;iodine content of household salt sample was determined by direct titration; iodine content of water sample was tested by cerium sulfate catalytic spectrophotometry.Results In low,moderate and high water iodine areas,240,242 and 242 urine samples of 8-10 years old children were tested; urinary iodine median was 135.49,183.62 and 448.99 μg/L,respectively,and the difference between groups was statistically significant (x2 =165.56,P < 0.05).In low,moderate and high water iodine areas,100,131 and 117 household salt samples were tested,and the median of salt iodine was 21.88,22.01 and < 5 mg/kg,respectively.Water samples collected in low,moderate and high iodine areas were 12,14 and 10 copies,respectively,and the median of water iodine was 8.89,38.52 and 189.25 μg/L,respectively,and the difference between groups was statistically significant (x2 =22.27,P < 0.05).Conclusions The iodine nutritional condition of 8-10 years old children in low,moderate water iodine areas in northern area of Jiangsu Province is appropriate.The new standard is suitable for iodine nutritional needs in the area.The iodine nutritional condition in high water iodine area is excess,iodine intake should be reduced.
10.Cardiovascular risk of anthracyclines in treatment of breast cancer and its strategy

Objective To evaluate the clinical cardiovascular reverse reactions of the patients with breast cancer who received chemotherapy with anthracyclines. Methods Two hundred and ninety-one cases of breast cancer who were treated with pirarubicin (40 mg/m2) + docetaxel (75 mg/m2) in Shanxi Provincial Cancer Hospital from January 2014 to December 2016 were collected. The changes of electrocardiogram (ECG), echocardiography and myocardial enzyme were recorded before and after chemotherapy. Results Before chemotherapy, there were 14 patients (4.8 %) with ST-T segment changes before ECG, 6 patients (2.1 %)with atrial extrasystole,8 patients(2.7 %)with ventricular extrasystole.After chemotherapy,cases with abnormal ECG were 62 (21.4 %), 47 (16.2 %) and 52 (17.9 %) respectively. The difference between the two groups was statistically significant (χ2= 6.249, P < 0.005). The average left ventricular ejection fraction after chemotherapy was significantly lower than that before chemotherapy (0.72±0.10 vs. 0.81±0.18, t= 3.480, P <0.05). The levels of creatine kinase (CK), aspartate aminotransferase (AST) and creatine kinase CK (CK-MB) were (68±20), (35±13) and (15.1±4.0) U/L after chemotherapy respectively, which were significantly higher than those before chemotherapy [(50 ±12), (28 ±12) and (7.9 ±2.6) U/L], the differences were statistically significant (t values were 9.721, 5.137 and 18.850 respectively, all P < 0.005). Conclusion The use of anthracyclines results in the increased risk of ECG abnormalities and myocardial damage in patients with breast cancer,and cardiac dynamic monitoring should be noted during chemotherapy.
