Objective:To study the eExecutive fFunctions Characteristics of Cchildren with Aattention dDeficit hHyperactivity Ddisorder in Cclock Ddrawing Ttest. Methods: 60 ADHD children and 60 normal children awere tested by CDT. Then analyze the data to detect the difference of Executive Functions of two growps. Results:ADHD group geot significant lower scores than control group on both time total score (data) and clock face structure score. (3.23?1.40/4.15?0.78,11.2?1.48/11.90?0.76,t=7.742、5.073,P0.05). Conclusions: the lower score in CDT showed that ADHD child have some weaknesses in planning , in attention regulating and in working memory. The core of these weaknesses is failing of execute function.

Objective To explore the specific brain regions involved in Chinese character radicals visualspatial processing,as well as to explore the relationship between visual-spatial identification and Chinese characters radicals identification. To provide an evidence for understanding the underlying neural mechanism of developmental dyslexic. Methods Twelve healthy Chinese fourth and fifth grade children(7 boys and 2 girls, aged from 10 to 11 years old)were chosen from a primary school in Guangzhou. The children were native Chinese speakers and right handed. The subjects participated in two fMRI experiments on a 3.0 T Siemens MR scanner. Two separate block-designed cognitive tasks were devised, one was figure directions judgement task and the other was Chinese characters radical identification task.A single-shot GE-EPI scan sequence were used for fMRI scans. The BOLD fMRI data were collected and were preprocessed and analyzed in SPM2.Results The subjects had longer reaction time during the two experiments tasks than in control states((879±127)ms vs(805±124)ms,P=0.029;(917±144)ms vs(750±101)ms,P<0.01),and had same response accuracy in both tasks and control states((96±5)% vs(95±8)%,P=0.515;(89±10)% vs(96±6)%,P=0.130).In figure direction judgement experiment, activations were found in bilateral occipital, right superior parietal, right precuneus and right middle frontal cortices. However, different and more extensive regions including left superior parietal, left precuneus, left hippocampus, right thalamus, right caudate cortices and bilateral occipital cortices were involved in identifying the radicals of Chinese characters. Conclusion Right hemisphere was dominant in judging the figure direction, and left hemisphere is dominant in identifying radicals of Chinese characters. These fMRI results show light on underlying partial similar neurophysiological mechanism between judging figure directions and identifying Chinese character radicals. The visual-spatial ability may be essential for Chinese character radical identification.
3.Mechanisms Underlying Brain Aging Under Normal and Pathological Conditions.
Neuroscience Bulletin 2023;39(2):303-314
Aging is a major risk factor for many human diseases, including cognitive impairment, which affects a large population of the elderly. In the past few decades, our understanding of the molecular and cellular mechanisms underlying the changes associated with aging and age-related diseases has expanded greatly, shedding light on the potential role of these changes in cognitive impairment. In this article, we review recent advances in understanding of the mechanisms underlying brain aging under normal and pathological conditions, compare their similarities and differences, discuss the causative and adaptive mechanisms of brain aging, and finally attempt to find some rules to guide us on how to promote healthy aging and prevent age-related diseases.

Objective:To investigate the distribution and related factors of lipoprotein(a) [Lp(a)] level in healthy Tajik and Kazak adults in China. Methods:A cross-sectional study was conducted from May to October 2021 and March to June 2022, and blood samples were collected from 2, 637 healthy Tajik adults [1 010 men, average age: (40.08±14.74) years; 1 627 women, average age: (38.27±12.90) years] in Tashkurgan Tajik Autonomous County and 1 911 healthy Kazak adults [720 men, average age: (42.10±12.26) years; 1 191 women, average age: (38.27±12.90) years] in Fuyun County of Xinjiang. Fasting blood glucose (FBG), creatinine (Cr), total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), and Lp(a) were measured. The distribution of Lp(a) levels in different sex and age groups was compared. The Lp(a) level of Tajik and Kazakh ethnic groups was compared by Mann-Whitney U test, and correlation factors of Lp(a) level were analyzed by multiple logistic regression. Results:The distribution of Lp(a) level in Tajik and Kazak population was skewed. Lp(a) levels of Tajik and Kazak ranged from the lowest 0.40 mg/L and 0.62 mg/L to the highest 1 229.40 mg/L and 2 108.58 mg/L, respectively, and the median Lp(a) level was 78.90 (38.60, 190.20) mg/L and 103.30 (49.57, 234.27) mg/L, respectively. Median Lp(a) level of Kazak was significantly higher than that of Tjik ( P<0.001). The median Lp(a) levels of Tajik males and females were similar: 77.45 (39.80, 187.10) mg/L and 79.90 (38.60, 192.30) mg/L ( P=0.948). The median Lp(a) levels of Kazakh males and females were also similar: 109.42 (50.49, 230.38) mg/L and 99.01 (49.11, 237.25) mg/L, respectively ( P=0.578). After pooling the data of Tajik and Kazak and adjusting for age, sex, BMI, smoking, drinking, blood pressure, blood glucose and other factors, Lp(a) level was correlated with ethnic (standard partial regression coefficient 0.066, P=0.008) and LDL-C level (standard partial regression coefficient 0.136, P<0.001). Conclusions:Lp(a) level in healthy Tajik and Kazak adults varied greatly among individuals, and Kazak residents had a higher Lp(a) level than Tajik residents. There was no significant sex difference in Lp(a) level among Tajik and Kazakh, and LDL-C and ethnicity are independent factors related to Lp(a) level.

The emergence of SARS-CoV-2 variants of concern and repeated outbreaks of coronavirus epidemics in the past two decades emphasize the need for next-generation pan-coronaviral therapeutics. Drugging the multi-functional papain-like protease (PLpro) domain of the viral nsp3 holds promise. However, none of the known coronavirus PLpro inhibitors has been shown to be in vivo active. Herein, we screened a structurally diverse library of 50,080 compounds for potential coronavirus PLpro inhibitors and identified a noncovalent lead inhibitor F0213 that has broad-spectrum anti-coronaviral activity, including against the Sarbecoviruses (SARS-CoV-1 and SARS-CoV-2), Merbecovirus (MERS-CoV), as well as the Alphacoronavirus (hCoV-229E and hCoV-OC43). Importantly, F0213 confers protection in both SARS-CoV-2-infected hamsters and MERS-CoV-infected human DPP4-knockin mice. F0213 possesses a dual therapeutic functionality that suppresses coronavirus replication via blocking viral polyprotein cleavage, as well as promoting antiviral immunity by antagonizing the PLpro deubiquitinase activity. Despite the significant difference of substrate recognition, mode of inhibition studies suggest that F0213 is a competitive inhibitor against SARS2-PLpro via binding with the 157K amino acid residue, whereas an allosteric inhibitor of MERS-PLpro interacting with its 271E position. Our proof-of-concept findings demonstrated that PLpro is a valid target for the development of broad-spectrum anti-coronavirus agents. The orally administered F0213 may serve as a promising lead compound for combating the ongoing COVID-19 pandemic and future coronavirus outbreaks.
